2.The immune function injury and its mechanism in drug abuser

Objective To explore the immune function injury and its mechanism in drug abuser.Methods The immune function changes in 50 drug abusers were compared with normal healthy populations by detection of the indexes of subgroups of Th cells, transformation rate of lymphocytes,IgA,IgM,IgG,IgE,compliment C3,C4,IL 1, IL 2,IL 6 TNF and NO.Result In peripheral blood the percentage of Th1 cell, transformation rate of lymphocyte, IgA, IgM, IgG, IgE content, compliment C4,C4, IL 1, IL 2,IL 6,and TNF levels were significantly lower than normal(P< 0.01).The value of Th1/Th2 was lower than normal as well(P< 0.05).NO content was significantly higher than normal(P< 0.001).Conclusion The mechanism of immune function injury in drug abuser might be correlative to direct injury of drugs and their inhibition effect on the thymus hypothalamus hypophysis adrenal axis.
3.Limb salvage surgery for malignant bone tumor of the extremities

Objective To analyze the clinical features of limb salvage surgery with epiphyseal preservation in children and adolescents, and to evaluate the recurrence rate, metastasis, complications, and the joint functional results after tumor resection. Methods From December 1995 to January 2003, we operated on 33 patients aging from 8 to 16 years (average 12.2 years) with diagnosis of primary malignant bone tumor of the extremities with preserving the epiphysis in the limb salvage surgery. In this group, the tumor located in distal femur in 24 cases and in proximal tibia in 9 cases. There were 23 osteosarcomas, 6 Ewing sarcomas, 2 chondrosarcomas, and 2 invasive osteoblastomas. The tumors were staged clinically, 2 cases inⅠA, 2 cases inⅠB, 17 cases in ⅡA, and 12 cases inⅡB. According to the relationship between the tumor and epiphyseal plate, the metaphyseal tumors in children were classified into 3 types: 18 cases were categorized as type Ⅰ, 13 as type Ⅱ, and 2 as type Ⅲ. The patients received 2-4 cycles of preoperative adjuvant chemotherapy, and another 6 cycles after surgery. Serious bone defects after tumor resections were repaired with massive allograft bone transplantation, followed by internal fixation by intramedullary nails and cancellous screws. Results Among the 33 cases, 3 cases were lost to follow up. 29 cases had complete clinical data. Postoperative follow-up was 12-72 months (average 37.6 months). Recurrences were seen in 3 cases, with one local recurrence in type Ⅲ case one year after tumor resection, other 2 recurrences around the femoral vessels in typeⅡcases 15 and 30 months after the tumor resection respectively. The recurrence rate accounted for 10.3%. Amputation was performed for the cases with recurrences. But pulmonary metastasis developed and the patients died. In this cohort, 9 cases died. Five-year survival rate was 57.9%. 4 cases reported 5 complications (17.2%). No patient reported skin necrosis, hematoma, infection, rejection to the allograft, and nonunion at the end of the follow-up. According to the functional evaluation criteria of Enneking, excellent were 11 cases, good 13 cases, fair 3 cases, and poor 2 cases. The excellent or good rate was 82.8%. Conclusion The limb salvage surgery with preservation of epiphysis for malignant bone tumors in children and adolescents guarantees the patient's satisfactory postoperative limb functional results. Preoperative effective adjuvant chemotherapy and prevention of postoperative complications deserve great attention.

5.Analysis of 50 cases of traumatic retinal detachment diagnosed by color Doppler ultrasound and two-dimensional ultrasound

AlM:To evaluate the value of color Doppler ultrasound in the diagnosis of traumatic retinal detachment.
METHODS: All 50 cases ( 56 eyes ) of suspicious traumatic retinal detachment from January to June in 2013 were enrolled in this study. All first received the two-dimensional ultrasound, then the color Doppler ultrasound, finally, these results were compared with the results of surgery.
RESULTS: All 56 eyes were observed intravitreal abnormal spots by the two-dimensional ultrasound, and 39 eyes were observed color blood signal by the color Doppler ultrasound. Forty eyes were approved retinal detachment by surgery.
CONCLUSlON: lt is better to display the intravitreal abnormal spots and color blood signal by the color Doppler ultrasound than by the two - dimensional ultrasound, so there is important clinical significance of the color Doppler ultrasound in the diagnosis of traumatic retinal detachment.
6.Comparison between overthick and overthin X ray film

