1.Involvement of mitochondrial pathways of apoptosis in hippocampus apoptosis in kainic acid induced epilepsy rat

Objective:To investigate the hippocampus neuron injury,mitochondrial function and mitochondrial apoptosis related protein expression in kainic acid(KA) induced epilepsy rat,and to explore the role of mitochondrial pathways of apoptosis in the neuron injury.Methods:KA induced epilepsy model was produced by injection of KA into the hippocampus.Forty SD rats were randomly divided into five groups: control and KA group(6 h,1 d,3 d and 7 d),n=32.The concentration of Glu in hippocampus CA3 area was detected by HPLC.The apoptosis of hippocampus neurons and the concentration of free calcium ion were assayed by flow cytometry.The expression of cytochrome C and caspase-9 in hippocampus CA3 area was determined by Western blot.Results: ①The concentration of Glu in hippocampus CA3 area was increased in the fourth day after KA injection.②The concentration of free calcium ion,mitochondrial membrane potential,and the number of apoptosis neurons was significantly changed after 6 h of KA injection.Seven days after KA injection,these changes were more significant.③The hippocampus neuron expressed low levels of cytochrome C and caspase-9,and the expression of cytochrome C and caspase-9 was dramatically increased in a time dependent manner which increased by 4.37and 3.20 fold 7 d after KA injection.Conclusion:Mitochondrial pathways of apoptosis involve in hippocampal neuron apoptosis in KA induced epilepsy rat.
2.The expression and significance of the cell cycle related genes in uveal melanoma

Objective To evaluate the role of the cell cycle related genes cyclinD1 and Bcl-2 protein expression in the pathogenesis and infiltration of the uveal melanoma. Methods Using immunohistochemistry to detect the cyclinD1 and bcl-2 protein expression in 96 cases of uveal melanoma. Results The expression content of bcl-2 was high in uveal melanoma, and there wasnt any relationship between bcl-2 cell positivity and tumor cell type and extrascleral extension. In contrast, cyclinD1 expression was higher in epithelial cell uveal melanoma than mix cell and spindle cell varieties. There was a positive correlation between cyclinD1 cell positivity and extrascleral extension. Conclusion The expression of bcl-2 protein is important for the survival of the uveal melanoma. CyclinD1 may serve as a sensitive index of its malignancy.
3.Biocompatibility of porous drug implant scaffolds based on 3D printing technology

Objective To evaluate the biocompatibility of porous drug implant scaffolds prepared by 3D printing technique. Methods Porous drug implant scaffolds were fabricated by 3D printing technique, and a series of tests were carried out to validate the biocompatibility, including acute systemic toxicity, hot source test, local irritation reaction, micronucleus test, muscle implant test and so on. Results The porous drug implant scaffolds showed no acute systemic toxicity, no pyrogenetic effect, no local erythema and edema in local irritation re-action, hemolysis rate of 0.29%, no cellular genetoxic. No local tissue denaturation, necrosis and exclusion were found in intramuscular implant test. Conclusion With good biocompatibility, the porous drug implant scaffold fabricated by 3D printing technique can meet the clinical requirement for biomaterial.
4.Effect of IKVAV peptide nanofiber on proliferation, adhesion and differentiation into neurocytes of bone marrow stromal cells.

This study examined the effect of IKVAV peptide nanofiber on proliferation, adhesion and differentiation into neurocytes of bone marrow stromal cells (BMSCs). IKVAV Peptide-amphiphile was synthesized and purified. Then, hydrogen chloride was added to the diluted aqueous solutions of PA to induce spontaneous formation of nanofiber in vitro. The resultant samples was observed under transmission electron microscope. BMSCs were cultured with IKVAV peptide nanofiber. The effect of IKVAV nanofiber on the proliferation, adhesion and induction differentiation of BMSCs was observed by inverted microscopy, calcein-AM/PI staining, cell counting and immunofluorescence staining. The results demonstrated that IKVAV peptide-amphiphile could self-assemble to form nanofiber gel. BMSCs cultured in combination with IKVAV peptide nanofiber gel grew well and the percentage of live cells was over 90%. IKVAV peptide nanofiber gel exerted no influence on the proliferation of BMSCs and could promote the adhesion of BMSCs and raise the ratio of neurons when BMSCs were induced to differentiate into neurocytes. It is concluded that BMSCs could proliferate and adhere well and yield more neurons during when induced to differente into neurocytes on IKVAV peptide nanofiber gel.
5.Two-dimensional Shear Wave Elastography in Predicting Prognosis of Patients with Acute-on-chronic Liver Failure

