1.In vitro expansion of hematopoietic stem/progenitor cells from human umbilical cord blood.

Objective To separate and cultivate homo umbilical cord blood (UCB) hematopoietic stem cell (HSC) in vitro, and u-tilize bone marrow desmohemoblast stem cell as trophoblastic layer combined with cytokine to amplify umbilical cord blood hematopoietic stem/progenitor cell. Methods Ficoll lymph-cell separating medium density gradient centrifugalization was used to segregate UCBHSC.Bone marrow desmohemoblast stem ceil and cytokine were added, and the sum of NC cells and CD34 + cells was counted. Results The sum of NC cells amplified 75.2±15.0 times, and the sum of CD 34 + cells amplified 18.7±12.3 times. Conclusions It has significant effect on amplification of hematopoietic stem cell with bone marrow desmohemoblast stem cell and eytokine when HSC are cultured in vitro.

The human venous valve of the brachial,femoral and long saphenous veins wereexamined with light,transmission and scanning electron microscopy.The observationshows that the venous valve is composed of three functional layers covered withendothelium on both surfaces.A loosely structured layer is located underlying theendothelium.A network mainly containing randomly oriented elastic fibers was foundnear the side towards the lumen.To the side towards the venous wall,there is adense layer composed of eircumferentially and transversly oriented collagen bundles.Some smooth muscle cells extend from the wall of the vein to the base of thevenous valve.The elastic fibers and smooth muscle cells together with the collagenfibers contribute to the mechanical load-bearing performance of the valve and to thepassive closing and openning mechanism.In addition,the smooth muscle cells mightplay an active role in the normal functioning of the valve.The scanning and transmission electron microscopy of venous valve showdifferent arrangement of the endothelium.On the surface of the valve next to the wallof the vein,the endothelial cells are transversely arranged,while on the othersurface over which the current of blood flows,the cells are longitudinally arrangedin the direction of the current.These accord with the role of fluid mechanics.12 normal venous valves were tested by universal testing instrument (Instrontype 1122).The mean value of the maximum tension of the valve is 1 N.Theaverage value of the tensile ultimate strength is 10N/mm~2.

Phaffia rhodozyma is a good strain for astaxanthin production. An over-producing mutant YB-20-29 was obtained by means of Cs137-?ray and N-methy1-N'-nitro-N-nitrosoguanidin (NTG) treatment. The biomass for this strain by shake culture was 36.32 g/L, the pigment content was 1216.0 ?g/g, an increase of 308% compare to o-riginal strain. The astaxanthin content in broth was 30.9?g /mL. It was a potential strain for astaxanthin over-production.

Carbon-nitrogen lyases (E.C.4.3) are a group of enzymes that release ammonia, amidine or amino group etc, and also show ability to form double bond or ring structure. Specifically, enzymes forming amino group are called amine-lyases (E.C.4.3.3), which are critical in the industrial production of many medicine intermediates. In this review is a summary of four major amine-lyases in terms of their source, enzymatic characteristics and their applications in preparation of pharmaceutical intermediates.

6.Using distance comparison method to build pharmacophore model of epidermal growth factor receptor inhibitors.

OBJECTIVETo build 3D-pharmacophore model of epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors using distance comparisons method and design novel EGFR inhibitors.
METHODSThirteen typical EGFR inhibitors were selected, and their biologically active conformations were obtained by using DOCK5.0 program, then 3D-pharmacophore model of EGFR inhibitors was built using distance comparisons method.
RESULTSValidation of the 3D-pharmacophore model was carried out and novel structures with potential inhibitory activity were selected by means of 3D-searching and docking method.
CONCLUSIONThis method can improve hit rate of lead compounds discovery and can be used to design novel EGFR inhibitors.

7.Synthesis and protective effect of ligustrazine intermediates against CoCl2-induced neurotoxicity in differentiated PC12 cell.

