1.Influence of preservation temperature on the characteristics of Anammox granular sludge.

To study the effect of preservation temperature on the characteristics of anaerobic ammonium oxidation (Anammox) granules and optimize the preservation temperature of Anammox granules, the Anammox granules were cultivated in an upflow anaerobic sludge bed reactor through adjusting the hydraulic retention times, and the inorganic carbon with KHCO3/NaHCO3 was alternately supplied. Subsequently, the enriched Anammox granules were preserved at -40, 4 and 35 °C, and ambient temperature of (27 ± 4) °C. NaHCO3 can be used as the inorganic carbon for the growth of anaerobic ammonium oxidizing bacterium (AnAOB). The best preservation temperature was 4 °C for maintaining Anammox biomass, Anammox activity, settleability, and the integrity of the Anammox granule and AnAOB cell structure. During the preservation period, the first-order exponential decay model can simulate the decay of Anammox biomass and activity, and the decay coefficients (bAN) of Anammox biomass and activity had positive correlation with the degree of AnAOB cell lysis. Meanwhile, the rate of Anammox biomass decay was larger than that of Anammox activity. The ratio of protein to polysaccharide in extracellular polymeric substances and heme c cannot effectively indicate the changes of Anammox granules settleability and activity, respectively, and the bioactivity has a negative association with the degree of AnAOB cell lysis.

2.Quality Control on Whole Process in Sterilization Supply Department

OBJECTIVE To investigate the method of quality control on the whole process in sterilization supply department. METHODS According to issued by Ministry of Health all kinds of criteria and inspections,the whole process in sterilization supply department was analyzed,and then the quality was controlled. RESULTS The whole process of sterilization was standardized,systematized and programmed. CONCLUSIONS The quality control on the whole process is an effective measure to ensure the rate of supply eligibility and support.
3.Antimicrobial resistance monitoring of bacteria isolated from blood culture in Xinjiang area during 2013

Objective To understand the distribution characteristics and drug resistance of bacteria isolated from blood cul‐ture in Xinjiang area during 2013 .Methods The identification of isolated bacteria were performed by adopting the France VITEK‐Compact and the ABI series bacterial identification instruments .The antimicrobial susceptibility test was carried out by using the minimum inhibitory concentration (MIC) and Kirby‐Bauer (K‐B) methods .Results A total of 3 962 strains of bacteria were isola‐ted from clinical blood culture ,in which Gram‐positive bacteria and Gram‐negative bacteria accounted for 50 .8% and 49 .2% respec‐tively .The most frequent strains were coagulase‐negative staphylococci (31 .7% ) ,Escherichia coli (23 .2% ) ,Staphylococcus aureus (9 .5% ) ,Klebsiella pneumoniae (8 .7% ) ,Acinetobacter baumannii (3 .6% ) ,Enterobacter cloacae (2 .5% ) ,Enterococcus faecium (2 .5% ) ,Pseudomonas aeruginosa (2 .2% ) ,Enterococcus faecalis (2 .0% ) and Streptococcus pneumoniae (1 .1% ) .The detection rate of extended‐spectrum beta‐lactamase (ESBLs) producing Escherichia coli ,Klebsiella pneumoniae and Proteus mirabilis were 69 .8% ,62 .6% and 66 .7% respectively .The detection rates of methicillin‐resistant Staphylococcus aureus(MRSA) and methicillin‐resistant coagulase‐negative Staphylococcus (MRCNS) was 36 .2% and 86 .3% respectively .The pan‐drug resistant (XDR) strains of Acinetobacter baumannii ,Pseudomonas aeruginosa ,Klebsiella pneumonia were 14 strains(9 .9% ) ,1(1 .2% ) ,2 strains(0 .1% ) ,16 strains(0 .6% ) .No strains resistant to vancomycin or linezolid were found in Staphylococcus and Enterococcus faecalis .Conclusion Among blood culture isolated bacteria in Xinjiang area ,the proportion of Gram‐positive bacteria and Gram‐negative bacteria have little difference .The diversity of bacterial species exist .The resistance to commonly used antibiotics is serious .The distribution situ‐ation of blood culture isolated bacteria should be timely understood .The bacterial drug resistance monitoring should be strengthened to control the nosocomial infections ,guide rational drug use in clinic and control the generation and spread of drug resistant bacterial strains .
4.Relationship between antiproliferation effects of aloe-emodin on growth of gastric cancer cells and cell cycle arrest

