1.Analysis of the Effects of Different Disinfectants on Results of Antibiotics Skin Test

OBJECTIVE:To analyze the effects of different disinfectants on results of antibiotics skin test,and to improve the accuracy of the judgment results of antibiotics skin test. METHODS:1 500 patients undergoing antibiotics skin test of penicillin and cephalosporin were randomly divided into group A and group B with 750 patients in each group. Group A was given 75% etha-nol disinfection,and group B was given iodine disinfection. The incidence of false positive results were compared after disinfected with 2 kinds of disinfectants. RESULTS:The incidence of false positive result in group B was lower than in group A,with statisti-cally significant difference (χ2=10.004,P<0.05). CONCLUSIONS:For skin disinfection of antibiotics skin test,iodine is better and safer than 75%ethanol.
2.Application of patient-reported outcome in Chinese medical therapeutic efficacy assessment of cardiovascular diseases and some considerations on it.

Patient-reported outcome (PRO) is one of the important measures for clinical therapeutic efficacy assessment. The current application and development of PRO in China and abroad were introduced in this paper, put stress on the application of PRO in Chinese medical therapeutic efficacy assessment of cardiovascular diseases. Moreover, some suggestions on its further application were offered.

3.Research of teaching methods on medical statistics of undergraduate nurse students studying traditional Chinese medicine

Objective To explore the cognitive level of medical statistics course in undergraduate nurse students studying traditional Chinese medicine and the methods to improve the present teaching method,and to develop statistical thinking of students with the view of promoting the research level of students.Methods With the nursing project and teaching method of enlightening interesting,A self-designed questionnaire was used to survey the cognitive level of 142 undergraduate nurse students studying traditional Chinese medicine in Nanjing University of TCM,all of them had completed the medical statistics curricula.Results Total of 142 students were collected,most students had confidence in studying the course.90.1% students had gasped the basic thought,51.4% students did not think about the future research work,80.3% students had realized the existence of variance and error,16.9% students thought nursing of Chinese medicine had disadvantages compared to Western medicine.Most students supported the application of statistical software.Among several examination methods,practice a specific data or distinguish data processing error was the methods that more accepted by students.Conclusions Nursing of Chinese medicine has broad and bright prospects,thus,it is essential to raise the scientific cognition and data procession level of nursing students.Data pertaining Chinese medicine is complicated and it is vital to deep the teaching of medical statistics course,to raise the studying interesting,to practice the concrete data and to teach the theory course combine with the software.
4.Construction and effect identification of MiR RNAi eukaryotic expression vectors of prohibitin.

AIMTo construct the RNAi eukaryotic vector of inhibitory member of the prohibitin (PHB-1) gene and observe the interfering effect in HEK293 cell line after the vector transfection.
METHODSThe specific Mi RNA sequence was designed according to the PHB-1 sequence in GenBank, complementary single-strand DNA oligonucleotides were designed and synthesized, and annealed the single-stranded oligonucleotides to generate a double strands oligonucleotides , cloned the oligonucleotides into pcDNATM6.2-GW/EmGFP-MiR-PHB to obtain an entry clone and then sequence analysis was performed. The recombinant plasmid was transfected into HEK293 cell by liposome. PHB-1 expression was detected by Western blotting.
RESULTSThe DNA sequence of interest clone to the vector was constructed to generate an entry clone and an expression clone successfully, which were proved by sequence determination. Western blotting analysis demonstrated that PHB-1 MiR RNA expression construction could suppress the expression of PHB-1.
CONCLUSIONA RNAi eukaryotic vector containing prohibitin gene was successfully constructed.

5.Effect of purified xuefu capsule on ultrasonographic figures in patients with carotid atherosclerosis.

OBJECTIVETo investigate the effect of Purified Xuefu Capsule (PXC) on ultrasonographic figures in patients with carotid atherosclerosis (CAS).
METHODSNinety-two patients with CAS were randomly divided into two groups, the 45 patients in the treated group were treated by conventional western medicine with additional PXC, the 47 patients in the control group were given conventional western medicine alone. One month's treatment was regarded as one course, and three courses of treatment were given successively to both groups. Carotid ultrasonography was performed before and after treatment, and the figures were compared.
RESULTSThe thickness of endangium, average number of plaques, plaque scores, plaque volume index and length of the biggest soft plaque or mixed plaques, stenosis rate of vascular area, and the resistant index of blood vessels were decreased after treated with PXC for 3 months in the treated group. The differences of these indexes in the treated group before and after treatment, and that between the treated group and the control group after treatment were significant (P < 0.01).
CONCLUSIONPXC can improve hemodynamics and eliminate atherosclerotic plaque to a certain degree, so it has definite effect for prevention and treatment of CAS.

