The core-shell nanopaticles of Au@polyvinyl-pyrrolidone ( PVP) with uniform size and controllabe shell-thickness were prepared by hydrothermal method. The core-shell nanoparticles could be assembled to be the monolayer array on Si substrate relying on the dispersion of core-shell nanoparticles arising from PVP shell. The malachite green ( MG ) absorbed by H-bond could be detected on the array under the electromagnetic enhancement of inner-core Au nanoparticles. Under the conditions of the optimum shell-thickness of Au@PVP and the appropriate absorbed time of MG, the detection of MG could be realized in the linear range from 1 × 10-10 mol/L to 1 × 10-5 mol/L with the correlation coefficient ( R2 ) of 0. 98. The detection limit was 10-12 mol/L. This method was applied to the determination of MG in tilapia fish fillets of Xiagang market. No MG was found in this real sample. The spiked recoveries of the sample ranged from 70. 8% to 126. 0%. This method is simple and accurate, and can be used for detection of MG in the fish.

Aim To explore the effect of picrodideⅡ on the expressions of Caspase-3 and poly ADP-ribose polymerase (PARP) in brain tissue following cerebral ischemic reperfusion injury in rats. Methods The middle cerebral artery occlusion reperfusion models were established with intraluminal thread methods in rats. PicrodideⅡ (10 mg·kg~(-1)) and salvianic acid A sodium (10 mg·kg~(-1)) were injected from tail vein for treatment. The neurological function was evaluated with Bederson's test and the cerebral infarction volume was observed with tetrazolium chloride (TTC) staining. The brain structure was observed by hematoxylin-eosin (HE) staining and the apoptosis was counted by TUNEL immunofluorescence assay. The expressions of Caspase-3 and PARP were detected with immunohistochemical and enzyme linked immunosorbent assay. Results After ischemia 2 h and reperfusion 22 h, the rats showed neurological function deficit and cerebral infarction in ischemic hemisphere. The expressions of Caspase-3 and PARP and the number of apoptotic cells in brain tissue increased compared with those in the sham operative group (P <0.05). In picroside and salvianic acid A sodium groups, the Bederson's scores and cerebral infarction volume, the expressions of Caspase-3 and PARP and the number of apoptosis cells were lower than those in the negative control group (P <0.05). While there was no significant difference in five indexes metioned above between picroside group and salvianic acid A sodium group (P >0.05).Conclusion PicrosideⅡ might reduce the expressions of Caspase-3 and PARP to inhibit the neuronal apoptosis induced by cerebral ischemia reperfusion injury and improve the neurological function of rats.

Objective To compare the influence of different admission modes on quality of life of patients with chronic schizophrenia. Methods 200 patients with chronic schizophrenia were randomly assigned to discontinuous group and continuous group with 100 cases in each group. All patients were treated with atypical antipsychotic drugs. A discontinuous hospitalization combined with home treatment was given to the discontinuous group and a continuous hospitalization to the continuous group for 1 year. The Positive and Negative Syndrome Scale (PANSS) and the Schizophrenia Quality of Life Scale (SQLS) were respectively used to evaluate the patient's condition and quality of life before and after the intervention. Results The intervention methods on scores of PANSS and SQLS in both groups had a change trend with time. The scores of PANSS and SQLS were significantly lower in the end of 6 months and 1 year after intervention than before in both groups (F=2119.513, 390.981, P=0.000). There was a interaction effect between intervention method and time in both groups on the scores of PANSS and SQLS (F=17.660, 11.709, P=0.000). There was significant difference between 2 groups on the score of PANSS (F=19.673,P=0.000). The decrease range of those scores was greater in the discontinuous group than in the continuous group (P=0.000). Conclusion A discontinuous hospitalization combined with home treatment is more beneficial than a continuous hospitalization for the patients with chronic schizophrenia in controlling the syndrome and improving or keeping the quality of life.

Objective To analyze the cost-effectiveness of the rehabilitation of intermittent hospitalization combined with home treatmenton chronic schizophrenics. Methods 100 patients with chronic schizophrenia were randomly assigned to study group and controlgroup, 50 cases in each group. All patients in both groups were treated with atypical antipsychotic drugs. An intermittent hospitalization combinedwith home treatment was given to the study group and a continuous hospitalization to the control group for a year. Positive and NegativeSyndrome Scale (PANSS), Activities of Daily Living Scale (ADL) and Schizophrenia Quality of Life Scale (SQLS) were used to evaluatethe patient's condition and activity of daily living and quality of life before and after the intervention. In the meantime, a cost-effectivenessanalysis was made. Results The interventions had significant influence on the scores of PANSS, ADL and SQLS in both groups. Thescores of PANSS, ADL and SQLS were significantly lower 1 year after the intervention than before in both groups (FB= 540.577, 318.148,326.751, all P=0.000). There was a significant interaction effect between intervention method and time in both groups on the scores ofPANSS, ADL and SQLS (FAB=13.710, 10.918, 6.578, P=0.000, 0.001, 0.012). The decrease value of those scores was significantly greater inthe study group than in the control group (t=3.703, 3.304, 2.565, P=0.000, 0.001, 0.012). The ratio of cost-effectiveness was larger in thestudy group than in the control group (t=7.035, P=0.000). Conclusion Intermittent hospitalization combined with home treatment is morebeneficial for the patients with chronic schizophrenia in controlling the syndrome, keeping or improving the activity of daily living and thequality of life. Therefore its rehabilitation benefit is better than that of continuous hospitalization.

This study was aimed to investigate the action mechanism ofHypoxis Hemerocallidea (African Potato, AP) on the AMPK signal pathway of skeletal muscles in diabetic rats. Among 40 male SD rats, 10 rats were used as the normal group, and the other 30 rats were fed with high-fat food for one month, and then injected with STZ for the model establishment. After the successful model establishment, rats were divided into the model group, pioglitazone hydrochloride group and the AP group. Intragastric administration was given for 5 weeks in each group. Then, the skeletal muscle tissues were embedded and sliced for immunohistochemistry test. The protein expression of p-AMPKα, p-AS16 and GLUT4 in skeletal muscles was detected by western blot. The 100 mmol·L-1 glucose was used in the establishment of C2C12 skeletal muscle cells insulin resistance model. AP drug-containing serum was used in the establishment of the treatment group. The control group was the normal cells. Glucose consumption, cell proliferation, SOD content, and MDA content were detected. And the protein expressions of p-AMPKα, p-AS160, GLUT4 were detected with the western blot and RT-PCR. The results showed that compared with the normal group, AP can up-regulate p-AMPKa protein express (P < 0.01), increase skeletal AS160 phosphorylation level (P < 0.01), and up-regulate the GLUT4 level (P < 0.01). Compared with the normal group, the high glucose caused the decrease of C2C12 skeletal muscle cell activity and the decrease of glucose consumption (P < 0.05), decrease of SOD, increase of MDA (P < 0.01), and the decrease of p-AMPKα, p-AS160, GLUT4 protein expression (P < 0.01). After 48 h intervention, the SOD of C2C12 skeletal muscle cells in the AP drug-containing serum group was significantly increased (P < 0.01), the MDA content was decreased (P < 0.05), the AMPKa and AS160 phosphorylation levels were increased (P < 0.01), the GLUT protein expression was increased (P < 0.01). It was concluded that the induced AMPKa and AS160 phosphorylation promoted GLUT 4 expression may be one of the action mechanism of insulin resistance of skeletal muscles in diabetes.
