1.Digital Image Measurement in Rhinoplasty and Its Reliability Confirmation

For developing Digital Image Expert System in rhinoplasty, the accurate and reliable Digital Image Measuring System must be first set up, and the data from the Digital Image Measuring System must be reliable with the data measured by hand. The measuring system mainly consists of a computer with an image processing board, a TV camera and a head fixing device. Ten adults were selected randomly and some indexes of head and face were measured with the measuring system and by hand respectively. The reliability of the measuring system was confirmed by the statistical analysis of data measured. Finally, the results were also discussed.
2.Treatment of acute perforation of gastric cancer:a report of 40 cases

Objective To investigate the clinical characteristics and therapy of acute perforation of gastric carcinoma(APGC) to improve the diagnosis,treatment and prognosis of APGC. Methods Forty cases of APGC admitted to Gong liao hospital of Zhenping county.Of the forty patients,ten underwent repair of perforation,twenty-one cases underwent palliative gastrectomy (six of them underwent lymphadenectomy three weeks after previous gastrectomy );five subjected to radical gastrectomy. two underwent gastrostomy or jejunostomy for nutrition.One underwent drainage and peritoneal lavage only;1 received non-operative therapy.Results Four patients died in this series.The 1-,2-,3-years survival rate was 83.3%(30/36),52.8%(19/36),27.8%(10/36)respectively. Conclusions Early diagnosis and adopting resection of gastric cancer plus comperhensive treatment can improve the prognosis of patients with APGC.
3.Protective effect of TQ0701-2 on cerebral ischemia reperfusion injury in rats

AIM:To investigate the protective effect of a new type nerves protectant TQ0701-2,as the derivate of edaravone free radical scavenger,on cerebral ischemia reperfusion injury rat.METHODS:120 male SD rats were randomly divided into sham group,model group,edaravone group(3.0 mg/kg)and TQ0701-2 high-dose group(6.0 mg/kg),middle-dose(3.0 mg/kg)and low-dose group(1.5 mg/kg).Animals in the latter five groups were subjected to transient focal ischemia by the middle cerebral artery occlusion(MCAO)for 2 h before reperfusion,while the sham group wasn't ischemic.The rats were injected with edaravone or TQ0701-2 30 min before ischemic and 0 min,2 h after reperfusion in edaravone and TQ0701-2 groups,sham group and model group were treated with normal saline as well.After 24 h of reperfusion,the infarct ratio,neurological and histological deficient and the changes of pathohistology were evaluated.RESULTS:The infarct ratios and neurological deficit scores in the model group were increased(P
4.Identification of human papillomavirus in vulvar intraepithelial neoplasia

Objective To evaluate the rate and types of human papillomavirus (HPV) in vulvar intraepithelial neoplasia (VIN).Methods We detected HPV DNA in 67 lesion tissues collected from 40 VIN patients with PCR based reverse line blot hybridization and DNA sequencing. The PCR using GP5/GP6 primers in HPV L1 region resulted in a 150-bp fragment. When the PCR did not amplify the target DNA,another PCR using SPF1/SPF2 primers was performed to amplify the 65-bp fragment.Results HPV DNA amplified with primers GP5/GP6 was positive in 52.2% (35/67) of the lesions. Of the 32 negative lesions,26 (81.2%) were positive for HPV DNA amplified with SPF1/SPF2 primers. The total positive rate was 91.0% (61/67). Ninety percent of the HPV belonged to high risk types. Sequencing data showed that the genotypes of 31 mono-infection lesions were the same as those of the reverse line blot results,yet the sequences in 4 multi-infection samples could not be determined. Of the 26 SPF PCR products,24 could be sequenced. 80.6% (25/31) patients with multiple lesions displayed the same genotype,suggesting HPV in the different lesions from the same patient was monoclonal.Conclusion The high risk type of HPV is associated with vulvar intraepithelial neoplasia and may play an important role in the development of invasive vulvar carcinoma.
5.The relationship between endothelin-converting enzyme-endothelin system and type 2 diabetes with macroangiopathy

Objective To determine the endothelial function, endothelin-converting enzyme (ECE) activity and ECE mRNA level in patients of diabetes with macroangiopathy, and to investigate the effect of the endothelial function change in type 2 diabetes and diabetes with macroangiopathy. Methods Sixty healthy persons and 40 patients of type 2 diabetes and 60 patients of type 2 diabetes with macroangiopathy were recruited for the study. The activity of ECE in white blood cells was measured by an established method. Endothelial function, ECE activity and expression of ECE mRNA were determined by ELISA, RIA and RT-PCR respectively. Results The expression of ECE mRNA and the ECE activity were increased in patients of type 2 diabetes, especially in diabetics with macroangiopathy. The damage of endothelial function in the diabetic patients was very common. Conclusion High level of ECE activity relates to the damage of endothelial function. High level of ECE activity and up-regulation of expression of ECE mRNA may play a key role in the development of diabetic macroangiopathy.
6.Expression of the Fas antigen and induction of apoptosis in human eosinophils in vitro

AIM: The expression of the Fas Antigen and induction of apoptosis by anti-Fas antibody in esoinophils in vitro were investigated. METHODS: Purified eosinophils from health donors were cultured for 72 h in the presence of human IL-5 and with or without anti-Fas monclonal antibody (MoAb) at various concentrations (1-1000 ?g/L). The expression of the Fas antigen on eosinophils was determined by immunocytochemistry. The changes of eosinophils viability and apoptosis were also studied. RESULTS: The Fas antigen was expressed on freshly isolated eosinophils, which had no significant changes after culture in the presence or absence of IL-5. The anti-Fas MoAb at different concentration suppressed significantly the IL-5-mediated eosinophils survival (78%?9%). 7.Effects of ultrasound-mediated SonoVue microbubbles destruction on the integrity and expression of hAng-1 gene

