2.Histopathological observation of acquired immunity in skin of hamsters elicited by Necator americanus third-stage infective larvae

Objective To obtain acquired immunity evidence in hamsters elicited by third stage hookworm larvae of Necator americanas(NaL3).morphology changes of NaL3 and inflammatory responses in the skin and undedying subcutaneous tissue and muscles of hamsters were observed.Methods Hamsters were immunized subcutaneously with one dose of 150 NaL3 at 2 weeks earlier,and then challenged pereutaneously with 900 NaL3.Skins were excised from post-challenge hamsters at 6,24,72 hours and 1,2 weeks,and then examined under light microscopy.Non-immunized hamsters served as negative controls.Results In non-immunized hamsters the number of NaL3 were 15,33,11.0 and 0 at 6,24,72 hours and 1,2 weeks post-infection.No damaged or dead NaL3 section was observed.All NaL3 exhibited no structural damage and infihrating inflammatory cells were absent from the sunDunding tissues.There were no cutaneous changes.In contrast.the total number of Nak sections in the skin of immunized hamsters were 25,53,15,5 and 4 at 6,24,72 hours and 1,2 weeks post-challenge.Among these NaL3 sections,damaged and dead section number were 0,24,6,0,0 and 0,0,7,5,4.At 24 hours post-challenge the Nak exhibited cutieular swelling and damage.By 72 hours post-challenge pyknosis of the somatic cells nuclei and sparseness or loss of definition in the internal structures of NaL3 were seen.One or two weeks after chanenge,the NaL3 showed severe damage or even dead with remnants.Inflammatory responses including macrophages,epithelioid cells and eosinophils infiltrating and granulomata forming were mainly seen around the NaL3 sections in the skin of immunized hamsters.Conclusions Hamsters initially immunized with NaL3 exhibited obvious acquired immunity protection against percutaneously challenged infection as evidenced by vigorous inflammatory responses in the skin and underlying subcutaneous tissue and muscle.
3.Cost-benefit analysis on the strategy of social health insurance regarding vaccination against influenza in Xi'an city.

OBJECTIVETo assess the economic implications of an annual vaccination strategy against influenza among people who were on a social-health program.
METHODSA retrospective cohort study was conducted. 1900 persons who had received the influenza vaccine were served as vaccine group, while 1049 persons who did not receive the vaccine were served as controls. Cluster random sampling method was used. Both of these two groups came from Donfang Company in which there were 12,109 employers in total and all of them joined the social health insurance program. The survey was carried out when the influenza vaccine was given one year ago.
RESULTSThe rates of vaccine group and control group for respiratory system diseases and cardiovascular diseases who were hospitalized, were 0.51%, 2.47% and 1.64%, 5.62% which showed 68.90% and 56.05% decrease, when compared with the control group. The crude inpatient rate among vaccinees and control group after receiving the vaccination for three and four month were 0.62%, 0.80% and 0.28%, 1.00% respectively. The inpatient rate of oldest-age group decreased by 53.59%, compared with control group. The cost-benefit ratio generated by the use of influenza vaccine in reducing the hospitalization rate was 6.48:1 for Social Health Insurants in Xi'an city.
CONCLUSIONThe Strategy to vaccinate the social-health-insured residents on influenza in Xi'an city had gained better economic benefits in reducing the hospitalization rate of respiratory system diseases and cardiovascular diseases for mild and old-aged persons.

4.Development of intelligent wound instillation-negative pressure therapeutic instrument

Objective To develop an intelligent wound therapeutic instrument based on active instillation and negative pressure suction.Methods SCM coordinated peristaltic pumps, solenoid valves and pressure receptor to infuse perfusate into the wound and retained it for a period of time. The perfusate then was eliminated by negative pressure and the wound was kept in the state of negative pressure. The instrument could give out alarm in case of bad tightness, over-high pressure and blocked negative-pressure pipeline.Results Peristaltic pump and central negative pressure were involved in to complete wound perfusion and irrigation, with perfusate controlled quantitatively and negative pressure kept reliable. The pressures in the wound and pipeline could be monitored at real time by the pressure receptor, and timely alarm was implemented when unexpected conditions occurred.Conclusion The instrument behaves well in automation, intelligence and safety, and thus is worth popularizing clinically.
5.AFLP analysis on genetic diversity of Haloxylon ammodendron in China.

