1.Low grade central osteosarcoma treated by wide resection-experience in 13 consecutive patients from one center

[Objective]To retrospectively study all the low grade central osteosarcoma(LGCO) cases treated in our center in about a past decade.[Method]Thirteen cases with LGCO treated from January 1998 to June 2008 were studied.The mean age at diagnosis was 31-year-old.The anatomical location of the lesions was similar to that of a conventional osteosarcoma.Pain and mass were common complaints with a mean duration of 16 months before diagnosis.Eight cases accepted the first operation in our center,while the other 5 cases had undergone operations in other hospitals with an intralesional resection in four and a marginal resection in one.Eleven patients had an Enneking stage I disease and two had a stage III disease.The first choice for patients with stage I disease was wide resection.[Result]In two patients with local recurrence,the postoperative pathological diagnoses were fibrosarcoma and malignant fibrous histocytoma respectively.The metastatic lesions were found to be a LGCO in both patients with stage III disease.The average follow up duration was 29 months.Lung metastasis occurred in one stage I patient 15 months after operation.Gama-knife was used to get rid of the lung lesions and then systemic chemotherapy was given.Now he was still alive with disease 27 months after the first operation.One of the 2 patients with stage III disease died of the disease 26 months after operation and the other one was failed to follow up 6 months after operation.The other 10 patients were alive without disease at the last follow-up.For those who received limb salvage operations,the mean MSTS 93 score was 79%.[Conclusion]For most patients with LGCO,wide resection of tumor is preferred.Good oncologic and functional results could be expected after a limb salvage operation without chemotherapy.Insufficient resection of tumor would lead to recurrence.Long time follow up is compulsory.High grade sarcomas would be found in some of the local recurrent lesions.Metastasis would occur in few patients and the metastatic lesions may still be a LGCO.

3.Relationship between serum uric acid and metabolic syndrome in middle-aged and elderly

Objective To explore the relationship between serum uric acid(SUA) levels and metabolic syndrome(MS) in middle-aged and elderly people,and to investigate the impacts of age and gender on the relationship between SUA and MS.Methods A cross-sectional study was carried out in general population aged over 35 years in rural area of Yiling District,Yichang City in 2007.All subjects were investigated by using the standardized questionnaire,physical and biochemical examinations.MS was defined according to the diagnostic criteria of the latest Chinese guidelines.All subjects were divided into different subgroups based on quartiles of SUA levels and age(35-44 years old,45-59 years old,over 60 years old).The relationship of MS with age and SUA levels were investigated.Results 9354 subjects consisted of 3883 males and 5471 females were included in this study.The percentage of subjects aged over 60 years was 21.1% in men and 17.4% in women.The prevalence of MS from the first quartile to the fourth quartile were 3.8%,5.5%,7.9%,15.2% in males,and 3.1%,5.5%,7.9%,16.7% in females,respectively.The SUA levels was positively correlated with MS in both sexes after adjusting for other confounding factors.By analyzing associated-subgroups based on age and SUA levels,logistic regression analysis showed that the risk of MS was significantly increased with aging and SUA levels in female subjects.The association between age and MS was significantly weaker in men than in women.Conclusions There is a positive correlation between SUA levels and MS.The associations of MS with age and SUA levels are more closely in women than in men.The risk of MS may be highest in middle-aged and elderly women with high level of SUA.
4.Effects of modified Xiaoqinglong decoction and its ingredient-reduced prescription on plasma levels of IL-5 and TNF-α

