1.Observation of improved extra panretinal photocoagulation for high risk proliferative diabetic retinopathy

AIM: To observe the therapeutic effect and safety of improved extra panretinal photocoagulation ( E - PRP) in the treatment of high risk proliferative diabetic retinopathy (hsPDR).
METHODS: A total of 88 consecutive cases (102 eyes) with hsPDR were diagnosed by fundus fluorescein angiography(FFA) from February 2011 to December 2014 in our hospital . Fifty two eyes had been treated by improved E - PRP with 532nm frequency - doubled laser. Fifty eyes had been treated by standard PRP. All cases were checked by FFA and fundus photocoagulation every 3mo. Patients with persisting neovascularization or non perfusion area were treated with laser again. All cases were followed up 6-36mo.
RESULTS: The postoperative visual acuity had no statistical difference between two groups ( P > 0. 05). In improved E - PRP group, retinal non perfusion area and neovascularization disappeared in 35 eyes ( 67%). Effective rate was 88%. Six eyes (12%) underwent pars plana vitrectomy because of vitreous hemorrhage, fiberosis and stretched retinal detachment. In standard PRP group, retinal nonperfusion area and neovascularization disappeared in 23 eyes ( 46%). Effective rate was 66%. Seventeen eyes(34%) underwent pars plana vitrectomy because of anterior retina bleeding or vitreous hemorrhage. The rate of neovascularization disappeared and effective rate had statistical difference between two groups (P<0. 05).
CONCLUSION: It is a safe and effective methods to treat hsPDR by improved E-PRP and it was more effective than traditional PRP.
2.Pharmacodynamical mechanism study of “Heat Asthma Formula” in treatment of bronchial asthma

Objective: In this paper the pharmacodynamical mechanism of Heat Asthma Formula(HAF)(Herba Ephedrae, Rhizoma pinelliae, Flos Lonicerae, etc) in the treatment of bronchial asthma was discussed. Methods: By means of the radio labelled aglycone, applying phlogistic medium TXB2, ? receptor on bronchus and cyclic nucleotide (cAMP/cGMP) in lung tissue used as observed targets. Results: The results showed HAF could inhibit the release of phlogistic medium TXB2, improve the conjugate rate of ? receptor, adjust the proportion between cAMP and cGMP. Conclusion: The treatment on bronchial asthma of “Heat Asthma Formula” was perhaps concerned with above mentioned contents.
3.Effect of acupuncture on cerebral amino acids and neurotransmitters in the immature rat model of cerebral palsy

BACKGROUND:Acupuncture is proved to have therapeutic effect on cerebral palsy of immature rats.
OBJECTIVE:To discuss the possible effect on cerebral amino acids and neurotransmitters in immature rats with cerebral palsy by using fire-needle.
METHODS:Cerebral palsy models were established in 45 Wistar rats aged 1 week by left common carotid artery ligation and randomly assigned into model, acupuncture and positive groups (n=15 per group). The other five rats were defined as controls, without any management. At 3 days, positive group rats were oral y administrated with brain enzyme solution, and acupuncture group given fire-needle treatment at points of Neiguan (P 6), Yongquan (K 1), Quchi (LI 11) and Baihui (Du 20) for 21 days. Hanging test, inclined plane test and Y maze test were performed 1 hour after the last treatment, and the cerebral cortex and hippocampus were extracted and homogenized after execution. Lelves of acethl choline, norepinephrine, 5-hydroxytryptamine, glutamic acid andγ-aminobutyric acid were recruited, and morphology of hippocampus was observed by pathological section.
RESULTS AND CONCLUSION:(1) Within 21-day treatment, positive and acupuncture treatment rats were more active than model group. Compared with the control group, the body mass in the model group was significantly decreased (P<0.05);compared with the model group, the body mass in the positive and fire-needle groups were significantly increased (P<0.05), while it showed no significant difference between latter two groups. (2) The hanging time of model group rats was significantly shorter than that of control group (P<0.05), and significantly lengthened in the positive and fire-needle groups compared with the model group (P<0.05). Compared with the control rats, the error numbers of model group rats were significantly increased (P<0.05), and conversely, significantly decreased in the positive and fire-needle groups (P<0.05). (3) In the model group, the levels of norepinephrine, 5-hydroxytryptamine andγ-aminobutyric acid were significantly decreased, while glutamic acid significantly increased compared with the control group (P<0.05);compare with the model group, the positive and fire-needle group rats held significantly increased levels of norepinephrine, 5-hydroxytryptamine andγ-aminobutyric acid and decrease of glutamic acid (P<0.05). (4) Hematoxylin-eosin staining showed that in the mode group, there were abundant neurons damaged, cel volume reduced, nucleus pycnosis and unclear border with nucleus;both positive and fire-needle groups could improve neuron structure induced by cerebral ischemia. (5) In conclusion, fire-needle can treat cerebral palsy effectively.

