1.Study on association premature ventricular contraction and myocardium injury with microvascular leakage in contrasted aided echocardiography

Objective To examine the relationship between premature ventricular contraction(PVC) and microvascular leakage in the myocardium due to microvascular contrast echocardiography(MCE). Methods Thirty eight rats were randomized into 5 groups: control, ultrasound exposure only, contrast agent only, real time MCE and trigger imaging MCE. Anesthetized rats with tail vein catheter were imaged in a 37℃ water bath at left ventricular parasternal short axis view. Optison was injected at a dosage of 5 ml/kg. Frequency was 1.7 MHz, MI= 1.7 and the depth of image at 10 cm. Results PVC was detected in all ultrasound exposure groups, together with petechial hemorrhages and Evans Blue leakage in the scan band. The triggered imaging showed worse effects than real time imaging. No PVC or microvascular leakage was noticed in controls or sham exposed rats. Conclusions Induction of PVC during contrast added echocardiography is associated with microvascular leakage.
2.The neuroprotective effects of grape seed proanthocyanidin on rat brain injury caused by chronic intermittent hypoxia

Chronic intermitted hypoxia including sleep breathing disorder leads to brain injury. This study explores
the potential therapeutic effects of grape seed proanthocyanidin as a neuroprotective agent. A rat model
of chronic intermittent hypoxia was employed, and the animals were given low or high doses of grape
seed proanthocyanidin. The ultrastructure changes in the brain, the biochemical components, and the
animal behavior were examined. The results showed that with hypoxia exposure, neuronal mitochondria
exhibited injuries at ultrastructural level, with increased malondialdehyde (MDA) content and reduced
superoxide dismutase (SOD) activity. Terminal deoxynucleotidyl transferase dUTP nick end labeling
(TUNEL) staining revealed increased cell apoptosis in hippocampus. In Morris water maze the animals
showed decreased learning abilities, when compared to normal control. The administration of grape
seed proanthocyanidin treatment reversed all these observed changes, and improved the learning
behavior. We concluded that grape seed proanthocyanidin could alleviate the brain injury caused by
hypoxia from sleep breathing disorder.
3.Therapeutic effect of psychological and drug intervention on anxiety and depression in patients after coronary artery bypass graft

Objective:To explore the therapeutic effect of psychological and drug intervention on anxiety and depres‐sion in patients after coronary artery bypass graft (CABG) .Methods :A total of 115 patients performed CABG were randomly divided into combined intervention group (n=60) and routine nursing group (n=55) .During peropera‐tive period ,combined intervention group received psychological intervention ,flupentixol and melitracen tablet and sertraline based on routine nursing .A total of 25 matching normal people were selected from community and regar‐ded as normal control group ,scores of Zung self‐rating anxiety scale (SAS) and self‐rating depression scale (SDS) were evaluated and compared among three groups before and after operation .Results :Before intervention ,there were no significant difference in standard scores of SAS and SDS between combined intervention group and routine nursing group ( P > 0.05) ,but they were all significantly higher than those of normal control group , P < 0.01 all . After operation ,standard scores of SAS and SDS in combined intervention group were significantly lower than be ‐ fore operation ( P < 0.01 both) ,and they were significantly lower than those of routine nursing group [SAS : (41.31 ± 6.13) scores vs .(51.35 ± 8.95) scores ,SDS : (40.20 ± 5.80) scores vs .(51.22 ± 8.78) scores , P < 0.01 both] . Conclusion :Psychological combined drug intervention could significantly relieve anxiety and depression in patients after coronary artery bypass graft ,which is helpful for improving postoperative prognosis .
4.Research about effect of spray drying conditions on hygroscopicity of spray dry powder of gubi compound's water extract and its mechanism.

OBJECTIVETo investigate moisture content and hygroscopicity of spray dry powder of Gubi compound's water extract obtained at different spray drying conditions and laying a foundation for spray drying process of Chinese herbal compound preparation.
METHODIn the paper, on the basis of single-factor experiments, the author choose inlet temperature, liquid density, feed rate, air flow rate as investigated factors.
RESULTThe experimental absorption rate-time curve and scanning electron microscopy results showed that under different spray drying conditions the spray-dried powders have different morphology and different adsorption process.
CONCLUSIONAt different spray-dried conditions, the morphology and water content of the powder is different, these differences lead to differences in the adsorption process, at the appropriate inlet temperature and feed rate with a higher sample density and lower air flow rate, in the experimental system the optimum conditions is inlet temperature of 150 degrees C, feed density of 1.05 g x mL(-1), feed rate of 20 mL x min(-1) air flow rate of 30 m3 x h(-1).

5.Biomechanical features of trabecular metal dental implants

BACKGROUND: Trabecular metal has been reported to provide enough physiologic support for new bone formation, to induce bone ingrowth and osseointegration so as to achieve the biological synosteosis; therefore, it is available for improving the initial and second stability of the dental implant.OBJECTIVE: To explore the stress distributional differences between porous tantalum trabecular metal (PTTM) dental implant and conventional titanium (TI) dental implant in the zone of mandibular premolar using three-dimensional finite element analysis.METHODS: Three-dimensional finite element models emulating PTTM and TI dental implants were established using Mimics16.0 and CATIA in the zone of mandibular premolar with three types of bone (II, III, IV). Then 100 N force was applied on the abutment at a 45° angle to the vertical axis of the implants in software of MSC Patran2010 and Nastran2010, then the representative von Mises stress nodes were calculated and sampled randomly, and finally the von Mises stress distributional differences between two types of implants were analyzed using SPSS 20.0 statistical software.RESULTS AND CONCLUSION: (1) The Von Mises stresses around the two types of implants both focused on the interface of the cortical bone-abutment-implant region, and the stress was relatively less for the corresponding trabecular bone. (2) If the bone quality was similar, the average von Mises stress of PTTM was higher than that of TI dental implants, but there was no significant difference (P > 0.05). As the bone density lowered (II-IV), the average Von Mises stresses were both increased, and the increasing rate of PTTM was higher than that of TI in bone of II-III, and was lower than that of TI in bone of III-IV. In this experiment, there were no significant stress distributional differences between PTTM and TI dental implant. However, with the bone density reduction, the increasing rate of average Von Mises stress for PTTM was lowered than that for TI dental implants in bone of III-IV, which suggests that PTTM dental implant may be more beneficial for the primary and secondary stability in osteoporosis patients undergoing dental implantation.
6.Expression and significance of aquaporin 11 in placenta and fetal membranes of term parturient with oligohydramnios