ObjectiveTo observe the modified Xiaoqinglong decoction and its ingredient-reduced prescription on plasma levels of IL-5 and TNF-ct in rats,and to explore its mechanism of treating AR.To make clear the law of combination in modified Xiaoqinglong decoction.MethodsThe modified Xiaoqinglong decoction was divided into six groups,namely,reinforcing qi group,warming yang group,expelling retained morbid fluid group,reinforcing qi and warming yang group,reinforcing qi and expelling retained morbid fluid group,warming yang to expel retained morbid fluid group.90 rats were randomly divided into nine groups,namely,normal control group,model group,reinforcing qi group,warming yang group,expelling retained morbid fluid group,reinforcing qi and warming yang group,reinforcing qi and expelling retained morbid fluid group,warming yang to expel retained morbid fluid group,and the whole decoction group,with 10 rats in each group.Rat model of allergic rhinitis was made by the use of adjuvant systemic antigen sensitization and local attack,the normal control group and model group were fed with normal saline,the other groups were treated with appropriate drugs,once a day oral administration,continuous for 4 weeks.Detected plasma IL-5 and TNF-α levels of each group in rats.ResultsThe contents of plasma IL-5 (16.0±2.7)mg/L and TNF-α (57.5±8.0)mg/L in the model group was significantly increased,while it was significantly reduced in the reinforcing qi group,warming yang group,expelling retained morbid fluid group,refinorcing qi and warming yang group,reinforcing qi and expelling retained morbid fluid group,warming yang to expel retained morbid fluid group with plasma IL-5[each group was (12.9± 3.1) mg/L、(11.8 ±2.8) mg/L、(12.0±2.3) mg/L、(12.3±2.3) mg/L、(11.1±2.1)mg/L、(11.2±2.5)mg/L、(8.42.3)mg/L respectively]and TNF-α[each group was (27.7±5.7)mg/L、(29.5 ± 3.7) mg/L、(31.2 ± 4.9) mg/L、(28.1 ± 2.8) mg/L、(33.4 ± 5.6) mg/L、(26.3 ± 3.9) mg/L、(21.6 ±4.9) mg/L respectively],the whole decoction group effect was significant compared with the other treated groups (P<0.05).ConclusionThe modified Xiaoqinglong decoction and its ingredient-reduced prescription can regulate plasma levels of IL-5 and TNF-α.The modified Xiaoqinglong decoction had best results in reinforcing qi,warming yang and expelling retained morbid fluid.
5.Expression of EphA2 and E-cadherin in colorectal carcinoma

Objective To explore the expression and clinical significance of EphA2 and E-eadherin in colorectal carcinoma.Methods The expression of EphA2 and E-eadherin in 67 cases of eoloreetal carcinoma and 28 cases of normal large intestine tissues were determined by immunohistochemieal method(S-P method),and their relationship to elinicopathological characteristics were analyzed.Results The positive expression of EphA2 and negative expression of E-cadherin in colorectul carcinoma tissues were significantly higher than those in normal intestine tissues(P<0.01).F111e positive expression of EphA2 and negative expression of E-eadherin were positively correlated with tumor histological grade,invasive depth and lymph node metastasis(P<0.01 orP<0.05),but not correlated with tumor~ross type(P> O.05).The expression of EphA2 was negatively related with E-cadherin.Conclusions The abnormal expression of EphA2 and E-cadherin may be together involved in the development and progression of eolorectal carcinoma,It may be a good marker for monitoring the malignant degree and the prognosis of colorectal carcinoma by combining E-eadherin and EphA2.
7.Pretreatment of ramie and kenaf stalk for bioethanol production.

Ramie and kenaf were traditional fiber crops in China, but their stalk after decorticating has not been used effectively. The stalk contains a lot of cellulose, and can therefore be used for the production of bioethanol. We studied the effects of different chemical pretreatment on enzymatic digestibility of ramie stalk and kenaf stalk. Ramie and kenaf stalks pretreated with alkali were chosen to produce ethanol using quasi-simultaneous saccharification and fermentation (Q-SSF) process. The results show that for the stalks pretreated with 4% NaOH and 0.02% anthraquinone-2-sulfonic acid sodium salt (AQSS) as catalyzer at 170 degrees C for 1 h, the ethanol concentration could reach 51 g/L after fermentation for 168 h at 18% of solid substrate concentration. By fed-batch to 20% of solid substrate concentration, the ethanol concentration could reach 63 g/L, 77% and 79% of the cellulose conversion could get for ramie stalk and kenaf stalk, respectively. For kenaf stalk pretreated with 5.2% NaHSO3 and 0.2% H2SO4 at 170 degrees C for 1 h, the ethanol concentration and cellulose conversion could reach to 65 g/L and 72%, respectively.

8.Application of PCR-RFLP technique on identification and genotyping of Brucella spp