To determine the genetic diversity of Haloxylon ammodendron collected from 14 sites in 5 provinces, 103 H. ammodendron samples of 12 wild populations and 2 cultivated which collected from 14 sites in 5 provinces were analyzed by amplified fragment length polymorphism (AFLP) DNA markers. PopGen32 and NTSYSpc2.1 was applied to evaluate genetic diversity of H. ammodendron populations. The average percentage of polymorphic loci (PPL) of total H. ammodendron populations was 94.13%, the average Nei's gene diversity index (H(e)) from 14 populations was 0.308 0, and the Shannon's genetic diversity index (I) was 0.467 6. The results indicated that the genetic diversity of H. ammodendron populations was high. Genetic differentiation index (G(st)) was 0.313 8, and the gene flow (N(m)) was 1.093 5 at the population level. The level of gene flow of H. ammodendron showed it possessed the feature of wind-pollinated outcrossing plants. AMOVA analysis indicated that genetic variation of H. ammodendron was much higher within groups (89.34%) than that among groups (10.66%), moreover genetic variation within groups mainly occurred among populations in different producing areas (84.80%). Cluster analysis (UPGMA) was applied to generate dendrogram based on Nei's genetic distances of 14 populations. Samples from Xinjiang and Qinghai were clustered respectively as a clade for their distant genetic relationship, while Samples from Gansu, Inner Mongolia and Ningxia were clustered together for their close genetic relationship. Genetic diversity of H. ammodendron populations is high in China, and genetic differentiation among regions is small, thus abundance within this specie is high at this stage. Therefore, wild nursery and artificial cultivating in different areas are effective measures for the conservation and sustainable utilization of H. ammodendron resources.

7.A new erythrodiol triterpene fatty ester from Scorzonera mongolica.

Two erythrodiol triterpene fatty esters, 3beta-dodecanoyl erythrodiol (1) and 3beta-tetradecanoyl erythrodiol (2), were isolated from Scorzonera mongolica. Their structures were elucidated on the basis of IR, MS and extensive 2D NMR spectroscopic analysis. Compound 1 was identified to be a new compound and 2 was confirmed to be a new natural compound. Their antitumor effects in vitro were evaluated with MTT and SRB assays, but compounds 1 and 2 only showed moderate cytotoxicities on A-549 cell line.

8.Therapeutic effect of PCI combined tirofiban on aged patients with ACS complicated diabetes mellitus and its safety/

Objective :To explore therapeutic effect of percutaneous coronary intervention (PCI ) combined hydro‐chloride tirofiban on aged patients with acute coronary syndrome (ACS) complicated diabetes mellitus (DM) and its safety .Methods :A total of 218 ACS + DM patients were selected .According to randomization method ,they were divided into routine treatment group (n=107 ,received standard PCI) and tirofiban group (n=111 ,received tirofi‐ban injection in coronary artery based on routine treatment group ).Operation indexes ,incidence rates of hemor‐rhage during hospitalization and major adverse cardiovascular events (MACE) within six months after PCI were ob‐served and compared between two groups .Results : There was no significant difference in door‐to‐balloon time , number and length of implanted stents between two groups , P>0.05 all.Compared with routine treatment group , there were significant rise in percentages of TIMI grade 3 blood flow (75. 70% vs.91.89%) ,MBG grade 2 ~3 (69.16% vs.85.56%) and ST segment regression >50% within 90min after PCI (77. 57% vs.92.79%) ,and sig‐nificant reduction in corrected TIMI frame count (CTFC)[(33. 05 ± 8.37) frames vs.(26.54 ± 5.47) frames]in tiro‐fiban group , P<0.01 all.There was no significant difference in incidence rate of hemorrhage events during hospi‐talization between two groups , P=0.375. Incidence rate of MACE within six months in tirofiban group was signifi‐cantly lower than that of routine treatment group ,P=0.001 .Conclusion :Intracoronary tirofiban injection based on routine PCI can significantly improve postoperative myocardial perfusion level ,reduce incidence rate of short‐term MACE after PCI without increasing incidence rate of hemorrhage in aged ACS patients .Its safety is good .
9.The study on cytokines of bronchoalveolar lavage fluid in children with mycoplasma pneumoniae pneumonia

