1.Inhibitory effect of cathepsin B-RNAi-lentivirus on mouse retinal neovascularization

Background Retinal neovascularization disease is a group of threatening-vision eye diseases.Researches showed that cathepsin B is involved in angiogenesis.Exploring a drug which inhibit retinal blood vessels will provide the basis for the molecular mechanism of these diseases.Objective This study was to investigate the inhibitory role of cathepsin B-RNAi-lentivirus on retinal angiogenesis.Methods Sixty 7-day-old C57BL/6J mice were raised together with maternal mice in the closed box with the oxygen concentration of (75-2)% for 5 days to establish the retinal angiogenesis mouse models.The mice were then taken into the normal air environment for continuous raise and were randomized into 3 groups.NC-GFP-LV of 1 μl and the equal volume of cathepsin B-RNAi-lentivirus was intravitreously injected respectively in 40 eyes in the control group and the gene treatment group,and no drug was administered in the 40 eyes of the model group.The mice were sacrificed and retinas were obtained.Expression of cathepsin B protein in the retina was detected by Western blot assay (cathepsin B/β-actin).Real-time PCR was used to detect and compare the expression level of cathepsin B mRNA (2△△Ct).FITC-dextran was used to perform heart infusion for the retinal stretched preparation 5 days after intravitreously injection.Retinal neovascularization was examined by fluorescent angiography.Results The expression level (2-△△Ct) of cathepsin B mRNA was 0.74 ±0.12 in the gene treatment group,showing a significant decline in comparison with 1.66±0.17 and 1.58±0.29 in the model group and control group (q--0.746,1.588,P< 0.01).The expression level of cathepsin B protein (cathepsin B/β-actin) in the retina was 0.64±0.06,0.93±0.09 and 0.96±0.09 respectively in the gene treatment group,model group and control group,indicating a significant reduce in the gene treatment group (q =0.637,0.894,P<0.01).Distorted vessels were seen in the mice retinas of the model group with more branches and vascular anastomosis,and fluorescine leakage was exhibited under the fluorescence microscope.However,the vessels were regular with less branches and angiogenesis.Conclusions Cathepsin B-RNAi-lentivirus can effectively inhibit oxygen-induced retinal angiogenesis in mouse.

7.Adipose-derived stem cells for repair of urinary system injury:current situation and problems in application

BACKGROUND:Although there is no report of adipose-derived stem cells in ureteral repair, but with the deepening of the research of adipose stem cells, the differentiation conditions of adipose-derived stem cells to the vascular endothelial cells, smooth muscle cells and urothelial cells are more mature, and the experimental research of adipose-derived stem cells in the repair and reconstruction of kidney and bladder is also increasing.
OBJECTIVE:To summarize the adipose-derived stem cells research and its application in damage and repair of urinary system in recent years.
METHODS:The first author retrieved PubMed database and CNKI databases for articles relevant to adipose-derived stem cells in the repair of urinary system published between January 2001 to September 2013 using the keywords of“adipose tissue-derived stem cel/adipose tissue-derived stromal cells/ADSCs;tissue engineering;kidney;ureter;ureathra;bladder;urology”in English and Chinese, respectively. Final y, 52 articles were included for further analysis.
RESULTS AND CONCLUSION:Adipose-derived stem cells which can be found easily and have the unique advantages of multi-directional differentiation ability have been used for repairing and constructing the urinary system. Adipose-derived stem cells provide a new model of treatment for the urinary tract, which solves the traditional problems, including immune rejection, source of organs and ethical issues, and become an ideal cellsource in repair of urinary system. Accumulated data related to adipose-derived stem cells and its experiment and clinical application in repair of urinary system injuries have been reported. But before the cells are widely used in clinic, the fol owing problems need to be solved:its specific surface marker identification, specific conditions and control of celldifferentiation, mechanisms of action.
8.Relationship Between Free Radical Content and Tar Content in Cigarettes

