2.Application of Xpert MTB/RIF assay on rapid detection of MTB and rifampin-resistant

Objective To investigate clinical value of Xpert M TB/RIF assay on rapid detection of M TB and rifampin‐resistant . Methods 408 cases of tuberculosis were detected by acid‐fast stain ,Solid culture and Xpert MTB/RIF ,the results were compared . Results All the samples were detected by acid‐fast stain ,culture and Xpert MTB/RIF assay ,the positive rates were 25 .00% (102/408) ,42 .16% (172/408) and 58 .09% (237/408) ,respectively .The positive rates of Xpert MTB/RIF assay were higher than acid‐fast stain and culture method ,and the difference is statistically significant(P<0 .05) .There are 168 cases of specimen which were tested positively by both culture and Xpert M TB/RIF ,the RIF resistance rates from standard drug susceptibility testing and Xpert MTB/RIF assay respectively were 26 .19% (44/168) and 27 .38% (46/168) ,and had no statistical significance(P>0 .05) .Accord‐ing to the result of drug susceptibility with proportion method ,the sensitivity and specificity of Xpert MTB/RIF in detecting RIF resistance were 97 .73% (43/44) and 97 .58% (121/124) .Conclusion Xpert MTB /RIF can quickly and accurately detect MTB and RIF resistance ,it has important clinical significance .
3.Human umbilical vein endothelial cells support hematopoiesis and expansion of hematopoietic stem/progenitor cells in vitro

Objective: To investigate the role of human umbilical vein endothelial cells (HUVEC) in supporting hematopoiesis and the expansion of hematopoietic stem/progenitor cells in vitro. Methods: According to the fact that HUVEC supernatant has colony stimulating activity shown by methylcellulose colony-forming assay and HUVEC can maintain the survival of mononuclear cells for at least four weeks in vitro, CD34+ cells from umbilical cord blood were seeded with (HUVEC group) or without (control group) HUVEC monolayer. Every week cells were collected and counted, the frequency of CFU-GM was measured by using methylcellulose colony-forming assay, and the percentage of CD34+ and CD41a+ cells was measured by flow cytometry. Results: In control group,all the CD34+ cells died in two weeks. However, in HUVEC group,most nucleated cells and CD34+ cells were expanded by 68.1±14.8 fold and 6.6±1.4 fold,respectively at the third week while CFU-GM expansion reached its peak (5.7±2.1 fold) at the week 2. Moreover, the percentage of CD41a+ cells was enhanced significantly, reaching a maximum (15.6%) at the week 3. Conclusions:HUVEC can support hematopoiesis in vitro and expand the hematopoietic progenitor cells and CD41a+ cells in direct contact coculture.

5.Effects of hydrogen sulfide preconditioning on myocardial ischemia reperfusion injury in rats.

OBJECTIVETo investigate the effects of hydrogen sulfide preconditioning on myocardial ischemia reperfusion injury in rats.
METHODSSprague-Dawley male rats were divided into 4 groups with 10 in each group: in S group rats received sham operation; in IR group rats were given with NS (1.0 ml/kg iv) 24 h before ischemia; in H group rats were treated with NaHS (0.05 mg/kg iv) 24 h before ischemia; and in D group, NaHS-treated rats received 5-hydroxydecanoate (5-HD) 15 min before ischemia. Rats in IR group,H group and D group were subjected to ischemia by occlusion of coronary artery for 30 min followed by 2 h of reperfusion. At the end of the reperfusion,myocardial infarct size was measured. SAM-s was measured by Western blotting. Plasma SOD activity and MDA were determined at the end of reperfusion.
RESULTSThe infarct size was significantly lesser in H group (25.40 % ± 3.54%) than that in IR group (38.27% ±5.64%,P<0.05). The SAM-s protein expression in myocardium was significantly lower in H group than that in IR group. The plasma MDA content was significantly lower and SOD activity was higher in H group than those in IR group,but there was no difference between IR group and D group.
CONCLUSIONThe hydrogen sulfide preconditioning attenuates myocardial IR injury possibly through down-regulating SAM-s expression,reducing the production of oxygen free radicals and enhancing anti-oxidize effect in rats.

6.Study on the status and quality of sleep-related influencing factors in medical college students.

