Objective: To clone the 5＇-flanking region of the human CD34 gene containing transcriptional regulatory sequence (TRS). Methods: According to the registered 5＇-flanking region of CD34 gene, two pairs of primers were designed and net-PCR was used to amplify 661 bp long TRS of CD34 gene. The CD34 TRS fragment was cloned into reported plasmid pEGFP-1. The role of the regulating the specific expression of recombinant plasmid pCD34 EGFP in hematopoietic and non-hematopoietic cells was observed. Results: Restrictive endonuclease identification and DNA sequencing provedthat the CD34 promoter cloned was consistent with the sequence reported to a large extent. It could induce the EGFP gene to express in hematopoietic cell line K562 specifically, while has no effect on hepatocellular carcinoma cell HepG-2. Conclusion: The cloned CD34 gene TRS has the effect of regulating gene expression specifically. The study established the fundament for the construction of specific gene expression vector used in hematopoietic system cells.

Orexin is a new neuropeptide related to diet intake and regulation of endocrine and metabolic functions. Rat model with insulin resistance (IR) was induced by high fructose diet and the extent of IR was assayed by hyperinsulinemic euglycemic clamp technique. The results showed that high fructose diet might induce IR and results in disturbance between orexin and leptin. Blood glucose, serum insulin, lipid metabolism and leptin may participate in the regulation of orexin.

OBJECTIVE:To ensure a long-term medication in children with asthma,prevent the attack of asthma and ensure safe,effective and correct drug use in these patients.METHODS:The pharmaceutical care for the asthma children was carried out through pharmacists’ involvement in assisting doctors to establish the treatment plan,carrying out medicine-use education and drug use consultation for patients,setting up the medicine-use record and so on.RESULTS & CONCLUSION:The practice of pharmaceutical care can markedly improve the compliance,safety and efficacy of drug use in asthma children as well as improving their quality of life.

Objective:To explore the interaction between gossypol and human serum albumin ( HSA) . Methods:The interaction of gossypol and HSA under physiological conditions was studied by fluorescence spectroscopy, and the molecular docking software was used to simulate the interaction. Results:The binding constant of gossypol and HSA at 293K and 303K was 2. 390 6 × 105 L·mol-1 and 3. 576 8 × 103 L·mol-1 , respectively. There was one binding site on HSA for gossypol. Hydrogen bond and Van Der Waals inter-actions were involved in the binding process. The binding of gossypol and HAS was closer to tyrosine residue in HSA. The molecular simulation analysis verified the above results. Conclusion: The gossypol-induced fluorescence quenching of HSA belongs to a static quenching procedure.

A strain named as WJ44 with capability of producing branched-chain aminotransferase was isolated from the soil, the enzyme activity is especially high when used leucine as a substrate. According to the physiological characteristics, biochemical characteristics and 16S rRNA sequence of WJ44, the strain was identified as Bacillus cereus. The optimal carbon source and nitrogen source were also determined as 20 g/L glucose, 15 g/L tryptone and 5 g/L beef extract, respectively. The optimal proportion of other component in the medium is corn steep liquor 15 g/L, KH2PO4 3 g/L, MgSO4?7H2O 0.5 g/L. The optimum original pH value is 7.0, the cultural temperature is 37℃, liquid volume is 20 mL/250 mL, the shaker’s rotating speed is 200 r/min. The highest leucine transferase activity of 45.787 U/mL was achieved after 10 h under the opti- mal condition, and the dry weight of cell is 8.643 g/L, increased by 54% and 10% respectively. This result shows that WJ44 is a branched-chain aminotransferase-production strain with fine capability.

Fed with high-fat diet and assessed by hyperinsulinemia-euglycemia clamp technique, rat models with insulin resistance were successfully induced. Compared with normal chow group ( NC ), serum concentrations of free-fatty acids(FFAs) and baseline insulin in high-fat diet group(HF) was higher( P＜0.05 ), the average glucose infusion rate from 60 to 120 min( GIR60-120 ) was lower( P＜0.01 ), and the expression of p-IRE-lα in the liver was higher( P＜0.05 ). Furthermore, the expression of p-IRE-1α in the liver was positvely correlated with the serum concentration of FFAs. All these data indicate that high-fat diet may induce endoplasmic reticulum stress in the liver by elevating serum concentration of FFAs, and may participate in the genesis of insulin resistance via p-IRE-1α.

To investigate the effects of Xinji' erkang (XJEK), a compound Chinese herbal medicine, on isoproterenol-induced ventricular remodeling in mice.

Objective To identify genes differentially expressed in the window of implantation and explore the molecular basis of the development of endometrial receptivity.Methods A subtracted cDNA library of the window of implantation was constructed by suppression subtractive hybridization(SSH) method.The screened clones of the subtracted library were sequenced and GenBank homology search was performed.The differential expression of ribosomal protein(RP)L7,RPL7 pseudogene(RPL7p),RPL19 and tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein zeta polypeptide(YWHAZ) were further confirmed by RT-PCR.Results After sequencing and GenBank homology search of 50 clones, 35 differentially expressed genes were detected in the window of implantation,of which 23 were known genes,and 12 were unknown genes.Some of the known genes have been proved to be associated with implantation,while others were firstly screened out by us.The results of RT-PCR confirmed that RPL7, RPL7p,RPL19 and YWHAZ were highly expressed in the window of implantation,0.75?0.21,1.72? 0.30,1.23?0.31,and 1.28?0.08,respectively.Conclusions SSH is a useful technique to detect differential expression genes and an effective method to clone novel genes.It provides a new method to investigate the molecular basis of the development of endometrial receptivity.
