1.Experimental Study on ATA Prescription in Treating Monkey with Chronic Infection of Simian Immunodeficiency Virus

[Objective] To investigate the effect of ATA, a herbal medicine compound, for the treatment of the chronic infection of simian immunodeficiency virus ( SIV) in monkeys. [ Methods ] Eight Rhesus monkeys were infected with SIVmac251 to establish monkey models with chronic infection of SIV. After 18 months, the monkeys were randomized into model group and ATA group, administered with saline and ATA 2 g?kg-1?d-1 for 56 days respectively. The changes of signs were observed, plasma levels of T-lymphocyte subsets CD4+ and CD8+ were detected by flow cytometry and the lymph node biopsy were observed under light microscope before and after treatment. [Results] After treatment, the body weight increased and the incidence of infective diarrhea decreased in ATA group, the difference being insignificant. ATA had no obvious effect on white blood cells counting and the replication number of plasma SIV, but increased the plasma levels of T-lymphocyte subsets CD4+ and CD8+(P
2.Determination of gallic acid,paeoniflorin and safflor yellow in Taohong Siwu Decoction

AIM: To determine gallic acid,paeoniflorin and safflor yellow in Taohong Siwu Decoction(Semen Persicae,Flos Carthamietc.) METHODS: To analyse three constituents by HPLC on a Hypersil BDS C_(18) column and gallic acid detection wavelength was set at 271 nm.The mobile phase was methanol-water(5∶95) (adjusted to pH=3.0 with H_3PO_4) at a flow rate of 1.0 mL/min. Safflor yellow detection wavelength was set at 403 nm.The mobile phase was methanol-water(80∶20) (adjusted to pH=3.0 with H_3PO_4) at a flow rate of 1.0 mL/min. Paeoniflorin detection wavelength was set at 230 nm.The mobile phase was methanol-KH_2PO_4 at a flow rate of 1.0 mL/min. RESULTS: Gallic acid showed a good linear relationship at the range of 4.547?10~(-3)-1.455?10~(-1) ?g/?L(r=0.999 3).Paeoniflorin showed a good linear relationship at the range of 0.010-0.200 ?g/?L(r=(0.999 8)) and safflor yellow showed a good linear relationship at the range of 2.34?10~(-2)-7.50?10~(-1) ?g/?L(r=(0.999 4)).The average recoveries were 98.0% for gallic acid,98.4% for paeoniflorin and 99.2% for safflor yellow.The RSD of the method were 1.8%,0.69% and 0.88%,respectively. CONCLUSION: The method is proved to be fast,feasible,simple,and can be used for quality control of gallic acid,paeoniflorin and safflor yellow in Taohong Siwu Decoction.
3.Circulatory sleep apnea: Preliminary report of clinical observation on sleep apnea in patients with chronic heart failure.

OBJECTIVEThe aim of this study is to investigate the occurrence and mechanism of Cheyne-Stokes breathing pattern in patients with heart failure.
METHODSFifty-six patients who performed polusomnography sleep testing at National Center of Cardiovascular Diseases Fuwai Hospital from March to May in 2015. We divided them into chronic heart failure (CHF) group and non-CHF group.
RESULTSThe occurrences of sleep apnea in two groups were high. In CHF group (n = 11) , there were 10 patients with apnea hypopnea index (AHI) > 5; and their AHI was 23.93 ±14.63. In non-CHF group (n = 45), there were 33 patients whose AHI > 5; and their AHI was 16.20 ± 18.76. The ratio of center sleep apnea to all gross sleep apnea ratio in CHF group was higher than that in non-CHF group (80.21% ± 30.55% vs 27.16% ± 35.71%, P < 0.01 ).
CONCLUSIONBased upon the new theory of holistic integrative physiology and medicine, we explain the mechanism of circulatory dysfunction induce the oscillation breathing in patients with CHF. The sleep apnea and C-S respiration in CHF should be called circulatory sleep apnea, rather than central sleep apnea.

4.The statin dosage for achieving goal of cholesterol-lowering based on risk stratification in patients with ischemic cerebrovascular diseases

