Objective To investigate the clinical significance in diagnosis and prognostic judgments of acute myelocytic leukemia with combined detection of morphology ,peripheral blood and immune typing of bone marrow cells .Methods Microscopic examina‐tion of bone marrow cell morphology ,automatic blood cell analyzer detection of peripheral blood parameter and flow cytometry anal‐ysis of immune markers of leukemia cells ,all these datas were analyzed statistically .Results Peripheral blood cell analysis showed that instruments may indicate the presence of abnormal or naive cells in about 85% patients ,different kinds of leukemia had signifi‐cant difference in total leukocyte count (majority higher in M1 - 2 ,M5 ;majority lower in M3) ;while anemia and thrombocytopenia were observed in most patients ,but the degree was different ;M1 - 2 and M5 can not be identified by classification of leukocyte by automatic blood cell analyzer ,CD7 ,CD10 and CD2 can cross express in myeloid leukemia ,which have a prompt effect with treatment and prognosis .Conclusion Detection of peripheral blood parameters has an important role in early screening ,differential diagnosis and prognosis judgment of leukemia ,immunological markers detection is the powerful supplement and support for leukemia diagno‐sis and typing ,sensitive markers have close contact with prognosis ,which significantly improves the effect of the diagnosis and treatment of leukemia .

Objective To evaluate the value of prenatal ultrasound in the interrupted aortic arch (IAA), and analyze the reasons of misdiagnosis and improve diagnostic ratio of this kind of defects. Methods Ultrasonic characteristics were analyzed in 16 fetuses with aortic arch anomalies, which were compared with ultrasound image and autopsy results of the normal fetus. The relativity of fetal IAA and abnormal chromosome was also analyzed. Results Among the 16 cases, 15 cases were confirmed by anatomy and the accuracy of ultrasound diagnosis was 94% (15/16), of which one case was serious constriction of aortic arch, one case was error type, three cases were obtained explicit type due to unsatisfactory results of prenatal ultrasound. The coincidence rate of ultrasonic type was 73% (11/15). All of the corrected diagnosed cases were detected with ventricular septal, of which 5 cases were detected with complete endocardial cushion defect, 9 cases were found extra cardiac malformations. Among the 7 karyotype check cases, 3 cases were diagnosed with Trisomy 18, 1 case Trisomy 13, while the rest 3 cases normal. The incidence rate of Trisomy 18 was 43 % (3/7). Conclusions Prenatal ultrasound shows a high accuracy in diagnosing fetal interrupted aortic arch (IAA ), but there’s a need to be improved in explicit typing. It is difficult to identify fetal interrupted aortic arch (IAA) and serious constriction of aortic arch (COA). The incidence rate of Trisomy 18 in the abnormal chromosome of fetal IAA is high.

Objective To propose a novel computer-aided segment algorithm, and retrospectively investigate its effect on radiologist's sensitivity and specificity for discriminating malignant masses from benign masses basing on ultrasound images. Methods Four hundred and sixty-six images of 100 masses obtained by conventional ultrasound were processed by a novel segment algorithm. Radiologists who were blind to the histology results were invited to analyse the original and computerized images respectively. The sensitivity and specificity were calculated by means of a binary outcome using receiver operating characteristic(ROC) analysis. Results By using the segment algorithm, the quantity of the image was obviously improved, especially the margin and calcification of the masses. The diagnostic performance of the radiologists was also improved. The sensitivity rose from 70.32% to 90.52% while the specificity was 74.31%. The area under the ROC curve increased significantly (P<0.01) from 80.8% to 90.5%. Conclusions The proposed segmentation algorithm can improve the diagnosis performance of the radiologists by meliorating the quality of the ultrasound image.

