Journal of Applied Clinical Pediatrics 1992;0(06):-
Objective To study the influence of interleukin-11 (IL-11) on graft versus host disease(GVHD) and graft versus leukemia(GVL) after allogenic bone marrow transplantation and related mechanism in acute lymphoblastic leukemic(ALL) junior mice. Methods The impact of IL-11 on CD4 and CD8 T cell after transplantation was evaluated by flow cytometry;it was observed that the GVHD pathology chance of internal after transplatation in ALL junior mice;the survial time after transplantantion was recorded;the level of tumor necrosis factor-?(TNF-?) which evidently related to GVHD was evaluated by ELISA.Results IL-11 could decrease the quantity of CD4 T cell and increase CD8 T cell;IL-11 could obviously increase the survival time and decrease the level of TNF-?. After ALL junior mice transplatated,IL-11 could delay and relieve GVHD.Conclusion IL-11 can alleviate GVHD and retain the effect of GVL after allogenic bone marrow transplantation.

Aim To explore the role of nicotinamide adenine dinucleotide phosphate ( NADPH ) oxidase in high glucose-induced injury in H9c2 cardiac cells. Methods H9 c2 cardiac cells were exposed to differ-ent concentrations of high glucose(5. 5 mmol·L-1 ,11 mmol · L-1 ,22 mmol · L-1 ,33 mmol · L-1 ,44 mmol ·L-1 , 55 mmol · L-1 ) for 24 h and different time pints of high glucose(0 h,12 h,24 h,36 h,48 h,72 h) . Cell viability was measured by MTT colorimetry, the protein expression of Bcl-2 , Bax and NADPH oxi-dase submits such as p22 phox , p47 phox and p67 phox were determined by western blotting. Results H9c2 cardi-ac cells exposure to high glucose for 24 h showed on decrease in cell survival and the Bcl-2 expression while an increase in the Bax expression ( P<0. 05 ) . Moreo-ver, high glucose could markedly up-regulate the activ-ity of NADPH oxidase characterized by the enhanced expression of p22 phox , p47 phox and p67 phox ( P<0. 05 ) . Conclusion Activating NADPH oxidase may play an important role in high glucose-induced injury in H9 c2 cells.

Objective To study the expression profile of microRNA (miRNA) and the roles in pathogenesis of acute liver failure in mouse model. Methods Eighty-five BALB/c mice were divided into four groups: 40 in model group of acute liver failure were intraperitoneally injected with Dgalactosamine (D-GalN) and lipopolysaccharides (LPS); 20 in D-GalN group were injected with DGalN only; 20 in LPS group were injected with LPS only; 5 in control group were injected with saline.Liver histology of mouse was observed at hour 0, 5, 7 of injection, and sera and liver tissues were collected at hour 0, 1, 3, 5, 7, 9 of injection. Meanwhile, levels of inflammatory factors [tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6)] in serum and liver tissue were detected by realtime polymerase chain reaction (PCR). Lock nucleic acid (LNA)-based miRNA microarray technology was used to detect the expression profile of hepatic miRNA, and the expression of miRNA was verified by real time quantification-polymerase chain reaction (RT-PCR). Mouse macrophage Raw264.7 cells were induced by LPS in vitro and the expressions of miRNA at different time points were detected. The comparison of means among groups was analyzed using one way ANOVA and the correlation were analyzed by Pearson and Spearman correlation. Results Microarray analysis found that the expression profile of miRNA during the acute liver failure changed dramatically. There were 97 miRNA in model group changed significantly compared with control group (P<0.01), including 21 up-regulated and 27down-regulated at hour 5 and 7 of injection. Furthermore, the expressions of miR 146a and miR-155were verified by RT-PCR and found they both increased progressively over time after injection.Correlation analysis showed that miR-155 was well correlated with both TNF-α and IL-6 expressions. It was further found that miR-146a and miR-155 were both up-regulated in activated Raw264.7 cells in vitro. Conclusions The expression profile of miRNA changes during acute liver failure in mouse model. Inflammation associated-miR-146a and miR-155 are both up-regulated significantly, which indicatcs that they may play an important regulatory role in pathogenesis of acute liver failurc.