OBJECTIVETo study the status and influencing factors on sleep quality in some medical college students.
METHODSStratified sampling, pittsburgh sleep quality index (PSQI), self-evaluation depression scale (SDS), self-evaluation anxiety scale (SAS) and self-developed questionnaire of influencing factors on the quality of sleep in medical college students were used. Cumulative odds logistic model was performed to analyze the related factors on the quality of sleep.
RESULTS19.17 percent of the medical college students showed poor quality of sleep and the difference between genders was not statistically significant (P > 0.05). Statistically significant (P < 0.05) difference was seen among different years of students and correlation was found between sleep quality and depression or anxiety (P < 0.0001). Factors influencing on the quality of sleep in medical college students would include: worry of sleep, irregular work/rest, worry on examination, stress, relationship with classmates, self-evaluated health condition, environments of the dormitory and late to bed.
CONCLUSIONInfluencing factors were identified and comprehensive measures should be taken to improve the quality of sleep.

7.Chemical constituents from safflower injection and their bioactivity.

The chemical constituents of Safflower injection were isolated and purified by polyamide, silica gel, Sephadex LH-20, ODS column chromatographies and preparative HPLC. As a result, sixteen compounds have been isolated. Based on the spectral data analysis, their structures were elucidated as scutellarin (1), kaempferol-3-O-β-rutinoside(2), hydroxysafflor yellow A(3), rutin (4), coumalic acid(5), adenosine(6), syringoside(7), (3E)-4-(4'-hydroxyphenyl)-3-buten-2-one(8), (8Z)-decaene-4, 6-diyne-1-Oβ-D-glucopyranoside(9), 4-hydroxybenzaldehyde (10), (2E, 8E) -tetradecadiene-4, 6-diyne-1, 12, 14-triol-1-O-β-D-glucopyranoside (11), kaem-pferol-3-O-β-sophorose (12), uridine (13), roseoside (14), cinnamic acid (15), and kaempferol (16). Compounds 1,2,7,9,11 and 12 were isolated from the Safflower injection for the first time. The anti-platelet aggregation activities of the isolated compounds were assayed. The results indicated all tested compounds exhibited potent activity except for 5, while 2, 3, 9 and 12 showed strong activity against platelet aggregation.

8.Research progress of Kruppel-like factor family in malignant tumors

Malignant tumors usually have no obvious clinical symptoms in the early stage. Most patients are already in the advanced stage when they are diagnosed. Some patients have lost the opportunity for operation, resulting in poor prognosis. Therefore, how to find the best therapeutic target for such patients and improve the prognosis of patients has gradually become the focus of scholar′s attention. Recently, Kruppel-like factor (KLF) is a transcriptional regulator that can bind to the target DNA, and its family plays an important role in the occurrence and development of malignant tumors. It has also been confirmed that the KLF family affects the proliferation, differentiation and migration of tumor cells, but the specific mechanism is still not fully elucidate. Consequently, in order to further explored the effect of the KLF family on tumors, this study intends to briefly review the roles and regulatory mechanisms of the KLF family in the cell proliferation, differentiation and migration of malignant tumors, hoping to provide new target for the biological treatment of tumors.
10.Analysis of mortality trend of cancer from 1980 to 2002 in Linqu County Shandong Province.

OBJECTIVETo investigate the trend of total cancer mortality in Linqu County Shandong Province from 1980 to 2002.
METHODSA retrospective survey on all causes of death in 1980 - 1982, 1990 - 1992 and 2000 - 2002 was conducted in Linqu County, a high risk area of gastric cancer in Northeast of China, respectively.
RESULTSThe cancer death, was found the third leading cause of death in 1980 - 1982 in Linqu County, and the second to that of vascular disease in 2000 - 2002. The cancer mortality (standardized mortality) was 108.97/100,000 (111.48/100,000), 132.38/100,000 (127.94/100,000) and 148.48/100,000 (105.53/100,000) in 1980 - 1982, 1990 - 1992 and 2000 - 2002, respectively. The trend of cancer mortality was significantly increased (Z = 13.42, P < 0.0001). The added cancer-eliminated life expectancy in three periods was 2.46 years, 3.29 years and 3.76 years in male (F = 13.99, P < 0.0001), and 1.67 years, 2.30 and 2.33 years in female (F = 13.61, P < 0.0001), respectively. The standardized mortality of gastric cancer (percentage in all cancer death) was 44.93/100,000 (40.29%), 41.37/100,000 (32.34%) and 27.73/100,000 (26.90%) in 1980 - 1982, 1990 - 1992 and 2000 - 2002, respectively. The trend of gastric cancer standardized mortality was significantly reduced (Z = 6.35, P < 0.01).
CONCLUSIONThe mortality of cancer in Linqu County has been increased from 1980 to 2002, but no such trend was found after adjusting ages. However, there was a decreased trend on standardized mortality of gastric cancer in the past 20 years.
