1.Clinical Observation of Homemade Medicated and Dietary Compound Granules in Treating Sub-health Insomnia with Deficient Cultivation of Heart-spirit

Objective To observe the clinical effects of the homemade medicated and dietary compound granules in treating sub-health insomnia with deficient cultivation of heart-spirit. Methods Sixty qualified patients were randomly divided into treatment group and control group, 30 cases in each group. The treatment group was given homemade medicated and dietary compound granules(mainly composed of Semen Ziziphi Spinosae, Radix Codonopsis, Sclerotiium Poriae Circum Radicem Pini, Radix Polygalae, Flos Rosae Rugosae, Plumula Nelumbinis, Fructus Ziziphi Jugubae, and Arillus Longan). The control group was given Oryzanol powder. The course of treatment lasted for half a month. Before and after treatment, the scores of Pittsburgh Sleep Quality Index(PSQI) and the traditional Chinese medicine(TCM)symptom rating scale were observed, and the clinical efficacy of the two groups were also evaluated. Results After treatment for half a month and after suspension of medicine for half a month , (1) the total effective rate of the treatment group was higher than that of the control group (P<0.05);(2) the total PSQI scores , sleep quality scores , falling asleep time scores , and daytime function disorder scores were decreased in the treatment group (P < 0.01 compared with those before treatment) , but the scores of each PSQI item in the control group stayed unchanged (P > 0.05). The total PSQI scores , sleep quality scores , falling asleep time scores, and daytime function disorder scores of the treatment group were lower than those of the control group (P<0.01);(3) the total scores of TCM symptoms as well as the scores of dreaminess and susceptibility to awakening, palpitation and amnesia, lassitude and weakness, poor appetite, pale complexion, vertigo and dizziness, abdominal distension and loose stools were declined in the treatment group (P < 0.05 or P < 0.01 compared with those before treatment). Conclusion Medicated and dietary compound granules are effective on improving sleep quality, shortening time to fall asleep, improving sleep disorder, increasing the efficiency of daytime function, improving patients’ mental state entirely, and maintaining the short-term curative effect.
2.Study on cone beam computed tomography for detecting bone thickness of skeletal class Ⅱ infrazygomatic crest in teenagers

Objective To adopt the cone beam computed tomography(CBCT) to analyze the bone thickness of infrazygomatic crest with different vertical skeletal facial types in teenagers.Methods Sixty teenagers of skeletal class were collected,including each 20 cases of high angle,average angle and low angle.The CBCT scanning data of oral maxillofacial region were collected.The bone thickness at different coronary slices in buccal side of infrazygomatic crest region at 13,15,17 mm above the maxillary occlusal plane was measured.The measured data were analyzed statistically.Results The bone thickness of infrazygomatic crest was gradually thinned from down to up,and the difference was statistically significant(P<0.05);the bone thickness(except 3 points at 13 mm from maxillary plane) at each measured point in the high,average and low angle groups showed the change trend of low angle >average angle>high angle,the difference was not statistically significant(P>0.05).Conclusion In teenagers,the bone thickness of infrazygomatic crest in skeletal class Ⅱ is gradually thinned from cranial direction and rearward,the high angle is thinnest and the low angle is thickest.
4.Subglottic secretion drainage for preventing ventilator-associated pneumonia: a Meta-analysis

ObjectiveTo assess the efficacy of subglottic secretion drainage for preventing ventilatorassociated pneumonia.MethodsData of relevant randomized controlled trials (RCT) from January 1991 to June 2010 were collected,and data were split into two groups,namely draining group and non-draining group.Meta analysis of ventilator-associated pneumonia was carried out for finding the incidence and mortality in patients with mechanical ventilation using the methods recommended by the Cochrane Collaboration.ResultsSeven RCTs met the inclusion criteria and 1647 patients were enrolled.Subglottic secretion drainage reduced the incidence of ventilator-associated pneumonia ( OR =0.45,95% confidence interval [CI]:0.32 - 0.63 ),primarily by reducing early-onset pneumonia.But the mortalities of ventilator associated pneumonia were not significantly different between the two groups ( OR =1.03,95% confi dence interval [CI]:0.75 - 1.41 ).ConclusionsSubglottic secretion drainage appeared effective in preventing ventilator associated pneumonia among patients expected to require >48 hours of mechanical ventilation,but the mortality was unchanged.
5.Age distribution of prostate specific antigen and its correlation with blood glucose and lipid in the male elderly

