1.Effects of electromagnetic radiation exposure on learning and memory function of rat pups

Objective To determine whether exposure to electromagnetic radiation results in behavioral effects of rat pups.Methods Wistar rats,either male or female,were divided randomly into four groups respectively:control(CTR),100 seconds(S1),1000 seconds(S10)and 3000 seconds(S30),then each of them were exposed to electromagnetic radiation of 100 kV/m field amplitude of corresponding time.For the day of exposure till 2 months later.they were mated with the rats in the same group and their offspring were divided into four groups(F-GTB,F-S1,F-S10,F-S30)correspondingly.Behavioral changes occur in 2-month old and 6-month old rat pups were found between radiation groups and control,and in Y-maze test,beth male and female pups in F-S10 significantly learned fewer times than their control(total study times:male 14.6±3.9 vs 21.1±7.8,female13.4±3.0 vs 25.8±8.8;false times:male 3.5±2.4 vs 7.8±5.4,female 3.4±2.6 vs 11.0±7.2).In open field test,both maze,male pups in F-S1 and F-S30 learned more times than control in total study time(24.2±8.9 vs 14.1±5.2.30.7±12.4 vs 14.1±5.2).In step throush test and open field test,no significant differences were found between radiation groups and control.Conclusion There was significant genetic effect exposed to electromagnetic radiation of 100 kV/m,mainly manifested in rat pups in growth period but without long-term effect.

3.Continuous aspiration of subglottic suction for preventing ventilator associated pneumonia: a meta-analysis

Objective To systemically analyze the effect of continuous aspiration of subglottic suction (CASS) on the incidence of ventilator-associated pneumonia (VAP) and other outcomes in mechanical ventilated patients.Methods PubMed,Cochrane Library,Ovid,EBSCOhost,ScienceDirect,Google scholar,CNKI,CBM and Wanfang Database were searched for the RCTs about the effect of CASS on the outcomes of mechanical ventilated patients from July 1994 to July 2014,and the relevant journals were also hand-searched.Data were extracted independently by two reviewers.Meta analysis was conducted using RevMan 5.1 software.Results Data extracted from 7 RCTs with a total of 1 566 patients were analyzed.The risks of developing clinically diagnosed VAP were significantly lower among the patients receiving CASS [RR =0.54,95% CI(0.43,0.65),P<0.01],episodes of VAP occurred later in patients receiving CASS [MD=3.52,95%CI(1.98,5.06),P<0.01],the duration of mechanical ventilation [MD=-2.03,95%CI(-2.23,-1.73),P<0.01]and length of ICU stay were reduced [MD=-2.99,95%CI(-3.75,-2.23),P<0.01],while no significant differences were detected between the two groups regarding the mortality rate [RR=0.98,95%CI (0.80,1.20),P>0.05].Conclusion This study proved that the CASS did have effect in reducing the incidence of VAP,but it is limited by lack of enough RCTs with high quality.Large sample of high quality RCTs is necessary to examine the wider applicability of CASS.
4.Qualitative study on burden of caregiver of children with asthma

Objective To describe the burden of caregivers of children with asthma,and to provide reference for the development of family care support program.Methods Qualitative research was used to carry out semi-structured interviews with 13 caregivers of children with asthma in the pediatric outpatient of the First Affiliated Hospital of Guangzhou Medical University.The data collected were analyzed with Colaizzi 7-step analysis method.Results The carcgiver burden was classified into 4 themes,including time and energy exhaustion,heavy economic burden,insufficient knowledge of care and negative emotions.Conclusions The burden of caregivers of children with asthma has a relatively great impact on the daily work and life,even the physical and mental health.Care workers should be concerned about the status of caregiver burden and provide targeted interventions,which could help caregiver adapt to their roles,maintain and promote the health of children patients and their families.
5.Effect of Electromagnetic Radiation on Oxidation Balance of Mice Brain and Therapeutic Effect of Curcumin

ObjectiveTo explore the changes of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), malondialdehyde (MDA) in the brain of mice after irradiated by electromagnetic radiation (EMR) and therapeutic effect of curcumin.Methods40 mice were divided randomly into the bare control group, simply EMR group, EMR+curcumin low dose, middle dose and high dose groups total 5 groups with 8 animals in each group. The mice except the bare control group received EMR irradiation and those in the EMR+curcumin groups were given various doses of curcumin at the same time. Five days later, EMR irradiation and medication stopped, and the levels of SOD, GSH-Px and MDA in the brain of mice were tested.ResultsCompared with the bare control group, the activities of GSH-Px and SOD, and level of MDA all increased in the mice irradiated by EMR ( P<0.05). Compared with the simply EMR group, the activities of GSH-Px and SOD, and level of MDA all decreased in the mice of EMR +curcumin groups ( P<0.05).ConclusionEMR irradiation can induce changes of GSH-Px and SOD and peroxidation of mice brain, curcumin can lighten these damages by its anti-oxidation with a dose-dependent effect.
6.The expression of plasmid DNaseI gene in mice bone marrow mesenchymal stem cells

