In 669 newly diagnosed T2DM patients the positive rate was 10.9% for GAD-Ab, 10.5% for ICA, 4.2% for both antibodies and 13.0% for anyone of the two antibodies. BMI was higher in antibodies-positive adult diabetic patients than that in control.

3.The effects of corticosteroid treatment on immune thrombocytopenia under new diagnostic criteria

Objective To address the standard first-line management under the new diagnostic criteria in adult immune thrombocytopenia (ITP). Methods A retrospective analysis was conducted involving 178 adult ITP patients treated with high-dose dexamethasone or prednisone in Qilu Hospital from March 2004 to November 2009 using new diagnostic criteria. Results The median age was 41 years with a male/female ratio of 0. 73: 1. Among the 178 ITP patients, 87 were newly diagnosed, 30 persistent ITP, 58 chronic ITP, and 3 unable to follow up. The efficacy rates among 167 patients able to assess in the three groups were 77.4% ( 65/84 ), 64. 0% ( 16/25 ) and 62. 1% ( 36/58 ) respectively, and their complete remission (CR) rates were 57. 1% (48/84), 36. 0% (9/25) and 32. 8% (19/58). The efficacy rate and CR rate of the newly diagnosed ITP category were significantly higher than those of the chronic ITP category (x2 = 3. 917, P < 0. 05 ;x2 = 8. 186, P < 0. 01 ). The patients treated with high-dose dexamethasone or prednisone therapy had no significant differences in sex, age or blood platelet count before treatment.Moreover, the short or long term response rates and the CR rates between the two therapies had no statistically significant differences while the former had a shorter onset time ( F = 10. 34, P < 0. 01 ).Conclusions The study sets up a basis for the application of the recommended new definition and outcome criteria for adult ITP. Dexamethasone therapy is favored as first-line therapy.
6.Climatic Characteristics of High Temperature and Muggy and Comfort Degree Assessment in Summer in Sichuan Basin

Objective To study the impact of the high temperature and muggy weather on the people’s life and work. Methods The data of summer temperature and relative humidity (Jun-Aug) from 1971 to 2000 and that of 2006 were used and the characteristic of muggy weather and distribution in main cities, Chengdu, Guangyuan, Yibing, Suining and Dazhou in Sichuan basin were compared, the influence of the thermal environment on people comfort degree was evaluated by using temperature-humidity index. Results 177 high temperature days, 17 high temperature processes, 167 muggy weather days and 12 muggy processes were seen in 5 cities in Sichuan basin in the summer of 2006. The thermal environment of Chengdu and Dazhou mainly manifested the lighter heat, heat and lower comfort degree. At 2 p.m., the relative humidity weakened the comfort degree in Chengdu, but enhanced it in Dazhou. The duration of non-comfort degree in Dazhou was longer than that in Chengdu. Conclusion The relative humidity has a strong impact on people comfort degree and the combined use of temperature and relative humidity will be useful to evaluate the city thermal environment and people comfort degree.
7.Molecular cloning of mouse peroxisome proliferator activated receptor ?_2 and characterization of its expressing product in eukaryotic cells

Objective To provide an approach to research of peroxisome proliferator activated receptor (PPAR) ? 2 function, mouse PPAR? 2 (mPPAR? 2) gene was cloned and its transient expression in eukaryotic cells was carried out. Methods mPPAR? 2 mRNA from epididymis fat pad of Chinese Kunming mice was amplified by RT PCR and subcloned into plasmid pcDNA3 to generate the recombinant plasmid pcDNA3/mPPAR? 2 which was confirmed to contain the amplified target gene segments with fluorescence sequencing. The recombinant plasmid pcDNA3/mPPAR? 2 was used to transfect COS 7 with lipofectamine and the expressing product was detected with immune fluorescence assay and Western blot. Results The sequencing results for amplified target gene showed that the sequence of mPPAR? 2 from epididymis fat pad of Chinese Kunming mice is similar to that of mouse PPAR? 2 in Genbank, only at the site of 383 amino acid where Ser (AGC) substitutes Asn (AAC). pcDNA3/mPPAR? 2 was efficiently expressed in eukaryotic cells in vitro. Conclusion This work is the experimental basis for further researching on PPAR? 2 function.
8.The Effect of immunomodulatory therapy On CD4+CD25+ tregs in peripheral blood of patients with Sepsis

Objective To investigate the percentage of CD4 + C125 +Tregs in peripheral blood of patients with sepsis and its effect on cell immunity so as to unravel the effect of immunomodulatory therapy on it. Method Fourty patients with sepsis in ICU were randomly (random number) divided into experimental group and control group . The patients of experimental group were treated with Ulinastatin and immunoregulation agent (Thymosin αl) as well. The blood specimens were collected just before treatment, 3 days and 8 days after treatment. The percentages of CD4 + CD25 + Tregs and lymphocyte subsets were detected by using FCM (flow cytometry), and TNF-α, IL-6 and IL-10 assayed by using ELISA, and APACHE Ⅱ scores were calculated. Results Before treatment, the percentage of CD4 + CD25 + Tregs increased, and the number of lymphocytes and the percentage of T lymphocytes decreased, especially the CD4 + T lymphocytes and CD4+/CD8+ decreased more markedly, and the levels of IL-6 and TNF-α increased. After treatment,the percentage of CD4+ CD25 + Tregs was decreased, the number of lymphocytes and CD4 +/CD8 + increased, and the levels of APACHE Ⅱ score, IL-6 and TNF-α decreased especially in the experimental group decreased more significantly (P < 0. 05). Conclusions The percentage of CD4 + CD25+ Tregs in peripheral blood can reflect the immune status of patients with sepsis and become a novel indicator to estimate the progress of sepsis, and the immunity and prognosis of patients. Treating the patients with Thymosin αl and Ulinastatin can raise their immunity, decrease the levels of IL-6, TNF-α and APACHE Ⅱ score and improve their prognosis.
10.23G vitrectomy outcomes of macular edema in retinal vein occlusion combined with vitreomacular traction or epiretinal membrane

AIM:To evaluate 23G vitrectomy for macular edema in eyes with retinal vein occlusion (RVO) combined with vitreoretinal traction (VMT) or epiretinal membrane (ERM).METHODS:Totally 22 patients (22 eyes) diagnosed with macular edema of RVO combined with VMT or ERM were retrospectively analyzed.Twelve cases performed with 23G vitrectomy together with peeling of inner limiting membrane (ILM) and/or ERM were considered as the observation group or intervention group.Ten cases without vitrectomy were recruited as control group.The best corrected visual acuity (BCVA) and central retinal thickness (CRT) at baseline, 1, 3 and 6mo were recorded and compared.RESULTS:At baseline, the difference of BCVA and CRT between observation group and control group was not statistically significant (P=0.645, 0.206).After vitrectomy, the BCVA and CRT of RVO patients in observation group were significantly improved compared with baseline at each follow-up (F=2.895, P=0.048;F=16.431, P<0.01).However, the BCVA and CRT in control group remained the same as baseline at every follow-up.Moreover, the BCVA and CRT in observation group were much better than that in control group at both 3 and 6mo after vitrectomy.However, the BCVA and CRT between two groups were not significantly different at 1mo postoperatively.CONCLUSION:The 23G vitrectomy could markedly improve BCVA and reduce CRT in RVO patients with macular edema combined with VMT and/or ERM.
