3.Effect of allergen on function of peripheral blood CD4+T cells in patients with asthma

Objective To investigate the relationship among the proliferation of CD4+T cells, the intracellular levels of interleukin-10 (IL-10) and transforming growth factor-?1(TGF-?1), and allergic bronchial asthma. Methods Dermatophagoides farinae antigen were prepared as allergen. Twenty-five patients with asthma and 15 healthy individuals were enrolled and divided into blank control group, allergen group and self control group, respectively, after venous blood sample collection. The proliferation of CD4+T cells and the distributions of CD4+/IL-10+, CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 were measuredby flow cytometry (FCM). Results The distributions of CD4+/IL-10+, CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 could hardly be detected in the peripheral blood samples of the blank controls of the patients with asthma and healthy ones. In the allergen group of the healthy individuals, the peripheral blood CD4+T cells were significantly proliferated, and the proportions of CD4+T cells andCD4+/IL-10+ cells were much higher than the self control group, while there was no significantly increase in the proportions of CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 subgroups. In the allergen group of those with asthma, the proportions of peripheral blood CD4+, CD4+/IL-10+, CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 cells were not found significantly increased compared with those self controls. 5.Identification of differentially expressed genes in endometrium during the window of implantation using suppression substractive hybridization

Objective To identify genes differentially expressed in the window of implantation and explore the molecular basis of the development of endometrial receptivity.Methods A subtracted cDNA library of the window of implantation was constructed by suppression subtractive hybridization(SSH) method.The screened clones of the subtracted library were sequenced and GenBank homology search was performed.The differential expression of ribosomal protein(RP)L7,RPL7 pseudogene(RPL7p),RPL19 and tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein zeta polypeptide(YWHAZ) were further confirmed by RT-PCR.Results After sequencing and GenBank homology search of 50 clones, 35 differentially expressed genes were detected in the window of implantation,of which 23 were known genes,and 12 were unknown genes.Some of the known genes have been proved to be associated with implantation,while others were firstly screened out by us.The results of RT-PCR confirmed that RPL7, RPL7p,RPL19 and YWHAZ were highly expressed in the window of implantation,0.75?0.21,1.72? 0.30,1.23?0.31,and 1.28?0.08,respectively.Conclusions SSH is a useful technique to detect differential expression genes and an effective method to clone novel genes.It provides a new method to investigate the molecular basis of the development of endometrial receptivity.

Objective:To highly express HIV-1p17 in E.coli and purify the protein.Methods:①Recombinant plasmid was constructed by inserting HIV-1p17 gene amplified by PCR into plasmid vector,pET28c;②The recombinant plasmid was expressed in BL21,BL21(DE3),BL21(DE3) plysS and HMS174(DE3) of E.coli separately;③The target protein were purified with Ni-NTA resin;④The purified protein was detected by western blot and ELISA.Results:The expression of the P17 protein in BL21(DE3) represented up to 32% of total protein in E.coli,which was the most amounts compared with other kinds of E.coli.The purity of the purified protein reached 95%.The purified protein was recognized by HIV-1P17McAb as well as by HIV-1 positive serum.Conclusion:The recombinant plasmid is highly expressed in BL21(DE3) of E.coli that can be proceeded to the immunocompetence and the bioactivity research.The method of Ni-NTA resin is simple with low protein losing and high purity.And the purified p17 can be employed in early detection of HIV-1 infection and prediction of the clinical progression.

9.HRCT and MRI image of bilateral large vestibular aqueduct syndrome.

OBJECTIVE:
To explore. HRCT and MRI three-dimensional fast imaging employing steady state ac-quisition(3D-FIESTA) imaging features and clinical characteristics of bilateral large vestibular aqueduct syndrome(LVAS).
METHOD:
The imaging and clinical features of 14 cases of bilateral LVAS identified over a 5-year periodwere retrospectively analyzed. All patients underwent HRCT and MRI 3D-FIESTA scanning of head and neck;MRI three dimensional reconstructions of internal acoustical meatus were also completed at the same time.
RESULT:
Audiogram showed mild to moderate hearing loss and was progressive. The cut-off values for the coronal midpointand operculum planes on the HRCT scan to diagnose an EVA were 1. 5 mm and 4. 3 mm respectively; the averagevalue was 2. 4 mm. VA expansion degree were not linked to the degree of hearing loss. MRI showed VA andlymph sac abnormalities. Concomitant image finding was cochlear hypoplasia.
CONCLUSION
HRCT and MRI 3D-FI-ESTA are important examinations for accurate diagnosis of LVAS. HRCT can acquire the specific size of reamedVA. MRI and 3D reconstructions of internal acoustical meatus can noninasive show more intuitive display ofLVAS and other inner ear malformations than HRCT.

10.Influence of different dose of atorvastatin on blood pressure ,blood lipids and vascular endothelial func‐tion in hypertensive patients with hyperlipidemia

Objective :To explore influence of different dose of atorvastatin on blood pressure ,blood lipids and vascular en‐dothelial function in hypertensive patients with hyperlipidemia .Methods :A total of 100 hypertensive patients with hyper‐lipidemia treated in our department of cardiology from Jan to Oct 2017 were randomly and equally divided into atorvastatin low dose group (atorvastatin 10mg/d) and high dose group (atorvastatin 20mg/d) ,both groups simultaneously received rou‐tine treatment for eight weeks .Another 50 healthy volunteers undergoing physical examination in our hospital were selected as healthy control group .Levels of blood pressure ,von Willebrand factor (vWF) etc.were measured and compared among three groups .Results :Compared with low dose group after eight‐week treatment ,there were significant reductions in levels of SBP [(147.33 ± 11.37) mmHg vs.(140.51 ± 10.85) mmHg] ,DBP [(96.35 ± 7.38) mmHg vs.(92.56 ± 6.83) mm‐Hg] ,TG [ (2.38 ± 0.59) mmol/L vs.(1.55 ± 0.46) mmol/L] ,TC [ (6.48 ± 0.58) mmol/L vs.(5.38 ± 0.52) mmol/L] ,LDL‐C [ (4.24 ± 0.41) mmol /L vs.(3.26 ± 0.42) mmol/L] ,hsCRP [ (6.38 ± 1.53) mg/L vs.(5.05 ± 1.38) mg/L] , ET‐1 [ (80.78 ± 18.54) pg/ml比(73.22 ± 10.98) pg/ml] and significant rise in HDL‐C level [ (1.13 ± 0.27) mmol/L vs.(1.29 ± 0.25) mmol/L] in high dose group , P<0.05 or <0.01. Conclusion :Different doses of atorvastatin possesses different therapeutic effect on hypertensive patients with hyperlipidemia .Therapeutic effects lowering blood pres‐sure and blood lipids ,improving vascular endothelial function and reducing levels of inflammatory factors of 20mg atorvas‐tatin is significantly better than those of 10mg atorvastatin ,which is worth extending.
