Objective To investigate the different expressions of adiponectin receptor 2(AdipoR2) mRNA in liver between diet-induced obese rats and controls.Methods The level of AdipoR2 mRNA expression was detected by reverse transcriptase polymerase chain reaction(RT-PCR).Insulin sensitivity was evaluated by insulin sensitivity index(ISI).Results Diet-induced obese rats showed higher expression of AdipoR2 mRNA compare to controls.The ISI of diet-induced obese rats was lower than that of controls.Conclusion High energy diet induceds AdipoR2 expression in liver.
4.Chemical constituents of Osmanthus fragrans.

By Silica gel, Sephadex LH-20 and other materials for isolation and purification and by physicochemical methods and spectral analysis for structural identification, 32 compounds were isolated and identified from ethyl acetate portion of alcohol extract of the Osmanthus fragrans. Their structures were identified as boschniakinic acid (1), ursolaldehyde (2), augustic acid (3), arjunolic acid (4), 5-hydroxymethyl-2-furancarboxaldehyde (5), isoscutellarein (6), 6, 7-dihydroxycoumarin (7), 2α-hydroxy-oleanolic acid (8), quercetin-3-0-β-D-glu-copyranoside (9), D-allito (10), 5, 4'-dihydroxy-7- methoxyflavone-3-0-β-D-glucopyranoside (11), 5,7-dihydroxychromone (12), lupeol (13), naringenin (14), acetyloleanolic acid (15), chlorogenic acid (16), kaempferol-3-0-β- D-glucopyranoside (17), oleanolic acid (18), kaempferol-3-0-β-D-galactopyanoside (19), 3', 7-dihydroxy-4'-methoxyisoflavon (20), ergosta-4,6,8 (14), 22-tetraen-3-one (21), p-hydroxycinnamic acid (22), syringaresinol (23), 3,4-dihydroxyacetophenonel (24), β-sitosterol (25), ethyl p-hydroxyphenylacetate (26), benzoic acid (27), caffeic acid (28), coelonin (29), p-hydorxy-phenylacetic acid (30), p-hydroxyacetophenone (31), and methyl-p-hydroxphenylacetate (32). Except for compounds 2, 4, 5, 8-11, 13, 15, 18, 20, 25, and 27, the rest were isolated from the Osmanthus fragrans for the first time.

BACKGROUNDTo study the composition and significance of the inclusion bodies of human cytomegalovirus (HCMV).
METHODSMicrodissection of inclusion bodies, PCR and Southern blot were adopted to detect DNA, and immunohistochemistry method and catalyzed signal amplification (CSA) were used to detect the different antigens of HCMV.
RESULTSThe inclusion bodies of HCMV were separated from the tissue section of human salivary gland. The fragments amplified by PCR from these dissected inclusion bodies were confirmed to be the DNA of HCMV. With the immunohistochemical method CSA, the immediately early and early antigens of HCMV were detected with monoclonal antibodies DDG9/CCH2, while matrix protein AAC10 was negative in the inclusion bodies.
CONCLUSIONThe ingredient of inclusion bodies of HCMV included the DNA and the antigens expressed in specific stage of the virus.

7.Changes in uncoupling protein-2, 3 mRNA expression in the scalded rats after escharectomy at different post scalding stages.

OBJECTIVETo investigate the expression of uncoupling protein (UCP)-2, 3 mRNA in skeletal muscle of the scalded rats after escharectomy at different post scalding stages.
METHODSOne hundred and twenty Wistar rats were employed in the study, in which 8 served as normal control (C) and 112 were subjected to 30% TBSA 3rd degree scalding and then again, divided into 4 groups. The rats in A group were sacrificed on 8th, 24th, 96th, 120th and 168th post scalding hours (PSHs) without escharectomy. The rats in B group underwent escharectomy at 8 PSH, and those in C group underwent escharectomy at 24 PSH. All the rats in both groups were sacrificed on 96, 120 and 168 PSHs after escharectomy, Escharectomy was performed at 96 PSH in rats of D group, and they were sacrificed on 120 and 168 PSHs after escharectomy. The serum levels of leptin and TNFalpha, and the expression level of UCP2 mRNA were determined at all time points in all groups of rats.
RESULTS(1) The serum levels of leptin in A group were obviously lower than that in C group (P < 0.01) during 24 approximately 168 PSHs, while those in B, C and D groups were much higher than those in A group (P < 0.01) during 24 approximately 168 PSH. (2) The serum TNFalpha levels in A group at all time points were higher than that in control group, while that in B group at all time points were lower than that in A group (P < 0.05 or 0.01), and that in C group at 168 PSH was lower than that in A group (P < 0.05). (3) The UCP2 mRNA expression in skeletal muscle in A group was increased evidently since 8 PSH (P < 0.01), peaking at 24 PSH and lowering thereafter, while that in B and C groups at 168PSH was significantly lower than that in A group at the same time points (0.32 and 0.35 vs 0.71, P < 0.05). The trend of the change in UCP3 mRNA expression in skeletal muscle was similar to that of UCP2.
CONCLUSIONThe postburn up-regulation of UCP mRNA expression might play important roles in the increase of metabolic rate. Escharectomy during shock stage could lower down the expression of UCP2 and UCP3 mRNA expression, and it could be beneficial by lowering metabolic rate.

9.Clinical research of Schatzker type IV tibial plateau fracture.

OBJECTIVETo study the characteristic, treatment and prognosis of tibial plateau fracture Schatzker type IV.
METHODSAccording to the roentgenogram and CT scan in 51 patients, Schatzker type IV injury is divided into 3 types: split, total condylar, depression. In the treatment of split and total condylar injury, if there was no articular depression on CT scan, reduction is done through medial approach; if CT scan demonstrated articular depression, middle approach was used to reduce the depression and bone graft. In split injury, buttress plate was fix on posterior-medial side. In total condylar injury, buttress plate is fix on medial side. In depression injury, fracture was reduced through medial approach, bone graft and buttress plate was fix on medial side. Thirty-three patients were followed up in 41 months averagely. And the statistical analysis was done.
RESULTSThe average Lysholm score of 33 patients was 83.2. Eight patients were excellent, 9 were good, 14 were fair, 2 were poor. The range of motion from 30 degrees to 147 degrees (average 110 degrees ). The factors that caused bad results were: the fracture was not anatomical reduced; tibial plateau is more than 4 mm wider than femoral condylar or lateral subluxation exceed 8 mm; varus deformity of knee. They also caused the increase of osteoarthritis.
CONCLUSIONSIn order to get anatomical reduction and stable fixation, the approach and fix pattern should be chosen on the bases of classification and CT scan of Schatzker IV injury. The factors which influence the prognosis are: (1) tibial plateau is more than 4mm wider than femur condylar, or lateral subluxation more than 8 mm; (2) Varus deformity. Attention should be paid to avoid these during operation.

10.Studies on germination characteristics of Prunella vulgaris seeds.

OBJECTIVETo understand the effect of illumination, temperature on the germination of the Prunella vulgaris seeds.
METHODThe general germination method.
RESULTIllumination showed no clearly effect on the germination rate of the P. vulgaris seeds under proper temperature condition; However, illumination was the main factor to determine the germination rate of the P. vulgaris seeds when the temperature condition was unsuitable. The germination rate of the P. vulgaris seeds decreased with the time and increased when the marinating time was from 12 to 36 hours. When temperature reached 5-25 9 degrees C, the trend of the germination rate increased at frst and then decreased.
CONCLUSIONThe optimal germination condition of P. vulgaris seeds is 12 h of marinating time at the temperature of 20 degrees C under illumination.