5.Anti-?2 integrin inhibits invasion and migration of osteosarcoma MG63 cells

Objective: Changes in the ?2 integrin of adhesion molecules between cells are closely associated with the invasion and migration of tumor cells.This study aimed at the effect of anti-?2 integrin on the invasion and migration of osteosarcoma MG63 cells. Methods: The osteosarcoma MG63 cell line was cultured in the DMEM medium.The effect of the anti-?2 integrin monoclonal antibody on the migration and invasion of tumor cells were measured by scratch assay and Transwell assay.The migration and invasion cells were stained by Crystal violet staining and counted under the hundredfold microscope.Results: Compared with the control group,the migration and invasion abilities of the MG63 cells were significantly decreased in the anti-?2 treatment groups.Conclusion: Anti-?2 integrin may inhibit the migration and invasion of osteosarcoma cells.
6.Combination of acupuncture with placenta derived mesenchymal stem cell transplantation for cerebral palsy in a rat model

BACKGROUND:Both of acupuncture and mesenchymal stem cel transplantation have therapeutic effects on cerebral palsy. OBJECTIVE:To explore the therapentic effect of acupuncture combined with placenta derived mesenchymal stem cel s on rat cerebral palsy. METHODS:Thirty-five Wistar rats were equivalently randomized into five groups, including control group, model group, acupuncture group, transplantation group and combination group. Rat models of cerebral palsy were prepared in al groups except the control group. At 7 days after modeling, in the transplantation group, 10μL placenta derived mesenchymal stem cel s were transplanted into the right cerebral cortex and striatum of rats;rats in theacupuncture group were given acupuncture at Shuigou (DU26), Dazhui (DU14), Baihui (DU20), Guanyuan (RN4) and Qihai (RN6) points;rats in the combination group given acupuncture and transplantation treatment as described above;the model and control groups received no treatment. Then, after 7-day intervention, the body mass, neurological function and pathological changes of rat brain tissues were measured and observed, respectively. . RESULTS AND CONCLUSION:The order of the growth rate of body mass was as fol ows:control group>combination group>acupuncture and transplantation groups>model group, and al groups had significant differences except the two groups of acupuncture and transplantation (P<0.05);findings from holding tract, foot error and water finding in maze tests showed the neurological function was best in the control group, fol owed by the combination group, as wel as acupuncture and transplantation groups, and the worst in the model group. Al groups had significant differences except acupuncture and transplantation groups (P<0.05). Besides, in the model group, cel s in the brain and hippocampus decreased, arranging disorderly with fuzzy structure, and inflammatory cel s appeared;pathological changes in the other groups were in between control group and model group, Additional y, brain tissues in the combination group exlibited better pathological changes than those in the acupuncture and transplantation groups, which were similar with the normal brain tissues. To conclude, acupuncture, placenta derived mesenchymal stem cel s transplantation, or their combination can improve neurological function in rats with cerebral palsy, especial y the combination treatment.
7.Surgical treatment for acute necrotizing pancreatitis:14 years' experience in a single Chinese Center

Objective:To investigate the reasonable approach and surgical indication for acute necrotizing pancreatitis(ANP)by analyzing the factors that affecting the mortality of ANP.Methods:One hundred and twelve patients with ANP were retrospectively divided into two groups-the dead and the survivors.Some parameters were analyzed statistically to reveal what's the reason for death.Results:The average age,sex ratio and onset of illness were similar between two groups.And the ratio of early shock,early adult respiratory distress syndrome (ARDS),high temperature,leukocytosis and high blood glucose between two groups were also similar between two groups(P>0.05,respectively).The important factors that affecting the mortality were:①severity of pancreatic necrosis,②improper surgical approach,③incorrect surgical indication.Conclusion:The patiets with mild or moderate ANP should mainly receive conservative treatment for 48~72 hours.The early shock and ARDS should be redressed before surgical intervention.If the operation is unavoidable,the swelling pancreas should be dissected fully,which will provide sufficient drainage after operation,and duodenostomy should be performed during operation.
8.Finite element mechanical analysis on fracture hip supporting joint for treatment of femoral neck fracture

