Tianjin Medical Journal 2014;(6):536-539
Objective To explore expression of Beclin1, autophagy-related protein 1 (Atg1), mammalian target of ra-pamycin (mTOR) in squamous cell carcinoma (SCC) and adenocarcinoma of lung and to analyse relationship among these three factors. Methods Immunohistochemical stainning was applied to detect expression of Beclin1, Atg1 and mTOR in 82 samples of SCC and adenocarcinoma of lung (lung cancer group) and 17 samples of paraneoplastic normal lung tissues (con-trol group). Correlations of expression of Beclin1, Atg1 and mTOR were analyzed in SCC and adenocarcinoma of lung. Re-sults The expression of Beclin1 was significantly lower in lung cancer group than that in control group(52.44%vs 94.12%, P<0.01). There were significant differences of the differentiation, TNM stages and lymph node metastasis in lung cancer group compared with those in control group. And Beclin1 expression showed a decreasing trend with malignant and invasive of tumors. mTOR expression in lung cancer tissue with lymph nod metastasis is higher than that in the tissue without lymph node metastasis(P<0.01). Atg1 expression in lung cancer tissue with larger mass is less than that in lung cancer tissue with small lumps(P<0.01). Beclin1 Expression was positively correlated with the Atg1 expression in SCC(r=0.609,P<0.01). Beclin1 and Atg1 expressions were both negatively correlated with mTOR expression in SCC(r=-0.617,-0.444,P<0.01). Beclin1 expression was positively correlated with Atg1 expression in adenocarcinoma(r=0.458,P<0.01). Beclin1 and Atg1 expression were both negatively correlated with mTOR expression in adenocarcinoma(r=-0.497,-0.541,P<0.01). Conclu-sion Beclin1, Atg1 and mTOR expression showed a strong correlation in lung cancer, and clinicopathological change of lung SCC and adenocarcinoma may be related to autophagy.

Chinese Journal of Experimental Ophthalmology 2016;34(1):56-59
Background Neovascular diseases such as retinopathy of prematurity often leads to irreversible vision loss. The study on oxidative damage mechanism is becoming more and more important.Whether brain derived neurotrophic factor (BNDF) has protection to retinal ganglion cells(RGCs) has few research reports. Objective The study was to investigate the expression changes of BDNFin mouse retinas of oxygen-induced retinopathy (OIR).Methods Thirty SPF C57BL/6J immature rats were divided into OIR group and normal control group and to fifteen rats for each group.OIR models were established by raising 7-day-old (P7) mice together with maternal mouse in (75±3) % oxygen environment for 5 days and then returned to the normal air environment, and the mice of the normal control group were raised in the normal air environment. The P17 mice were sacrificed for retinal histopathological examination by hematoxylin and eosin staining, and the number of vascular endothelial cell nucleus extending the inner limiting membrane was counted.Whole retinal mounts were prepared after fluorescein isothiacyanate-dextran (FITC-dextran) (9 ml/kg) was retrosorbitally injected, and the distribution of retinal vessels was observed in P17 mice. The relative expression levels of BNDF in retinas were detected in P13, P15, P17 mice, and the results were compared between the normal control group and the OIR group. Results Histopathological examination showed that retinal inner limiting membrane was smooth in the normal control group, but a lots of vascular endothelial cell nucleus extending the inner limiting membrane were seen under the optical microscope in the OIR group. The number of the vascular endothelial cell nucleus extending the inner limiting membrane was 1.70±0.68 in the normal group and 45.3±3.13 in the OIR group, showing significant difference between them (t =86.5, P =0.00).Whole retinal mount revealed that normal retinal vessels and network-like capillaries were exhibited in the mice in the normal group, while tortuous vessels, capillary loss and non-perfusion areas were revealed in the OIR group on the whole retina mounts. The relative expressing levels of BDNF in retinas were 263.992±9.451 and 218.432±9.710 in P15 and P17 mice in the OIR group, which were significantly higher than 230.324±7.779 and 115.846±7.305 in the normal control group (t=14.2,42.3 ,P<0.05).Conclusions OIR can be inhibited by increasing the expression of BNDF.

BACKGROUND: Color Doppler flow imaging can exam the early myocardial disorder in type 2 diabetic patients. What does exercise tolerance test work for the examination of such disorder in combination with color Doppler flow imaging (CDFI) ?OBJECTIVE: To analyze in comparison the early evaluation on reduced diastolic function in left ventricle in patients with type 2 diabetes between the examinations of exercise tolerance test combined with color Doppler flow image and simple color Doppler flow imaging.DESIGN: Cases-controlled comparison and self-comparison.SETTING: Department of electrodiagnosis and department of Endocrinology in a municipal hospital.PARTICIPANTS: Thirty-six cases of inpatients with type 2 diabetes were selected from Department of Endocrinology of Shenyang Red Cross Hospital from March to December in 2004, of which, 25 cases were males and 11cases females. The diabetic patients included had no cardiac vascular complications and participated in the study in volunteer. Thirty-two patients who received annual routine health check at the same period were selected as the control, of which, 20 cases were males and 12 cases females.METHODS: Metronics treadmill exercise test equipment was used for exercise tolerance in two groups. Before exercise(at quiescent state) and after exercise tolerance, Vivid 4 CDFI was used to determine flow velocity at E and A peak values respectively, ratio between flow velocities at E peak value and A peak value as well as isovolumtric relaxation time (IRT).MAIN OUTCOME MEASURES: Comparison of cardiac functional indexes before and after exercise tolerance in two groups.RESULTS: Thirty-four diabetic patients accomplished exercise tolerance test, of which, 1 case presented frequent ventricular extrasystole, another one presented precordial pain and stopped the test. In the control, all of 32 cases ratio between flow rates of E peak value and A peak value was remarkably lower than that in the control (0. 90 ± 0. 25, 1.40 ± 0.30, P < 0.05 ); IRT was remarkably longer than that in the control [ (112. 07 ± 20. 16),imental group, the ratio between flow rates of E peak value and A peak value was remarkably lower than that in the control (0.62 ±0. 12, 1.28 ±0.87, P< 0.01 ); IRT was remarkably longer than that in the control[ (138. 10± 19.21), (97.37±9.61) ms, P <0.01].CONCLUSION: CDFI can supervise and evaluate at early stage the cardiac functional changes in type 2 diabetic patients. Due to the induction of exercise tolerance, the combination of exercise tolerance test and CDFI provides more accurate, objective and valuable conclusions at early stage.