Objective To amplify the 16S RNA fragments of 7 clinically isolated strains of Brucella spp. by PCR-RFLP technique, so as to provide experimental basis for the studies on diagnostics, genetics and epidemiology of Brucella spp. Methods According to the gene sequence of ATCC 25840 standard strain in GenBank, special primers for the 16S RNA conservative area in the Brucella spp. were designed. DNA extraction and PCR amplification of the 16S RNA fragments were performed with the 7 isolated strains. PCR products were then sequenced and RFLP analysis was conducted with appropriate restricted enzymes to study the homology and the mutation sites in those strains. Meanwhile, the clinical data of infected patients were retrospectively analyzed to evaluate the relationship between the clinical features and genotypes of Brucella infection. Results The amplified target fragments were about 1500bp in length and consistent with what was expected. The sequencing and homology analysis showed a 98.88% homology and 11 mutation sites among the 7 isolated strains. Four genotypes were identified by RFLP. Retrospective analysis of the clinical data indicated that no obvious relationship existed between the genotypes and the clinical features. Conclusions Amplifying 16S RNA fragments by PCR technique is a feasible method to make an early diagnosis of Brucella infection. The 7 clinically isolated strains are different in genotypes and 16S RNA fragment is a highly conservative fragment in bacterial genome with some mutations. The research provides evidence for the genetics and epidemiology of brucellosis.
9.The expression of miR-101 in pancreatic cancer and its effect on proliferation of pancreatic cancer cell line ASPC1

Objective To investigate the expression of miR-101 in pancreatic cancer and the effect of down-regulation miR-101 on proliferation of pancreatic cancer cell line ASPC1. Methods Real-time PCR was used to determine the expression of miR-101 in pancreatic cancer, adjacent tissues and pancreatic cancer cell line ASPC-1. The miR-101 over-expression vector (peGFPc1-miR-101) was constructed and was transfected into ASPC-1 cell. Transfection efficiency was measured by fluorescence microscope. The expression of miR101in the transfected cells was detected by real-time PCR. Cell viability analysis was performed by MTT. The targeted genes of miR-101 in pancreatic cancer were scanned by the online targeted gene prediction software (target Scan). Results The expression of miR-101 was in pancreatic cancer tissues, adjacent tissues and ASPC-1 cell line, respectively. The expressions in pancreatic cancer tissues and ASPC-1 cells were significantly lower than that in adjacent tissues ( P < 0.01 ). The expression of miR 101 in transfected cells increased to 19.8 folds as much as that in the control group (P <0.01 ). Proliferation rate of transfected cells was significantly decreased, which was only 26% of primary cells ( P < 0.01 ). EZH2 was the potential targeted gene of miR-101 in pancreatic cancer. Conclusions miR-101 was weakly expressed and it may affect the proliferation of pancreatic cancer cell by inhibiting the EZH2 expression.
10.Correlation between serum testosterone level and changes of levels of inflammatory factors in male patients with acute coronary syndrome

Objective: To measure serum testosterone level, plasma levels of interleukin-18 (IL-18) and IL-10 and explore their correlation in patients with different types of coronary heart disease (CHD), and their possible role in occurrence and development of CHD. Methods: A total of 96 male CHD patients were divided into acute myocardial infarction (AMI) group (n=35), unstable angina pectoris (UAP) group (n=32) and stable angina pectoris (SAP) group (n=29). Another 30 patients who were excluded for CHD by coronary angiography were enrolled as non CHD control group. Enzyme linked immunosorbent assay (ELISA) was used to measure serum testosterone level and plasma levels of IL-18 and IL-10 in all groups. Results: Compared with non CHD control group, there were significant decreases in serum testosterone level [(13.46±1.99) mmol/L vs. (6.89±1.35) mmol/L vs. (5.02±1.87) mmol/L, t=1.917~2.365, P<0.05 both] in UAP group and AMI group, and that of AMI group was significantly lower than that of UAP group, t=1.034, P<0.05; there were significant increases in IL-18 levels [(146.72±79.36) pg/ml vs. (209.32±80.49) pg/ml vs. (316.78±75.63) pg/ml vs. (457.78±83.21) pg/ml, t=2.016~3.167,P<0.05 all] in SAP group, UAP group and AMI group, and those of UAP group and AMI group were significantly higher than that of SAP group, t=2.173, 2.596, P<0.05; there were significant increases in IL-10 levels [(48.46±18.27) pg/ml vs. (116.45±42.76) pg/ml vs. (85.64±27.33) pg/ml vs. (70.26±18.55) pg/ml, t=2.997~2.018, P<0.05 all] in SAP group, UAP group and AMI group, and those of AMI group and UAP group were significantly lower than that of SAP group (t=2.034, 2.291, P<0.05 both). Pearson linear regression analysis indicated that serum testosterone level was negatively correlated with levels of IL-10 (r=-0.678, P<0.01) and IL-18 (r=-0.579, P<0.01) in CHD group. Conclusion: There are significant changes in serum testosterone level and plasma IL-18, IL-10 levels, and testosterone level is significantly negatively related with IL-18, IL-10 levels, and they can be regard as new indexes assessing coronary atherosclerotic lesion.
