AIM To investigate the modification of GABA A receptor subunits in audiogenic seizure rat cortex and hippocampus when rendered tolerant to clobazam. METHODS Rats were administrated with clobazam for two weeks, resulting in tolerance to clobazam. GABA A receptor subunit ?1, ?3, ?5, ?2L and ?2S were assayed using quantitative competitive RT PCR in rat FrPaM and hippocampus. RESULTS In FrPaM,the content of mRNA encoding for ?1,?2L and ?2S were all significantly decreased (27%,43% and 37% respectively),whereas that of ?5 was significantly increased (73%) compared with the control. In hippocampus, ?1, ?2L, and ?2S mRNA content was significantly decreased (28%, 32% and 31% respectively). CONCLUSION The accomodated change in GABA A receptor subunit ?1, ?5,?2L and ?2S in FrPaM and hippocampus may be associated with the mechanism for tolerance to clobazam in audiogenic seizure rat.

Methods： Rats were administrated with flurazepam for two weeks, which resulted in tolerance to flurazepam, in the absence of behavioral signs of withdrawal. GABAA receptor subunits α1, α3, α5, γ2L and γ2S were assayed using quantitative competitive RT-PCR in rat frontparietal motor(FrPaM) and hippocampus. Results： In FrPaM, the contents of mRNA encoding for α1, α3,　γ2L and γ2S were all significantly decreased (24%,17%,35% and 45% respectively),whereas that of α5 was significantly increased (33%) compared with the control. In hippocampus, α1, γ2L, and γ2S mRNA contents were significantly decreased (33%, 35%and 27% respectively). Conclusion： The accomodated change in GABAA receptor subunits α1, α3, α5,γ2L and γ2S in FrPaM and hippocampus may be associated with the mechanism of flurazepam tolerance in audiogenic seizure rats.

Objective To observe the effect of acupuncture and herbs on risk factors and prognosis of transient ischemic attack (TIA).Methods 120 patients received treatment from March to May 2011 in Gaomi People's Hospital and the Hospital of Gaomi were randomly divided into three groups,with 40 cases in each.Acupuncture and Chinese medicine group was treated with needling Neiguan and administered with modified Tianmagouteng decoction,3 times/d; Chinese medicine group was treated with modified Tianmagouteng diction exclusively,3 times/d; Western medicine group was treated with Aspirin,0.1 g,1 times/d.The three groups were treated for 30 d of two therapeutic courses.TCM syndrome score,blood pressure,blood lipids,blood rheology,blood sugar changes in the three groups of patients were observed before and after the treatment.And follow-up of elapse rate and the incidence of stroke in a year were observed.Results After treatment,① in the acupuncture and Chinese medicine group: the total cholesterol was (5.21 ± 0.97)mmol/L,whole blood viscosity was [(4.26±0.66) 200 mPa · s,(5.69±0.15) 30 mPa · s,(9.72±2.33) mPa · s],fibrinogen was (2.12± 0.46)g/L,systolic blood pressure was (124.13±9.96) mm Hg,diastolic blood pressure was (79.23±7.50) mm Hg,blood glucose was (5.59 ±0.78) mmol/L; in the Chinese medicine group,the total cholesterol was (5.43± 1.04) mmol/L,whole blood viscosity was [(4.25±0.25) 200 mPa · s,(5.46±0.55) 30 mPa · s,(9.81± 2.01) 5 mPa · s],fibrinogenwas (2.32±0.33) g/L,systolic blood pressure was (129.78±14.25) mm Hg,diastolic blood pressure was (79.54± 7.65) mm Hg,blood glucose was (5.60± 0.75) mmol/L; in the western medicine group,the total cholesterol was (5.52± 1.16) mmol/ L,full blood viscosity was [(4.12±0.54) 200 mPa · s,(5.45± 0.65) 30 mPa · s,(9.09 ± 1.11) mPa · s],fibrinogen was (2.41 ± 0.44) g/L,systolic blood pressure was (129.83 ± 14.91) mm Hg,diastolic blood pressure was (80.72±7.47) mm Hg,blood glucose was (5.75 ± 0.61) mmol/L.all these results were improved significantly than those before the treatment in the same group (P＜0.05),and the acupuncture and Chinese medicine group improved better than the other two groups (P＜0.05).② In 1-year follow-up,TIA recurrence rate of stroke in the acupuncture group were lower than the other two groups.Conclusion Acupuncture and Chinese medicine treatment were not only be able to improve the clinical symptoms of patients with TIA and blood pressure,blood lipids,blood theology,blood sugar and other risk factors for TIA patients significantly,but also can effectively reduced the relapse rate and the incidence of stroke.