Objective To investigate the expression and clinical significance of intergrin linked kinase (ILK) in pancreatic carcinoma.Methods ILK protein was detected by immunohistochemistry and Western blotting in 60 cases of pancreatic carcinoma and 32 cases of normal pancreatic tissue,and the relationship with the clinicopathological characteristics were analyzed.Results Immunohistochemistry showed ILK was expressed in cytoplasm and membrane of pancreatic carcinoma cells and the positive rate was 65% (39/60),which was significantly higher than 18.75% (6/32) of normal pancreatic tissue(P ＜0.05 ).Western blotting showed the expression of ILK in pancreatic carcinoma tissue was 303933±195116,which was significantly higher than 144613±30074 of normal pancreatic tissue(P＜0.05 ).In pancreatic carcinoma,the expression of ILK was correlated with clinical stage and lymph metastasis(P＜0.05 ),but not correlated with tumor cell differentiation(P＞0.05 ).Conclusions ILK protein was highly expressed in pancreatic carcinoma tissue and it was correlated with the degree of malignancy.

OBJECTIVETo observe the effect of Chinese herbal extract HuNan A-1 (HNA-1) on the thymic output function in Simian immunodeficiency virus (SIV) chronically infected rhesus macaques.

METHODSEight Chinese rhesus macaques had been infected by SIVmac239 for 16 to 21 months, and then they were randomly divided into the treatment group and the control group, 4 in each group. Monkeys in the treatment group were administered with HNA-1 by gastrogavage, once daily for 2 successive months, while those in the control group were administered with equal volume of normal saline by gastrogavage, once daily for 2 successive months. The general condition and body weight of monkeys were observed. Plasma viral loads were detected using real-time fluorescent quantitative PCR assay. CD4 percentages and counts, as well as naive CD subsets were detected using flow cytometry. T-cell receptor excision circles (TREC) were detected using real-time fluorescent quantitative PCR assay. The thymus tissue was pathologically observed using routine HE staining. The correlation between lesions of the thymus tissue, CD4 counts, naive CD counts, and TREC were analyzed.

RESULTSThere was no statistical difference in body weight, viral loads, absolute CD ratios between the two groups after treatment (P > 0.05). The altered TREC multiple showed an obvious decreasing tendency in the control group, while it showed an increasing tendency in the treatment group (P < 0.05). In both groups, destroyed structures of the thymus tissue could be seen, filled with pink unstructured material. Increased connective tissues, lowered connective cell density, and confused arrangement could also be seen in the two groups, with no obvious difference. TREC contents were positively correlated with naive CD4 counts after removing extremum (r = 0.926, P = 0.001). Naive CD4 counts were positively correlated with CD4 counts (r = 0.961, P = 0.005).

CONCLUSIONSTREC content determination, as a marker of newly thymic emigrants, could be taken as a testing method for evaluating the thymic output function. Besides, HNA-1 treatment increased the thymic output significantly in SIV chronically infected monkeys. Correlation existed among TREC contents, naive CD4 counts, and pathologies of thymus tissues, especially in late infection stage.

The contents of two lignans, namely 4'-demethylpodophyllotoxin and podophyllotoxin in cultivated and wild Sinopodophyllum hexandrum plants were extracted by ultrasonicaction and determined by HPLC. According to the result showed, the order of parts of cultivated plants containing 4'-demethylpodophyllotoxin from high to low is as follows: stem > root, no 4'-demethypodophyllotoxin was detected in leaves of cultivated plants; The order of parts of wild plants 4'-demethylpodophyllotoxin from high to low is as follows: lateral root > petiole > rhizome > leaf, no 4'-demethypodophyllotoxin was detected in fruit. The order of parts of cultivated and wild S. hexandrum containing podophyllotoxin from high to low is as follows: root > petiole > leaf ( > fruit). Both of the lignan contents in different parts of cultivated plant varied in a " W" curve with the changes in seasons, with the highest content in July.

