1.Effects of Zafirlukast on Emotions and Th1/Th2 Cytokines in Asthmatic Children

Objective To investigate the effects of Zafirlukast on emotions and Th1/Th2 cytokines in asthmatic children.Methods One hundred and eight asthmatic children were randomly allocated to 3 groups.The children in group A inhaled Pulmicort aerosol 200 ?g twice every day.On the basis of inhaled Pulmicort aerosol,the children in group B took orally Zafirlukast orally 10 mg once every day.The children in group C only took orally Zafirlukast 10 mg once every day.The course of treatment of all children were 4 weeks.Anxiety,depression status and the levels of IFN-? and IL-4 in blood serum were respectively detected before and after treatment.Results Scores of anxiety and depression after treatment in 3 groups were significantly lower than those before treatment(Pa0.05).Conclusions Zafirlukast or Glucocorticoid inhalation has the same effect on improving emotional status and accommodating disequilibrium of Th1/Th2 for asthmatic children,but effect of Zafirlukast combined with Glucocorticoid inhalation are better.
2.Curative effect of simple volar or dorsal plating in the treatment of intra-articular distal radius fracture

Objective To compare the clinical effect of simple volar or dorsal plate fixation of intra-articular distal radius fracture.Methods This retrospective study included 42 patients with closed intra-articular distal radius fracture treated surgically using the dorsal or volar plate.Out of the 15 patients in dorsal plating group 5 were males and 10 females at age of (55 ± 7)years (range, 48-62 years), 13 were injured from falls and 2 traffic accidents, 10 were classified as AO type C3 and 5 AO type C2.Out of the 27 patients in volar plating group 8 were males and 19 females at age of (56 ± 6)years (range, 50-62 years), 24 were injured from falls and 3 traffic accidents, 17 were classified as AO type C3 and 10 AO type C2.Between-group differences were compared with respect to wrist range of motion, postoperative radiographic parameters, postoperative complications, disabilities of the ann, shoulder and hand (DASH) score and Gartland-Werley score.Results All the patients were followed up for 11-25 months.There were no significant differences in the wrist range of motion and radiographic parameters between the two groups (P > 0.05).Volar plating group resulted in a significantly better Gartland-Werley score compared to dorsal plating group [1 vs 4 points, P < 0.05], but no significant difference was noted in DASH score (P > 0.05).Four patients (27%) in dorsal plating group developed tendon adhesions and tenolysis was in demand, but one patients (4%) in volar plating group was complicated by median nerve symptoms (P < 0.05).Conclusions Although the DASH score of the two methods was similar, volar plating yields better results in Gartland-Werley score and complication incidence.Thus the volar plating is recommended for intra-articular distal radius fracture.
3.Interaction of Mina and IL-4 gene and their relevance to asthma children

Objective:To study the relevance of Mina rs4857304 (G/T) gene locus, IL-4 gene locus rs2243248 (T/G), rs2070874 ( T/C ) single nucleotide polymorphism and allergic asthma pathogenesis of Chinese children , and further analysis of the in-teraction among three loci.Methods:Based on the case-control study, we have analyzed the different distributions , frequencies, inter-action of alleles and genotypes between asthma patients and patients in control group .Results: The Mina rs4857304 sites of patients were compared, and there was statistically significant difference between two groups ( P=0.001, OR =1.773,95%CI =1.256 -2.503);the rs4857304T allelic frequency of patients with asthma was higher than that of the patients in control group , but there was no statistically significant difference about the distribution of different locus genotypes of patients between two groups (P>0.05);MDR interaction analysis have showed that Mina gene loci , rs4857304 and IL-4loci rs2243248, consisting of 2 sites optimal model;Mina loci rs4857304, IL-4loci rs2070874 and rs2243248 that consisting of 3 sites optimal model.There was statistically significant difference be-tween two groups(P<0.05).Conclusion:Allergic asthma would be associated with Mina loci rs4857304, while there might be sig-nificant interactive effects among Mina rs4857304, IL-4loci rs2243248 and rs2070804.

5.Optimal induction conditions for adipose-derived stem cells differentiating into lymphatic endothelial cells

