3.Effects of extracts of Dragon's blood on fibroblast proliferation and extracellular matrix hyaluronic acid.

OBJECTIVETo investigate the effects of Dragon' s blood extract on proliferation and secret extracellular matrix function of fibroblasts in vitro.
METHODSDragon' s blood was extracted by chloroform, acetoacetic ester, alcohol. Human fibroblast were cultured in vitro in media containing gradient dilutions of Dragon' s blood extracts (0.002, 0.02, 0.2, 2, 20 mg/ml) , which was followed by cell proliferation assessed with MTT assay on 0, 12, 24, 36, 48, 60, 72 h. Under the optimal concentration, the cell growth curves were drawn and the flow cytometry (FCM) was used to determine the changes of cell cycle. On 0, 12, 24, 36, 48, 60, 72 h, the concentration of hyaluronic acid in the supernatant of fibroblast culture was measured by radioimmunoassay.
RESULTS0.2-2 mg/ml Dragon' s blood extracts enhanced the proliferation of fibroblasts in a dose-dependent manner. 2 mg/ml was the optimal dilution of Dragon's blood extract, and it increased the ratio of S cells in cell cycle [(25.80 ± 3.10)%] than control group [(7.50 ± 0.70)%, P < 0.01]. From 12 h to 72 h, in 2 mg/ml Dragon's blood group, concentration of Hyaluronic acid secreted by fibroblasts gradually increased, but were less than control (P < 0.01).
CONCLUSIONSDragon's blood acetoacetic ester extract improved the proliferation of cultured human fibroblasts in vitro, might be beneficial to promote wound healing.

4.Study on expression of caspase-3 and free radical injury in hypoxic-ischemic brain damage of neonatal rats

Objective:To study the expression of caspase-3 and free radical injury in hypoxic-ischemic brain damage ( HIBD) of neonatal rats.Methods:All seven-day-old Wistar rats were randomly divided into HIBD group and sham operation group.Brains was obtained at time of 6 h,12 h,24 h,48 h,72 h,96 h after HIBD.Neuronal cell apoptosis was detected by terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling ( TUNEL).The expression level of caspase-3 protein was detected by immunohisto-chemistry.The level of malondialdehyde ( MDA) and supemxide dismutase ( SOD) were measured by thiobarbitic acid colorimetry and xanthin oxidase,respectively.Results:The number of neuronal cell apoptosis and the expression of caspase-3 protein began to increase at 6 h and reached the peak at 48 h in HIBD group,they were both significantly higher than those in the sham operation group at each time point (P<0.05).The level of MDA began to increase at 6 h and reached the peak at 24 h in HIBD group,it was significantly higher than the sham operation group at each time point (P<0.05).The level of SOD began to decrease at 6 h and reached the lowest level at 24 h in HIBD group,it was significantly lower than the sham operation group at each time point ( P<0.05 ) .The number of neuronal cell apoptosis and the expression of caspase-3 protein were positively correlated with the level of MDA, but they were negatively correlated with the level of SOD.Conclusion: Free radical injury promotes the expression of caspase-3 and neuronal cell apoptosis in HIBD.
5.IMMUNOFLUORESCENCE ANALYSIS OF MANCHETTE DURING MOUSE SPERMIOGENESIS

Objective To investigate the localization and the morphological changes of manchette during mouse spermiogenesis.Methods Immunofluorescence staining with FITC and costaining with DAPI were used to demonstrate the cellular localization of the manchette at different stages during mouse spermiogenesis.The structural changes of the manchette were observed during the maturing of the spermatid.Results Immunofluorescence staining showed that manchette existed exactly around the nuclei of the spermatids.Manchette began to form,when the shape of the nucleus changed from spherical to slightly elongated.While the nucleus of the spermatids condensed and elongated at later stages,manchette moved gradually to the caudal position of the spermatids.At last,the manchette diminished as the spermatids became mature.During mouse spermiogenesis,manchette underwent a transition from a cap-like to a tubular configuration.ConclusionThe formation and diminishment of the manchette is in step with the condensation and elongation of the nucleus of the spermatid.Both the structural and positional changes of the manchette coincide with the changes of the nucleus.These results imply that manchette might play an important role in mouse spermiogenesis.
6.Effects of Dragon’s blood extracts on fibroblast proliferation and procollagen type III

BACKGROUND:Dragon’s blood is the main ingredient of traditional medicine prescription for promoting granulation, which has been used in clinical treatment of a variety of refractory wounds and achieved the exact effects. But the Dragon’s blood effect on colagen secretion from normal fibroblasts has not been reported. OBJECTIVE: To investigate the effects of Dragon’s blood extract on the proliferation and secret function of fibroblastsin vitro. METHODS: Dragon’s blood was extracted by extracts chloroform, acetoacetic ester, and alcohol in turn. Normal human fibroblasts were respectively cultured in Dragon’s blood extracts of chloroform, acetoacetic ester, and alcohol, DMEM containing 1% dimethyl sulfoxide, and normal culture medium. Then, the fibroblasts were cultured in vitro in different media containing gradient dilutions of Dragon’s blood extracts (0.002, 0.02, 0.2, 2, 20 g/L), which was folowed by cel proliferation determination assessed with MTT assay. Under the optimal concentration, the cel growth curves were drawn and the flow cytometry was used to determine the changes of cel cycle. The concentration of procolagen type III in the supernatant of the fibroblast culture systems was measured by radioimmunoassay. RESULTS AND CONCLUSION:0.2 g/L-2 g/L dilution of Dragon’s blood extracted by acetoacetic ester enhanced the proliferation of fibroblasts in a dose-dependent manner. The 2 g/L was the optimal dilution of Dragon’s blood extracted by acetoacetic ester, and it increased the ratio of S cels in cel cycle than control group and decreased procolagen type III. These findings indicate that Dragon’s blood acetoacetic ester extract can improve the proliferation of cultured human fibroblastsin vitro, and decrease the secretion of procolagen type III of fibroblasts, and it can be beneficial to improve wound healing and inhibit hypertrophic scar.
7.Objective Collection and Analysis on Voice Signals of 231 Patients with Three Common Traditional Chinese Medicine Syndrome Types