6.Experimental study of mandibular periosteal distraction in rabbits.

OBJECTIVETo investigate a novel technique for new bone formation--periosteal distraction osteogenesis.
METHODSA custom made periosteal distraction device was fixed to bilateral surface of the mandible in three rabbits. Periosteal distraction was performed on the left side of the mandible, the right side of the mandible served as the control. The animals were sacrificed at the end of distraction process. All the specimens were X-rayed and histologically examinated.
RESULTSAll three animals survived with no obvious complications. Both in mass specimens and X-rays, there showed new bone formation on the distracted side of the mandible. In histological examinations, there was osteoblast-like cell infiltration and bone tissue formation in the distracted area.
CONCLUSIONPeriosteal distraction osteogenesis can provide a novel technique for the repair of bone defects.

7.Effect of autocontrol micromotion intramedullary interlocking nail on fracture healing: an experimental study.

OBJECTIVETo investigate the effect of autocontrol micromotion locking nail (AMLN) on experimental fracture healing and its mechanism.
METHODS16 goats undergoing both sides of transverse osteotomy of the femoral shafts were fixed intramedullary with AMLN and Gross-Kempf (GK) nail, respectively. The follow-up time was 7, 14, 28 and 56 days. Roentgenographic, biomechanical, histological, scanning electromicroscopic and biochemical analyses were done.
RESULTS(1) The strength of anticompression, antiflexion and antitorsion in the fractural end in the AMLN-fixed group was higher than that of GK nail-fixed group; whereas, the rate of stress shelter in the fractured end decreased significantly (P<0.01). (2) The content of the total collagen, insoluble collagen, calcium and phosphate in the AMLN-fixed group was higher than that in the GK nail-fixed group (P<0.05). (3) Histological observation and quantitative analysis of calluses revealed that AMLN could promote the growth of bridge calluses and periosteum calluses. Hence the fracture healing and remolding process achieved early, which was much better than traditional GK nail fixation. (P<0.05). (4) 7-14 days postoperation, the calluses of AMLN-fixed group was flourish and camellarly arranged and the collagen fibril formed constantly in the absorption lacuna of bone trabecula. 28-56 days postoperation, the collagen fibril was flourish around the absorption lacuna and was parallel to the bone's longitudinal axis. Active bony absorption and formation were seen, so was remolding and rebuilding. Haversian system was intact and the bony structural net was very tenacious because of the deposition of calcium salt. None of the above findings was observed in the GK nail-fixed group.
CONCLUSIONSThe design of AMLN accords well with the plastic fixation theory. As the geometry ametabolic system constituted by the intramedullary fixation instruments and the proximal and distal end of the fracture is very firm and stable, the disturbance to the physical stress distributed in the fractural end is light. The generation and conduct of the intermittent physical stress between the fractural parts could reach the balance between stress conduct and stress protection. The feature that the healing and remolding take place at the same time speeds up the fractural healing process.

8.Regulation of proliferation and apoptosis of human vascular endothelial cell by Acheron.