7.Decreased c-kit and miR-21 gene expression in rat heart failure

Objective To explore mechanism ( s) is dynamic expression and internal relations of c-kit gene and miR-21 in rat ventricular remodeling .Methods SD rats( n=140) were randomly divided into normal control group ( n=15 ) and heart failure model group ( n=125 ) .Heart failure model:4 mg/kg intraperitoneal injection of adria-mycin.To detect the cardiac function of rats in eight weeks , proving the heart failure .Then harvest the rat heart , frozen section , using immunohistochemistry and immunofluorescence color to detect the expression changes of miR-21 and c-kit in myocardial tissue .Results The emergence of the pathological changes of cardiac muscle cells after myocardial infarction in the heart failure groups and the control group didn't appear the myocardial infarction and heart failure .Immunohistochemistry and immunofluorescence show that miR-21 positive cells in normal and heart failure myocardium mainly express in vascular endothelium ,a few myocardial cells and stem cells , and endocardial expressing quantity fell more than epicardial;c-kit positive cells tend to cluster together , mainly gather in the epi-cardial and its nearby , and the expressing quantity decrease significantly after heart failure .A small number of cells exsit miR-21 and c-kit common expression in both groups ( P<0.05 ) .Conclusions The decreased expression of c-kit and miR-21 is highly related to rats heart failure and left ventricular remodeling .
9.Effects of deep brain stimulation on expression of DARPP-32 and its phosphorylated proteins in corpus striatum of rats with dyskinesia

Objective To investigate the effects of subthalamic nucleus (STN)-deep brain stimulation (DBS) on expression of dopamine and adenosine 3'5'-monophosphate-regulated phosphor-protein(DARPP-32) and its phosphorylated proteins in corpus striatum of rat models with levodopa-induced dyskinesia. Methods The rat models of levodopa-induced dyskinesia were set up and were given STN-DBS (stimulation group). The expression of DARPP-32 and its phosphorylated proteins in corpus striatum (damage side and normal side) were detected and compared with sham-stimulation group and sham-operation group. Results There was no significant difference in the expression of DARPP-32 total protein in corpus striatum of rats with dyskinesia among three groups (P>0.05). The expression of Phosphor-Thr34-DARPP-32 protein in the damage side of corpus striatum in stimulation group was significantly lower than that in sham-stimulation group and sham-operation group (P<0.05), while the expression of Phosphor-Thr75-DARPP-32 protein in the damage side of corpus striatum in stimulation group was significantly higher than that in sham-stimulation group and sham-operation group (P<0.05). Conclusion DARPP-32 and its phosphorylated proteins play an important role in the pathogenesis of levodopa-induced dyskinesia.
10.Expression of NogoA in 21a area of visual cortex in visual developing normal cat and strabismic amblyopia cat

BackgroundThe study of the molecular mechanism of visual plasticity is helpful for the explaining and prevention of strabismus and amblyopia.The effect and significance of NogoA in the strabismus and amblyopia formation are attracting more attention.ObjectiveThe present study was to investigate the expression of NogoA in 21a area of visual cortex in strabismic-induced amblyopia cats and explore the possible molecular mechanism of strabismic-induced amblyopia.MethodsSixteen 4-week old clean cats were randomizedly divided into normal group and strabismic-induced amblyopia group and eight for each group.The strabismic-induced amblyopia models were created by cutting off the external rectus in 8 cats.Pattern visual evoked potentials ( P-VEP ) were recorded 1 week after operation and compared with normal cats,and depression of amplitude and prolongation of implied time of P100 wave were as the successful criterion of model.The 200 ml paraformaldehyde was infused via heart to fax the brain under the deep anesthesia and then the cats were sacrificed and the brain cortex sections were prepared.The morphology of 21a zone of cat visual cortex was examined by haematoxylin and eosin staining,and the expressions of NogoA in 21a area of visual cortex were detected by immunohistochemistry with a monoclonal antiNogoA antibody.The use of the animals complied with the Regulations for the Administration of Affair Concerning Experimental Animals by State Science and Technology Commission.ResultsThe implied time and amplitude ratio of VEP P100 wave were (98.10±7.07)ms and (0.83±0.14) in normal group,and those in strabismic-induced amblyopia group were (108.50±6.95 )ms and (0.35 ±0.09 ),showing significant differences between two groups (t=4.450,P=0.005 ; t =5.970,P =0.005 ).The numbers of neurons were similar in 21a area of visual cortex between the two groups,but the volume of the neurons was lessen in strabismic-induced amblyopia group.The positive cell densities for NogoA were ( 387.37±2.01 ) cells/mm2,( 354.58± 1.85 ) cells/mm2 and ( 289.68± 1.81 ) cells/mm2 in layer Ⅱ/Ⅲ,Ⅳ and V/V[ respectively in strabismic-induced amblyopia group,and those in normal group were ( 161.39± 1.98 ) cells/mm2,( 128.93 ± 1.26 ) cells/mm2 and ( 96.25 ± 1.49 ) cells/mm2,indicating a significant increase in model cats ( t =- 160.400,- 201.890,- 164.740,P =0.000 ).Conclusions NogoA may play an important role in the regulation of the sensitive developmental period of visual sense and its plasticity.
