Objective To observe the effect of healthy education on BMD and the risk of bone fracture in pa- tients with primary osteoporosis.Methods 628 patients with primary osteoporosis were randomly divided into two groups under the same conditions,and healthy education were conducted in treated group but were not conducted in control group.Two years later,BMD and the rates of bone fractures were collected and analysed.Results Compared with control groups,the bone loss was significantly lower at the feroral neck,Ward's triangle and the great trochanter,and the risk of bone fractures was remarkably reduced in treated group(P
2.Study on Mechanism and Regulation of CD3AK Cytotoxic Activity

Human CD3AK cells were prepared from peripheral blood mononuclear cells by culturing with recombinant IL-2 and antiCD3AK McAb. The mechanism and regulation of CD3AK cytotoxic activity with cytokines (rhIFN-?, rhIFN-?, TNF) and chernotherapeutic agents (CDDP or ADM) were observed by LDH-release assay, ABC-CELISA and the flow cytometric assay. The results showed: (1) Adhesion molecules ICAM-l/LFA-1 participated in CD3AK-mediated killing of tumor cells, hrlFN-? and TNF enhanced cytotoxicity of CD3AK through this pathway. (2) CD3AK could indirectly kill tumor cells by releasing soluable cytotoxic factors. (3) The membrane-associated TNF may be involved in CD3AK-mediated cytotoxicity. (4) CD3AK cells could induce the apoptosis of tumor cells. (5) Pretreatment of tumor cells with CDDP or ADM resulted in the increased vulnerability of tumor cells to CD3AK-mediated killing, the enhancement of CD3AK-mediated cytotoxicity by CDDP was relative to the increased expression of ICAM-1, HLA-ABC on tumor cell membrane.
3.The Evaluation of Organ Damage in Systemic Lupus Erythematosus

Objective To study the organ damage in patients with systemic lupus erythematosus (SLE) and its significance in prognosis. Methods The SLE damage index (SDI) was used to evaluate 121 female and 15 male patients. Results Among these patients, the lowest and the highest SDI was 0 or 9, separately (average: 1.79 ? 2.22). Most damaged organ was renal (39.0%), followed by muscle/skeletal system (19.9%) and cardiovascular system (18.4%). There was statistically significant difference of SDI in different groups of prognosis, life condition or disease duration ( 10 years). However, there was no significant difference of SDI in different sex groups. The correlation analysis revealed that the age at onset or diagnosis was not related to SDI. Conclusion SDI is effective in evaluating the organ damage in predicting prognosis of SLE patients.
4.Predictive value of MRA, DWI and ABCD2 score for secondary cerebral infarction after transient ischemic attack

Objective To evaluate the association of magnetic resonance angiography (MRA),diffusion-weighted imaging (DWI) and the ABCD2 score assessments with secondary cerebral infarction after transient ischemic attack (TIA).Methods Intracranial vascular MRA,cranial DWI and ABCD2 score were retrospectively analyzed in 162 cases with TIA.The impact of TIA on survival time was assessed using the univariate Kaplan-Meier curve by Log-rank test.Hazard ratio (HR) and 95 % confidence interval (CI) of secondary cerebral infarction after TIA predicted by MRA,DWI and ABCD2 score were analyzed by Cox multivariable regression.Results Among the 162 patients with first attack of TIA,86 cases (53.1 %) developed cerebral infarction within 90 d,of which 22 cases (13.6%) developed secondary cerebral infarction within 0 7 d,27 cases (16.7%) within 8~30d and 37 cases (22.8%) within 31-90 d.Single factor analysis by Kaplan-Meier curve showed that moderate to severe intracranial vascular stenosis diagnosed by MRA,positive DWI and moderate to high ABCD2 score were obviously related to cerebral infarction after first attack of TIA (all P<0.001 or 0.01).Cox multifactor risk model indicated that age ≥70 y,moderate to severe intracranial vascular stenosis,positive DWI,moderate to high ABCD2 score were the risk factors for secondary cerebral infarction within 90 d after TIA (HR=1.782,2.245,1.964,1.204,95%CI:1.171-2.256,1.627 3.097,1.273-3.031,1.050-1.381,respectively,P<0.05,0.01 or 0.001).Conclusions Intracranial artery stenosis examination may be more valuable than DWI and ABCD2 score in evaluating the outcome of TIA.
5.Effects ofYixintai Granules on Expression of C-Myc mRNA and Its Protein in Myocardial Tissues of Rabbits with Chronic Heart Failure