Objective To explore statin dosages for targeting goal of LDL-C lowering on the basis of stroke risk stratification and the dosage-effective relation of statin and LDL-C lowering in Chinese patients with ischemic stroke and transient ischemic attack (TIA).Methods This is a prospective and open clinical trial patients with ischemic stroke/TIA within 6 months were enrolled and the dosages of atorvastatin were calculated based on risk stratification according to "Chinese Consensus for Prevention of Ischemic Stroke/TIA with Statin" (Chinese Consensus).A dose of 10 mg of atorvastatin daily to target LDL-C goal was takenas the standard dosage targeting goal (SDTG).Patients taking this dosage of atorvastatin constituted a SDTG group.Those who needed a daily dose of 20 mg or more of atorvastatin were randomized into an intensive dosage targeting goal (IDTG) group ( atorvastatin 20-80 mg/d) and a standard dosage non-targeting goal (SDNTG) group (atorvastatin 10 mg/d without targeting goal).All patients took atorvastatin for 12 weeks.The primary outcome was the rate of targeting goal for LDL-C lowering at 2,4 and 12 weeks,respectively and the secondary outcome was the occurence of recurrent stroke and other vascular events within 12 weeks.The main safety endpoint was serial adverse events including symptomatic intracranial hemorrhage.Results Altogether 102 cases were enrolled and 99 cases were followed up for 12 weeks.According to the Chinese Consensus,the rate of high risk,very high risk- Ⅰ and very high risk- Ⅱ was 44% ,28% and 28%,respectively.Targeting rate for LDL-C lowering was 77% -85% at each time point in the SDTG and IDTG groups ,being significantly higher than those in the SDNTG group ( 12% -16%,P < 0.01 ).No significant difference was found concerning the occurrence of recurrent stroke,other vascular events and safety endpoints among the three groups.The amplitude of LDL-C lowering was 32%-35% ,46%-49% ,51%-52% and 60%-65% with corresponding to daily dosage of 10 mg,20 mg,d0 mg and 80 mg atorvastatin.Conclusions At least more than half of the patients after iscbemic stroke/TIA need intensive statin therapy to target the LDL-C lowering goal.The dosage- effective relation of atorvastatin and LDL-C lowering in Chinese is similar to the reported data in other races.
5.Nasal Colonization by Opportunistic Pathogens among Health Care Workers:A Survey

OBJECTIVE To investigate the prevalence of nasal colonization among health care workers(HCWs).METHODS Nasal swabs from 93 ICU workers and 98 other clinical workers were cultured and isolated and the tests of antibiotic susceptibility were performed by using paper diffusion method.RESULTS In total,214 isolates of 8 species from 191 health care workers were recovered,of which 187 isolates were coagulase-negative Staphylococcus(CNS)(the carriage rate of 93.71%) and 8 isolates were Staphylococcus aureus(the carriage rate of 4.19%).While the total Gram-negative bacteria carriage rate was 14.14%(27 isolates).The most frequent CNS species were S.epidermidis and S.haemolyticus.The antibiotic susceptibility profiles of S.aureus and CNS differed sharply: all 8 S.aureus strains were resistant to penicillin but were fully susceptible to oxacillin,in contrast,most of CNS were resistant to both penicillin and oxacillin.The carriage rate of CNS(60.2%)and Gram-negative bacteria(26.9%)in HCWs of ICU were higher than other HCWs(P
6.Effects of manganismus on proliferation of neural stem cells in mice's hippocampus.

OBJECTIVETo explore the effects of manganese poisoning on the proliferation of neural stem cells (NSCs) in mice's hippocampus.
METHODSThe mice (weight 8 approximately 10 g) were divided into control group(CG) low-dose group(LDG) middle-dose group(MDG) and high-dose group(HDG)by intraperitoneal injection of 0, 5, 20, 50 mg x kg(-1) x d(-1) of manganese chloride dissolved in physiological saline. The ability of learning and memory was detected by Morris Water Maze, and the proliferation of NSCs in subgranular zone (SGZ) in these mice's hippocampus was also detected by immunohistochemistry.
RESULTS1) Compared with the CG, the ability of learning and memory in all manganism group decreased significantly (P < 0.01) and this phenomenon in HDG was most notable (P < 0.01). Meanwhile, the ability of memory was negatively correlated with the dose of manganese chloride (r(s) = -0.598, P < 0.01), but the difference of swimming speed in every group was of no statistic significance. (2) The numbers of NSCs in proliferation period in SGZ of all manganism groups was much lower than that of CG (P < 0.01) negatively correlated with the dose of manganese chloride (r(s) = -0.666, P < 0.01). (3) The reduction of NSCs had a positive correlation to the depression of learning and memory (r(s) = 0.734, P < 0.01).
CONCLUSIONSManganismus can affect the ability of learning and memory, which is probably caused by the inhalation of manganese on NSCs in hippocampus.

7.Cloning and expression of spinach glycolate oxidase in Escherichia coli.

The cDNA coding spinach glycolate oxidase (GO) was amplified by RT-PCR using the total RNA of spinach leaves as the template, and was cloned into cloning vector pMD18-T. After the DNA sequence was determined, the go gene was subcloned into E. coli expression vector pBV220, pET-22b(+), pTIG-Trx and pThioHisC. SDS-PAGE analysis revealed that the recombinant E. coli BL21 (DE3) (pTIG-Trx-GO) and E. coli BL21 (DE3) (pET-22b(+)-GO) expressed the predicted 38 kD glycolate oxidase, and the enzyme activity was also detected.