Objective To study the cytokine levels of TNF-α, IFN-γ, IL-2, IL-4, IL-6 and IL-10 in bronchoalveolar lavage fluid (BALF) in children with mycoplasma pneumoniae pneumonia (MPP). Methods Clinical data of 90 patients with MPP (MPP group), who were admitted to Tianjin Children's Hospital from January to December 2017, were collected in this study. The 90 cases were divided into group A1 (<7 days), group A2 (7-14 d) and group A3 (15-30 d) according to the course of disease on admission, with 30 cases in each group. Children who were without MPP and needed bronchoscopy examination or treatment at the same time were used as the control group (B group). The expression levels of cytokines (TNF-α, IFN-γ, IL-2, IL-4, IL-6 and IL-10) were detected by enzyme-linked immunoassay (ELISA). Results The serum and BALF levels of TNF-α, IFN-γ, IL-2, IL-4, IL-6 and IL-10 increased significantly in A group compared with those in B group (P<0.05). The highest levels of cytokines (TNF-α, IFN-γ and IL-2) were found in Group A1. The highest levels of cytokines (IL-4 and IL-6) were found in Group A2. Conclusion The levels of TNF-α, IFN-γ and IL-2 are increased significantly in the disease course of < 7 days. In the disease course of 7 to 14 days, the IL-4 and IL-6 are elevated significantly. Our data suggest that these cytokines may be involved in the pathogenesis of MPP.
10.Expression of DOG-1 in gastrointestinal stromal tumors and its significance.

OBJECTIVETo identify the expression of DOG-1 in gastrointestinal stromal tumors (GIST) and to explore its potential association with clinicopathological features of GIST.
METHODSTwo tissue microarrays (TMA) were used for the study. Each TMA contained 80 tissue samples of GIST from 80 different patients, with each tumor represented by one core, and paraffin-embedded sections of 40 samples from normal gastrointestinal tissue were used as control. Immunohistochemistry staining (SABC method) was performed on TMA and paraffin-embedded sections to detected the expression of c-Kit (CD117) and DOG-1.
RESULTSImmunohistochemistry showed that in 80 GIST patients, 76 cases (95.0%) were DOG-1 positive and 67 cases (83.8%) were CD117 positive. The positive rate of DOG-1 was higher than that of CD117 (P<0.05). In 13 GIST samples of negative CD117, the positive rate of DOG-1 was 100%. Expressions of both DOG-1 and CD117 were negative in all the 40 samples of normal gastrointestinal tissue. The positive expression of DOG-1 and CD117 was not significantly different in spindle cell type (96.0% vs. 96.0%, P>0.05) and in mixed cell type (91.7% vs. 75.0%, P>0.05). While in the epithelioid cell type, the DOG-1 expression was higher than CD117 expression (94.1% vs. 52.9%, P<0.05). The positive expression of DOG-1 and CD117 was not associated with age, gender, location and risk stratification of the tumors (all P>0.05).
CONCLUSIONSDOG-1 expression is up-regulated in gastrointestinal stromal tumors, especially in epithelioid cell GIST, and may be used as a new marker in the diagnosis of GIST.
