Chinese Journal of Radiology 2010;44(1):65-69
Objective The study was to evaluate DWI for quantifying liver fibrosis. Methods A total of 12 volunteers, 47 patients who had chronic HBV or HCV hepatitis and underwent liver biopsy [Scheuer score for fibrosis(S) and inflammation(G)] were enrolled in this study. They were scanned using a 1.5 T MR unit with b value of 0,250,500,750, 1000 s/mm~2. ADCs at b_(250-1000) and b_(500-1000) were the average ADCs of b=250, 500, 750, 1000 s/mm~2 and b=500, 750, 1000 s/mm~2. The studied the correlation between Scbeuer scores and ADC values, and conducted Mann-Whitney U test and Logistic regression to evaluate ADC for prediction of fibrosis scores. Results The average ADCs were (1.41± 0.11),(1.37±0.09), (1.27±0.05), (1.26±0.04), (1.22±0.06) mm~2/s respectively from SO to S4, stage at b=750 s/mm~2 (F=18.31, P<0.01). With the increase of fibrosis score, the average ADC decreased gradually, the two were better negatively correlated at b_(250-1000)(r=-0.727, P<0.01) than other b values. Using b_(750) and the two combined b values, the found significantly lower ADCs in S2 or greater versus S1 or less and in S3 or greater versus S2 or less fibrosis (P<0.01). The best predictor for S2 or greater was b_(750) with the largest AUC of 0.909, sensitivity of 85.7%, and specificity of 100.0% (ADC ≤1.35×10~(-3) mm~2/s). The best predictor for S3 or greater was b_(250-1000) with the largest AUC of 0.864, sensitivity of 69.6%, and specificity of 95.8% (ADC≤1.53×10~(-3) mm~2/s). Conclusion DWI can be a good predictor for scoring liver fibrosis for S2 or S3 stage above, while b_(750) and the combined b values are suitable for evaluation.

Chinese Journal of Radiation Oncology 2016;25(4):407-411
Objective To investigate the significance of modified gold nanocages ( GNCs ) in photothermal therapy and radiosensitivity enhancement for murine mammary carcinoma 4T1 cells. Methods A molecule probe, CD44-PEG-GNCs, was constructed on the basis of GNCs.Inductively coupled plasma mass spectrometry ( ICP-MS) and transmission electron microscopy ( TEM) were used to analyze Au uptake by 4T1 cells. The cell counting kit-8( CCK-8) assay was used to determine the impact of the probe on the viability of 4T1 cells. The CCK-8 assay and the Hoechst/PI double staining were used to evaluate the photothermal killing effects of near infrared laser (NIR).The colony forming assay and the Hoechst/PI double staining were used to measure the viability of 4T1 cells after irradiation with 6 MV of X-ray.Results ICP-MS and TEM showed that 4T1 cells had an uptake of a large number of GNCs, in which the number of CD44-PEG-GNCs taken up by 4T1 was 3-4 times larger than that of PEG-GNCs.The cell toxicity assay revealed that in a certain range of concentration, GNCs showed little effect on the viability of 4T1 cells, which was reduced to 81.2%in 48 hours. The CCK-8 assay, the Hoechst/PI double staining, and the colony forming assay showed that after NIR irradiation and radiotherapy, large-scale apoptosis was induced in CD44-PEG-GNCs-contained tumor cells by photothermal therapy combined with radiotherapy, which had substantially better outcomes than other groups. Conclusions The GNC-based molecular probe actively targets 4T1 cells with little cytotoxicity, indicating that it enhances the sensitivity of tumor towards photothermal therapy and radiotherapy.

Objective To observe changes of the structure and function of microvessels during the formation and development of the rat cerebral glioma with steady-state susceptibility contrast-enhanced MR imaging. Methods A total of 30 bearing-tumor rats were divided into 3 groups (1-week group, 2-week group and 3-week group) and underwent MR examination. Blood volime (BV) and vessel size index (VSI_(MRI)) of peripheral tumor areas, central tumor areas and normal cerebral tissue in the opposite side were measured, and compared with histologic findings. Results With time of bearing-tumor increasing, BV of central tumor area present an up-and-down curve-like change, which related to the host blood vessel regression in tumor central area, the decreasing of blood vessel density and histopathologically obvious necrosis. BV of peripheral tumor areas increased because of active blood angiogenesis in this area, while VSI_(MRI) increased gradually, especially in the central tumor area. There was obviously correlation between the VSI_(MRI) and VSI _(histo) (P<0.01). Conclusion BV and VSI_(MRI) can exactly reflect the morphology and functional information of microcirculation of tumor during the growth and development of gliomas.

Chinese Journal of Medical Imaging Technology 2010;26(3):425-428
Objective To explore the of value steady-state susceptibility contrast-enhanced MRI (SSCE-MRI) in assessment of anti-angiogenesis therapy in rat C6 glioma. Methods Seventeen rats with the bearing-tumor were randomly divided into the therapy group (n=8) and the contrast group (n=9). Human-recombination endostatin (10 mg/kg) was injected to animals in therapy group, and saline of the same dose was injected to the contrast group for 7 days. Blood volume (BV) and vessel size index (VSI) were measured of tumors in peripheral, central and normal areas with MR pre- and post-therapy. Results After therapy of endostatin, the tumor volume was obviously different between therapy group and contrast group (t=5.26, P<0.05). BV and VSI_(MRI) of tumor decreased in both peripheral and central areas, particularly in the peripheral area (t=4.14, 3.66, all P<0.01). Conclusion Assessment of therapeutic effect of anti-angiogenesis in C6 glioma is feasible with SSCE-MRI, and it can reflect the changes of BV and VSI_(MRI) pre- and post-therapy.
