1.Scientific research management promotes the development of translational medicine

Translational medicine appears as a new branch of medicine, aiming at quickly transforming the basic research products into clinical application. Nowadays there is a trend that laboratory researches do not have a close relationship with clinical reality, which is facing the present medical research and education field. Herein translational medicine appeared to solve this fundamental conflict.In this paper, we will analyze the essence and status quo of translational medicine, and provide suggestion on the development of translational medicine from the perspective of medical research management.
2.The expression of Calbindin and Parvalbumin in auditory pathway of kit gene mutated C57BL/6J mouse.

OBJECTIVETo observe the expressions of Calbindin(CB) and Parvalbumin (PV), the two calcium-binding protein, in auditory pathway in mice of wild type C57BL/6J and kit⁺/kitW⁻ ²Bao, a kit gene mutant.
METHODSSix mutated kit gene kit⁺/kitW⁻ ²Bao mice and 6 wild type C57BL/6J (B6) mice were anaesthetized i. p. with chloral hydrate. After the mice were fixed by heart perfusion, the brains were removed and coronal sections were cut with a freezing microtome.
RESULTSWe found that wild type mice had significant expressions of PV on ventral cochlear nucleus, anterior part (AVCN), ventral cochlear nucleus, posterior part (PVCN), inferior colliculus (IC) and auditory cortex (AC). CB was expressed in wild type mice on PVCN and nucleus of the trapezoid body (Tz). The mutant of kit gene induced the less expression of PV on PVCN, IC and AC (P < 0.01), but increased the expression of Tz (P < 0.01). CB could not be observed on PVCN in mutant mice, and the expression of AC was increased( P < 0.01).
CONCLUSIONCB and PV has differential expression level in auditory pathway. Since mutated kit gene can affect expression of PV on PVCN, IC, Tz and AC, as well as CB on PVCN and AC, it suggests that the mutation of kit gene can affect the advanced function of central nervous system in auditory pathway.

3.Dynamic Changes of Mineral Element in The Cell Wall of Growth Cells Detected by CSEM-EDX

Aerenchyrna formation has been described in depth in a number of species at a histological level. But large gaps remain in our understanding of its regulation as a developmental process. It is attempted to analyse essential mineral elements like K, Mg, Cu, Zn, Ca and P in the cell wall of aerenchyma cells in petioles ofS. trifolia at five different developmental stages by CSEM-EDX technique. At early stage, K and Cl concentrations in cell wall were high up to 36% and 4.3% of dry weight, respectively. It supported the hypotheses that aerenchyma spaces are filled with liquid at early developmental stages of aerenchyma in S. trifolia petiole. Mg concentration was high at stage 2, up to 0.86% of dry weight. Zinc and Cu were detected only at rapid expansion stages, during which the concentrations were up to 1.5% and 2.5%, respectively. Calcium was detected in the cell wall only at mature stages, the concentration was high up to 1.3% of dry weight at stages 4 and 5. These results confirmed that the element concentration of aerenehyma cell wall undergoes dynamic changes during different developmental stages, and a low Ca with high Zn and Cu concentration are needed for cell expansion. Copper and Zn deposition in the cell wall showed a significant positive linear correlation, suggesting that these two elements share same or similar uptake and transport mechanism in plants.
4.Carbon Nanotubes-based Drug Delivery to Cancer and Brain

Current treatments for cancer and the central nervous system diseases are limited,partly due to the difficulties posed by the insolubility,poor distribution of drugs among cells and lack of selectivity of drugs,the inability of drugs to cross cellular barriers and blood brain barrier (BBB).Carbon nanotubes (CNTs) possess many distinct properties including good electronic properiies,remarkably penetrating capability on the cell membrane,high drug-loading and pH-dependent therapeutic unloading capacities,thermal properties,large surface area and easy modification with molecules,which render them as a suitable candidate to deliver drugs to cancer and brain.CNTs as a drug delivery could achieve a high efficacy,enhance specificity and diminish side effects.Whereas CNTs have been primarily employed in cancer treatment,a few studies have focused on the treatment and diagnosis of the central nervous system diseases using CNTs.Here,we review the current progress of in vitro and in vivo researches of CNTs-based drug delivery to cancer involving CNTs-based tumor-targeted drug delivery systems (DDS),photodynamic therapy (PDT) and photothermal therapy (PTT).Meanwhile,we also review the current progress of in vitro and in vivo researches of CNTs-based drug delivery to brain.
5.Multi-slice Helical CT Virtual Cholangioscopy:Preliminary ClinicalApplications in the Detection of Biliary Calculus

Objective To investigate the methods and clinical value of multi-slice CT virtual cholangioscopy in the detection of biliary calculus.Methods Volume scanning was performed in 38 patients with suspected biliary calculi after injection 50% Meglumine adipiodone injection 20 ml through vien.All data sets were transferred to ADV3.1 workstation.MSCTVC reconstruction were done with Navigator soft-ware.All cases were proved by ultrasound,axial CT and MSCT 3 D results or operations.Results MSCTVC could well demonstrate 3D fashion of biliary calculi and it was similar to ultrasound,axial CT and MSCT 3D results.Conclusion MSCTVC combined with CT 2D 3D results has a high diagnosis accuracy and clinical usefulness,thus,improve the diagnosis of biliary calculi.
6.Clinical Application of Multi-slice Helical CT Virtual Endoscopy in Colorectal Carcinoma