Sleep deprivation (SD) has been taken as an independent predictor for cardiovascular risks, which was closely related to the increased morbidity and mortality in coronary heart disease (CHD). In this article, after reviewing the impact of modern medical method sleep deprivation on CHD and studies on principle method recipe medicines for preventing and treating CHD, the authors observed the autonomic nerve dysfunction, hormonal metabolism dysfunction, endothelial dysfunction and inflammatory responses after sleep deprivation, which can cause or aggravate CHD. On the basis of the traditional Chinese medicine theories of "heart dominating the blood and vessels and the mind", the authors considered that traditional Chinese medicines can tonify heart and soothe the nerves, reducing all of the risk factors through multi-target and multi-pathway, and improve sleep and decrease the risk factors caused by sleep deprivation, which provides a new idea for the prevention and treatment of CHD.

Objective To study the relations between the susceptibility to AIDP and AMAN, two forms of Guillain-Barr? syndrome (GBS), and the frequency of HLA-A, B alleles. Methods A case-control research was done in 31 cases of AIDP, 33 cases of AMAN and 132 health individual controls. DNA was extracted from peripheral blood leukocytes by improved fast saltingout. HLA-A, B antigens were typed by DNA-based technology, PCR-sequence specific primers (PCR-SSP) method. In determination of allelic polymorphism by PCR amplification with SSP, oligonucleotide primers are designed to obtain amplification of specific alleles or groups of alleles. Assignment of alleles is based on the presence or absence of PCR amplified product, which may be detected by agarose gel electrophoresis. Results On research of HLA-A, B alleles polymorphism, it showed that HLA-A33 frequency was increased significantly in AIDP patients [22.6% vs 4.5%,corrected probability (Pc)=0.011]; related risk (RR) was 6.1; HLA-B15, B35 frequencies were increased (51.7% vs 20.8%, Pc =0.015; 34.5% vs 6.9%, Pc=0.000 8); RR was 4.1 and 7.1, respectively. Conclusions There are different distribution of HLA-A, B alleles in AIDP and AMAN, two forms of GBS. AIDP susceptibility is associated with HLA-A33, while AMAN is with HLA-B15, B35.

Qi in TCM is the most essential substance that makes up the body and maintains life activities. All vital substances in the body are transformed by constant motion and changes of qi. Qi in TCM mainly means full of functions. What is the basic material attribute of qi? We don't have a systematic study on it. Therefore, we combined the achievement of modern medicine, and explored further from the origin, functions, pathogeneses, and therapies of mitochondria and qi. Surprisingly, we found out that the origin of mitochondria was similar to that of qi. They are tiny substance constituting the human body. Secondly, the function of mitochondria is similar to that of qi. When the disorder of qi and mitochondria occurs, similar vital signs occur or the same reactions occur. These results suggested that the basic material attribute of qi might be mitochondria.

Objective: To clone and express the preferentially expressed antigen of melanoma (PRAME) gene in E.coli. Methods: The cDNA encoding human PRAME gene was extracted from human AML cell line HL-60 and amplified by RT-PCR, then the PRAME gene was inserted into plasmid PGEM-T-easy. After sequenced, it was cloned into the prokaryotic expression vector pGEX-4T-2 to construct a clone with high level expression and the recombinant plasmid was cloned in E.coli. Results:The protein product reached the highest level at 5 h after IPTG induction. Conclusion: Since PRAME is only expressed at low levels in a few normal tissues and encodes an antigen recognized by autologous cytotoxic T lymphocytes, while expressed at high levels in patients with leukemia, it might be a new targeting candidate for tumor immunotherapy.

Objective To understand the mental health status of residents aged 15 years old or above in Hainan Province,and and to analyze its influencing factors to provide a scientific basis for improving the psychological health level of populations.Methods A multistage randomsampling method was used to extract 3 295 residents in Hainan Province and perform the investigate on the mental health status.Results Among 3 295 surveyed residents,410 respondents had mental disease,accounting for 12.44％ of respondents total number.The total score of SCL-90 in the residents of Hainan Province was lower than that of the national norm(t=14.65,P=0.00).(2)The morbidity of mental disease had no statistical difference between different genders(wald x2 =0.27,P=0.60);which had statistical difference among marital status,environmental satisfaction and family harmony (wald x2 =46.53,P =0.00;wald x2 =51.98,P =0.00;wald x2 =89.47,P =0.00).(3) The non-conditional Logistic regression analysis results showed that the influencing factors entering the regression equation were marital status(OR=0.52),family type(OR =0.57;0.53),environment satisfaction(OR=1.26),family harmony(OR=1.31),health self-examination(OR=0.72),borrowing 5 000 yuan within 2 d when urgent need(OR=1.31) and general well-being(OR=0.86).Conclusion The factors influencing mental illness in residents are multiple,so synthetic measures should be taken for its prevention.
