Objective To evaluate the predictive value of electrocardiogram in localizing the occlusion site in the left anterior descending coronary artery(LAD) in acute anterior myocardial infarction(AMI).Methods According to the coronary angiogram,all patients with AMI were divided into two groups:one(the PS group) was a collection of patients(n=61) whose occlusion sites were proximal to the first septal branch(S1) and the other(the DS group) was a collection of patients(n=40) whose occlusion site were distal to S1.Diagnostic sensitivity and specificity in prediction of the occlusion site in LAD were calculated based on the measurements and incidence of ST segments deviation on electrocardiogram(ECG).Results The sensitivity and specificity for prediction of occlusion site being proximal to S1 in LAD using ECG were as follow:43% and 85%(P=0.004) for ST elevation in lead aVR,16% and 97%(P=0.031) for ST elevation≥1.5 mm in lead aVL,39% and 85%(P=0.009) for ST depression≥1.0 mm in lead Ⅱ,23% and 98%(P=0.005)for ST depression ≥2.0 mm in lead Ⅲ,38% and 88%(P=0.006) for ST depression ≥1.0 mm in lead aVF,20% and 86%(P=0.037) for ST depression in lead V5,18% and 100%(P=0.005) for ST elevation in lead aVR together with ST depression in leads V5,30% and 93%(P=0.008) for ST elevation in lead aVR together with ST depression in leads V6.The sensitivity and specificity for diagnosis of occlusion site being distal to S1 were 53% and 90%(P=0.000)ST elevation or unchanged in lead Ⅲ,50% and 82%(P=0.001) for ST elevation in lead V5.There were no significant difference in infarction size and left ventricle ejection fraction between the two groups.Conclusion(1) ST elevation in lead aVR together with ST depression in leads V5 and V6,obvious ST depression in the inferior leads,and ST elevation in leads I and aVL were all indication of occlusion in LAD proximal to S1.(2) ST elevation or unchanged in inferior leads(especially in lead Ⅲ),and marked ST elevation in lead V5 indicated that the occlusion site in LAD was distal to S1.

Objective To study the impact of the high temperature and muggy weather on the people’s life and work. Methods The data of summer temperature and relative humidity (Jun-Aug) from 1971 to 2000 and that of 2006 were used and the characteristic of muggy weather and distribution in main cities, Chengdu, Guangyuan, Yibing, Suining and Dazhou in Sichuan basin were compared, the influence of the thermal environment on people comfort degree was evaluated by using temperature-humidity index. Results 177 high temperature days, 17 high temperature processes, 167 muggy weather days and 12 muggy processes were seen in 5 cities in Sichuan basin in the summer of 2006. The thermal environment of Chengdu and Dazhou mainly manifested the lighter heat, heat and lower comfort degree. At 2 p.m., the relative humidity weakened the comfort degree in Chengdu, but enhanced it in Dazhou. The duration of non-comfort degree in Dazhou was longer than that in Chengdu. Conclusion The relative humidity has a strong impact on people comfort degree and the combined use of temperature and relative humidity will be useful to evaluate the city thermal environment and people comfort degree.

OBJECTIVETo explore the feasibility of the transdiferentiation of human breast adipose-derived stem cells (hbASCs) into mammary epithelial-like cells after co-culturing in Transwell in vitro.

METHODSThe third passage hbASC and the HBL-100 cell line were co-cultured in a Transwell culture system for 15 days. The hbASCs were observed and identified by inverted phase contrast microscope and transmission electron microscopy, and immunocytochemistry staining in the induced and control groups.

RESULTSBoth the third passage hbASCs and the HBL-100 cell line cells could adhere and grow rapidly after co-culture in the Transwell system. After co-culture for 15 days, the morphology of some induced hbASCs changed into epithelial-like cells. Some induced hbASCs showed positive expression of CK18, CK19 by immunocytochemistry staining, and typical epithelium cells with microvilli, desmosomes and tonofilaments observed under TEM. The positive rate of CK18 and CK19 was (24.4 +/- 12.0)% and (21.6 +/- 16.4)% in experimental group, and (1.8 +/- 1.7)% and (1.1 +/- 0.6)% in control group.

CONCLUSIONThe data suggests that hbASCs may have the potential to transdifferentiate into human mammary epithelial-like cells after co-culturing in Transwell in vitro.

The starting material L-tryptophan reacted with 4-chlorobenzaldehyde via Pictet-Spengler condensation and followed by oxidation and decarboxylation to afford the 1-(4-chlorophenyl)-beta-carboline. The intermediate was further reacted with alkyl halogenide by N(9)-alkylation and N2-quaternarization to obtain 12 novel 1-(4-chlorophenyl)-beta-carboline derivatives. The chemical structures of all target compounds were characterized by elemental analyses, MS and 1H NMR spectra. The antitumor activities of the target compounds were evaluated by MTT method. The results demonstrated that N2-quaternarized compounds enhanced the antitumor activity significantly. In particular, compound 15 was found to be the most potent compound with IC50 values lower than 5 micromol x L(-1) against 6 human tumor cells. These results confirmed that the N2-alkyl or aralkyl substituent on the beta-carboline ring played an important role in the modulation of the antitumor activities.