Objective To investigate the relationship between the antiproliferation effects of aloe-emodin on growth of gastric cancer cells and cell cycle arrest.Methods Human gastric cancer SGC-7901 cells were treated with 2.5,5,10,20,and 40 ?mol/L aloe-emodin for 1—5 d.The cell growth was determined by MTT assay.Cell proliferation and cycle distributions were analyzed by flow cytometry.Western blotting assay was used to detect the changes of cell cycle regulators,cyclins,and cyclin-dependent kinases(CDK).Results Aloe-emodin inhibited the growth of gastric cancer cells in a dose-dependent manner.Treatment of aloe-emodin resulted in cell cycle arresting at G2/M phase.Its molecular mechanisms involved the decrease of the expression of cyclin A and CDK2,the increase of the expression of cyclin B1 and CDK1.Conclusion One of the antitumor mechanism of aloe-emodin on the growth of gastric cancer SGC-7901 cells is to arrest the cell cycle,which indicates that aloe-emodin has a potential value for the treatment of gastric cancer in clinic.
5.Preparation and evaluation of Shedan in situ forming gel based on ocular characteristics.

To develop an ophthalmic preparation of Shedan, an in situ forming gel was prepared with the formulation containing 18% of poloxamer 407 and 5% of poloxamer 188 by response surface designs plus central composite designs. The rheology results showed that LVE range gamma should limited within 0.5%, Shedan high-frequency region, and the thixotropy recovery time is less than 5 seconds. The phase transition temperature was 33.25 °C according to curve of storage modulus and loss modulus determined by temperature scanning. Surface tension and osmometer of it determined by surface tension meter and dew point osmometer were 36.43 mN · m(-1), and 320.6 mOsm · kg(-1), respectively. Fluorescein sodium was selected as the marker to monitor the corneal residence time, and the results showed that Shedan gel could prolong drug residence for 180 min. In line with zero-order kinetics, releases of muscone and salvianolic acid B in vitro depends on gels erosion. The results of rabbit ocular irritation experiments suggested that Shedan in situ forming gel was biocompatible and nonirritant. In conclusion, a novel Shedan in situ forming gel was developed and characterized for potential drug treatment of retinal vein occlusion.

6.Application and obstacles of ANAMMOX process.

Anaerobic ammonium oxidation (ANAMMOX), as its essential advantages of high efficiency and low cost, is a promising novel biological nitrogen elimination process with attractive application prospects. Over the past two decades, many processes based on the ANAMMOX reaction have been continuously studied and applied to practical engineering, with the perspective of reaching 100 full-scale installations in operation worldwide by 2014. Our review summarizes various forms of ANAMMOX processes, including partial nitritation-ANAMMOX, completely autotrophic nitrogen removal over nitrite, oxygen limited autotrophic nitrification and denitrification, denitrifying ammonium oxidation, aerobic deammonification, simultaneous partial nitrification, ANAMMOX and denitrification, single-stage nitrogen removal using ANAMMOX and partial nitritation. We also compare the operating conditions for one-stage and two-stage processes and summarize the obstacles and countermeasures in engineering application of ANAMMOX systems, such as moving bed biofilm reactor, sequencing batch reactor and granular sludge reactor. Finally, we discuss the future research and application direction, which should focus on the optimization of operating conditions and applicability of the process to the actual wastewater, especially on automated control and the impact of special wastewater composition on process performance.

