2.Cloning and Regulating Expression of Human CD34 TRS

		                        			Objective: To clone the 5'-flanking region of the human CD34 gene containing transcriptional regulatory sequence (TRS). Methods: According to the registered 5'-flanking region of CD34 gene, two pairs of primers were designed and net-PCR was used to amplify 661 bp long TRS of CD34 gene. The CD34 TRS fragment was cloned into reported plasmid pEGFP-1. The role of the regulating the specific expression of recombinant plasmid pCD34 EGFP in hematopoietic and non-hematopoietic cells was observed. Results: Restrictive endonuclease identification and DNA sequencing provedthat the CD34 promoter cloned was consistent with the sequence reported to a large extent. It could induce the EGFP gene to express in hematopoietic cell line K562 specifically, while has no effect on hepatocellular carcinoma cell HepG-2. Conclusion: The cloned CD34 gene TRS has the effect of regulating gene expression specifically. 3.Demands on the technology of prevention and treatment of hypertension and evaluation on training effect among doctors in county and countryside hospitals of Xinjiang

		                        			Objective To analyze the demands and to evaluate training effect on the knowledge and technology of prevention and treatment about hypertension among doctors in county and countryside hospitals of Xinjiang.Methods 336 doctors were trained about knowledge of prevention and treatment of hypertension and technology of useful therapeutic regimen,and were investigated for demands of the knowledge and technology and self-evaluation of training effect by questionnaire.Results The knowledge of prevention and treatment about hypertension and the technology of useful therapeutic regimen were needed urgently by 94.8% subjects.91.3% trained doctors thought that the technology suited practically in county and countryside hospitals,and 96.7% hospitals had condition of carrying out the technology.97.9% trained doctors expected this kind of technology on hypertension prevention and treatment to be trained again.74.1% trained doctors comprehended the technology,and the percent of doctor practically using the technology increased from pre-training 12.3% to post training 62.1%.Conclusion The knowledge of prevention and treatment about hypertension and technology of useful therapeutic regimen were needed urgently and were comprehended with training for the most doctors in county and countryside hospitals of Xinjiang.
		                        		
		                        		
		                        		
		                        	
4.Release of hepatocyte growth factor mediated by heparin

		                        			Objective: To observe the effects of sodium heparin and low molecular weight heparin on the release of plasma hepatocyte growth factor (HGF) in senior coronary heart disease patients.Methods: Fifty-four senior patients with coronary heart disease were divided into three groups: intravenous sodium heparin, subcutaneous sodium heparin, and subcutaneous low molecular weight heparin (LMWH).  Plasma HGF and vascular endothelial growth factor (VEGF) were measured before and after injection.Results: Plasma HGF was increased rapidly and significantly after intravenous injection of sodium heparin, reaching its peak level (about 48 fold) after approximately 10 minutes.  Plasma HGF was also increased rapidly and significantly after subcutaneous injection of sodium heparin and LMWH, reaching its peak level (about 4 and 5 fold in sodium heparin and LMWH respectively) after approximately 2-3 hours.  Conclusion: The rise of plasma HGF after heparin treatment suggests that heparin has some other biological effects in addition to its anticoagulant property through HGF. 6.Value of soluble epithelial cadherin in differential diagnosis of pleural effusion

		                        			Objective To investigate the value of soluble epithelial cadherin(sE-cad)in the differential diagnosis of pleural effusion. 7.Effect of whole-body hypothermia on cardiac hemodynamics neonatal piglet after hypoxia-ischemia

		                        			Objective To determine the effect of mild hypothermia on neonatal piglet cardiac hemodynamic function after hypoxia-ischemia (HI).Method Twenty five 7-day-old piglets were used for hypoxic ischemic brain damage (HIBD) model by the method of temporary occlusion of the bilateral carotid arteries and followed by mechanical ventilation with low concentration of oxygen (FiO_2=6%) for 30 minutes.The piglets were randomly divided into three groups:group A (normothermia with body temperature to 39℃,n=9),group B (body temperature to 36℃for 72 hours,n=8),and group C (body temperature to 34℃for 72 hours,n=8).Mild hypothermia was initiated at 4 hours after HI,the systolic and diastole function were evaluated by Doppler echocardiography at pre-HI,post-Hi 4 hours and post-HI 72 hours.Results There were no significant differences in left ventrieular ejection time/left ventrieular ejection time (LPEP/LVEF),right ventricular ejection acceleration time/right ventricular ejection time (RACT/RVET) and CO at post-HI with hypothermia 72 hours in three groups,but the heart rate decreased in B and group C group.Compared with nonnothermia,mild hypothermia treatment showed no significant differences in MAP,LPEP/LVET,RACT/RVET,CO,SV at post-HI with hypothermia 72 hours.Conclusions Body temperature decreased by 3~5℃for 72 hours will not aggravate hemodynamic abnormity.
		                        		
