Through genetic recombination technique, the rat glial cell line-derived neurotrophic factor (rGDNF) cDNA was in-serted into polylinker site of retroviral vector pLXSN, to generate a recombinant plasmid pLXSN-GDNF as transfer vector. Therecombinant plasmid was verified with restriction analysis, PCR, dot blot hybridization and Southern blot hybridization. The re-sults showed that GDNF cDNA was cloned correctly into retroviral vector pLXSN, recombinant retroviral vector was construct-ed. It is concluded that the eukaryotic cell expression vector was constructed successfully for gene therapy of Parkinson's,Alzheimer's and other central nervous system diseases.

Objective To summarize theskill practice as the coremethods and training effect in tumor specialist nurse training , and to explore the suitable effective methods and pattern of China′s national conditions of specialist nurse training in China, for providing a reference for conducting cancer specialist nurse training. Methods Reviewed and analyzed tumor specialist nurse training method to make clear the training effect from 2012 to 2013, 100 students was investigated by using questionnaires , which contain 15 aspects, before and after the training respectively. designskill practice as the coreof the training content (160 hours) and theoretical classes (80 hours), established training evaluation criteria and the organization management system. Results All 100 students had grasped maintenance technology, observation of the complications of treatment, infusion port of the maintenance, the use of chemotherapy drugs, health education for patients with PICC catheter, and the use of effective order, the grasp rate was 100%. Complications rate was decreased significantly after using PICC catheter, (the rate of catheter related blood stream infections and the rate of thrombosis was reduced from the top 35%and 12%before training to 4%and 1%after training respectively (P<0.05). Conclusions In order to train more practical clinical nurse specialists, reduce the complications, raise the overall level of nursing profession, and improve the quality of nursing after using PICC catheter. Skill practice as the core methods in tumor specialist nurse training should be advocated.

OBJECTIVE To realize the disinfectant effect of medical instrument by acidic electrolyzed oxidizing water(EOW).METHODS Two groups of subjects participated in this study,and chlorinated disinfectant is used in the control group. Adopting stability test to measure effective chlorine and pH,and to compute the bacterium elimination rate after disinfection. Contrast the result with the experimental group.RESULTS The pH mean value and the effective chlorine mean value of two groups before and after instrument immersing in had no remarkable differences. Medical instrument sterilized by EOW for 5 minutes,the bacterium eliminative rate was 100.0%,while sterilized by the chlorinated disinfectant needed 30 minutes. Sterilizing effects of two groups were the same,but sterilizing time was obviously short in experimental group.CONCLUSIONS The acidic electrolyzed oxidizing water can be used in medical instrument's disinfection. The effect is good,and the process is fast. EOW is better than chlorinated disinfectant in saving of resources economizing.

Objective] From the analysis of Zhu Danxi's life with its clinical diagnosis treatment characteristics, in order to learn, inherit Zhu's academic thought essence and clinical characteristics of diagnosis and treatment. Objective To determine the serum level of KL-6 in patients with polymyositis(PM)and dermatomyositis(DM),and to investigate the possible diagnostic value for interstitial lung diseases(ILD)in pa- tients with PM/DM.Methods Serum KL-6 concentrations were measured by ELISA in 53 adult PM/DM pa- tients,the control groups of 22 patients with infectious lung disease,and 50 healthy subjects.The association with clinical features and serum KL-6 levels was analyzed.Results The serum levels of KL-6 were(1543?761)、(429?106)、(336?196)and(289?105)U/ml in PM/DM patients with ILD and without ILD,patients with infectious lung disease and healthy controls,respectively.Serum KL-6 levels in the PM/DM patients with ILD were significantly higher compared with PM/DM patients without ILD,patients with infectious lung disease and healthy controls(all P＜0.01 ).However,no significant differences of serum KL-6 levels was found among PM/ DM patients without ILD,patients with infectious lung disease and healthy controls(P＞0.05).Significant cor- relations were found between the elevated levels of serum KL-6 and the presence of ILD in patients with PM/ DM(P＜0.01).The sensitivity and specificity of serum abnormal KL-6 levels for ILD in patients with PM/DM were 90.9% and 80.6%.Additionally,follow-up study showed the mean serum levels of KL-6 in six patients who died were significantly higher than that in other PM/DM patients with ILD(P＜0.01).Conclusion Serum KL-6 level is a reliable serum marker for ILD in idiopathic inflammatory myopathies,which may contribute to early differentiate ILD from lung infectious disease.Increased serum level of KL-6 may predict a poor out- come.