ObjectiveTo investigate the age distribution of serum prostate specific antigen (PSA) and explore its correlation with fasting blood glucose(FBG)and lipid in the male elderly.MethodsThe levels of FBG,triglyceride (TG),low-density lipoprotein (LDL-C),high density lipoprotein (HDL-C) and PSA were reviewed in 2903 elderly men (aged ≥ 60 years) in our hospital.The subjects were classified into 6 groups by age,and PSA levels were compared among groups.The correlations of FBG,TG,LDL-C,HDL-C with PSA were analyzed using multiple linear regression analysis.Results The level of PSA had the trend to increase with age.Positive association of TG,LDL with serum PSA level,but negative correlation between HDL and serum PSA level were noted in the group aged 60-64 years (r=0.10,0.15,0.12,P<0.05).In the group aged 66-69 years,there was negative relationship between FBG and serum PSA(r= 0.10,P<0.05).No relationship could be found in other groups (P>0.05).ConclusionsPSA level may increase with age,but without relation with FBG,TG,LDL-C and HDL-C.
6.Expression of drculatory and pulmonary angiotensinⅡin rats with acute lung injury

Objective To investigate the changes in circulatory and pulmonary monary angiotensinⅡin rats with acite lung injury(ALI)and explore the role of angiotensinⅡin ALI.Method Thirty S-D rats were randomly divided into control group(n=6)and ALI group(n=24).The ALI group was further divided into four subgroups of observation at various intervals,3,6,9 and 12 hours after administration of lipopolysaccharide (LPS)into the femoral vein(each n=6).The indices rate,blood gas analysis,wet weight/dry weight(W/D)ratio of lung lobes,and pathological changes were successively observed at 3,6,9,and 12 hours after injury.The content of angiotensinⅡin lung tissue and blood plasma were detected at above set intervals by radioimmunoassay.Data of these assays were analyzed by one-factor analysis of variance.Results Compare with the control group,pH and PaO2 of arterial blood in ALI group decreased significantly(P<0.05)at all intervals and PaCO2 of arterial blood in ALI group decreased significanfly(P<0.05).at all intervals and PaCO2 of arterial blood and lung W/D weight ratio increased significantly(P<0.05),and scores of lung histopathology denoted the lung injuried(P<0.01).After injury of lung,angiotensin Ⅱ content increased markedly in lung homogenate and blood plasma (P<0.05).Angiotensin Ⅱ content in blood plasma reached peak value at 9 hours,and content of angiotensin Ⅱ in lung homogenates kept on increasing at allintervals of observation.Conclusions A large amount of angiotensin Ⅱ releases into lung tissue and blood plasma during ALI,suggesting systemic and pulmonary rennin-angiotensin systems are activated.
7.Clinical observation of qingre shugan tuihuang tang in the treatment of newly diagnosed hepatitis B with jaundice

Objective To study treatment effect of qingre shugan tuihuang tang in the treatment of newly diagnosed hepatitis B with jaundice.Methods88 patients newly diagnosed hepatitis B with jaundice in our hospital from August 2015 to December 2016 were randomly divided into control group and treatment group according to different modes of treatment, control group was given protecting liver and reducing enzyme, antivirus and other conventional therapy, treatment group added qingre shugan tuihuang tang on the basis of control group, changes of liver function indexes, changes of bilirubin, changes of clinical syndromes and treatment effect in the two groups were compared.ResultsSerum total bilirubin (TBIL), direct bilirubin (DBIL), Alanine transaminase (ALT) and prothrombin time (PT) in the two groups were significantly decreased (P<0.05), albumin was significantly increased (P<0.05), and rangeability of TBIL, DBIL, ALT and albumin in treatment group after the treatment were significantly higher than control group, the difference was statistically significant (P<0.05);hepatitis B virus DNA copy numbers in the two groups were significantly decreased (P<0.05), treatment group was significantly lower than control group, the difference was statistically significant (P<0.05);yellow tint of sclera and skin, gastric distention, rib-side pain, anorexia, drowsiness and fatigue symptom scores in the two groups were significantly decreased (P<0.05), decrease of treatment group was significantly higher than control group, the difference was statistically significant (P<0.05);total effective rate of treatment group was 97.73%, while the control group was 81.82%, the two groups had significant difference (P<0.05).ConclusionQingre shugan tuihuang tang in the treatment of newly diagnosed hepatitis B with jaundice can improve symptoms of jaundice significantly, improve liver function injury, effect is remarkable, its clinical application is very important.
8.Effect of glibenclamide on viability and acid-base equilibrium of glioblastoma cells

AIM: To investigate the effect of glibenclamide (Glib) on the viability and acid-base equilibrium of glioblastoma cells.METHODS: U251 cells and U87 cells were treated with Glib at different concentrations.The inhibitory rates were detected by CCK-8 assay.The effective dose was screened and the experiment was divided into control group and drug treatment groups.The migration ability was monitored by wound healing assay, and intracellular pH was detected by pH indicator fluorescent probe.The protein expression levels of inwardly-rectifying potassium channel 4.1 (Kir4.1) and monocarboxylate transport protein 1 (MCT1) were determined by Western blot.RESULTS: The half maximal inhibitory concentrations (IC50) of Glib for 48 h exposure of U251 cells and U87 cells were 400.20 μmol/L and 553.70 μmol/L, respectively.The effective inhibition doses of Glib for U251 cells were from the ranges of 100 μmol/L to 1 600 μmol/L, and those for U87 cells were from 50 μmol/L to 1 600 μmol/L in a concentration-dependent manner (P<0.05).Glib not only inhibited the migration (P<0.05) of U251 cells and U87 cells, which was negatively correlated with drug concentration (P<0.05), but also reduced the intracellular fluorescence intensity in experimental group (P<0.05), suggesting that with the increase in drug concentration, the intracellular pH decreased gradually (P<0.05).The protein expression of Kir4.1 and MCT1 was down-regulated by treatment with Glib, and was negatively correlated with concentration of Glib.CONCLUSION: Glib, a kind of potassium channel blocker, induces intracellular acidification via down-regulating the expression of Kir4.1 and MCT1, thus inhibiting the growth of glioblastoma in a certain dose range.
9.Establishment a standard real-time PCR for rapid detection of duck virus enteritis