ObjectiveTo explore the effects ofYixintaiGranules on expression of C-Myc mRNA and its protein in myocardial tissues of rabbits with chronic heart failure (CHF).Methods CHF rabbit models were established by ear marginal vein injection of adriamycin. Successfully modeled rabbits were divided into the model group, the Losartan Potassium group, the high-, medium-, and low doseYixintaiGranules groups. Besides, a normal control group was set up. Administration groups were given relevant medicine for gavage, equal volume of physiological saline was administered to rabbits of the model group and the normal control group by gastrogavage. The intervention lasted for 4 weeks, once per day. Echocardiographic indexes and mRNA and protein expression levels of C-Myc in myocardial tissue were detected after 4 weeks of medication.Results Compared with the normal group, the LVEF, LVFS, and E/A of the model group decreased significantly (P<0.01), but mRNA and protein expression levels of C-Myc in myocardial tissues increased significantly (P<0.01). Compared with the model group, the LVEF, LVFS, and E/A of YG groups and Losartan Potassium group increased significantly (P<0.01), but mRNA and protein expression levels of C-Myc in myocardial tissues decreased significantly (P<0.05,P<0.01).ConclusionYixintaiGranules can effectively inhibit the expression of mRNA and protein expression of C-Myc, and improve cardiac function.
6.Effect of Yixintai granule on myocardial remodeling in rabbits with chronic heart failure

Objective To explore the effect of Yixintai granule on myocardial remodeling in rabbits with chronic heart failure ( CHF) .Methods The rabbit model of CHF with adriamycin was established. These successful CHF models of rabbits were divided into model group, high,middle, and low dose of Yix-intai groups, losartan potassium group, and normal control group.Echocardiographic indexes and myocardi-al remodeling indexes were measured after 4 weeks of medication.Results Compared to the model group, the left ventricular posterior wall (LVPW), left ventricular internal diameter at end-systole (LVIDs), left ventricular internal diameter at end-diastole ( LVIDd) , cardiac index ( CI) , and left ventricular mass index (LVWI) of the treatment groups were decreased ( P <0.05 or P <0.01), and the interventricular septum ( IVS) of the middle and high Yixintai groups was decreased significantly ( P <0.01 ) .Compared to the low dose of Yixintai group, the IVS, LVPW, CI, and LVWI of the middle and high Yixintai groups were decreased ( P <0.05 or P <0.01) , and the LVIDs and LVIDd of the high Yixintai group were decreased ( P <0.05 ) .Conclusions Yixintai granule can improve myocardial remodeling in rabbits with CHF. Meanwhile, the curative effect of medial and high groups of Yixintai is better than the low dose group.
7.Computational Identification of microRNAs and Their Targets

The discovery of microRNAs (miRNAs) has introduced a new paradigm into gene regulatory systems. Since inception, computational methods have been an invaluable tool complementing experimental approaches, and many discoveries have been obtained through combination of experimental and computational approaches. The knowledge that has been accumulated about the principles of miRNAs and target recognition were reviewed. The currently available computational methodologies and software for prediction of miRNA and their target genes also have been discussed.
8.Dynamic contrast-enhanced MRI for the detection of testicular ischemia in experimental induced testicular torsion of rabbits