Ligustrazine, one of the major effective components of the Chinese traditional medicinal herb Ligusticum Chuanxiong Hort, has been reported plenty of biological activities, such as protect cardiovascular and cerebrovascular, neuroprotection and anti-tumor, et al. Because of its remarkable effects, studies on structural modification of ligustrazine have attracted much attention. Ligustrazine synthetic derivatives reported in recent decades are mainly derived from four primary intermediates (TMP-COOH, TMP-OH, TMP-NH2, HO-TMP-OH). To explore the neuroprotection activitiy of ligustrazine intermediates, six ligustrazine intermediates (2, 5, 8, 11, 12, 13) were synthesized and their protective effects against CoCl2-induced neurotoxicity in differentiated PC12 cells were studied. The target compounds were prepared via different chemical methods, including oxidation, substitution, esterification and amidation without changing the structure nucleus of ligustrazine. Compared with TMP (EC50 = 56.03 micromol x L(-1)), four compounds (2, 5, 12 and 13) exhibited higher activity (EC50 < 50 micromol x L(-1)) respectively, of which, compound 2 displayed the highest protective effect against the damaged PC12 cells (EC50 = 32.86 micromol x L(-1)), but target compounds 8 and 11 appeared lower activity (EC50 > 70 micromol x L(-1)). By structure-activity relationships analysis, the introduction of carboxyl, amino to the side chain of ligustrazine and appropriately increase the proportion of ligustrazine may contribute to enhance its neuroprotective activity, which provides a reference for the design, synthesis and activity screening of relevant series of ligustrazine derivatives in the future.

8.Study on time-series analysis and forecast models on lung cancer incidence in Qidong, 1972 - 2001.

OBJECTIVETo explore the lung cancer incidence rates from 1972 to 2001 and utilize varieties of models in forecasting trend up to 2010 in the city of Qidong, Jiangsu in order to provide baseline data for its control and prevention.
METHODSUsing data from the cancer registry office in Qidong, we tried to reveal the trends of lung cancer incidence by analyzing the time-series on trends extrapolation, exponent smoothness, Box-Jenkins model etc. We also compared the prognostication precision, endow differ power, and established assembled forecast model.
RESULTSData showed that there had been a rising trend of lung cancer from 1972 to 2001 and would still probably be on the increase in the future. The rate of male and female attained to 70 per 100 000 and 20 per 100 000, predicting that there would be a respective 33 percent and 10 percent increase in 2010.
CONCLUSIONSAccording to analysis of forecast models, it was right to prognosticate lung cancer incidence from time-series and improve forecast precision through developing combination models. The result of combination seemed close to ARIMA models which suggested that it could serve as a chief way to forecast the incidence of lung cancer.

9.A study on the integrated application of risk matrix and analytic hierarchy process in risk assessment of vaccination operation

Objective To evaluate the risks of vaccination operation in order to provide scientific basis for policy makings of vaccination operation.Methods The risk matrix and analytic hierarchy process approach were used to evaluate the risks of vaccination operation,including the possibility,severity,risk weight,and risk level.Results Seven items for the first level and thirty one items for the second level vaccination risk factors were determined.In the first level,E level risk (Extremely serious risk ) was vaccination implementation risk,and H level risk (high risk ) was the risks of pre -notification and health education and risks of vaccine and cold chain management.Vaccination implementation risk accounted the largest weight (weight coefficient=38.95%).In second level,E level risk was three inspection and seven verification before vaccination,and H level risk were personnel professional quality,personnel responsibility,pre -inspection,informed before vaccination, vaccination route, site and dose, vaccination operation, post vaccination notification and retention,and three inspection and seven verification before vaccination,accounted the largest weight (weight coefficient=10.14%).The weight of the logical consistency of the test results were satisfactory (consistency ratio<0.1 ).Conclusion The integrated application of risk matrix and analytic hierarchy process in risk assessment of vaccination operation helps to further regulate vaccination services and has application and promotion value.
10.SNP marker and allele-specific diagnostic PCR for authenticating herbs of Perilla.

AIMTo authenticate all the varieties of Perilla (single-species genus), to analyze sequences of rDNA ITS regions and single nucleotide polymorphism (SNP) within them and based on these, to design allele-specific diagnostic PCR primers.
METHODSThe rDNA ITS regions of the perilla varieties were sequenced and analyzed by Clustal X 1.8, MEGA 3.0. Allele-specific diagnostic PCR primers that can authenticate all the perilla varieties were designed based on SNPs loci.
RESULTSThe length of rDNA ITS sequences of perilla varieties ranged from 612 to 615 bp in size, including ITS1 (230 -232 bp), 5.8S (179 bp) and ITS2 (203 -204 bp). The GC content is about 61.5% - 61.9%. There is not only SNPs in non-coding region ITS1 and ITS2 (ncSNP), but also three coding SNPs (cSNP) loci in the conservative region of 5.8S. All the SNPs have only two allele loci polymorphism. The cSNP in 5.8S is related to the morphology variation among the varieties. Allele-specific diagnostic PCR primers have been designed according to SNPs loci to authenticate accurately all the seeds and leaves of Perilla varieties.
CONCLUSIONSNPs in rDNA ITS region can be used as an effective molecular markers to authenticate all the varieties of Perilla.