Chinese Journal of Dermatology 2014;47(3):157-159
Objective To determine the gene polymorphism and serum concentration of mannose-binding lectin (MBL) in patients with psoriasis, and to analyze the relationship between MBL and psoriasis. Methods Totally,67 patients with psoriasis vulgaris and 69 healthy human controls were enrolled in this study.Venous blood samples were obtained from all the subjects.Genomic DNA was extracted, and PCR-restriction fragment length polymorphism (PCR-RELP) analysis was conducted to determine the polymorphism at codon 54 of the MBL gene.Enzyme-linked immunosorbent assay was performed to measure the serum level of MBL.A chi-square goodness-of-fit test was carried out to evaluate Hardy-Weinberg equilibrium,t test to compare the serum concentration of MBL,and chi-square test to compare the frequency of genotypes and alleles of MBL gene codon 54.Results The patients with psoriasis showed higher frequency of GGC/GAC heterozygote but lower frequency of GGC/GGC homozygote (x2 =10.36,P < 0.05),together with increased frequency of GAC allele but decreased frequency of GGC allele (x2 =8.31,P < 0.05),at codon 54 of the MBL gene compared with the healthy controls. The variant allele GAC at codon 54 of the MBL gene was markedly associated with psoriasis (OR =3.383,95% CI 1.585-7.211,P < 0.05).The serum concentration of MBL was (2.193 7 ± 0.816 3) mg/L in patients with psoriasis, significantly lower than that in the healthy controls ((3.269 5±1.205 8) mg/L,t=6.11,P< 0.05).Conclusion MBL might be associated with the pathogenesis of psoriasis to some degree.

?AlM: To study the correlation between monocular mild myopic juvenile myopia degree and dominant eye.
? METHODS: Totally 158 patients with juvenile monocularly mild myopia in our hospital from December 2012 to December 2013 were retrospectively analyzed, and cylindrical mirror astigmatism was used for spherical equivalent conversion. On the basis of the myopic degree, they were divided into three groups, 30 cases in group A (-0. 25~-0. 75D), 92 cases in B group (-1. 0~-2. 0D), 36 cases in group C (-2. 25 ~ -3. 0D). The card hole method was selected to measure dominant eye for subjectsat nearly 33cm and 5m. After glasses correction of ametropia, the far and near dominant eyes were received measurement again.
?RESULTS:The monocular mild myopia of dominant eye was compared with the non-dominant eye adjustment function, and there was no significant difference ( P>0. 05 ) . The dominant eye, non - dominant eye mean diopter and other correlations of eyes in the three groups were compared, and there was no significant difference (P>0. 05). The dominant eye of three groups at 5m was compared, and there was significant difference (P<0. 05). The dominant eye of three groups at 33cm was compared, and there was significant difference (P<0. 05). Dominant eye at different visual distance was compared, and there was significant difference (P<0. 05). Monocular uncorrected eye was compared with dominant eye mild afte myopia glassesr, and there was significant difference (P< 0. 05).
?CONCLUSlON: The formation of myopic anisometropia is related to the degree of clearly seeing objects, although glasses correction can improve the clarity and visual, but affect the choice of the dominant eye, because the myopic anisometropia appears most early in the dominant eye, therefore optician correction stage, which should be taken into consideration, and avoid the severity of the adolescent myopia.

Abstract? AIM: To investigate changes of anterior chamber parameters after posterior chamber phakic implantable collamer lens ( ICL ) surgery and its correlation with intraocular pressure ( IOP) .?METHODS: This was a retrospective case series study. Seventy four eyes in 43 myopia patients were examined by Allegro Oculyzer anterior segment tomography to obtain the changes of anterior chamber volume ( ACV ) , anterior chamber angle ( ACA) , central anterior chamber depth ( ACD) and vault, meanwhile, to measure the IOP to analyze the correlation with anterior chamber parameters.?RESULTS: Compared with preoperative, ACV, ACA, ACD all decreased apparently ( P <0.01 ) in the postoperative period of 1, 3, and 6mo.ACV, ACA and ACD showed their stability in each period after the surgery. There were changes of vault in 1 and 6mo ( t=27.66, P=0.01).IOP had not altered when compared with the preoperative. This research revealed that IOP had no relation with ACV, ACA, ACD and vault in every phase( all P>0.05).?CONCLUSION:For patients underwent ICL, the anterior chamber parameters all decreased which included ACV, ACA, ACD, and had stabilized since early postoperative period. Correspondingly, IOP was stable and had not correlate with ACV, ACA, ACD and vault, however the long-term observation is still necessary.