7.Expression and purification of HgbA from Haemophilus ducreyi and its partial fragment for development of a Sandwich ELISA to detect infection of Haemophilus ducreyi using specific polyclonal antibodies

Objective:To express and purify hemoglobin receptor(HgbA) and its partial fragment(HgbAF) from Haemophilus ducreyi and to develop a sandwich ELISA for the detection of H.ducreyi infection.Methods:The HgbA,a hemoglobin-binding outer membrane protein of H.ducreyi and its partial fragment(HgbAF) were expressed by cloning the genes of hgbA and its 705bp fragment into pET30a and pET28a respectively,and the expressing products were purified from E.coli BL21 with Ni-NTA-His affinity chromatography.The polyclonal antibodies were developed by immunizing rabbits with the rHgbA and rHgbAF.The anti-rHgbA IgG and anti-rHgbAF IgG were purified respectively by saturated amine sulphate precipitation,and their immunoactivity with rHgbA and rHgbAF was tested by Westen blot and ELISA analysis.A Sandwich ELISA was developed for the detection of chinical infection of H.ducreyi using the specific polyclonal antibodies.Results:The HgbA and its partial fragment,HgbAF of H.ducreyi,were expressed and purified successfully by cloning their genes respectively.The results obtained by Western blot analysis showed that each of the antibodies could react with both antigens,rHgbA and rHgbAF.The results of the ELISA analysis showed that H.ducreyi strain was strongly positive,and all other bacteria,including H.influenzae and the bacteria known to relate to genital ulcers were negative.The results of the ELISA analysis showed that the minimum amount of rHgbA detected was 1.56 ng/ml and the minimum number of CFU of H.ducreyi detected was 2?105 cfu/ml in buffer and 1?106 cfu/ml in pus.Conclusion:HgbA and its partial fragment,HgbAF of H.ducreyi are expressed and purified successfully.The polyclonal antibodies developed by immunizing rabbits using rHgbA and rHgbAF could react not only with rHgbA and rHgbAF,but also with H.ducreyi specifically.They do not react with other bacteria,including H.influenzae and the bacteria known to relate to genital ulcers.So the ELISA based on the polyclonal antibodies was specific for the detection of H.ducreyi.Although the level of sensitivity of the ELISA may not be sufficient to detect H.ducreyi in all clinical specimens,further work to increase the sensitivity could potentially make this assay a valuable tool in areas where chancroid is endemic.
8.Open-Monotherapy Study on Different Types of Epileptic Seizures in Children Treated by Lamotrigine for 10 Years

Objective To assess the long efficacy and safety of Lamotrigine(LTG) monotherapy and add-on therapy different types of epileptic seizures in children.Methods According to the classification of the 1981 and 1989 International Union of Antiepileptic for epileptic seizure,a total of 124 cases with epilepsy were included in the study and divided into non-refractory group with 93 cases and refractory group with 31 cases.LTG treatment only or add-on were used.Original drug dosage was not changed and LTG was added slowly and carefully untill the terrible side effect appeared.The average monthly seizure frequency with baseline in the last 3 months was compared and the side effect was observed.Results Total efficiency was 72.6%,control rate was 51.6%.Total efficiency and control rate of the non-refractory group was 81.0% and 61.3%,which was significantly higher than those of refractory group(48.4%,22.6%).Total efficiency and control rate of the combination group with LTG and valproate sodium(VPA) was 78.4% and 54.5%,which was significantly higher than those of the group of LTG only(61.0%,44.0%).Clinical results was different significantly between the course of the observation period within and over 5 years(P
9.Comparative analysis of nursing adverse events between psychiatric and general departments with SHEL model

Objective To compare and analyze the causes that occurred in psychiatric and general department using SHEL models, in order to provide evidence of preventing nursing adverse events.Methods We analyzed causes of 136 nursing adverse events using SHEL models, and compared the classification, causes, incidence of nursing adverse events between psychiatric and general departments. Results The first three nursing adverse events in psychiatric department were medication error,going outside, suicide and self harm, and those in general department were medication errors, specimens being wrong, improper operation and pipeline off. 91.18% of nursing adverse events accounted for quality of nursing services, 43.38% accounted for nursing environment, 41.18% accounted for clinical environment,and 57.35% accounted for parties and others. Incidence of nursing adverse events in psychiatric department were higher than that in general department. Conclusions SHEL model can objectively and comprehensively analyze the causes of nursing adverse events, and make nursing managers take the appropriate management strategy and the best safeguard against the risk of development of response.