		                        		
		                        		
		                        	
8.The Clinical Observation on the Influence of Guo's MaQian Decoction on Joint Flexion and Extension of Elbow and Knee Joints of Moderate Skeletal Fluorosis

		                        			Objective To observe the degree of joint flexion and extension of elbow and knee joints of moderate skeletal fluorosis patients before and after treatment and to assess the effects of Gun's MaQian decoction in treating moderate skeletal fluorosis. Methods 54 moderate skeletal fluorosis patients were randomly recruited into a treatment group and a control group. The treatment group was treated with Gun's MaQian decoction initially, and changed to be treated with GuKangNing capsule after 8 weeks' treatment. The control group was treated with placebo at first, and also changed to be treated with GuKangNing capsule after 8 weeks. Results Joint flexion and extension of elbow was improved greatly in both groups after the treatment, while the treatment group showing better effects than the control group (P<0.05). The improvement of joint flexion and extension of knee had not showed a statistical significance after the treatment. A 24 weeks follow-up to the joint flexion and extension of both elbow and knee revealed obvious improvement and the difference between the two groups were significance (P<0.05). Conclusion Gun's MaQian decoction can improve the flexion and extension of elbow and knee in moderate skeletal fluorosis patients.
		                        		
		                        		
		                        		
		                        	
9.Role of dynophin and substance P in end stage renal disease rats with pruritus

		                        			Objective To investigate the role of dynophin and substance P in pruritus with 5/6 subtotal nephrectomy rats (STNx). Methods 5/6 subtotal nephrectomy rots were prepared. After 24 weeks , STNx rats entered in end stage renal disease(ESRD). Sixty male SD rats were divided into 4 groups, STNx+P group(n=15), STNx+dynophin group( n=15), STNx control group(n=15) and sham operation group(n=15). Substance P (SP), dynophin and saline were intradermal injected separately. After injection, scratch reaction of rats in 30 minutes were recorded. And then SP concentration in blood was estimated by ELISA and SP expression in skin was examined by immunohistochemisty. Results SP concentration in blood of STNx rats were (1010.2±103.5)pg/ml, which increased to (2530.0± 236.3) pg/ml in STNx+SP group and decreased to (612.4±72.2)pg/ml in STNx+dynophin group, and it was the lowest in sham opera- tion group (240.2±36.5)pg/ml. SP expression in skin was similar to that of in blood. The scratch times in STNx+SP group were highest (7.3±1.9 times), there was no significant difference between STNx + dynophin group and STNx control group. Conclusion Pruritus in ESRD rats was correlated to the increase of SP in blood and skin. 10.Radio-frequency ablation for multiple hepatic cancer

		                        			Objective To study the effects of intraoporative radio-frequency ablation on immune functions and survival of patients with multiple large hepatic cancer. Methods Forty five admitted patients with multiple large hepatic cancer from January 2003 to January 2007 were devided into: simple hepatic artery embohzation chemotherapy group (TACE group, n = 20) , local resection of multiple lesion + TACE (LR group, n = 13), and TACE + intraoperative radio-frequency ablation (IRFA group, n = 12). The changes of peripheral blood T-cell subsets were evaluated using flow cytometry, and a comparison of the complete remission rate and survival rate between the 3 groups was made and the survival rate analyzed with Kaplan-Meier method, the validity check with long-rank method. Results CD4+ , NK, and CD4+/ CD8+radio significantly increased 4 weeks after treatment only in IRFA group. The immune function was suppressed during the first week after treatment in local resection group. Tumor complete remission rate in IRFA group, local resection group and simple TACE group were 41.70%, 46. 20% and 25.50% respectively, the difference was not statistically significant between the 3 groups (x2 = 1.81, P > 0.05). the 1.5 year and 2.0 year survival rate in the 3 groups were 75.00%, 69. 20%, 30% (x2 = 7.96, P < 0.05) and 50.00%, 23.10%, 10. 00% respectively (x2 = 18.98 ,P <0.05), the mean survival period of patients in the 3 groups was 26. 56 months, 21.04 months, and 16.41 months respectively (x2 = 14.69, P < 0.001). Kaplan-Meier survival curve showed the overall survival rate in the IRFA group was significantly higher than that of the other 2 groups (x2 = 4.635, P < 0.05). The prolongation of the survival period in patient with multiple macronodular hepatic cancer after IRFA treatment was mainly due to the prolongation of survival period in tumor bearing patients (IRFA group vs LR group, x2= 4.615, P < 0.05).