BACKGROUND: For treatment of femoral neck fracture, all therapies with the exception of joint replacement encounter the problems including slow healing, poor prognosis, various complications, and unable to bear weight for long time. Fracture hip supporting joint (FHSJ) is a novel unlimited hip support implement that possesses the double functions of fracture fixation and joint supporting and can be used to prevent and treat the complications of femoral neck fracture in young people.OBJECTIVE: To investigate the mechanical effects of FHSJ on treatment of femoral neck fracture.METHODS: Three types of two-dimensional finite element models were constructed by AutoCAD: normal hip (group A), femoral neck fracture fixed with two screws (group B), and femoral neck fracture fixed with two screws and FHSJ (group C). The grids of two-dimensional four nodal point elements were divided by ANSYS (PLANE82). Under the identical condition, the calculations were performed respectively.RESULTS AND CONCLUSION: The stress peak value of femoral head weight-bearing zone was 1.029 and 1.63 MPa in group A and group B, respectively, and that in the group C was 0.1-0.4 MPa. The stress peak value of the screws was 37.186 and 7.474 MPa in the group B and group C, respectively. These results indicate that FHSJ installation based on fixation of multiple screws could promote the recovery of femoral head and neck, which exhibits promising prospects in treatment of femoral neck fracture in young people.
9.Ventricular synchronicity: observations comparing pulse flow and tissue Doppler assessment in a Chinese healthy adult cohort.

BACKGROUNDMechanical asynchrony is an important parameter in predicting the response to cardiac resynchronization therapy, but detailed knowledge about cardiac timing in healthy persons is scarce. Therefore, in the current study, we sought to investigate the physiological status of interventricular synchronicity using pulse wave flow and tissue Doppler imaging in a healthy Chinese population.
METHODSEighty-eight healthy volunteers underwent standard flow and tissue Doppler echocardiographic examinations. Ventricular inflow and outflow pulse wave flow Doppler patterns were recorded together with annulus pulse tissue Doppler imaging. Time intervals from the beginning of the QRS complex to the onset, peak and end of each wave were measured.
RESULTSThe onsets of systole between left and right ventricles were highly synchronized by both imaging modalities. However, the left ventricle reached the peak flow ejection and peak mechanical contraction earlier than the right ventricle, (165.61 ± 26.23) ms vs. (204.3 ± 34.55) ms (P < 0.01) and (133.62 ± 26.19) ms vs. (191.25 ± 38.47) ms (P < 0.01). Time to peak early diastolic relaxation was earlier in the left ventricle than in the right heart, (500.23 ± 56.52) ms vs. (524.94 ± 47.42) ms (P < 0.01).
CONCLUSIONSLeft and right ventricles were well synchronized at the onsets of systole and diastole even though interventricular peak systolic and peak early diastolic dyssynchrony was observed in healthy people by pulse wave Doppler imaging. In addition, diastolic timing events were slightly affected by age and gender.

10.Effect of nordihydroguaiaretic acid on expressions of VEGF and its receptor KDR in vitro

Objective To explore the effect of nordihydroguaiaretic acid (NDGA) on the expressions of vascular endothelial growth factor (VEGF) and its receptor, kinase-inserted domain containing receptor(KDR) and the possible mechanism. Methods The expression of VEGF in human malignant glioma cell line SHG-44 and that of KDR in human umbilical vein endothelial cell (HUVEC) line ECV-304 were observed 1~3 d after NDGA treatment with immunohistochemistry, in situ hybridization and image analysis. Results The expression of VEGF was declined at protein or mRNA levels in SHG-44 cells after treated with 100 μmol/L NDGA for 1 to 3 d. The expression of KDR in endothelial cells with 100 μmol/L NDGA treatment for 1 to 3 d was decreased too, in a more obvious way compared with the decline of VEGF expression in SHG-44 cells. Conclusion The results suggest that NDGA inhibits the expression of VEGF in glioma cells as well as that of VEGF receptor KDR in endothelial cells, which may be the important molecular mechanism of anti-angiogenesis of NDGA.