Objective To study the methods of preparing samples, analyse free radical in smoke of cigarette and the relationship between free radical and the other harmful components in the cigarette smoke. Methods The contents of two types of free radical in smoke of 13 kinds of cigarettes were determined with electron spim resonance(ESR) and the self-made equipment. Results The analytic results showed that the main component was the free radical in gas state in all cigarette samples and it was about 10-30 times higher than the content of free radical in solid state. The content of free radical in gas state of cigarette made in China was lower than that made in foreign country. Conclusion The analysis method using ESR and the self-made equipment will be easily used in the related experiment; The level of free radical in gas and solid state is positively related to the level of tar and nicotine in the cigarette sample of same trademark but it is not the same in the samples of different trademark. Therefore, we can not to infer the content of free radial in cigarette smoke by the content of tar and nicotine in it,especially for the content of free radial in gas state. Using charcoal filter is benefit to reduce the level of free radical in cigarette smoke.
9.Effect of acupuncture and herbs on risk factors and prognosis of patients with transient ischemic

Objective To observe the effect of acupuncture and herbs on risk factors and prognosis of transient ischemic attack (TIA).Methods 120 patients received treatment from March to May 2011 in Gaomi People's Hospital and the Hospital of Gaomi were randomly divided into three groups,with 40 cases in each.Acupuncture and Chinese medicine group was treated with needling Neiguan and administered with modified Tianmagouteng decoction,3 times/d; Chinese medicine group was treated with modified Tianmagouteng diction exclusively,3 times/d; Western medicine group was treated with Aspirin,0.1 g,1 times/d.The three groups were treated for 30 d of two therapeutic courses.TCM syndrome score,blood pressure,blood lipids,blood rheology,blood sugar changes in the three groups of patients were observed before and after the treatment.And follow-up of elapse rate and the incidence of stroke in a year were observed.Results After treatment,① in the acupuncture and Chinese medicine group: the total cholesterol was (5.21 ± 0.97)mmol/L,whole blood viscosity was [(4.26±0.66) 200 mPa · s,(5.69±0.15) 30 mPa · s,(9.72±2.33) mPa · s],fibrinogen was (2.12± 0.46)g/L,systolic blood pressure was (124.13±9.96) mm Hg,diastolic blood pressure was (79.23±7.50) mm Hg,blood glucose was (5.59 ±0.78) mmol/L; in the Chinese medicine group,the total cholesterol was (5.43± 1.04) mmol/L,whole blood viscosity was [(4.25±0.25) 200 mPa · s,(5.46±0.55) 30 mPa · s,(9.81± 2.01) 5 mPa · s],fibrinogenwas (2.32±0.33) g/L,systolic blood pressure was (129.78±14.25) mm Hg,diastolic blood pressure was (79.54± 7.65) mm Hg,blood glucose was (5.60± 0.75) mmol/L; in the western medicine group,the total cholesterol was (5.52± 1.16) mmol/ L,full blood viscosity was [(4.12±0.54) 200 mPa · s,(5.45± 0.65) 30 mPa · s,(9.09 ± 1.11) mPa · s],fibrinogen was (2.41 ± 0.44) g/L,systolic blood pressure was (129.83 ± 14.91) mm Hg,diastolic blood pressure was (80.72±7.47) mm Hg,blood glucose was (5.75 ± 0.61) mmol/L.all these results were improved significantly than those before the treatment in the same group (P<0.05),and the acupuncture and Chinese medicine group improved better than the other two groups (P<0.05).② In 1-year follow-up,TIA recurrence rate of stroke in the acupuncture group were lower than the other two groups.Conclusion Acupuncture and Chinese medicine treatment were not only be able to improve the clinical symptoms of patients with TIA and blood pressure,blood lipids,blood theology,blood sugar and other risk factors for TIA patients significantly,but also can effectively reduced the relapse rate and the incidence of stroke.
10.GC-MS analysis of ether extracts from three species of Chaenomeles fruits

Objective:To analyze the chemical constituents of the ether extracts from the fruits of 3 species of Chaenomeles . Methods: The chemical constituents were analyzed by GC MS. Results: Sixty one constituents were firstly isolated and identified from the fruits of 3 species of Chaenomeles . Forty compounds in Chaenomeles lagenaria , thirty four compounds in Xuan mugua ( C.lagenaria from Xuanzhou) and 23 compounds in C.sinensis were isolated and identified respectively. There were 10 compounds in all 3 species of Chaenomeles , but their contents were very much different. They were acetic acid, benzaldehyde, n caproic acid, 14 methyl pentadecanoic methyl ester, glycerin, benzoic acid, 6,9 octadecadienoic methyl ester, squalane, stearic acid and oleic eicosyl ester. Conclusion: The composition and content vary with different species and district of Chaenomeles . The result may be useful for the evaluation of Chaenomeles fruits quality. [