ObjectiveTo explore the changes of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), malondialdehyde (MDA) in the brain of mice after irradiated by electromagnetic radiation (EMR) and therapeutic effect of curcumin.Methods40 mice were divided randomly into the bare control group, simply EMR group, EMR+curcumin low dose, middle dose and high dose groups total 5 groups with 8 animals in each group. The mice except the bare control group received EMR irradiation and those in the EMR+curcumin groups were given various doses of curcumin at the same time. Five days later, EMR irradiation and medication stopped, and the levels of SOD, GSH-Px and MDA in the brain of mice were tested.ResultsCompared with the bare control group, the activities of GSH-Px and SOD, and level of MDA all increased in the mice irradiated by EMR ( P<0.05). Compared with the simply EMR group, the activities of GSH-Px and SOD, and level of MDA all decreased in the mice of EMR +curcumin groups ( P<0.05).ConclusionEMR irradiation can induce changes of GSH-Px and SOD and peroxidation of mice brain, curcumin can lighten these damages by its anti-oxidation with a dose-dependent effect.

Objective To investigate the clinical curative effect of Prepenem for pathogen from cranial neu-rosurgieal patients with lower respiratory tract infection after traeheotomy and to provide basis for treatment of such pa-tients. Methods Thirty-two cranial neurosurgical patients with lower respiratory tract infection after tracheotomy were enrolled in the study with original empiric therapy with Prepenem. And the pathogenic bacteria in sputum from patients with lower respiratory tract infection after traeheotomy were stricdy identified and analyzed by automatic mi-croorganism analyzers. The clinical efficacy of Prepenem was identified after treatment. Results The straim of bacte-rial species(48) and funni(5) from 32 specimens were isolated. 15 strains were infected by one kind of bacteria and 38 strains were infected by two kinds of bacteria. Among the pathogen, gram-negative bacilli were about 64. 1% and Klebsiela pneumoniae (18. 8%)was the most predominant, gram-positive coccobacteria were about 26. 4% and Staph-ylococcus aureus(13.2%)was the most predominant,funni were about 9.4% and C. albicans (5.7%)was the most predominant. The drug sensitivity test showed that the ratio of drug resistance of bacteria isolated from sputum was high,but gram-negative bacilli were highly sensitive to imipenem. Staphylococcus aurens was sensitive to Nitrofuran-toin and Rifampicin. Satisfied clinical curative effect was shown by the de-escalation therapy for all patients. Conclu-sions Gram-negative bacili in cranial neurosurgical patients with lower respiratory tract infection after tracheotomy are the main pathogenic bacteria from lower respiratory tract infection, and Staphylococcus aureus are the main gram-positive cocci. So the drug sensitivity test of sputum should be done more often and antibiotics must be selected ac-cording to the drug sensitivity test. In general, this kind of infection can be controlled by prepenem effectively.

