2.On the Conditionality of "Disease" in Treating Different Diseases with the Same Method.

Treating different diseases with the same method is a unique original thinking in Chinese medicine, which embodies the spirit of treatment based on syndrome differentiation. We briefly reviewed the origin and development of this concept. We also reviewed that thinking of combination of disease and syndrome is its premise and foundation. We put forward the conditionality of diseases in treating different diseases with the same method by cutting-in modern biological basic researches, that is to say, one kind specific diseases or disease of one specific system. We emphasized the importance of diseases conditionality in treating different diseases with the same method, which was of great significance in studies on combination of disease and syndrome, correspondence of prescription and syndrome, and modern biological basic researches of treating different diseases with the same method.

4.Relationship between plasma substance P and calcitonin gene-related peptide concentrations and intraoperative cardiovascular events in diabetic patients

Objective To evaluate the relationship between plasma substance P (SP) and calcitonin gene-related peptide (CGRP) concentrations and intraoperative cardiovascular events in diabetic patients.Methods Twenty-two patients of either sex with type 2 diabetes mellitus and 22 non-diabetic patients of either sex,of American Society of Anesthesiologists physical status Ⅰ or Ⅱ,aged 50-77 yr,undergoing elective knee arthroplasty under neuraxial anesthesia,served as diabetes mellitus group (group DM) and non-diabetes mellitus group (group NDM),respectively.The patients of either group were further divided into 2 subgroups (n =11 each) according to whether or not the patients had cardiovascular diseases before operation:no cardiovascular disease subgroups (NDM-NCVD subgroup and DM-NCVD subgroup) and cardiovascular disease subgroups (NDM-CVD subgroup and DM-CVD subgroup).The plasma SP and CGRP concentrations were dertermined by enzyme-linked immunosorbent assay at 30 min before operation and at the end of operation.The plasma cardiac troponin Ⅰ (cTnI) concentrations were measrued by enzyme-linked immunosorbent assay at 30 min before operation and 24 h after operation.The development of intraoperative cardiovascular events was recorded.Results Compared with group NDM,the incidence of intraoperative cardiovascular events and plasma cTnI concentrations at each time point were significantly increased,and the plasma concentrations of SP and CGRP were decreased in group DM (P<0.05).Compared with group DM-NCVD,the incidence of intraoperative cardiovascular events and plasma cTnI concen trations at each time point were significantly increased,and the plasma concentrations of SP and CGRP were decreased in group DM-CVD (P<0.05).Conclusion The development of intraoperative cardiovascular events is related to the decrease in plasma concentrations of SP and CGRP in diabetic patients.
5.The culture and identification of astrocytes of human optic nerve in vitro

Objective To culture astrocytes of human optic nerve and establish the cell lines for further study of healing process after optic nerve trauma. [WT5”HZ]Methods [WT5”BZ]Astrocytes of infantile optic nerve were cultured by tissue inoculation or tissue digestion with 0.25% trypsin and 0.06% EDTA. The second and fourth passage cells were stained with HE and anti GFAP, S 100 protein, vimentin, and CD34 antibodies. [WT5”HZ]Results The trypsinized astrocytes of infantile optic nerve reached confluence in 7 days. The cultured cells were in polygonal shape with processes and the cytoplasm was abundant. These cells were positive in GFAP, S 100 protein and vimentin staining, and negative in CD34 staining. Conclusions Astrocytes of human optic nerve can be successfully cultured by trypsinization rather than tissue inoculation.

This paper mainly introduces the characteristics and mathematical models of solid state fermentation and summarizes the development situation of various bioreactors, the design standards and applications.
7.Effects of pravastatin on the proliferation of vascular smooth muscle cells and cell adhesion induced by lysophosphatidylcholine

Aim This study was designed to investigate the effects of pravastatin,a potent 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitor,on vascular smooth muscle cells (VSMCs) proliferation and leukocyte-endothelial cell adhesion induced by lysophosphatidylcholine (LPC).Methods Cultured VSMCs from rabbit thoracic aorta were incubated with various concentrations of LPC in the absence or presence of different concentrations of pravastatin. MTT was used to evaluate the proliferation of VSMCs. We determined the effects of LPC and pravastatin on neutrophil K562 adhesion to endothelial cells ECV304 by directly counting non-adhesive K562 cells.Results Incubation of VSMCs with LPC (1~10 ?mol?L -1) stimulated proliferation of VSMCs in a time- and dose-dependent manner,while pravastatin (0.3~1 mmol?L -1) treatment prevented the proliferation of VSMCs caused by LPC. Moreover, incubation of ECV304 with 3 ?mol ? L -1 LPC for 12 h significantly enhanced K562 cells adhesion to endothelial cells, whereas pretreatment with pravastatin reduced leukocyte-endothelial cell adhesion. Conclusion These results suggest that pravastatin can antagonize the effects of VSMCs proliferation and leukocyte-endothelial cell adhesion induced by LPC.
8.Research Progress of the Modern Evaluation Methods of De Qi Sensation in Acupuncture and Moxibustion

Objective To introduce the research progress of the modern methods in evaluating de qi sensation in acupuncture and moxibustion, and to analyze the current research situation and major problems. Method The objective evaluation methods majorly used to estimate qi sensation were summarized. Result The currently used evaluation methods basically include scales and cerebral function imaging, while the rest methods are still in the beginning stage. Conclusion As a subjective feeling, de qi sensation is difficult to determine and quantify objectively, and there lacks an objective standard. The study on the mechanism of de qi sensation is relatively insufficient and superficial.
9.Effect of human papilloma virus16 E6 shRNA mediated by lentivirus on infection ratio and invasive ability of cervical cancer cell

Objective To investigate the influence of lentivirus mediated short hairpin RNA(shRNA)target to human papilloma virus(HPV)16 E6 on invasive ability of cervical cancer Caski cells.Methods Lentivirus was produced after shRNA target to human papilloma virus(HPV)16 E6 and to nonsense was cloned to lentivirus work vector.Infection ratio was assessed by assay of EGFP positive cells of Caski.Total mRNA of E6 was determined by RT-PCR after Caski cells were infected by lentivirus.The change of E6 protein expression was analyzed by Western blot.The invasive ability of Caski cells was assayed employing Transwell.Results The optimal MOI (Multiplicity of infection)of lentivirus to Caski was 2.5.Total mRNA and protein of E6 were decreased (by 70%and 63%)in interfering group compared with control group.The invasive ability of Caski cells also reduced after infected by lentivirus.Conclusion shRNA mediated by lentivirus can inhibit expression of HPV16 E6 and invasive ability of cervical cancer cells.
10.Effects of Low-Molecular-Weight Heparin on Respiratory Syncytial Virus Nephropathy in Rats

0.05).3.Under light microscope, there was no change of the glomeruli detected in all of the LMWH-treated groups compared with the controls,while congestion and swel-ling in part glomeruli of group RSV were observed significantly. 4.Under electron microscope,glomerular structures of the LMWH-treated groups were almost normal compared with the control, while extensive foot process effacement was observed in group RSV under an electron microscope. 5.RSV RNA signal expressed weaker in the LMWH-treated groups than in group RSV.Conclusions Positive charged RSV destroys the glome-rular filtration barrier through its electrostatic interaction with the negative charged heparan sulfate(HS) of GBM. LMWH, as the analog of HS, charged with anion, competes with GBM HS to combine with RSV to keep the glomeruli from being infected and destroyed, and then reduce the proteinuria.