Objective To explore the application of clustering analysis in the study for iodine deficiency disorders(IDD)surveillance.Methods System cluster method of average linkage was applied to study the data obtained from national IDD surveilance in 2005 and a clustering analysis' tree chart was drawn.Such data as the rates of consuming qualified iodine salt,coverage rate of iodine salt,median of iodine salt,median urinary iodine,goiter rate detemined by palpation and B-uhrasonography,Child intelligence quotient were investigated.Results Thirty-one provinces or autonomous regions including the Xinjiang Production and Construction Coms had beed assorted into three types.Tibet was claasified into the type Ⅰ;Guangdong,Qinghai,Hainan,Sinkiang were assorted into the type Ⅱ,other areas were assorted into the typeⅢ.The rates of consuming qualified iodine salt,coverage rate of iodine salt and median urinary iodine in the type Ⅰ area were 21.00%,33.20%and 96.70 μg/L,while they were 71.95%,78.50%and 145.40μg/L,respectively,all being lower than that of type Ⅲ(93.20%.97.70%and 245.20μg/L)with statistical significance(P＜0.05).11.50%of the children in the type Ⅰ area were detected by palpation to have goiter;the child intelligence quotient in three types of areas,was 77.00,95.95±4.46 and 104.59±6.77,the differences being statistical significant between any two types(P＜0.05).Conclusion Multiple areas of IDD could be assorted by clustering analysis into distinct types with different epidemic value.More attentions of the government must be paid to the type Ⅱ areas,the prevention and cure work must be enhanced in the second-class areas while it is carried on in the typeⅢareas.The results are helpful for decision making for epidemic supervisors in the government.

Objective: To investigate the role of human umbilical vein endothelial cells (HUVEC) in supporting hematopoiesis and the expansion of hematopoietic stem/progenitor cells in vitro. Methods: According to the fact that HUVEC supernatant has colony stimulating activity shown by methylcellulose colony-forming assay and HUVEC can maintain the survival of mononuclear cells for at least four weeks in vitro, CD34+ cells from umbilical cord blood were seeded with (HUVEC group) or without (control group) HUVEC monolayer. Every week cells were collected and counted, the frequency of CFU-GM was measured by using methylcellulose colony-forming assay, and the percentage of CD34+ and CD41a+ cells was measured by flow cytometry. Results: In control group，all the CD34+ cells died in two weeks. However, in HUVEC group,most nucleated cells and CD34+ cells were expanded by 68.1±14.8 fold and 6.6±1.4 fold，respectively at the third week while CFU-GM expansion reached its peak (5.7±2.1 fold) at the week 2. Moreover, the percentage of CD41a+ cells was enhanced significantly, reaching a maximum (15.6%) at the week 3. Conclusions:HUVEC can support hematopoiesis in vitro and expand the hematopoietic progenitor cells and CD41a+ cells in direct contact coculture.

Objective To review the efficacy and safety of combination of iodine-125 brachytherapy and concurrent chemoradiotherapy for middle and advanced stage non-small cell lung cancer (NSCLC).Methods Eighty-two cases of peripheral NSCLC patients were divided into the experimental group (n =32)and the control group (n =50) by different treatment methods.The patients in experimental group were treated by iodine-125 brachytherapy combined with concurrent chemoradiotherapy,and the patients in control group were treated by concurrent chemoradiotherapy.The two groups were treated with helical tomotherapy,and the chemotherapy regimen was PC regimen (paclitaxel + cisplatin) or TP regimen (taxotere + cisplatin).The response rate (RR),progression free survival (PFS),overall survival (OS) and adverse reactions were observed.Results The RR of experimental group and control group was 87.5％ and 76.0％,and the difference was not statistically significant (x2 =0.992,P =0.319).The median PFS was 16 months and 11 months,and the difference was statistically significant (x2 =5.216,P =0.022).The median OS was 26 months and 19 months,and the difference was not statistically significant (x2 =1.085,P =0.298).There were no statistically significant differences of the occurrence rates of adverse reactions in the experimental group and the control group such as radioactive pneumonia (18.75％ vs.24.00％,x2 =0.314,P =0.575),radiation esophagitis (25.00％ vs.30.00％,x2 =0.242,P =0.623),digestive tract reaction (21.88％ vs.26.00％,x2=0.180,P =0.671),bone marrow suppression (12.50％ vs.16.00％,x2 =0.014,P =0.907) and hemopneumothorax (6.25％ vs.0,P =0.149).Conclusion The combination of iodine-125 brachytherapy with concurrent chemoradiotherapy is a safe and effective treatment for middle and advanced stage NSCLC,which is worthy of further research and promotion.
