OBJECTIVE: To investigate factors that affected the distribution of rehydrated particle size of freeze-dried docetaxel liposome and develop a technique of producing docetaxel liposome that can meet clinical requirements. METHODS: The single factor method was utilized in this study. Docetaxel liposome were prepared using different types of negatively charged phospholipids, or lecithins with varying phosphatidylcholine (PC) content, or various cryoprotectants at different gradient concentrations, followed by freeze-drying with different protocols. The particle sizes of corresponding rehydrated products were compared and the effects of different factors were analyzed. RESULTS: The optimal liposomal membrane was composed of dipalmitoyl phosphatidic acid (DPPA), 20% trehalose and lecithin containing 92% PC. The best freeze-drying protocol included following steps; flash freezing, 19 h of sublimation-drying and 9 h of desorption-drying. CONCLUSION: The parameters, including negatively charged phospholipids, cryoprotectants, PC content of lecithin and lyophilization protocols, had various effects on the particle sizes of reconstituted docetaxel liposome.

A catheter(0.3 mm O.D.)was introduced into the external carotid artery(ECA) lumen into the lumen of the internal carotid artery(ICA), then it was gently advanced to the intracranial segment of ICA until the intraluminal catheter was 2~3mm from the origin of MCA. At this point thrombin were injected into ICA to induce thrombosis. Behavior disorders were observed at 3, 6, 12h after the operation. Willis circle, infarct volume and histopathologic changes were examined 12 hours after operation. The results showed that the model had stable behavior disorder score and infarct volume. Mixed thrombus could be observed at the origin of the MCA. Infarction volume of this model was similar with that of the model developed by linear occlusion of MCA in our laboratory. The model has the advantages of easy operation, reliable ischemia and reproducible infarct volume. It may be useful in documenting the safety and efficacy of thrombolytic therapies.

Objective To observe the effect of treadmill training on metabotropic glutamate receptor of a middle cerebral artery occlusion model in rat and its underlying mechanisms. Methods Sprague-Dawley male rats of 2 or 3 months old were randomly divided into 3 groups: a sham operation group, an iscbemia-reperfnsion group and a treadmill training group (subject to 2 weeks of training after the ischemia-reperfusion). All the animals were sacri-ficed after 2 weeks of training and their brains were sampled for measurement of the expression level of the striatum mGluR Ⅰ-mRNA, using RT-PCR technique. Results The striatum mRNA of mGluR1 and mGluR5 was signifi-cantly elevated in the ischemia-reperfusion group(P<0.01), but treadmill training significantly suppressed the ele-vation of the expression of mRNA of mGluR1 and mGluR5 (P<0.01). Conclusions Treadmill training can signif-icantly downregulate mGluR Ⅰ-mRNA expression. This might be one of the important mechanisms for inhibition of the excitatory glutamate production.

Objective To explore the protective effect of Red Sage Root on bronchopulmonary dysplasis(BPD) induced by hyperoxia in newborn rats.Methods On the 2nd postnatal day,SD newborn rats were randomly assigned to 4 groups:air and NS group(group Ⅰ),air and Red Sage Root group(group Ⅱ),hyperoxia and NS group(group Ⅲ),hyperoxia and Red Sage Root group(group Ⅳ).The rats in group Ⅲ and group Ⅳ were exposed to hyperoxia(the level of oxygen was 900-960 mL/L).The rats in group Ⅱ and group Ⅳ were injected with Red Sage Root intraperitoneally(10 mg/kg)daily.On 14 days after birth,6 rats in each group were killed.Lung histologic changes,radical alveolar counts(RAC)were monitored.Thiobarbituric acid method,nitrite method,2-nitroben zoic acid method were used to determine the concentration of malony ldialdengde(MDA),superoxidedismutase(SOD),glutathione peroxidase(GSH-Px) were monitored.Results 1.Group Ⅲ and group Ⅳ showed the inhibition of lung development and the evident lung fibrosis.In contrast to group Ⅰ and group Ⅱ,RAC in group Ⅲ and group Ⅳ decreased dramatically(Pa
