Objective To study the growth inhibitory and apoptosis inducting effect of paclitaxel on human osteoblastic cell line U-2 OS. Methods U-2 OS cells were treated with various concentrations of paclitaxel. Proliferation was determined by cell count in a Neubauer cytometer chamber. Viability was assessed by trypanblau dye exclusion. Paclitaxel induced morphologic alterations were visualized, using light and transmission electron microscopy. The extent of paclitaxel induced apoptosis was determined by flow cytometry and immunohistochemical detection (TdT mediated dUTP nick end labeling technique, TUNEL). Results Paclitaxel had a growth inhibitory and apoptosis inducting effect on U-2 OS cell. The cell treated with paclitaxel initially show G2/M arrest; follow by apoptosis. A characteristic apoptosis change including nuclear disintegration and chromatin agglomerate were displayed. Lots of multinucleate cells appeared, which was not seen on the cell treated with other chemotherapeutic agents such as cisplatin and adriamycin. Also, extensive DNA cleavage was detected by immunohistochemical technique. Conclusion Paclitaxel has an obvious growth inhibitory and apoptosis inducting effect on osteosarcoma cell line by induce a G2/M arrest and inhibit the mitosis. The effect of paclitaxel displays a time dependent and dose dependent manner.

Objective To evaluate the changes of right ventricular (RV) volume and function by using single beat real-time three dimensional (3-D) echocardiography in patients with atrial septal defect (ASD) before and after percutaneous closure. Methods During the period from July 2011 to Oct. 2013, a total of 45 patients with pure ostium secundum defect were admitted to authors’ hospital to receive percutaneous transcatheter closer. The patients were divided into ASD without pulmonary hypertension (PH) group (group A, n = 28) and ASD with PH group (group B, n = 17). By using 3-D echocardiography and right cardiac catheterization, the right ventricular end-diastolic volume (RVEDV), right ventricular end-systolic volume (RVESV), right ventricular stroke volume (RVSV), right ventricular ejection fraction (RVEF), right ventricular cardiac output (RVCO), pulmonary artery systolic pressure (PASP) and the mean pulmonary artery pressure (mPAP) were determined before and after the percutaneous transcatheter closer. The results were compared between the two groups. Results After the treatment a statistically significant reduction in RVEDV, RVESV, RVSV and RVCO were seen in all patients (P < 0.05). In group A, RVEF decreased significantly after ASD closure when compared with that determined before transcatheter closer (P < 0.05), while no significant reduction in RVEF was seen in group B (P > 0.05). Pulmonary artery pressure (PAP) decreased significantly in group B after ASD closure when compared with that obtained before Objective To evaluate the changes of right ventricular (RV) volume and function by using single beat real-time three dimensional (3-D) echocardiography in patients with atrial septal defect (ASD) before and after percutaneous closure. Pulmonary artery pressure (PAP) decreased significantly in group B after ASD closure when compared with that obtained before transcatheter closer (P < 0.05). Conclusion Single beat real-time 3-D echocardiography is a newly-developed technique. This technique can quickly and accurately assess the right ventricular volume and function. Right ventricular volume will decrease after ASD closer. In ASD patients without PH the right ventricular function will decrease after ASD closer, while in ASD patients with PH the right ventricular function shows no changes after ASD closer although their PAP will decrease.

Chinese Journal of Urology 2006;0(S1):-
Objective To investigate the feasibility of tissue-specific gene therapy for bladder cancer using human UroplakinⅡ(UPⅡ) promoter. Methods The recombinant adenoviruses,Ad-hUPⅡ-TNF carrying tumor necrosis factor (TNF-?) under control of the hUPⅡ,were generated.ELISA showed the production and secretion of TNF by bladder cancer cells infected with Ad-hUPⅡ-TNF.The level of TNF in urine was identified with ELISA. Results ELISA showed that production and secretion of TNF by bladder cancer cells infected with Ad-hUPⅡ-TNF was distinctly higher than by non-urothelium cells infected with Ad-hUPⅡ-TNF and MTT showed that proliferation of bladder cancer cells was obviously inhibited.Conditioned medium from bladder cancer cells apparently inhibited cells of L929 proliferation, compared with conditioned medium from non-bladder cells by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay.Intravesical inoculation of Ad-hUPⅡ-TNF also caused decreased tumor growth in orthotopic human bladder cancer model. The sustained high level of TNF in urine could be identified with ELISA. Conclusions TNF driven by hUPⅡ promoter has effective active in the inhibition of bladder cancer growth both in vivo and in vitro. These will undoubtedly yield a new approach of therapy for bladder cancer.

Objective:To investigate auricular lobule index difference in healthy young female of different height or weight, and to provide an additional reference for otoplasty of auricular lobule. Methods:Subjects were made up of 266 healthy young females of Henan residents. Their height, weight, physiognomic ear breadth, morphological ear breadth, physiognomic ear length, morphological ear length, auricular lobule length, auricular lobule breadth were measured and calculated. All the data were analyzed by SPSS 15.0 software. Results:We obtained the means, standard deviation of each items, and data were treated statistically. In the 266 subjects, average height was (162.9±4.1) cm, weight (55.3±6.5) kg, morphological ear breadth (4.9±0.6) cm, morphological ear length (2.9±0.3) cm, physiognomic ear breadth (6.2±0.4) cm, physiognomic ear length (3.3±0.3) cm, auricular lobule breadth (1.6±0.3) cm, auricular lobule length (1.8±0.2) cm, lobule physiognomic ear breadth index (25.6±4.9)%, lobule-physiognomic ear length index (53.69±7.69)%, and auricular lobule index (91.4±19.3)%. The differences among the indices mentioned above were of statistical significance ( P<0.05); the indices of subjects of different height were significantly different ( P<0.05); the indices of subjects of different weight were of no statistical difference. ( P>0.05). Conclusions:The subjects of different height or weight have no different morphological ear breadth, morphological ear length, physiognomic ear breadth, physiognomic ear length, auricular lobule length, lobule-physiognomic ear length index and auricular lobule index; hence, the indices are no related to body height and weight. However, the subjects of different height have different auricular lobule breadth and lobule-physiognomic ear breadth index.

Objective: To observe the clinical efficacy of warm needling moxibustion plus spine subtle adjusting manipulation for cervical radiculopathy. Methods: A total of 70 patients with cervical radiculopathy were randomized into an observation group and a control group, with 35 cases in each group. The observation group was treated with warm needling moxibustion plus spine subtle adjusting manipulation, while the control group was treated with warm needling moxibustion alone. The treatments were performed three times a week, and for four weeks in total. The visual analog scale (VAS) was scored before and after treatment. And the clinical efficacy of the two groups was compared after treatment. Results: The total effective rate was 97.1% in the observation group, versus 88.6% in the control group. The difference between the two groups was statistically significant (P<0.05). After treatment, the VAS scores in both groups significantly decreased (P<0.01), and the score in the observation group was significantly lower than that in the control group (P<0.05). Conclusion: Warm needling moxibustion plus spine subtle adjusting manipulation has a better effect in the treatment of cervical radiculopathy than warm needling moxibustion alone.
