1.A novel experimental system for red blood cell-modulating globulin in patients with cancer

Objective To study the effect of red blood cells on the secretion of white blood cells modulating globulin in patients with cancer with a new immunological reaction experimental system. Methods Cancer cells (S180, 5?10~6/ml) or 0.2ml of NS or 0.2ml of white blood cells and 0.3ml of plasma were added to anticoagulant suspension of white blood cells treated by citric acid and incubated for 1h at 37℃. The content of globulin (g/L) was determined by Olympus AU100 detector method. 0.2ml of cancer cells was added to 0.2ml of whole blood cells and 0.3ml of plasma in the whole blood experimental group. 0.2ml of NS was added to 0.2ml of whole blood cells and 0.3ml of plasma in the control group. 0.2ml of cancer cells was added to 0.2ml of white blood cells and 0.3ml of plasma in the white blood cell isolation experimental group. 0.2ml of NS was added to 0.2ml of white blood cells and 0.3ml of plasma in the control group. Results Activation rate (0.31?0.09) of globulin in whole blood cell experimental group of patients with cancer was significantly decreased compared to that (0.39?0.14) in normal people. Red cell adhesion rate (0.09?0.08) of globulin in control group was also significantly decreased compared to that (0.29?0.14) in normal people. Conclusion The results of our study indicate that the new experimental system can be used for detection of red blood cell-modulating globulin. The function of red blood cells in modulating globulin of patients with cancer is decreased.
2.Experimental observation on visceral nociception involving nitric oxide in the interstitial nucleus of spinal trigeminal tract

BACKGROUND: Nitric oxide synthase(NOS) is extensively distributed in the central nervous system and modulates neuronal activities. However,there are few reports about NOS' s involvement in nociceptive information transmission at the supraspinal level.OBJECTIVE: To investigate the relation between the primary afferent visceral terminials and the NOS-containing neurons which project from the interstitial nucleus of spinal trigeminal tract(INV) to the parabrachial nucleus(PBN).DESIGN: A controlled experimental observation based on the experimental animals.SETTING: Department of anatomy in a military medical university of Chinese PLA.MATERIALS: The experiment was conducted in the Department of Anatomy,Fourth Military Medical University of Chinese PLA,from January to December 2002. Ten adult male SD rats were selected in this experiment.INTERVENTIONS: Tetramethyl rhodamine-dextran(TMR) and biotinylated dextran-amine(BDA) were injected into the PBN and vago-glossopharyngeal nerves respectively combining with the immunohistochemical method for NOS and the observation with laser confocal scanning microscope.MAIN OUTCOME MEASUREES: Overlapping of TMR retrogradely labeled neurons,NOS positive labeled neurons and transganglionic tracing labeled terminals,and their relationship in INV.RESULTS: After injection of TMR into the PBN,the retrogradely labeled neurons,mostly small and medium-sized(below20 μm),were found in the INV ipsilateral to the injection side. The distribution of the NOS positive neurons overlapped with retrogradely labeled neurons in INV. The proportion of the number of double-labeled neurons with NOS/TMR was 55% (17/31)and 34% (17/49) in the whole population of the NOS positive neurons and retrogradely neurons,respectively. Some transganglionic tracing labeled terminals after BDA injection had close contact with the double-labeled neurons.CONCLUSION: The visceral nociceptive pathway exists from INV to PBN and NO may act as the transmitter or neural signal molecule in the transmission and regulation of visceral nociceptive process.

Hyperhomocysteinemia is an important independent risk factor for cardio-cerebrovas- cular diseases.Studies in recent years have suggested that hyperhomocysteinemia is also an independent risk factor for vascular dementia and Alzheimer's disease.Mild vascular cognitive impairment is the prophase of vascular dementia.Mainly through the damage of vascular wall structure and its function,homocysteine may has correlation with mild vascular cognitive impairment.
4.Using Metallic Stent to Treat Malignant Stricture of Esophagus and Cardia

Purpose:To evaluate the effect of metallic stent in the treatment of malignant stricture in esophagus and cardia.Materials and Methods:Treated by metallic stent:14patients with malignant stricture in esophagus and cardia,13 males and 1 female aged ranged from 57 to 81,were discussed in this article.Among 12 patients,8 were proved as esophageal squamous carcinoma,4 as esophagus adenocarcinoma,and 1 was complicated with esophagotrachial fistula,l0 of the pa- tients could take fluid food and 4 only water.After catheterizing from mouth,the strictured portions dilated with hallon catheter,15 selfmade Z stents coveted with dacron and 1 strecker stent were in 13 and 1 patients respectively.Results:All stems were successfully placed.Patients could eat more easily after the operations.The esophogotrachial fistula was completely sealed.The average lifetime after the operation was 5.5 months.Conclusions:Metallic stent prosthesis is an effective method to treat malignant stricture in esophagus and cardia,and could elevate the life quality of patients sig- nifieantly.
6.Application research of TBL based on evidence-based medicine PICOS model in practice teaching of Neurology