Objective To evaluate the methodology and the clinical usefulness of multi-slice helical CT virtual endoscopy(MSCTVE).Methods Twenty-six cases by suspected colorectal carcinoma underwent multi-slice helical CT virtual endoscopy after colon was cleaned and insufflated with air.The parameters were used:120 kV,250 mA,pitch 6,reconstructure thickness 2.5 mm,interval 1.0.All data sets were transferred to ADV 3.1 workstation,CTVE reconstruction were done with Navigator and used CTVE to view colorectal and rectum carcinoma.Of 26 cases,16 cases were proved by colonoscopy and 22 cases by operation.Results 14 cases carcinoma in rectum,8 cases in sigmoidcolon,1 case involved both rectum and sigmoidocolon.MSCTVE results were similar to colonography.Conclusion Multi-slice helical CT virtual endoscopy has clinical usefulness for detecting colorectal and rectum carcinoma as it is non-invasive,safe,feasible.MSCTVE is a supplement to colonoscopy especial in incomplete colonoscopy.
7.The roles of TANK-binding kinase-1 in chronic hepatitis B virus infection induced interferon antiviral immunity

Objective To elucidate the roles of TANK-binding kinase-1(TBKl)in hepatitis B virus (HBV)infection induced interferon antiviral immunity.Methods Peripheral blood monocytes were separated by CD14 magnetic microbeads from healthy volunteers(HV)and chronic hepatitis B(CHB)patients.Purified mDCs were induced and proliferated in the culture medium with human granulocyte-macrophage concentration of 25 mg/L were stimulated.The mRNA expressions of TBK1,interferon regulatory factor (IRF)3 and interferon(IFN)-βwere quantified by real time polymerase chain reaction(PCR).The levels of IFN-β in supernatants were determined by enzyme-linked immunosorbent assay(ELISA).Reslllts The mRNA levels of TBK1,IRF3 and IFN-β did not change significantly at 0,12,24 and 48 h after the significantly at 0, 12, 24 and 48 h in CHB group, whereas, it was significantly up-regulated at 12 h in HV group. Conclusions Our results suggest that there may be some disorders in host antiviral signal transduction pathways downstream the binding between ligands and receptors on mDC surface. The insufficient IFN-β expression after HBV infection may result in persistent chronic infection.
8.Clinic applications of vascularized plantaris tendon grafting.

OBJECTIVETo study a new method of the vascularized plantaris tendon transferring.
METHODSThe vascularized plantaris tendon was elevated with the pedicle of the fascial perforating vessel or a small segment of the posterior tibial artery, which was transplanted to the wrist to repair the tendon defects of the flexor pollicis longus or the flexor digitorum muscle resulted from electric injury. According to the defects, the plantaris tendon can be used together with the skin, fascia, or both as a composite flap. 7 cases received the operation with complete survival of the flap.
RESULTSPostoperative follow-up showed satisfactory results in the 7 cases. Through physical exercises, the patients obtained increased joint motion and better function of the hand.
CONCLUSIONThe vascularized plantaris tendon has many advantages, including easy to harvest, easy to plerosis the donor site, little affect to a main vessel, able to repair the complex defects. This method has great potential for applications.

9.Genotypes of β-lactamases produced by Klebsiella pneumoniae

Objective To investigate the genotypes of β-lactamases produced by Klebsiella pneumoniae.Methods Plasmid conjugation,PCR amplification,gene cloning and DNA sequencing,isoelectric focusing electrophoresis and extended-spectrum β-lactamase(ESBLs)confirmatory test were carried out for analyzing the encoding gene of β-lactamases in clinical strains of Klebsiella pneumoniae collected from hospital wards.Results Totally 75 clinical strains of Klebsiella pneumoniae were collected,in which 48 strains were confirmed to produce genotype of β-laetamases(64.0%),including 39 ESBLs-producing Btraim(52.0%).Among 48 strains,17 isolates(35.4%)carried 2 types of ESBLs genes,7(14.6%)carried 3 types of ESBL8 genes,and 5(10.4%)carried 4 types of ESBLs genes.CTX-M was the most comon type(30/48,62.5%),followed by TEM(26/48,54.2%)and SHV(25/48,52.1%).Among 9 isolates with DHA-1 AmpC β-laetamase,8 produced AmpC β-lactamases and ESBLs.Class A carbapenemase KPC-2 was produced in 3 isolates.False negative rate of ESBLs confirmatory test was 23.1%(9/39).Condusion Genotypes of β-lactamases produced by Klebsiella pneumoniae are complicated,which results in multi-drug resistance in clinic.
10.Effect of p-nitrophenol Shock on Sludge Activity and Microbial Populations and in UASB Reactor

Effect of p-nitrophenol shock on microbial populations and sludge activity in UASB reactor was studied by DGGE-PCR of 16S rDNA fragments and detection of COD removing and biogas yield.The results showed that p-nitrophenol seriously inhibited the sludge activity,resulting in the drop of biogas and COD removing rate.The 40mg/L p-nitrophenol had more inhibition than 20mg/L p-nitrophenol.It would take 27 and 16 days respectively for reactor to recover after 40mg/L and 20mg/L p-nitrophenol shock.The diversity of eubacteria and methanogens were also effected by the p-nitrophenol shock.The variation of eubacteria was more than that of methanogens after p-nitrophenol shock.The drop of biogas was mainly related to the variation of Methanosaeta sp.and Methanomicrobia sp.after p-nitrophenol shock.Among the eubacteria the population of Chloroflexi sp.、Bacteroide sp.and Anaerovibrio sp.decreased greatly after p-nitrophenol shock.And more,the Rheinheimera sp disappeared after 40mg/L p-NP treatment.But the Flavobacteria sp.appeared after p-nitrophenol shock,which was probably related to the degradation of p-NP.