Objective To test the efficiency of gene transfer and expression mediated by ultrasound and microbubble strategy in 293T cell. Methods SonoVue microbubbles were mixed with pla.smid DNA encoding hAng-1 and green fluorescent protein. The mixture of the DNA and microbubbles was administer to cultured 293T cells by ultrasound exposue. The ultrasound condition was 1. 5 W/cm~2 and 30 s. Forty eight hours later, transfer rate was assessed by fluorescence microscopy and flow cytometry. Cell viability was assayed by Trypan Blue staining. RT-PCR and Western blot analysis was used to examine the expression of hAng-1 mRNA and protein. Agarose gel electrophoresis was used to evaluate the integrity of the plasmid. Results The transfection expression rate of eGFP in 293T cells was markedly increased with the additon of 20% microbubbles and 15 mg/L DNA. Fetal calf serum had no influence on the gene transfer rate. Ultrasound irradiation combined with microbubbles couldn't destroy the integrity of plasmid. Conclusions Ultrasound-mediated microbubbles destruction can increase the transfection and expression of hAng-1 gene in 293T.
8.The effects of different fenofibrate preparation on the hyperlipidaemic rats

AIM To compare the effects of micronised fenofibrate with those of standard fenofibrate on regulating serum lipid. METHODS Wistar rats were fed by the hyperlipids food to induce hypolipidemia, and then orally treated with 20,30,40 mg?kg -1 per day micronised fenofibrate and 20,30,40,60,80 mg?kg -1 per day standard fenofibrate for 10 days. At the tenth day, serum total cholesterol and triglycerides were measured. RESULTS 1 In the same experimental conditions,the minimal efficacious dose of the micronised fenofibrate is 30 mg?kg -1 , but that of the standard fenofibrate is 80 mg?kg -1 ; 2 Using efficacious doses, both kinds of fenofibrate could decrease the content of triglyceride to normal level. They also could decrease the level of total cholesterol by 36 69%~51 56%. CONCLUSION The effects of micronised fenofibrate are better than those of standard fenofibrate, which mainly decreases the level of triglyceride. Micronised fenofibrate also decreases level of serum total cholesterol.
9.Practice and thinking of online teaching of pediatrics in undergraduate clinical medicine

Objective:To explore the application method and effect of online teaching of pediatrics in undergraduate clinical medicine.Methods:From March to August, 2020, two classes of pediatrics were selected as research objects: 67 students from Batch 2016 "5+3" clinical medicine class were taught by providing recorded teaching resources and online communication and answering questions (hereinafter referred to as "recorded courses"); 39 students from Batch 2015 "5+3" clinical medicine class were taught by live courses and online communication and answering questions (hereinafter referred to as "live courses"). Through the questionnaire survey on the two groups of students, the effect of the two online teaching methods were compared and the medical students' views and suggestions on them were collected. SPSS 26.0 was used for statistical analysis.Results:Most of the students in the two groups (98.1%) agreed with the effect of the online teaching of pediatrics and thought it could meet their learning needs. There was no significant difference in the online teaching effect between recorded courses group and live courses group ( P > 0.05). However, they had a poor recognition with the implementation of complete online teaching in the future, while they preferred the combination of online teaching with practical exercises in offline hospitals. Conclusion:Online teaching of pediatrics has been recognized by medical students. The two online teaching methods have their own advantages: recorded courses are more planned and the content is more rigorous; live courses are more interactive and flexible. However, considering that medicine is a highly practical subject, it still needs to be supplemented by offline practice while giving full play to the advantages of online teaching.
10.Expression and significance of signaling pathway of miR-134/CREB/pCREB in patients with epilepsy and in epileptic rats

Objective To investigate the expression of microRNA-134 ( miR-134 ) , CREB and pCREB in the temporal lobe tissue of patients and epileptic rats and to explore their roles in pathogenesis of epilepsy.Methods Tempo-ral lobe tissue samples of 14 patients with refractory epilepsy and 10 non-epileptic patients, and hippocampus and brain tis-sue samples of 42 rats were used in this study.Forty-two healthy adult male Sprague-Dawley rats were randomly divided in-to 6 epilepsy groups (24 h, 72 h, 7 d, 14 d, 30 d, and 60 d after kindling epilepsy) and a normal control group (n=6 for all groups) .The rat model of epilepsy was generated by intraperitoneal injection of 127 mg/kg lithium chloride and 16-20 h later, 35 mg/kg pilocarpine.In the temporal lobe tissue of patients and hippocampal tissue of rats, the expression level of miR-134 was detected by real-time polymerase chain reaction.The expression levels of CREB and pCREB were de-termined by Western blot, and CREB and pCREB localization was assessed by immunohistochemistry.Results Compared with the control rats, the expression of miR-134 was significantly decreased in the temporal lobe tissue of experimental rats at 72 h,7 d,14 d, 60 d after kindling (P<0.05),and no significant change at 24 h and 30 d after kindling (P>0.05). Expression of miR-134 in patients with refractory epilepsy was significantly lower than that of the controls ( P<0.05 ) , while up-regulation of CREB expression was at the same time points (P<0.05).Up-regulation of pCREB expression was at all the time points after kindling (P<0.05).CREB and p-CREB expressions were seen in the nuclei of neurons, and significantly higher in patients with refractory epilepsy and epileptic rats.Conclusions The expression of miR-134 is sig-nificantly decreased and that of CREB and pCREB was significantly increased in the temporal lobe tissue of patients with re-fractory epilepsy and the hippocampal tissue of epileptic rats.These findings indicate that the signaling pathway of miR-134/CREB/pCREB may play an important role in the pathogenesis of epilepsy.