BACKGROUND:Our previous studies have shown that adipose-derived stem cels under vascular endothelial growth factor C (VEGF-C) can be induced to differentiate into lymphatic endothelial cels that are confirmed by lymphatic vascular endothelial hyaluronan receptor-1 staining. However, its optimal induction program is not clear.
OBJECTIVE:To investigate the best condition for the differentiation of adipose-derived stem cels into lymphatic endothelial cels under induction of VEGF-C156s.
METHODS: Adipose tissues from healthy adults were colected to isolate adipose-derived mesenchymal stem cels using trypsin digestion method. Flow cytometry was employed to detect cel surface markers, andin vitro differentiation capacity was identified by adipogenic and osteogenic induction. Passage 3 cels at good growth state were selected and divided into six groups: cels in control group were cultured in low-glucose DMEM, and those in the rest five groups were treated with 25, 50, 100, 200, 300 μg/L VEGF-C156s, respectively.
RESULTS AND CONCLUSION:Adipose-derived stem cels were successfuly obtained by trypsin digestion and purification, and then differentiated into lymphatic endothelial cels under the induction of VEGF-C156s, basic fibroblast growth factor and other growth factors. No cels were positive for lymphatic vascular endothelial hyaluronan receptor-1 in the control group. After 8 days of induction, few cels were positive in the 25 μg/L VEGF-C156s group; a great amount of positive cels were visible in the 50 and 100 μg/L VEGF-C156s groups; 200 and 300 μg/L VEGF-C156s resulted in a large number of deaths in the cels. These findings indicate that it is optimal for adipose-derived stem cels to differentiate into lymphatic endothelial cels under 8-day induction of 50 μg/L VEGF-C156s.
6.One-staged release and reduction by posterior approach to treated basilar invagination with irreducible atlantoaxial dislocation

Objective To evaluate the safety and effectiveness of one stage surgery of release and bone reduction by posterior approach to treat basilar invagination (BI) with irreducible atlantoaxial dislocation (IAAD),and to explore the indications and crucial techniques of posterior approach.Methods All of 17 Consecutive patients (8 males and 9 females) with BI and IAAD who underwent release and reduction by posterior approach from July 2000 to June 2015 were enrolled in the present study,the mean age was 35.2±13.8 years with a range of 12-56 years.The clinical symptoms and signs was recorded,and preoperative imaging examination,including anteroposterior,lateral,dynamic films,MRI and CT of cervical spine,were performed to identify the series.There were 14 cases with atlanto-occipital fusion,7 cases with C2,3 fusion,6 cases with Chiari malformation,6 cases with Syringomyelia,and 8 cases with myelomalacia.The clinic symptoms include occiput/neck pain in 15 cases,cervical movement limitation in 13 cases,short neck in 9 cases,torticollis in 12 cases,Paresthesia in 14 cases,weakness in 13 cases,tendon reflexes hyperfunction in 16 cases and ataxia in 13 cases.The postoperative X-rays,MRI or CT were used to observed the results of decompression,fixation and fusion.Neurological function was assessed by JOA scale and Ranawat's score before,after surgery and at final follow-up.Pre-and post-operative Chamberlain (CL),Wackenheim (WL),McGae (ML),atlantodental interval (ADI) and cervico-medullary angle (CMA) were analyzed by student t-test.Results The average operation time was 145 mins (90-210 mins) and blood loss was 175 ml (150-350 ml).The average follow-up was 44.47 months (9-94 months).JOA score was increased from 8.06 preoperatively to 15.20 postoperatively,the improvement rate was 77.2%.Preoperative Ranawat's score was Ⅱ in 1 case,Ⅲla in 12 cases,ⅢB in4 cases.Postoperative score was Ⅰ in 13 cases,Ⅱ in 4 cases.The preoperative CL,WL,ML,ADI and CMA were (12.52±5.17) mm,(6.59±3.04) mm,(6.96±4.32) mm,(9.88± 1.93) mm,115.35°± 12.40°,respectively.and the postoperative CL,WL,ML,ADI and CMA were (2.0±3.67) mm,(-3.06±1.85) mm,(-1.76±2.88) mm,(1.17± 1.18) mm,136.76°±11.44°,respectively.The perioperative complications were discovered in 2 cases,including 1 case of infection and1 case of cerebrospinal fluid(CSF) leakage.Conclusion Primary surgery of nerve release and bone reduction by posterior approach may be safe and efficient for the treatment of BI and IAAD.Preoperative evaluation,proper surgical indications and advanced surgical techniques are important for treatment results.
7.Effects of recombinant human TNFR: Fc fusion protein on rat acute liver injury induced by lipopolysaccharide