Objective To explore the effect of TBL based on evidence-based medicine PICOS model in practice teaching of neurology. Methods Totally 47 medical undergraduates in our department were chosen and randomly divided into 2 groups during March 2016 to April 2016. 24 students in the trial group were taught with TBL based on PICOS, while other 23 students in the control group were only taught with conventional TBL method. After three times clinical practices, the test referring to disease pathogenesis, clinical manifestations, auxiliary examinations, diagnosis and treatments of related diseases was performed, and meanwhile, questionnaires were distributed to students in order to survey their satisfaction degree of teaching methods. All the evaluation results and scores of two groups were compared. Statistical data were analyzed by using t-test or Chi-square test with SPSS 17.0 software. Results The score of the theoretical test of trial group was significantly superior to the control group [(89.08±3.60) vs. (79.09±7.75), t=5.707, P=0.03 ]. Survey showed that in the experimental group , the number of students in the trial group who thought teaching method could help understand clinical thinking of neurology and could help integrate the theory into clinical practicewas significantly higher than that in the control group (P<0.05). Conclusion In the clinical teaching of neurology, the TBL based on PICOS model is more effective than conventional TBL method for medical undergraduates.
7.Expression and significance of ABCG2 in lung cancer

Objective ABCG2(ATP-binding cassette superfamily G member 2)is a member of ABC transporter superfamily,and participates in the mechanisms of high resistance to chemotherapeutic agents in tumor treatment.The aim of present study is to investigate the expression and significance of ABCG2 in lung cancer tissues.Methods Lung cancer tissues were obtained from 83 patients(64 NSCLC,19 SCLC)by bio-autopsy or surgery.No patients had received chemotherapy or radiotherapy before bio-autopsy or surgery.83 lung cancer specimens were analyzed for ABCG2 protein expression by using immunohistochemistry.Negative controls had the primary antibody eliminated.Human placenta tissues were used as positive control.In addition,five specimens of normal lung tissues were included for comparative study.Results The present study confirmed the predominant localization of ABCG2 transporter in plasma membrane.Some of ABCG2-positive tumors showed mixed membranous and cytoplasmic staining.ABCG2 expression was found in 71.9%(46/64)of NSCLC and 10.5%(2/19)of SCLC.The level of ABCG2 expression was significantly higher in NSCLC than in SCLC(P
8.Optimization of cDNA-AFLP reaction system for Carthamus tinctorius

Objective To construct for the heredity linkage map and to study the functional gene research of Carthamus tinctorius,the factors affecting cDNA amplified fragment length polymorphism(cDNA-AFLP) of C.tinctorius were investigated with developing and optimizing the cDNA-AFLP reaction system.Methods Improved Trizol method was used to extract RNA from compounds in new petals specific to safflower.With the help of M-MLV RTase without RNase activity combined with replacement synthesis method,double-stranded cDNA was synthesized from total RNA;cDNA was digested by restriction enzyme MseI/EcoRI and ligated by two steps.Then the products were provided for pre-amplification and selected amplification of different concentration gradients.After tiny modifications of system concentration,finally PAGE electrophoresis and silver-staining were performed.Results High purity and integrated total RNA for later cDNA synthesis were obtained and high quality cDNA was synthesized with the help of M-MLV RTase without RNase activity combined with replacement synthesis method.The cDNA-AFLP reaction system in C.tinctorius was as follows: 250 ng integrity cDNA was digested thoroughly at 37 ℃ for 6 h,and ligated 12 h at 16 ℃.Furthermore,the sample dilution multiplication was 10 fold for pre-amplification and 150 fold for selected amplification under the proper system concentration.According to the above reaction system,the polymorphous strips with high resolution power in PAGE electrophoresis were clear and stable.Conclusion The cDNA-AFLP reaction system established and optimized in this experiment is suitable for the functional gene analysis of C.tinctorius.

Objective To approach the capacity of erythrocyte regulating IL-8 in patients with primary hepatocarcinoma (PHC) by a new experimental system of hemaimmune. Methods 0.2ml suspension of cancer cells (S180: 5?10~6/ml) or NS were added into 0.2ml anticoagulant suspension of whole blood cells or leukocytes and 0.3ml plasma, then incubated for 1 h at 37℃. The content of IL-8 was determined by ELISA. Results In the patients with PHC, the IL-8 levels (pg/ml) in experimental and control groups of whole blood cells, and in experimental and control groups of leukocytes were 376.35?243.96, 353.64?271.92, 461.27?277.11 and 424.97?278.93, respectively; while in the normal human, they were 11.36?6.93, 4.98?4.35, 29.41?30.66 and 20.77?24.20, respectively. In the patients with PHC, the activation rates of cancer cells in the experimental groups of both whole blood cell and leukocyte were 0.22?0.24 and 0.25?0.53, respectively; while in normal human, they were 2.49?2.33 and 0.75?0.21, respectively. In the patients with PHC, the IL-8 adsorption rate of erythrocyte in both experimental and control groups of whole blood cell were 0.22?0.18 and 0.17?0.33, respectively; while in normal human, they were 1.18?2.29 and 0.86?0.49, respectively.