OBJECTIVETo investigate regulatory effect of Acheron (Achn) on proliferation and apoptosis of human vascular endothelial cell.
METHODS(1) Eahy926 cells were cultured in serum-free DMEM medium (96-well plates) and were divided into Achn inhibition group (transfected with plasmid psi-Achn), psi4.1 group (transfected with psi4.1 empty vector), Achn induction group (transfected with pcDNA-Achn), pcDNA3.1 group (transfected with pcDNA3.1 empty vector), cotransfection group [cotransfected with pcDNA-Achn + psi-calcium/calmodulin-dependent serine protein kinase (CASK)], blank control group (treated with PBS) according to the random number table (the same method below). The cell proliferation was determined by MTT assay at post transfection hour (PTH) 1, 24, 48, 72, with expression of absorbance value. (2) Total protein of Eahy926 cells were extracted and quantitated by BCA assay, and then they were divided into Achn antibody precipitation group (100 µg protein), CASK antibody precipitation group (100 µg protein), IgG antibody group (100 µg protein), Western blot group (20 µg protein). Achn and CASK protein levels were determined by immunoprecipitation and Western blot. (3) Synchronously cultured Eahy926 cells were divided into LPS induction group (treated with 5 mol/L LPS), Achn transfection group (transfected with pcDNA-Achn), cotransfection group (cotransfected with psi-CASK and pcDNA-Achn), KCl group (treated with 5 mol/L KCl), and blank control group (treated with 5 mol/L PBS). Cells in transfection groups were stimulated by LPS for 12 hours after PTH 24. Caspase-3 protein level was detected by immunohistochemistry. (4) Synchronously cultured Eahy926 cells were divided into Achn inhibition group (transfected with psi-Achn vector), Achn induction group (transfected with pcDNA-Achn vector), and blank control group (treated with PBS). Apoptosis rate was determined by FITC/PI with flow cytometry. Data were processed with one-way analysis of variance and t test.
RESULTS(1) The cell proliferation in Achn inhibition group was lower than that in psi4.1 group from PTH 24, and the differences were statistically significant at PTH 48, 72 (with t value respectively 10.777, 6.112, P values all below 0.05). The cell proliferation in Achn induction group during PTH 24-72 were higher that in pcDNA3.1 group (with t value respectively 5.367, 6.053, 9.831, P values all below 0.05). The cell proliferation in cotransfection group at PTH 48, 72 were significantly lower than that in Achn induction group (with t value respectively 5.481, 9.517, P values all below 0.05). (2) Achn protein was detected in CASK antibody precipitation group while CASK protein was also detected in Achn antibody precipitation group. (3) Caspase-3 level in Achn transfection group was lower [(15.6 ± 0.5)%] as compared with that in LPS induction group [(32.8 ± 2.6)%, t = 10.083, P < 0.05], and that in cotransfection group showed further inhibition [(7.0 ± 2.0)%, t = 9.827, P < 0.01]. (4) Apoptosis rate in Achn inhibition group [(45.6 ± 10.9)%] was higher than that in blank control group [(13.2 ± 4.3) %, t = 7.043, P < 0.05]; while that in Achn induction group [(5.3 ± 2.9)%] was lower than that in blank control group (t = 6.499, P < 0.05).
CONCLUSIONSAchn can promote human vascular endothelial cell proliferation, and inhibit its apoptosis induced by LPS or burn serum, and the effect is related to CASK.

9.Therapeutic effect of dense-packing autologous hair grafting technique for restoration of seborrheic alopecia

Objective To investigate the therapeutic effect of dense-packing auto hair grafting technique on the restoration of seborrheic alopecia. Methods With local anesthesia, a scalp strip was harvested from the back of the head. Under operating microscope (Various graft was created from the scalp strip, including micro-grafts with 1-2 hairs, mini-grafts with 3-4 hairs and sliver graft with 5-6 hairs. In the alopecia recipient area, micro slots were made with a small triangle-edged needle for the micro-grafts,mini slits were made with mini blade for the mini-grafts and foramen ovale were made with a slot punch. The grafts were then implanted into these holes. Results 32 cases of seborrheice alopecia were treated with the above mentioned technique from March 2007 to July 2009. Postoperative following up for 12-24 month showed that the grafted hairs were growing well with average 90 % survival of the hair. 81 % of the patients obtained satisfactory results with only one stage operation. Six patients needed the second operation to improve the appearance. All of the patients were satisfied with the appearance. Conclusions The dense-packing hair grafting technique with various grafts not only saves time of operation, but also obtains dense grafted hair and well appearance. The results are satisfactory to most patients with only one stage operation.
10.Clinical study of transcrestal maxillary sinus floor elevation with the disk-up sinus reamer.

OBJECTIVETo evaluate the clinical effect of the disk-up sinus reamer (DSR) applied to transcrestal maxillary sinus floor elevation with simultaneous placement of implants.
METHODSThirty-seven patients underwent transcrestal maxillary sinus floor elevation with fifty-one implants placed simultaneously using the DSR. The residual bone height (RBH) was 3 to 8 mm, (5.61 ± 1.61) mm on average. The safety of this technique and the pain index during the operation was evaluated. The final prostheses were restored in 3-6 months postoperatively. The follow-up period was 3 to 24 months. The stability and osseointegration of the implants were clinically evaluated, and the endo-sinus bone gain around the implants were measured.
RESULTSThe elevation height ranged from 2 to 8 mm, with an average of (4.75 ± 1.55) mm. There was no detectable sinus membrane perforation, no serious suffering or uncomfortable subjective sensation in any patients during operation with a pain index of (2.22 ± 0.98). During the follow-up period, no sinus complication was observed. Favorable osseointegration was obtained. There were no implants or prostheses which were loose or lost. The survival rate was 100%. The radiographic results demonstrated that the endo-sinus bone gain tended to reach stabilization after 6 months and the marginal bone loss was (1.20 ± 0.72) mm after 12 months.
CONCLUSIONSTranscrestal maxillary sinus floor elevation with simultaneous implant placement by DSR is a safe, invasive and handy technique, with higher elevation height, fewer clinical complications and less pain. It shows satisfactory clinical results in short term and a long-term observation is still needed.