8.Evaluation of the diagnostic value of (18)F-FDG PET-CT and enhanced CT for staging of lymph node metastasis in non-small cell lung cancer.

OBJECTIVETo evaluate the clinical value of (18)F-FDG PET-CT and enhanced CT imaging for staging of regional lymph node metastasis in non-small cell lung cancer (NSCLC) patients.
METHODS122 patients with proven or suspected NSCLC underwent integrated PET-CT and enhanced CT scan before surgery. The results of lymph node metastasis diagnosed by PET-CT and CT were compared and analyzed according to the results of histopathological examination.
RESULTSPET-CT showed correctly lymph node staging in 80.3% of cases, overstaged in 13.1%, and understaged in 6.6%, while 55.8%, 26.2% and 18.0% by CT, respectively. The sensitivity, specificity, and accuracy of PET-CT for lymph node staging was 86.3%, 85.0% and 85.3%, respectively, while the corresponding data were 68.6%, 71.0% and 70.4% by CT, respectively (P < 0.01). 81.3% of false-negative and 71.6% false-positive lymph nodes by CT were interpreted correctly by PET-CT, while 57.1% of false-negative and 45.2% of false-positive lymph nodes by PET-CT were correctly diagnosed by CT. 5.9% of PET-CT-diagnosed negative lymph nodes were pathologically proven to be positive with small cancer foci and below 10 mm in diameter, while 8.2% of pathologically proven negative lymph nodes with inflammation, high FDG uptake and exceeding 10 mm (15 mm in subcarnial nodes) in diameter were false-positive on both PET-CT and CT imaging, therefore, these lymph nodes were still in the common blind area of diagnosis by both of PET-CT and CT.
CONCLUSIONCompared with enhanced CT, integrated PET-CT improves the accuracy and is helpful to correct some CT-diagnosed false-positive and false-negative lymph nodes. But CT is also beneficial supplementation to PET-CT for assessment of regional lymph node metastasis. The combination of PET-CT and CT can make up the shortage of both of them in staging of regional lymph nodes in NSCLC patients.

9.Posttranscriptional induction of p21Waf1 mediated by ectopic p16INK4 in human diploid fibroblast.

BACKGROUNDBoth p16(INK4) and p21(Waf1) are tumor suppressors with similar biological functions in the regulation of cellular senescence. Previous reports showed that p16(INK4) could be activated by p21(Waf1) through transcriptional factor Sp1 in HeLa cells. This study was undertaken to determine the effects of p16(INK4) on the expression and functions of p21(Waf1).
METHODSHuman diploid fibroblast 2BS cells were stably transfected with sense (2BS/p16(INK4)), antisense p16(INK4) (2BS/asp16(INK4)) or empty vector (2BS/neo). Then they were assayed by reverse-transcription polymerase chain reaction (RT-PCR), fluorescence activated cell sorting (FACS) and Western blot.
RESULTS2BS/p16(INK4) cells exhibited cell cycle arrest in both G1 and G2/M phases. Endogenous p21(Waf1) protein levels increased twofold in the 2BS/p16(INK4) cells, but not decreased in the 2BS/asp16(INK4) cells. p21(Waf1) mRNA levels were not affected in neither 2BS/p16(INK4) nor 2BS/asp16(INK4) cells.
CONCLUSIONp16(INK4) may play an important role in the regulation of cellular senescence by modulating the p21(Waf1) protein level via the posttranscriptional mechanism.

10.Construction of recombinant adenovirus vector carrying human miR-133a and its expression in vitro

AIM:To construct recombinant adenovirus vector carrying human miR-133a and study its expression in human mesenchymal stem cells(hMSCs).METHODS:The PCR product containing miR-133a was amplified from human genomic DNA and inserted into the adenoviral shuttle vector pAdTrack-CMV.Then the recombinant shuttle plasmid linearized by pmeⅠwas cotransformed into competent E.coli.BJ5183 with the adenoviral backbone plasmid pAdEasy-1 to generate the recombinant adenovirus vector rAd-mir-133a.rAd-mir-133a was then packaged and amplified in human embryonic kidney 293(HEK293) cells.The purified rAd-miR-133a was used to infect the hMSCs and the expression of miR-133a was detected by non-quantitative RT-PCR and real-time PCR.RESULTS:The recombinant adenovirus shuttle vector pAdTrack-CMV-miR-133a was constructed and verified by restriction endonuclease analysis and DNA sequence analysis.rAd-miR-133a was successfully packaged and amplified in HEK293 cells.The transcriptions of primary miR-133a and mature miR-133a were over-expressed in the hMSCs infected with rAd-miR-133a.CONCLUSION:The recombinant adenovirus vector carrying human miR-133a is successfully constructed,which lay a foundation for miR-133a function study.
