1.CONNEXIN43 EXPRESSION WITH AGE ADVANCING IN RAT HEART

Objective To investigate the change of Connexin43 expression in rat heart as age advancing. Methods The expressions of Connexin43 in different age of rat heart were determined by immunohistochemistry. Results 1\^ The expression of Connexin43 was only seen in ventricular myocardium. 2\^ The Connexin43 immunolabelling of infant rat heart lie much more within the transversely orientated intercalated disk, besides, a fewer have a punclate distribution over the entire surface of the ventricular myocytes, and Connexin43 distributed mainly on the myocyte surface of the parpillary muscles. Connexin43 immunolabelling of young and old rat heart are typically confined to the site of intercalated disk.Conclusion The expressions of Connexin43 are different in different age and different parts of rat heart.
2.Clinical observation in 6 patients with cervical cancer after ovarian transposition surgery

Objective To explore the impact of ovarian function in young patients of early cervical cancer after ovarian shift surgery. Methods 6 young patients with cervical cancer were given radical surgery for ovarian transposition at the same time, before and 1 and 6 months after operation, blood FSH, LH, E2 were measured. B-ultrasound was used to monitor ovarian ovarian function. Results 4 patients after 1 month postoperative, 1 case after postoperative 6 months with normal ovarian function, 1 case of pre-operative radiotherapy after 6 months the resumption the ovarian function restoration. Conclusion Young cervical cancer patients with ovarian transposition can maintain good ovarian function.
3.Distribution of free Ca~(2+) and characterization of Ca~(2+) channel in the cultured early-stage human embryonic retina

Objective To investigate the distribution of free Ca 2+ and characterization of Ca 2+ channel in the cultured 11~15 week human embryonic retina. Methods The cells from 11~15 week embryonic retina were dissociated enzymatically and cultured on coated cover slides. Three weeks later, Fluo3, a Ca 2+ indicator, was added in Ca 2+ containing or no Ca 2+ Hepes buffer and incubated with retinal cells for 30 min, meanwhile with or without verapamil, perdipine, or dexamethasone adding. Ca 2+ staining and Ca 2+ transit before and after 10 ?mol/L or 50 ?mol/L KCl stimulation was observed and recorded using Zeiss LM510 confocal laser scanning microscopy. Results In response to K + exposure, a rapid rise of free Ca 2+ in cytoplasma and nuclei of neuron and glial cell was observed, but was repressed with the treatment of verapamil, perdipine, or dexamethasone. The obvious Ca 2+ influx from cytoplasma into nuclei was observed even in the absence of external Ca 2+ . Conclusion L type Ca 2+ channel was expressed and functionally matured in the cultured early stage human embryonic retina.
4.Detection of nucleic acids in Borna disease virus in patients with viral encephalitis

Objective To explore the relationship between viral encephalitis(VE)and Borna disease virus(BDV)infection,and discuss the clinical features of viral encephalitis patients infected by BDV.Methods The p24 gene fragment of BDV in cerebrospinal fluid mononuclear cells(CSFMC)from 32 patients with viral encephaliris and 34 healthy individuals were examined by fluorescence quantitative nested reverse transcriptase polymerase chain reaction(FQ-nRT-PCR),in which, β-actin was used as internal reference. The clinical manifestations of patients with positive BDV p24 were watched. And the gene sequence,homology and amino acid sequence of positive products were analyzed. Results The positive rate(4/32,12.5%)of BDV p24 in CSFMC of patients with viral encephalitis was significantly higher than that of healthy individuals(0/34,0%),P<0.05,and the numbers of copies of BDV p24 in CSFMC of VE patients were more than 102/μl. Compared with the sequences of the standard strain Ⅴ and the virus strain H1766 of BDV from horse supplied by GenBank,the homology of the gene sequence of positive product of CSFMC was 95.35% and 98.84% with gene mutation in 4 sites(nt1649 T→C, nt1656 G→A,nt1670 C→T, nt1676 C→T). The genetic relationship between the positive product and the virus strain BDV from horse was the nearest. The clinical features of VE patients with positive BDV p24 were mainly by mental and behavior disorders. Conclusion The occurrence of VE patients in the city of Zunyi in Guizhou province may be associated with BDV infection. The clinical features of VE patients with positive BDV p24 were mainly mental and behavior disorders.
5.Optimization of Reverse Osmosis Process of Oil-Bearing Water Bodies Ultrafiltrate from Compound Chuanxiong Capsules

Objective To investigate the effect of liquid temperature, pressure, concentration, pH parameters on the membrane process and rejection in the process of using reverse osmosis membrane to concentrate oil-bearing water bodies ultrafiltrate of Compound Chuanxiong Capsules, and optimize the process. Methods With oil-bearing water bodies ultrafiltrate of Compound Chuanxiong Capsules volatile oil as the research object, the changes in flux under different operating parameters was measured. Results For this system, suitable operation parameters was as follows: pressure was 1.6 MPa, temperature was 35~ 40 ℃, pH was 9~10. Conclusion Using reverse osmosis membrane to concentrate oil-bearing water bodies ultrafiltrate of Compound Chuanxiong Capsules volatile oil, better flux and rejection can be obtained at the appropriate operating conditions.
6.ENZYMOLOGY OF MICROBIAL DEGRADATION OF ORGANOPHOSPHATE CHEMICALS

Organophosphate chemicals are widely used as agricultural pesticides and war reagents, their biodegradation is emphasized on the theoretical and practical aspects. Organophosphate hydrolases play important roles in the biodegradation of organophosphate chemicals. Great advancement was achieved recently in the determination of crystal structure and catalytic mechanisms of the hydrolase. This paper reviewed the research progresses in the enzymology, protein structure, catalytic mechanisms and application of the organophosphate hydrolase, and predicted the future research in this field.
10.The significance of changes in thyrotropin receptor antibody and soluble intercellular adhesion molecule-1 levels in patients with thyroid-associated ophthalmopathy during intravenous methylprednisolone pulse treatment

Objective To explore the relationship of thyrotropin receptor antibody (TRAb) and soluble intercellular adhesion molecule-1 ( sICAM-1 ) in patients with thyroid-associated ophthalmopathy ( TAO), and the role played by TRAb and sICAM-1 in the pathogenesis of TAO. Method Twenty-three TAO patients were assigned to groups according to the clinical activity score and EUGOGO. All patients were treated with intravenous methylprednisolone pulse. The levels of serum TRAb and sICAM-1 were evaluated by a competitive radioimmunoassay and enzyme-linked immunosorbent assay respectively before treatment and by the end of each methylprednisolone pulse. Results The differernce in serum TRAb levels was associated with activity scores of TAO (P=0. 020). The change in serum sICAM-1 was associated with durations of TAO ( P = 0.015). During methylprednisolone treatment in active TAO patients, the levels of TRAb kept on decreasing gradually and markedly declined after the third methylprednisolone pulse in active TAO patients (P<0.05). The trends of changes in serum TRAb and sICAM-1 levels were both different in active and inactive TAO patients by tendency analysis. Conclusion TRAb level was related to the activity of TAO and might serve as a significant predictor of response to methylprednisolone therapy. The negative correlation between sICAM-1 levels and duration of TAO corroborates the role played by ICAM-1 during the early stage of TAO. Higher sICAM-1 levels are not expected to be specific to TAO and may not predict a response to methylprednisolone therapy.