Objective To explore the expression and localization of aquaporin 11(AQP11) in human term pregnancies with oligihydramnios, and its role in amniotic fluid balance.Methods We studied 55 patients who underwent elective cesarean sections, consisting of 25 patients with isolated oligohydramnios and 30 with normal amniotic fluid volume.Immunohistochemistry and reverse transcription polymerase chain reaction were employed to determine the expression and localization of AQP11 in the amnion, chorion and placenta.Results AQP11 protein was detected in expressed in the full-term pregnant women`s amnion, chorion and placenta.The expression in the amnion was positively correlated with amniotic fluid amount;the expression in the placenta was negatively correlated with amniotic fluid amount.The expression increased in the chorion with different amniotic fluid amount.Conclusion AQP11 plays an important role in regulating amniotic fluid balance.
7.Evolution of hepatitis B virus quasispecies during antiviral therapy in one chronic hepatitis B patient.

OBJECTIVETo investigate the evolution of hepatitis B virus (HBV) quasispecies in one patient during lamivudine (LAM) monotherapy and switching to entecavir (ETV) rescue treatment.
METHODSSerum samples were taken at seven different time points during antiviral therapy (0, 24, 48, 60, 72, 96, 152 weeks, respectively), the HBV DNA polymerase gene was amplified, cloned and sequenced to analyze the amino acid substitutions within HBV DNA polymerase gene and distribution of virus quasispecies. Quantitative detection of the HBV wild strains and total virus was performed by amplification refractory mutation system real-time PCR (ARMS-PCR).
RESULTSThree mutation patterns detected during antiviral therapy in the patient: rtM204V, rtM204V+rtL180M and rtM204I. The HBV quasispecies were found always in dynamic variation. The HBV populations were completely replaced with the LAM-resistant variants when the viral breakthrough was encountered during LAM monotherapy. Interestingly, the wild-type variants presented gradually dominant (79.3%) with the decline of HBV DNA load after switching to ETV rescue administration. ARMS-PCR results showed that the wild-type variants account ed for 68.55% of the HBV populations at baseline and this proportion declined to 0.21% when the viral breakthrough emerged under LAM therapy. The wild-type variants gradually increased from week 24 after switching to ETV rescue therapy and the proportion of HBV wild-type variants in the population fluctuated between 16.01% to 26.93%.
CONCLUSIONSThe distribution of virus quasispecies were always in dynamic variation during sequential therapy with nucleotide analogs in chronic hepatitis B patients. Different patterns of dynamic HBV quasispecies may have different contribution in ETV resistance in LMV refractory patients with ETV administration.

8.Determination of three constituents in Polygoni multiflori Radix Praeparata in plasma of atherosclerosis rats

AIM To establish an HPLC method for determining the contents of three constituents in Polygoni multiflori Radix Praeparata in plasma of atherosclerosis rats.METHODS After the rats were intragastrically administrated with Polygoni multiflori Radix Praeparata CMC-Na solution,the plasma was collected.The HPLC analysis was carried out on a 30 ℃ thermostatic Hypersil C1s column (250 mm ×4.6 mm,5 μm),with the mobile phase comprising of acetonitrile-0.03% phosphoric acid flowing at 0.9 mL/min in a gradient elution manner,and the detection wavelength was set at 280 nm.DAS2.0 software was applied to drawing concentration-time curves and calculating pharmacokinetic parameters.RESULTS Stilbene glucoside,emodin and physcion showed good linear relationships within the ranges of 61.25-6 125 μg/L (r =0.999 8),12.6-3 150 μg/L (r =0.999 3) and 24.1-6 030 μg/L (r =0.999 5),respectively.The method recoveries were 99.5%-105.8% with the RSDs of 1.3%-3.3%,while the extraction recoveries were 87.2%-96.3% with the RSDs of 3.2%-5.9%.The pharmacokinetic behaviors of three constituents all accorded with two-compartment model,but their contents in plasma were much lower than those in medicinal material.CONCLUSION The bioavailabilities of stilbene glucoside,emodin and physcion are relatively low in plasma of atherosclerosis rats,which may be related to constituents' intestinal absorption after intragastric administration with Polygoni multiflori Radix Praeparata.
9.Study of the gene expression pattern in the Kkay mouse with type 2 diabetes by genomic-scale gene expression analysis.

OBJECTIVETo study the genes differentially expressed in the liver of Kkay diabetic and normal mice by genomic-scale gene expression analysis.
METHODScDNA microarray chips containing 8,192 cDNAs were used to explore the gene expression pattern of Kkay mouse liver.
RESULTSOne hundred and fifty-four genes were screened out, including 68 complete cDNAs and expressed sequence tags, and among them 40 genes were up-regulated and 114 genes were down-regulated respectively.
CONCLUSIONMost of the gene expression analysis results were consistent with previous study, and the gene expression pattern of Kkay mouse based on cDNA microarray could be used for high-throughout screening out the genes associated with type 2 diabetes.