Objective To investigate the protective effects and the undedying mechanism of recombinant human tumor necrosis factor receptor: Fc fusion protein (Yisaipu, rhu TNFR: Fc) on the lipopolysaccharide (LPS) induced acute liver injury of rats. Method Totally48 SD rats were randondy divided into four groups , in-cluding control gronp (n = 12), Yisaipu group(n = 12), LPS gronp(n = 12) and Yisaipu + IPS group(n = 12). The models of acute liver injury were produced by injection of LPS intravenously. Being fasted for 12 h, the rats were anaesthetized (60 mg/kg pentobarbital sodium, i.p.) and cannulated into carotid arteries. The cannula was connected with the multi-channel creature signal analysis system. The rata in control group and LPS group were injected with normal saline or LPS in dose of 5 mg/kg through rats' sublingual vein respectively. While the rats in Yisaipu group and Yisaipu + LPS group was pretreated with Yisaipu in dose of 0.4 mg/kg subcutaneously 24 h be-fore normal saline or LPS infusion. Six rats of each goup were randomly selected and mean arterial pressure (MAP) were monitored for 6 h via multi-channel creature signal analysis system, and rats' survival rate was calcu-lated. The rats whose MAP less than 10 mmHg were considered to die and the alive rats during period of observa-tion sacrificed by exsanguination. The liver tissue at the same site was removed, fixing in 10% formalin or stored at -80 ℃. To detect serum TNF-α, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) level, 0.2 mL blood samples were collected from the carotid artery 2 and 3 h after the injection of saline or LPS. The serum was collected from centrifuged blood samples and stored at -80 ℃. Enzyme-linked immunosorbent assay (ELISA) and flow cytometry were used to assess serum TNF-α level and bioactivity respectively. We also measured the serum ALT and AST levels, the myeloperoxiase (MPO) and superexide dismutase (SOD) activity, and malon-dialdehyde (MDA) content in liver tissue The pathology of hepatic tissue was evaluated by HE staining. Statistical-ly,the data of TNF-α level and bioactivity, ALT and AST release, and MDA content were analyzed by ANOVA, and rat survival rate were analyzed by Chi-square Tests. Results The rats in control group and Yisaipu group were all survived. Rat survival rate was significantly higher in Yisaipu + LPS group (67%) than in LPS group (17%) (P < 0.05). Serum TNF-α bioactivity was significantly lower in Yisaipu + LPS group than in LPS group [(7.3±2.8)% vs.(51.3±6.4)%, P <0.05]. Compared with IPS group, Yisaipu pretreatment decreased MDA content [(1.40±0.10)vs. (2.81±0.11) nmol/mgprot, P <0.05]and MPO acticity [(0.38±0.04) vs. (0.54±0.02) U/g, P <0.05]in hepatic tissue, while SOD activity [(188.4±20.2) vs. (142.5 ± 18.3) U/mgprot, P <0.05]was increased. The serum AST level, ALT level and the pathology in the liver were also ameliorated correspondingly. Conclusions These data suggest that Yisaipu could protect rats from LPsinduced a-cute liver injury by inhibiting TNF-α bioactivity and by enhancing anti-oxidation.
8.Protection effects of soluble TNFR: Fc fusion protein on acute kidney injury induced by lipopolysaccharide in rats

Objective To investigate the protective effects of recombinant human tumor necrosis factor receptor:Fc fusion protein (rhuTNFR:Ice)on the acute renal injury induced by lipepelysaccharide(LPS)in rats.Methods Models ofacute renal injury in rats were constructed by intravenous injection of LPS 10 m/kg.Forty-eight SD rats weighing 180~240g were randomly divided into 4 groups with 12 animals each group,including control group,rhuTNFR:Fc group,LPS groupand rhuTNFR:Fc+LPS group.Mean arterial pressure(MAP)wKs continuously monitored for 6 h.The levels of blood tLrea nitrogen(BUN)。Creatinine(Cr),TNF-αas well as TNF-α bioactivity were assessed.The myeloperoxinse(MPO)and superoxide dismutase(SOD)activity,content ofmalondialdehyde(MDA)were also measured.Pathologic changes of lung tissue in each group were observed by HE staining.Results Compared with LPS group,the status of hypotension and pathological manifestation in kidneys were ameliorated,and MPO activity significantly decreased in rhuTNFR:Fc+LPS group(P<0.05).Conclusion These data suggest that rhuTNFR:Fc can ablate the rise in serum TNF-α bioactivity that occurs in response to LPS,and rhuTNFR:Fc could in part protect rats from the acute renal injury induced by LPS.

10.Optimization of Fermentation Medium for Honggumycin Using Response Surface Methodology

Under the shaking-flask culture, fermentation medium of honggumycin produced by Streptomyces 702 were studied.The experiment was used response surface methodology to optimize the shaking-flask fermentation medium.Firstly, we applied full factorial design to screen important factors soybean meal and industrial peptone which affected hongmycin produced by Streptomyces 702.Furthermore, we designed experiment to obtain the steepest ascent path and optimal level by the central composite design.The optimum medium consisted of (g/L): maize starch 20, maize meal 20, glucose 20, soybean meal 23, industrial peptone 9, KNO_ 3 2.5, (NH_ 4 )_ 2 SO_ 4 2.5 KH_ 2 PO_ 4 0.3, NaCl 3, CaCO_ 3 6, bean oil 5mL/L.Under the optimal medium, the yield of honggumycin was up to 1500 g/mL, which was increased by 308% than the original medium.