This study was aimed to assist the clinic diagnosis and analysis with the application of modern ausculta-tion diagnosis technology. Meanwhile, this study provided certain objective evidences for traditional Chinese medicine (TCM) auscultation diagnosis in syndrome differentiation according to Zang-Fu. The Voice Sampling System of TCM was used in the collection of voice signals of patients with liver-depression and spleen-deficiency, deficiency of both heart and spleen, disharmony between heart and kidney (231 cases). Meanwhile, voice signals of healthy peo-ple (100 cases) were used as the control group. Based on wavelet packet decomposition and reconstruction technique, four kinds of featured parameters, including energy proportion, generalized energy proportion, energy gradient and generalized energy gradient, were extracted. The results showed that there were significant differences in multiple fre-quencies on energy proportion, generalized energy proportion, energy gradient and generalized energy gradient be-tween the liver-depression and spleen-deficiency, deficiency of both heart and spleen, disharmony between heart and kidney, and healthy people (P< 0.05). There were significant differences in multiple frequencies of four features a-mong samples from each type (P < 0.05). The generalized energy proportion of the deficiency of both heart and spleen type was higher than patients from other types and healthy people. The generalized energy gradient of healthy people was lower than the other three types. Differences were mainly posterior to the middle frequency segment. The recognition rate of three types and healthy people was ideal after feature screening. It was concluded that modern auscultation diagnosis technology can be used in the analysis of TCM phonetic feature parameters among common TCM syndrome types. The research results provided references for the objective study of TCM auscultation diagnosis. It provided theoretical evidences for the objective detection of TCM auscultation diagnosis and intelligent computer.
8.Effects of Neonatal Handling and Enriched Environment Stimulation on Brain Damage in Neonatal Rats Following Hypoglycemia

Objective To explore the effects of the neonatal handling and enriched environmental stimulation on brain damage in neonatal rats with hypoglycemia.Methods Thirty-six neonatal rats were randomly divided into the normal group,hypoglycemia intervention group and the hypoglycemia non-intervention group.Those rats in hypoglycemia intervention and hypoglycemia non-intervention groups were weaned for 12 h,then the blood sugar of both groups were monitored.After neonatal rat models with hyperglycemia were prepared,the rats in hypoglycemia intervention group received the neonatal handling for 14 d and then were kept in an enriched stimulation environment for another 14 d.Rats in normal group and hypoglycemia non-intervention group were fed in the routine way.Neonatal handling was done when the rats were born for 24 h.The rat was rubbed with the brush from head to tail softly.Rats in the hypoglycemia non-intervention group was not handled.The enriched environment stimulation was used after 15 d when the rats were born.Rats in the hypoglycemia intervention group was put into the enriched environment for 1 h per day until 28 d when the rats were born,and rats in the hypoglycemia non-intervention group was put into the normal environment.Then the body weight was scaled at 0 d,7 d,14 d,21 d and 28 d when the rats were born.Space learning and memory were tested with Morris earter at their third month's age.After that,changes of pathology was observed in their occipital cortex.Results The weight increase,the ability of space learning,memory and the number of survival pyramid neurons of occipital cortex in normal group were better than those in hypoglycemia intervention and hypoglycemia non-intervention group(Pa
9.Fribrinolysis kinetics and its application.

A new, convenient, and rapid method for kinetic measurement of human fibrinolysis was established. The alteration of absorbance (A) in the process of blood coagulation and lyses was automatically scanned and recorded using a UV2000 spectrophotometer connected to a computer. The parameters of human fibrinolysis kinetics were established. Urokinase at 20 U/mL was the optimal concentration used. There was significant difference in fibrinolysis kinetics and plasma plasminogen concentration between 22 normal subjects and 27 patients with acute myeloblastic leukemia (P<0.05 and <0.01 respectively). The coefficience of variation was (5.24+/-1.51)%. This method could also be used to measure the plasma fibrinogen concentration at the same time. It was concluded that this method was stable and was capable of providing dynamic, direct experimental data and multiparemeters for clinicians. It was also valuable in evaluating the anti-and pro-fibrinolytic capcity of patients' plasmas, allowing for monitoring of therapy, choice of drugs and adjustment of drug concentrations.
10.Echocardiography diagnosis of coronary artery lesion in Kawasaki disease

The main complications of Kawasaki diseases(KD) is relative to cardiovascular,especially coronary artery.Coronary artery lesions can be showed as arteritis,artery dilatation,aneurysm,arterial stenosis and so on.As a noninvasive,convenient,fast and well reproducible inspection method,echocardiography can offer dynamically observa tion of the evolution process of coronary artery lesions in each stage,accurately measure the coronary artery diameter,evaluate heart function in children,find other cardiac complications and which has become a standard diagnosis technics.With upgrades and new technology,the echocardiography plays a more and more important role in the diagnosis of acute period of KD and observation of curative effect,prognosis and long-term follow-up of recovery period of KD.