Objective To evaluate the dynamic contrast-enhanced MRI (DCE-MRI)in detecting experimentally induced testicular ischemia. Methods Thirty healthy male New Zealand rabbits were randomly assigned into 6 groups. There were 5 rabbits in each of the following experimental groups: ( 1 ) Normal control, (2) Sham-operated, (3) ischemia of 3 h group, (4) ischemia of 6 h group, (5) ischemia of 12 h group, (6) ischemia of 24 h group. In all experiment groups, the right testis served as the internal control while the left testis served as the experimental side. DCE-MRI for each animal lasts about 10 minutes. Signal enhanced ratios (SERs) of ROI for both sides of each group were calculated by a computer, and parameters of SERs of 30 s, 75 s, 120 s and maximal SER were used for statistical analysis.Time intensity curves (TICs) were made for two sides of each group via Excel 2003 software and classified into 4 types. Statistical analysis was performed to compare the differences of SERs between left and right testis by two independent Kolmogorov-Smirnov test. Results In group I and 2, significant enhancement was observed on both testes of 10 rabbits. The enhancement decreased gradually with the elongation of ischemia in torsion groups. Three cases of type Ⅰ and 2 cases of type Ⅱ were observed in group 1,5 cases of type Ⅰ in group two, 2 cases of type Ⅰ and 3 cases of type Ⅱ b in group three, 2 cases of type Ⅰ and 2 cases of type Ⅱ b in group four, 5 cases of type Ⅱ b in group five and 5 cases of type Ⅲ in group six were noticed in the left testes. And in TICs of right testes, all cases showed TICs of type Ⅰ except 2 cases of type Ⅱ a in group six. In four torsion groups, the values for SER75 of the left side were 0. 084%, 0. 076%, 0.164% and 0.065%, while the right side were 0.255%, 0.410%, 0.586% and 0.302% (P <0.05). The values for SER120 in group three, five and six were 0.221% , 0.158% and 0.059% for the left side, and 0.405%,0.522% and 0.207% for the right side(P <0.05). The values for MSER in group three, five and six were 0.217% ,0.164% and 0.072% for the left side, and 0.405%, 0.586% and 0.302% for the right side(P <0.05). Conclusion DCE-MRI technique may be useful in the diagnosis of testicular torsion, which shows potential in the clinical application.
9.Silencing of MGMT with small interference RNA reversed resistance in human BCUN-resistant glioma cell lines.

BACKGROUNDOur previous study had cloned two glioma cell lines SWOZ1 and SWOZ2 isolated from parental glioma cell line SWO38. The 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) resistance of SWOZ1 was higher than that of SWOZ2. Since O6-methylguanine-DNA methyltransferase (MGMT) was thought to be closely related to BCNU resistance in glioma, this study aimed to explore the function of MGMT in glioma resistant to BCNU.
METHODSA BCNU resistant glioma cell line SWOZ2-BCNU was established. The expression of MGMT was detected in SWOZ1, SWOZ2 and SWOZ2-BCNU. Small interferencing RNA targeting MGMT was used to silence the expression of MGMT in resistant cell lines SWOZ1 and SWOZ2-BCNU. The cytotoxicity of BCNU to these cells was measured using the cell counting kit-8 assay. Statistical analysis was carried out by one-way analysis of variance in statistical package SPSS 13.0.
RESULTSThe resistance of SWOZ1 and SWOZ2-BCNU against BCNU was 4.9-fold and 5.3-fold higher than that of SWOZ2. The results of quantitative RT-PCR and Western blotting confirmed that MGMT was both significantly increased in SWOZ1 and SWOZ2-BCNU compared to SOWZ2. After transfection with small interferencing RNA targeting MGMT, a decreased level of MGMT mRNA expression in SWOZ1 and SWOZ2-BCNU for more than 75% compared to negative control was found and confirmed by Western blotting. As a result, the resistance against BCNU was reversed for about 50% both in the BCNU-resistant cell lines SWOZ1 and SWOZ2-BCNU.
CONCLUSIONSSilencing MGMT with specific small interferencing RNA can reverse the BCNU resistant phenotype in these glioma cell lines. MGMT may play an important role both in intrinsic and acquired BCNU-resistance in glioma.

10.Reduction and fixation of fracture of pubic rami by inserting construction plate through the punctiform-incision approach.

OBJECTIVETo observe the effect of pubic fractures reducted and fixed thorough the punctiform incision approach.
METHODSFrom 2002 to 2005, 10 cases with 18 fractures of pubis rami (8 male and 2 female) were treated with inserting construction plate by the punctiform incision approach. The average age of these patients was 37.2 years (range, 24 to 56 years). The mean duration between injury and operation was 8.7 days (range, 4 to 14 days).
RESULTSInternal fixation for eighteen pubis fractures were accomplished by 28 punctiform incisions. The blood loss for each incision was averagely 30 ml, operation time of each pubic was about 45 minutes. Function restoration was evaluated by Majeed' score and all patients gained excellent result.
CONCLUSIONThe fracture of pubic rami can be fixed sucessfully by punctiform incision approach. It provides smaller incision, less postoperative complications and excellent function rehabilitation.