BACKGROUND: The neurons in the medial septum (MS), vertical and horizontal limbs of diagonal band of Broca (vDB and hDB) in the basal forebrain contain rich androgen receptors (ARs) and estrogen receptors (ERs) by which androgen and estrogen can act dramatically on the neurons in the basal forebrain, subsequently affecting learning and memory processes.OBJECTIVE: To qualitatively and quantitatively investigate the effects of androgen replacement therapy on the nitric oxide synthase (NOS)-positive and nestin-positive neurons in the MS, vDB and hDB of castrated adult male rats.DESIGN: A randomly controlled study on experimental animals.SETTING: Department of Anatomy and Brain Research Laboratory of Zhngshan Medical College of Sun Yat-sen University.MATERIALS: The experiment was performed at Department of Anatomy and Brain Research Laboratory of Zhongshan Medical College of Sun Yatsen University from June 2001 to June 2002. Totally twenty-eight adult male Sprague-Dawley rats were randomly divided into four groups with seven rats in each group: androgen replacement therapy for 4 weeks following 24 hours of castration (ART1), androgen replacement therapy for 2 weeks following 2 weeks of castration (ART2), vehicle replacement therapy for 4weeks following 24 hours of castration (VRT), sham-operated group (Sham).INTERVENTIONS: ① ART1 group: The castrated rats received subcutaneous injection of testosterone proprionate (25 mg/kg) dissolved in 100 μL of sterile sesame oil every other day from 10:30 am to 11:00 am for 14 times (4 weeks). ② ART2 group: The castrated rats received subcutaneous injection of testosterone proprionate with the same dosage and method as ART1 group for 7 times (2 weeks). ③ The rats in VRT group received subcutaneous injection of 100μL of sterile sesame oil for 14 times (4 weeks) by the same regime as described above. ④ Rats in Sham group only received sham-operated treatments, and testes were intact and lived for 4 weeks.MAIN OUTCOME MEASURES: Morphology and counts of NOS-positive and nestin-positive neurons were observed in the MS, vDB and hDB with immunohistochemical method at various time points.RESULTS: Data of totally 28 rats were involved in the final analyses. ①Morphological features of both NOS-positive and nestin-positive neurons in the MS, vDB and hDB were not significantly changed among four groups. ② The number of NOS-positive and nestin-positive neurons in the MS and vDB of VRT group were significantly higher than those of Sham group (P＜ 0.05 or 0.01), whereas the numbers of the NOS-positive and nestin-posi-tive neurons in the MS and vDB of ART1 and ART2 groups was significantly lower than those of VRT group (P ＜ 0.05 or 0.01), which nearly reached the levels of sham group (P ＞ 0.05).CONCLUSION: Androgen replacement therapy produces no significant effects on the morphological features of NOS-positive and nestin-positive neurons, but the therapy can selectively decrease the numbers of NOS-positive and nestin-positive neurons in different subregions of the basal forebrain, which may be closely related to androgen downregulation of expressions of NOS and nestin by ARs-mediated mechanisms, thereby producing complex effects on learning and memory processes.

Objective To observe the effect of different needling retaining times in clinical curative effect of acupuncture on the treatment of periarthritis humeroscapularis.Methods A total of 60 patients, who met the inclusion criteara, were randomly divided into 2 groups according to random number table method, 30 patients in each group. Both groups were treated with the same acupuncture therapy for 20 times, but the needling retaining times were different as 20 minutes and 40 minutes. The curative effect rates, Visual Analog Scale (VAS) and Constant-Murley Score (CMS) were evaluated.Results After the treatment, the VAS scoresin 40mins group was significantly lower than that in the 20 mins group (2.67 ± 1.03vs.3.60 ± 1.48,t=-3.251, P<0.01); the CMSin 40 mins group was significantly higher than the 20 mins group (73.20 ± 10.88vs.66.47 ± 12.62,t=-2.199,P<0.05). The curative effect rates of the 40 mins group was 26.7% (8/30), which was significantly higher than 3.3% (1/30) in the 20 mins group (χ2=4.706,P=0.030). The total effective rates in the 40 mins group was 90.0% (27/30), which was significantly higher than 96.7% (29/30) in the 20 mins group (χ2=0.268,P=0.605).Conclusions Acupuncture treatment for the patients with periarthritis humeroscapularis showed that the 40 minutes of needling retaining times had better symptom improvement and restore function effects than 20 minutes, however the total effective rate was no significant difference.