Objective To identify the isolated rat bone marrow derived mesenchymal stem cells(MSCs),and evaluate the efficiency of adenoviral vector expressing human urokinase type plasminogen activator(uPA) in transfection of rat MSCs and its effect on proliferation of MSCs. Methods MSCs were isolated and purified by pasted wall purification,and were identified by immunicytochemistry.The transfection efficiency of uPA was detected by fluorescent microscopy,the expression of uPA in MSCs was detected by Western blotting,and the proliferation of MSCs was evaluated by MTT. Results The harvested MSCs exhibited the typical appearance of MSCs,and it was revealed by immunohistochemistry that the expression of MSCs markers CD29 and CD90 was positive,while that of CD34 and CD45 was negative.A tendency of increase in expression of green fluorescent protein(GFP) was observed with increase of multiplicity of infection(MOI).After transfection with AduPA for 72 h,the transfection efficiency reached(94.0?1.5)% at MOI of 80,and positive GFP cells could still be observed even after 7 d.The transfected uPA had no effect on the proliferation of MSCs. Conclusion MSCs are favourable genetic vectors to express uPA,and can be used for treatment of liver fibrosis.

Objective To review the clinical data of reduction mammoplasty by central pedicle technique,and to summarize all kinds of the complications to modify the technique for improvement of long term aesthetic effects.Methods The postoperative complications were analyzd and then an approach was used to investigate the pattern of the blood supply and the nerve distribution of breast.Based on the anatomical study,a modified double-circle reduction mammoplasty technique was designed to treat patients with hypertrophical breasts. Results With a follow-up for 3 months to 3 years,the patients who underwent this modified central pedicle technique,had an invisible scar,good projection,the better shape of breast and preserved their sensation of nipple-areola complex.Conclusions Modified central pedicle technique is a safe and reliable technique,especially ideal for Chinese women.The blood supply is rich and the sensation of nipple-areola complex is preserved.The fixation of the gland tissue is more important than the dermal-bra.

Methods The patients were randomly assigned to a control group and a treatment group. 30 patients in the treatment group were treated by SJG and 30 patients in the control group were given Cilostazo. Both groups were treated for 3 months. Lower extremity vascular color Doppler image, blood levels of interleukin-6 (IL-6) were measured before and after the treatment. Results The thickness of atherosclerosis (AS) plaque and arterial media were obviously reduced (t=3.376, 3.468, P<0.05) , serum levels of IL-6 were also be lowered obviously (t=3.345, P<0.05) in the treatment group. While these indexes remained unchanged in the control group. Conclusion SJG has remarkable action in reducing the thickness of AS plaque and media of artery. The mechanism might be related with its reducing serum IL-6 level.

Objective To observe the expressions and distribution of transient receptor potential cation channel 6 (TRPC6) and integrin-linked kinase (ILK) in the glomeruli of renal biopsytissue of patients with proteinuric kidney diseases,and to investigate the effect of TRPC6 over-expression on ILK in vitro.Methods The archival histological specimens of patients admitted to Tangdu hospital from 2012 to 2013,with 24-hour urinary protein over 1 g,were collected.The expressions and distribution of TRPC6 and ILK in the glomeruli of renal biopsy tissue were observed by immunohistochemistry.MPC5 podocytes were cultured in vitro and they were stimulated with 10-7 mol/L ADR for 12,24 and 36 h.The pcDNA3.1(+)-TRPC6 plasmid and pcDNA3.1(+) were transfected into MPC5 podocytes by liposome 2000 reagent to establish the TRPC6 overexpression group and the negative control group respectively.Western blotting was used to detect the expressions of TRPC6 and ILK protein.Results There were 14 cases of membranous nephropathy,13 cases of focal segmental glomerulosclerosis (FSGS),15 cases of membranoproliferative glomerulonephritis,12 cases of mesangial proliferative glomerulonephritis,10 cases of hyperplastic sclerosis nephritis,15 cases of IgA nephropathy,13 cases of purpura nephritis,15 cases of lupus nephritis,13 cases of hypertensive renal injury,14 cases of diabetic nephropathy and 9 cases of normal renal tissue included.In glomerulus,TRPC6 was expressed mainly in podocytes,and the expressions of TRPC6 in these renal tissues were higher than that in normal renal tissues (all P ＜ 0.05),except for hypertensive nephropathy.ILK was expressed in podocytes and the mesangial areas.The expressions of ILK in FSGS,lupus nephritis and diabetic nephropathy were higher than that in normal kidney tissue (all P ＜ 0.05),while the other renal tissues was high but showed no statistical difference with normal kidney tissue (all P ＞ 0.05).The expressions of TRPC6 and ILK were positively correlated in renal tissues of FSGS and diabetic nephropathy (r=0.906,P ＜ 0.001;r=0.783,P=0.001 respectively).The expressions of TRPC6 and ILK protein in 24 and 36 h stimulating with ADR were significantly higher than that in the control group (all P ＜ 0.05).The expression of ILK in the TRPC6 overexpression group was significantly higher than that in the normal control group (P ＜ 0.05).Conclusions The expressions of TRPC6 and ILK increase in the glomeruli of patients with kidney diseases with proteinuria being the main manifestation,especially in FSGS and diabetic nephropathy.The up-regulation of TRPC6 can increase the expression of ILK protein,which may be involved in podocyte injury.