Objective To investigate the expression and secretion of mice DNaseI gene plasmid transfected into bone marrow (BM-MSCs) mesenchymal stem cells. Methods The plasmids of mouse DNaseI gene had been transfected into the BM-MSCs of mice by liposomes. The expression of DNaseI gene in the BM-MSCs was detected by western blotting and the DNaseI activity was measured by DNA-methyl green substrate colorimetry. Results About 30% BM -MSCs were transfected with mice plasmid DNaseI gene, DNaseI was expressed in the transfected BM-MSCs and active DNaseI could be detected in the supernatant of cell culture. Conclusion The mice DNaseI gene plasmid can be transfected into mice BM -MSCs by liposomes and DNaseI gene can be expressed by the transfected BM-MSCs and active DNaseI can be secreted. This may provide potential target for the treatment of SLE.
7.Effect of eoeulture with mouse dermis-derived mesenehymal stem cells on the secretion of collagen and expression of transforming growth faetor-betal by hnman dermal fibroblasts

Objective To explore the role of mouse dermis-derived mesenchymal stem cells (mdMSC) on skin repair. Methods mdMSC and human dermal fibroblasts were isolated and identified. Human dermal fibroblasts were cultured alone or eoeultured with mdMSC in Transwell chambers with the density ratio of human dermal fibroblasts to mdMSC being 2/5, 1/1, and 2/1. On day 4 and 8 of culture, the expression levels of hydroxyproline and transforming growth factor-beta (TGF-beta) 1 were measured in the supematant of monoculture and coculture by alkaline hydrolysis and ELISA respectively. Results The level of hydro-xyproline was significantly higher in the supematants of coculture system with a density ratio of 2/5 and 1/1 than that in monoculture supematants of human dermal fibroblasts on day 8 (both P < 0.05). Elevated level of TGF-betal was observed in all coculture supematants on day 8 (all P < 0.01) and in the supernatants of coculture system with a density ratio of 1/1 on day 4 (P < 0.05). There was no significant correlation between the expression level of TGF-betal and hydroxyproline in the coculture supernatants (r = 0.108, P > 0.05). Conclusion In vitro coculture with mdMSC can increase the production of hydroxyproline and TGF-betal by fibroblasts, which may be a mechanism underlying the facilitation of skin repair by mdMSC.
8.Application effectiveness of clinical pathways for pediatric asthma: A systematic review

Objective To evaluate the effectiveness of clinical pathways for pediatric asthma.Methods The literatures on the application of clinical pathways for pediatric asthma were collected in the databases such as PubMed,Cochrane Library,Ovid,ScienceDirect,EBSCOhost,CNKI,Wanfang and CBM while a manual search was added.The search was performed from 1980 to 2014.Two reviewers independently screened references,extracted data and assessed the risk of bias.Data was pooled and analyzed by using Revman 5.2 version.Results Eleven studies involving 2 207 participants from nearly 1 048 relevant studies met the inclusion criteria.1 174 participants were in the clinical pathway group while the other 1 033 participants were in the control group.Meta-analysis result showed that patients with asthma in a clinical pathway had a shorter length of stay,less medical cost,reduced rate of emergency room visits,and improved health education.However,clinical pathway did not appear to reduce readmission.Conclusions Clinical pathway can significandy reduce the length of hospital stay,cost,rate of emergency room visits and improve health education.
9.Effects of anti-MDM2 siRNA on radiation-mediated cell killing in lung cancer cell line A549.

BACKGROUNDLung cancer cell lines A549 and NCI-H446 with different radiosensitivity to ionizing radiation (IR) have different MDM2 gene expression status, which may contribute to the radioresistance of cells. The aim of this study is to use small interfering RNA (siRNA) targeting MDM2 to investigate the influence of MDM2 gene silencing on radioresponse of A549 cell.
METHODSPlasmid targeting MDM2 was constructed with pPUR/U6 vector and oligonucleotide designed according to the sequence of effective antisense oligonucleotides and principles of siRNA design. A549 cells were transfected by Lipofectamine™ 2000. MDM2 expression in A549 cells was detected by RT-PCR and Western blot. Radiation-mediated cell killing was detected by flow cytometry.
RESULTSTwo out of three siRNA plasmids were constructed successfully. siRNA transfection resulted in downregulaton of MDM2 expression of A549 cells on mRNA and protein levels. After treated with siRNA, radiation-mediated cell killing of A549 cells was significantly increased (P < 0.01).
CONCLUSIONSThe results support the hypothesis that MDM2 gene is a candidate for radioresistance in A549 cells. siRNA targeting to MDM2 can enhance the radiation-mediated cell killing of A549 cells.
10.Effect of EMP on sex ratio of the offspring of male BALB/c mice

Objective To investigate the effects of EMP exposure to male BABL/c mice on the reproduction and its sex ratio.Methods Thirty-three male BALB/c mice were randomly divided into sham-exposed group,10 000 pulses/d exposed group,and 100 000 pulses/d exposed group with 50 Hz static magnetic field of 35 kV/m for 2 weeks consecutively,then the male mice were mated with unexposed females (1 male vs.2 female).Mated males were sacrificed to examine the exposure effects on sperm number and testis index after 7 days of mating.The sex ratio of embryos collected from unexposed mated females was investigated by PCR.Results The sex ratio of offspring was 0.298 (14/47) in the 10 000 pulses/d exposed group,significantly lower than 0.871 (27/31) in the sham-exposed group.No statistical changes were observed in the sex ratio of offspring,sperm number,the weights of testis and testis index after 100 000 pulses/d exposure.Conclusions The exposure of male mice to 35 kV/m EMP at 10 000 pulses/d for 2 weeks consecutively may change the sex ratio of mice's offspring.
