1.Comparison of detection sensitivity of RDA and SHDD methods in cloning differential genes

Objective To establish matched-pairs of DNA samples with copy-number controlled differential target genes, and to compare the detection sensitivity of typical Representational Difference Analysis (RDA) method and Subtractive Hybridization Difference Display (SHDD) method in isolating differential genes.Methods Two gene fragments (376 bp and 869 bp in length respectively) cloned by PCR using Human Papilloma Virus (HPV) DNA as template were used as differential target genes, and mixed with human genome DNA. Five matched, pairs of human genome DNA samples with gradually increased difference in copy numbers of target genes were established and RDA and SHDD methods were performed to clone differential target genes and compared their detection sensitivity. Results By using RDA method, 376 bp fragment with 6-fold difference and 869 bp fragment with 8-fold difference were cloned.However, both of these two target fragments with 4-fold difference were isolated using SHDD method.Conclusion The SHDD method adopts balanced bi-directional subtractive hybridization between two sample difference products and avoids loss of differential target genes caused by unbalanced subtractive hybridization of RDA method, and thus outweighs RDA method in isolating target genes, especially long-length target fragments, with small difference.
2.Correlation between abnormal heterogeneous nuclear ribonucleo-protein expression and tumors

Heterogeneous nuclear ribonucleoprotein (hnRNP) is a family of multifunctional nuclear RNA-binding proteins that regulate the alternative splicing of pre-mRNA and the transport, translation, and stability of mRNA. The most abundant and best charac-terized proteins of this group are hnRNP A1 and hnRNP A2, which share a high degree of sequence homology and functional similarity. HnRNP A1 and hnRNP A2 are upregulated in multiple human tumors and modulate the alternative splicing and mRNA stability of vari-ous tumor-related genes critical to tumor cell growth, apoptosis, inflammatory and immune reactions, and epithelial-to-mesenchymal transition. Therefore, hnRNP A1 and hnRNP A2 have potential diagnostic, prognostic, and therapeutic values.
3.The effect of overexpression of sphingosine kinase on biological characters of gastric carcinoma cells

Objective To investigate the roles of sphingosine kinase (SPK) on the regulation of proliferation, migration and apoptosis of gastric carcinoma cells. Methods BGC-823 cells were infected with adenoviral carriers bringing the wild type SPK gene and SPK mutant (SPK DN ), and their effect on the migration and 5-FU-induced apoptosis of BGC-823 cells was evaluated. The adenovirus-mediated expression of SPK was detected by Western blot, and cellular SPK enzyme activity was assayed. The cell migration was investigated by Transwell Migration Technology. Results The adenovirus-mediated wild type SPK gene increased the cellular SPK activity, thus leading to enhanced migration and resistance to 5-FU-induced apoptosis of BGC-823 cells. Whereas overexpression of SPK mutant (SPK DN ) decreased the cellular SPK activity, thus inhibited the migration and sensitized BGC-823 to 5-FU-induced apoptosis. Conclusion SPK plays important roles in the regulation of migration and apoptosis of gastric carcinoma cells, therefore it may be of a new target for tumor treatment.
4.Impact of sleep deprivation on coronary heart disease and progress in prevention and treatment with traditional Chinese medicines.

Sleep deprivation (SD) has been taken as an independent predictor for cardiovascular risks, which was closely related to the increased morbidity and mortality in coronary heart disease (CHD). In this article, after reviewing the impact of modern medical method sleep deprivation on CHD and studies on principle method recipe medicines for preventing and treating CHD, the authors observed the autonomic nerve dysfunction, hormonal metabolism dysfunction, endothelial dysfunction and inflammatory responses after sleep deprivation, which can cause or aggravate CHD. On the basis of the traditional Chinese medicine theories of "heart dominating the blood and vessels and the mind", the authors considered that traditional Chinese medicines can tonify heart and soothe the nerves, reducing all of the risk factors through multi-target and multi-pathway, and improve sleep and decrease the risk factors caused by sleep deprivation, which provides a new idea for the prevention and treatment of CHD.

5.Interaction of GSTM1, GSTT1 Polymorphism and Air Pollution in Asthma

0.05). In stratified studies, risk of asthma in individuals with null genotype of GSTM1 is 2.667 times of that with wild genotype after exposure to light air pollution. Risk of asthma in individuals living in heavy air pollution area is 2.125 time of that in light pollution area for all wild genotype of GSTM1 individuals, but without statistical significance. Conclusion It was not found that the relationship between GSTM1, GSTT1 polymorphism and asthma. Synergism of genotype of GSTM1, GSTT1 and air pollution was not also seen in this study.
6.Infectious arthritis caused by human parvovirus B19:case report and literature review

Objective To understand the clinical profiles of arthritis after human parvovirus B19(B19) infection.Methods Four cases were described and the clinical and laboratory characteristics were analyzed. Results The median onset age of the 4 patients(2 females and 2 males)was 34 years old.Typical clinical manifestations included joint symptoms,flu-like malaise and erythematous rash.Anti-Bl9 IgM antibodies were all positive in 4 patients.B19 DNA was detected in blood samples of 3.Other types of arthritis were excluded. No relapses were noticed after followed-up of 1~3 years.Conclusion The Anti-B19 lgM and B19 DNA should be detected in patients suffer from arthritis with unknown origin.
7.Application and development of non contact angle - wide viewing system in vitreous retinal surgery

Wide- angle viewing system as an important auxiliary device can clearly observe the whole fundus field of vision in vitreous surgery, which enable vitreoretinal surgery more efficient, safer and more effective. So it has very high application value in ophthalmologic operation. In this paper, we studied the development and application of wide - angle viewing system in vitreoretinal surgery in recent years, from which we summed up the advantage of non - contact wide - angle viewing system in clinical field, and pointed out the shortcomings. The ultimate goal is to make the non -contact wide - angle viewing system better applied in vitreous surgery.
8.Lipoprotein lipase gene polymorphism in simple obese patients

Objective To investigate the impact of lipoprotein lipase (LPL) gene Hind Ⅲ polymorphisms on plasma lipid and body adipose tissue distribution in simple obese patients. Methods The Hind Ⅲ site of LPL gene intron 8 was detennined by PCR-RFLP in 98 simple obese patients and 51 normal controls. Anthropometry and blood lipid levels were also measured. The parameters for regional adipose tissue distribution were measured by computerized tomography (CT). Results In both groups, H + was the major allele. There was no statistically significant difference in frequencies of genotypes or alleles between the two groups. 9.Content Determination of Astragaloside Ⅳ in Qingshen Jianfei Tablets by HPLC-ELSD

Objective To establish a HPLC-ELSD method for determining the content of astragaloside Ⅳ in Qingshen Jianfei Tablets. Methods Stationary phase was C_(18) column (4.6 mm?250 mm, 5 ?m), mobile phase was acetonitrice-water (35 : 65). The evaporation temperature was 120 ℃, the pulverization temperatire was 80 ℃, the flow rate was 1.6 mL/min and column temperature was 35 ℃. Results The standard curve was linear within the range of 1.03~8.24 ?g, r=0.9999. The average recovery was 99.35% with RSD = 1.36% (n = 6). Conclusion The established method is simple and accurate, with good reproducibility and high precision, and suitable for the determination of astragaloside Ⅳ in Qingshen Jianfei Tablets.