[Objective]To evaluate the two-dimensional shear wave elastography(2D SWE)in predicting the prognosis of the patients with acute-on-chronic hepatitis B liver failure(ACLF-HBV).[Methods]312 patients diagnosed with ACLF-HBV at the De-partment of Infectious Diseases in the Third Affiliated Hospital of Sun Yat-sen University from October 1st 2013 to December 31th 2015 were recruited. The baseline data of all patients,including 2D SWE,laboratory data,ultrasonographic data,Model for End-stage Liver Disease score(MELD),Child-Turcotte-Pugh score(CTP),and King′s College Hospital score(KCH),were collected when enrolled. All patients were followed up for at least 90 days and their clinical outcomes(recovering or worsening)were recorded. 2D SWE values of all patients were dynamically collected every 2~4 weeks during the follow-up until death,liver transplantation, or discharging from hospital.[Results]The worsening patients showed higher 2D SWE values than the recovering ones[(44.0 ± 7.5) kPa vs(36.8 ± 10.3)kPa,t=-6.5,P=0.000],2D SWE value less than 39.2 kPa could be a prediction of the patient′s recovery in 90 days.The predictive value of 2D SWE(AUROC=0.73)was higher than that of KCH(AUROC:0.65,z=2.1,P=0.033). Among all the dynamically measured patients,2D SWE rose from(42.1 ± 8.0)kPa to(47.5 ± 9.3)kPa in the worsening group during the 90-day follow-up,while nearly stable in the recovery group.[Conclusion]Noninvasive liver stiffness measurement by 2D SWE is a use-ful method in evaluating the prognosis of ACLF-HBV patients. Taking a cutoff of 39.2 kPa,patients with higher 2D SWE values could have worse clinical outcomes ,otherwise better. It seems that dynamically measuring 2D SWE values could also be helpful ,but more research in the future should be needed.
6.Isolation, culture and identification of neural stem cells in new-born rats.

The cortexes were obtained from new-born rats and dissociated to single cells by triturating. The cells were cultured in neural stem cell (NSC) culture medium (DMEM supplemented with bFGF, EGF and B27) and formed primary neurospheres after 7 days. Single cells dissociated from neurosphere were cultured in 96-well plates and formed single-cell cloning neurosphere 7 days later. The primary and single-cell cloning neurospheres were both positive for the immunofluorescent staining of nestin and were identified as NSC. It was proved that NSC can be expanded in vitro and provide seed cells for neural tissue engineering.

7.Neuron-specific enclose and myelin basic protein in cerebrospinal fluid of patients with first episode schizophrenia.

In order to study whether patients with schizophrenia have cerebral injury, neuron-specific enolase (NSE) and myelin basic protein (MBP)in cerebrospinal fluid (CSF) of 33 patients with first episode schizophrenia and 9 from the control group were determined by double antibody sandwich enzyme immunoassay method. The results showed that there was significant difference in the NSE contents between the experimental group and control group (P<0.01). The NSE contents in CSF in the experimental group were positively correlated with MBP in schizophrenia patients (P< 0. 05). These findings suggested that patients with schizophrenia had cerebral injury.
8.Reoperation of papillary thyroid carcinoma

Objective To investigate the procedures and therapeutic effects of reoperation of papillary thyroid carcinoma with lymph node metastasis.Methods The clinical data of 10 papillary thyroid carcinoma patients with lymph node metastasis received reoperation were retrospectively studied. Results Reoperation was successfully performed in 9 patients except one patient with tumor surrounding common carotid artery and brachial plexus who had partial tumor removed. Three patients had simple lymphectomy,2 had complete thyroidectomy, 2 had complete lobectomy plus ipsilateral cervical lymph node dissection and 2 had tumor resection plus cervical lymph node dissection. There were no new complications. Conclusions Papillary thyroid carcinoma has the characteristic tend to spread to spread to cervical lymph nodes. Rreoperation offers good outcome for recurrent papillary thyroid carcinoma.

10.Design of Non-Invasive Blood Oxygen Measurement Based on AFE4490.

From the perspective of portable monitoring devices,we use an analog front-end AFE4490 design a module of Non-invasive blood oxygen measurement, used to collect human pulse wave signal and peak (valley) value detection and then use the principles of non-invasive oximetry calculated oxygen saturation (SPO2). This design of noninvasive oximetry module has the characteristics of small size, low power consumption, and the results of test show that the measurement of oxygen saturation are correct.
