3.Interference effect of picrosideⅡon cerebral ischemia reperfusion injury in rats

Objective To investigate the neuroprotective effects of picrodideⅡ on cerebral ischemic reperfusion injury in rats. Methods Intraluminal thread methods were applied to establish the left middle cerebral artery occlusion reperfusion models (MCAO/R) in rats. PicrodideⅡ (10mg/kg) and salvianic acid A sodium (10mg/kg) were injected from tail vein for treatment. The neurological behavioral function was evaluated with Bederson's test. The cerebral infarction volume was observed with tetrazolium chloride (TTC) staining. The structure of cells was observed with histopathology. The apoptosis positive cells were counted by terminal deoxynucleotidyl transferase midiated dUTP nick end labeling (TUNEL). Results The neurological behavioral malfunction appeared in all rats with MCAO/R. The infarction focus showed in the ischemic hemisphere following cerebral ischemia reperfusion injury. In the picrodideⅡ and salvianic acid A sodium treatment groups, the number of apoptosis positive cells decreased and the cerebral infarction volume reduced, while the neurological behavioral function was significantly improved than those in the model control group (P<0.05). The cerebral infarction volume in the picrodideⅡ group was smaller than that in the salvianic acid A sodium group (P<0.05).Conclusion PicrodideⅡ might reduce cerebral infarction volume and improve the neurological behavioral function through inhibiting the neuronal apoptosis induced by ischemia reperfusion injury.

Objective To observe the influence of neuregulin-1?(NRG-1?) on neuronal apoptosis and the expressions of signal transducer and activator of transcription(STAT3) and glial fiberillary acidic protein(GFAP) following cerebral ischemia/reperfusion in rats.Methods The animal models of middle cerebral artery occlusion/reperfusion(MCAO/R) was established by the intralumianl filament method from left external-internal carotid artery in adult healthy male Wistar rats.The rat models were treated by injecting 1.5% NRG-1? 5?l from internal carotid artery.The neuronal apoptosis was detected by DendEnd fluorometric TUNEL assay,and the expressions of STAT3 and GFAP were determined by immunohistochemical and immumofluorescent assays.Results The cerebral ischemia/reperfusion injury could induce neuronal apoptosis and the expressions of STAT3 and GFAP in brain tissue.In control group,the number of neuronal apoptosis increased gradually and the STAT3 and GFAP were expressed highly along ischemia times in the cortex,striatum and hippocampus areas.After treatment with NRG-1?,the number of neuronal apoptosis reduced and the expression level of STAT3 and GFAP increased when compared to those in the control group at different ischemia times and corresponding areas(P
6.Paracrine effect of endothelial colony-forming cells on biological functions of human umbilical vein endothelial cells

BACKGROUND: Endothelial colony-forming cells (ECFCs) transplantation exerts beneficial impact on angiogenesis of impaired tissues caused by ischemia. However, whether this impact is related to the paracrine effect of ECFCs needs to be studied further.OBJECTIVE: To detect the cytokine secretion profile of human umbilical cord blood derived-ECFCs conditioned medium (ECFCs-CM) and explore the effect of ECFCs-CM on the proliferation, migration and tube formation ability of human umbilical vein endothelial cells (HUVECs).METHODS: Human cord blood derived-ECFCs were isolated, cultured in vitro, and then identified based on the previous studies. The cytokines in serum free ECFCs-CM were detected using a cytokines antibody array. HUVECs were cultured with ECFCs-CM, serum free EBM-2 as control. The proliferation, migration and tube formation abilities of HUVECs were examined by cell counting kit-8, scratch test and Matrigel assay, respectively.RESULTS AND CONCLUSION: The cultured cells demonstrated typical characteristics of ECFCs, which showed cobblestone appearance and were positive for CD34, KDR and CD144, but not CD45 or CD133, uptook Dil-acLDL and bond FITC-UEA-I, and tube-like structures were formed on Matrigel. The cytokine antibody array showed that ECFCs-CM significantly upregulated the expressions of 30 kinds of angiogenic factors. Compared with the control group,the HUVECs cultured with ECFCs-CM showed significantly improved proliferation ability at 24, 48 and 72 hours (P <0.01). The migration rate of HUVECs in the experimental group was significantly higher than that in the control group at 12 and 24 hours (P < 0.01). There were more tubular structures in the experimental group than those in the control group (P < 0.05). These results indicate that ECFCs can promote the bioactivity of mature endothelial cells through paracrine action.

Despite different opinions on its definition and classification in the past,a consensus has gradually been reached regarding the na-ming,classification,and clinical diagnosis of liver failure.The classification of liver failure is described,and the definition and natural his-tory of severe exacerbation of hepatitis B are summarized.Antiviral treatment and artificial liver support in the early stage are beneficial for clinical outcomes and prognosis.

A moderately thermoacidophilic iron-oxidizing bacterium,designated as strain MLY,was isolated from a coal spoil heap in China.The optimum of temperature for growth is 50℃~54℃.The optimum of pH is 1.2~1.4.The strain MLY is facultative autotroph and grows heterotrophically on yeast extract.It is able to oxidize ferrous iron(Fe 2+ ),pyrite(FeS 2),and elemental sulfur(S 0) autotrophically and mixotrophically in the presence of yeast extract.Autotrophic oxidation of elemental sulfur is relative weak.The comparison of ferrous iron and pyrite oxidation between strain MLY and A10 Thiobacillus ferrooxidans,strain indicated that MLY is one time faster than A10.
9.Effect of Physiotherapy on Knee Osteoarthritis

Objective To observe the effect of physiotherapy on knee osteoarthritis. Methods 40 knee osteoarthritis patients were dividedinto treatment group (n=20) and control group (n=20). The control group accepted conventional therapy, acupuncture and moxibustion,fumigation, massage, etc. The treatment group accepted physiotherapy in addition, including strength training and joint activity training.They were assessed with Lysholm Knee Scoring Scale (LKSS) and Visual Analogous Scale (VAS). Results The scores of LKSS and VASimproved in both groups after treatment, and improved more in the treatment group than in the control group (P<0.01). Conclusion Physiotherapycan further improve the stability of knee and release the pain in knee osteoarthritis patients.
10.Cloning of human preferentially expressed antigen of melanoma gene and its expression in E.coli

Objective: To clone and express the preferentially expressed antigen of melanoma (PRAME) gene in E.coli. Methods: The cDNA encoding human PRAME gene was extracted from human AML cell line HL-60 and amplified by RT-PCR, then the PRAME gene was inserted into plasmid PGEM-T-easy. After sequenced, it was cloned into the prokaryotic expression vector pGEX-4T-2 to construct a clone with high level expression and the recombinant plasmid was cloned in E.coli. Results:The protein product reached the highest level at 5 h after IPTG induction. Conclusion: Since PRAME is only expressed at low levels in a few normal tissues and encodes an antigen recognized by autologous cytotoxic T lymphocytes, while expressed at high levels in patients with leukemia, it might be a new targeting candidate for tumor immunotherapy.
