A catheter(0.3 mm O.D.)was introduced into the external carotid artery(ECA) lumen into the lumen of the internal carotid artery(ICA), then it was gently advanced to the intracranial segment of ICA until the intraluminal catheter was 2~3mm from the origin of MCA. At this point thrombin were injected into ICA to induce thrombosis. Behavior disorders were observed at 3, 6, 12h after the operation. Willis circle, infarct volume and histopathologic changes were examined 12 hours after operation. The results showed that the model had stable behavior disorder score and infarct volume. Mixed thrombus could be observed at the origin of the MCA. Infarction volume of this model was similar with that of the model developed by linear occlusion of MCA in our laboratory. The model has the advantages of easy operation, reliable ischemia and reproducible infarct volume. It may be useful in documenting the safety and efficacy of thrombolytic therapies.

Objective To observe the effect of treadmill training on metabotropic glutamate receptor of a middle cerebral artery occlusion model in rat and its underlying mechanisms. Methods Sprague-Dawley male rats of 2 or 3 months old were randomly divided into 3 groups: a sham operation group, an iscbemia-reperfnsion group and a treadmill training group (subject to 2 weeks of training after the ischemia-reperfusion). All the animals were sacri-ficed after 2 weeks of training and their brains were sampled for measurement of the expression level of the striatum mGluR Ⅰ-mRNA, using RT-PCR technique. Results The striatum mRNA of mGluR1 and mGluR5 was signifi-cantly elevated in the ischemia-reperfusion group(P<0.01), but treadmill training significantly suppressed the ele-vation of the expression of mRNA of mGluR1 and mGluR5 (P<0.01). Conclusions Treadmill training can signif-icantly downregulate mGluR Ⅰ-mRNA expression. This might be one of the important mechanisms for inhibition of the excitatory glutamate production.

OBJECTIVE: To investigate factors that affected the distribution of rehydrated particle size of freeze-dried docetaxel liposome and develop a technique of producing docetaxel liposome that can meet clinical requirements. METHODS: The single factor method was utilized in this study. Docetaxel liposome were prepared using different types of negatively charged phospholipids, or lecithins with varying phosphatidylcholine (PC) content, or various cryoprotectants at different gradient concentrations, followed by freeze-drying with different protocols. The particle sizes of corresponding rehydrated products were compared and the effects of different factors were analyzed. RESULTS: The optimal liposomal membrane was composed of dipalmitoyl phosphatidic acid (DPPA), 20% trehalose and lecithin containing 92% PC. The best freeze-drying protocol included following steps; flash freezing, 19 h of sublimation-drying and 9 h of desorption-drying. CONCLUSION: The parameters, including negatively charged phospholipids, cryoprotectants, PC content of lecithin and lyophilization protocols, had various effects on the particle sizes of reconstituted docetaxel liposome.

Securin is a multifunctional protein which is encoded by PTTG gene.In the metaphase to anaphase transition during mitosis,Securin is involved in sister chromatid separation.The aberrant expression of Securin is found in many endocrine and related cancers,such as pituitary tomour,thyroid cancer,ovarian cancer,colon cancer et al.The function of Securin in cancer is related to its gene transactivation and association with other poteins like p21,p53.

Objective To find the rule of the distribution of H antigens on AB subgrouperythrocytes.Methods ABO subgroups were confirmed by using serological and molecular biology (PCR-RFLP) methods. AB subgroup with strong H was defined as red cell agglutination by anti-H of 2 scores or more higher than that of B cells.Results Strong H was only found in certain AB subgroups ,CisAB(100%),B(A)(100%), AxB(46.2%) and A2B(43.6%), but seldom in others among Chinese population.Conclusion The fact that H type-3, which comes from A type-2, can hardly be transferred by B and weak A glycosyltransferase can help to explain why some ‘strong’ H combines with ‘weak’ A in AB erythrocytes. Why only little H can be found in 53.8% AxB, 56.4% A2B and all A3B, AmB subgroup samples still cannot been explained.

Objective To analyze the relation between glycosylated hemoglobin (HbA1c) and coronary heart disease (CHD) among diabetic and non-diabetic individuals.Methods A total of 1190 in patients from January 2009 to September 2009 were selected and divided into two groups:225 patients with diabete mellitus and 965 patients without.We collected the age,sex,triglyceride ( TG),total cholesterol ( CHO),high-density lipoprotein ( HDL-C),low-density lipoprotein ( LDL-C),fasting blood-glucose (FBG),glycosylated hemoglobin (HbA1c),high-sensitivity CRP (hs-CRP) and analyzed the relation between these indexes and CHD in diabetic and non-diabetic individuals respectively,then,analyzed the difference of HbA1c between patients with and without CHD after correcting the differentiating factor with a multivariable-adjusted model.Meanwhile,according the HbA1c level,we divided all participants into four groups:4.0％ -5.4％( Ⅰ),5.5％ -5.9％ ( Ⅱ),6.0％ -6.4％ ( Ⅲ),≥6.5％ ( Ⅳ),and observed the distribution of HbA1c and coronary heart disease in diabetic and non-diabetic individuals respectively.Results ( 1) In non-diabetic individuals,statistically significant difference of male( 80.5％ υs 62.7％),age [ ( 59 ±11) years υs (55 ± 11) years],FBG [(5.62 ± 1.48) mmol/L υs (5.30 ±0.84) mmol/L],HbA1c [(5.98±0.92)％ υs (5.65 ±0.53)％],CHO [(4.48 ±1.01) mmol/L υs (4.77 ±1.04) mmol/L],LDL-C [(2.59±0.87) mmol/L υs (2.79 ±0.86) mmol/L],HDL-C [(1.08 ±0.26) mmol/L υs(1.21 ±0.32) mmol/L] was observed between patients with and without CHD (P ＜0.01),however,in diabetic individuals,only LDL-C [ ( 2.56 ± 0.81) mmol/L υs ( 3.07 ± 0.90) mmol/L],CHO [ (4.44 ±0.95) mmol/L υs ( 5.08 ± 1.16) mmol/L] were observed ( P ＜ 0.0 1).(2) In all participants,the higher of HbA1c,the higher of CHD percent,compared to patients without CHD,the percent of high HbA1c in CHD patients was significantly higher.After adjusting for age,sex,HDL-C,HbA1c was an independent risk marker for CHD in non-diabetic individuals ( OR:1.935,95％ CI:1.356 - 2.762,P ＜ 0.05),however,links between FBG in the non-diabetic range and CHD appeared weaker after controlling indexes listed above( OR:1.507,95％CI:1.082 -2.098,P＜0.05).Conclusion HbA1c is an independent risk factor for CHD in nondiabetic individuals and prior to FBG.