The specific primers and Taqman probe were designed and synthesized according to the UL31 gene sequence available in GenBank(EF417996).The fragment is 76 bp.A Taq Real-time fluorescent quantitative PCR assay was established for detection of Duck Plague Virus DNA based on DNA copy obtained from attenuated vaccine strain as a positive template.The sensitivity of the test for DEV is 2.1 × 100-2.1 × 109 copies.The minimum detection limit is 2.1 copies(2.1 fg).Tissues,excrement and blood of 4 ducks challenged with DEV were detected repeatly 3 times by this method.All results were positive (3/3).The specificity of the assay was proven based on no amplification by detecting DNA from normal duck cells,duck viral hepatitis virus,Pasteurella,E.coli,S.gall inarum,Newcastle disease virus,goosling plague virus et ai.The whole process of the test could be completed within 4 hours.It brings about quantification detection of DEV DNA.
10.HOXB6-mRNA and its gene expression in the differentiation process of human cytomegalovirus-infected hematopoietic stem progenitor cells into granulocyte and erythrocyte progenitor cells

BACKGROUND: Is the inhibition of the hematopoietic stem progenitor cell (HSPC) proliferation and differentiation after human cytomegalovirus (HCMV) infection associated with abnormal expression of infected cell proliferated gene?OBJECTIVE: To observe the HOXB6-mRNA expression in the process of proliferation and differentiation of HCMV-infected HSPC into colony-forming unit granulocyte-macrophage (CFU-GM) and colony-forming unit erythroid (CFU-E).DESIGN: A controlled observation.SETTING: Laboratory for Molecular Biology, Affiliated Hospital of Luzhou Medical College, Lanzhou, Gansu Province, China.MATERIALS: All cord blood (CB) specimens were provided by the Obstetrics Department of Affiliated Hospital of Luzhon Medical College. They were collected from the umbilical vein of normal term neonates delivered spontaneously. All neonate mothers were healthy and HBS-Ag-negative. HCMV-IgM antibody revealed by routine ELUSA and HCMV-DNA checked by PCR were undetectable. Written informed consent for the laboratory measurements was obtained from each neonate mother, and the protocol was approved by the hospital's Ethics Committee. HCMV-AD169 strains were obtained from the Institute of Virology, Chinese Academy of Preventive Medicine. All-trans retinoic acid (ATRA, lot No. 20010126) was provided by Chongqing Huapont Pharm. Co., Ltd., China.METHODS: This study was performed at the Laboratory of Molecular Biology (state-level), Affiliated Hospital of Luzhou Medical College of Luzhou Medical College from April 2006 to April 2007. Cord blood mononuclear cells were separated for HSPC culture. According to different interventions, the study consisted of 4 groups. Control group: no HCMV virus solution was added and equal volume of culture medium was added instead. HCMV group: 105 PFU/mL HCMV-AD169 virus solution was added to the culture system. ATRA group: ATRA was added into the cultivation system at the final concentration of 60 μ mol/L. HCMV+ATRA group: ATRA was added into the HCMV group, and its final concentration was also 60 μ mol/L.MAIN OUTCOME MEASURES: In each group, cells were harvested on days 3,7 and 12. HOXB6 mRNA expression levels in CFU-GM and CFU-E were detected by real-time fluorescent-based quantification PCR.RESULTS: In the control group, both CFU-E and CFU-GM expressed HOXB6-mRNA. The HOXB6 mRNA expression was increased as a function of time. The HOXB6-mRNA expressed by CFU-E reached its peak level on day 12, while that expressed by CFU-GM reached its peak level on day 7. Compared to control group, the expression levels of CFU-E and CFU-GM HOXB6-mRNA genes in normal cord blood were significantly lower in the HCMV group (P<0.05)and significantly higher in the ATRA group (P<0.05) at each time point after HCMV infection. Furthermore, the expression levels were significantly higher in the ATRA+HCMV group than in the HCMV group at each time point(P<0.05-0.01).CONCLUSION: HOXB6-mRNA expression is stable and lasting in the proliferation and differentiation of HSPC into CFU-GM and CFU-E. HCMV could down regulate HOXB6 gene expression, and ATRA could up regulate HOXB6 gene expression.
