1.Tumor lysis syndrome in solid tumors

Tumor lysis syndrome (TLS) occurs frequently in the chemotherapy of patients with hematologic malignancies; however, it is rarely reported in solid tumors. Because of the latent incidence, TLS is vulnerable to misdiagnosis or missed diagnosis, leading to a poor prognosis. TLS is characterized by hyperuricemia, hyperkalemia, hyperphosphatemia and hypocalcaemia, with some major complications such as acute renal failure and cardiac arrhythmias. Therefore,the key treatment strategies usually refer to appropriate prophylactic measures for high-risk patients, early diagnosis and aggressive therapy. This paper reviews 87 cases of TLS reported in the English literature and discusses its incidence, prevention and treatment.
2.Display of human decay-accelerating factor on the surface of yeast

Objective To display efficient folding hDAF on the surface of yeast. Methods The hDAF open reading frame was cloned by PCR from DAF- pBluescript M13-(Amp+)plasmid, then subcloned into the yeast surface displayed vector pYD1.The recombinant vector was transformed into yeast cells EBY100.Flow cytometric analysis was carried out to evaluate direct binding of anti-DAF mAbs onto the surface of yeast cells displaying DAF. Results Three mAbs against DAF different epitopes could bind onto DAF displayed on the surface of yeast.Conclusion Efficient folding DAF can be displayed on the surface of yeast potentially leading to the development of novel applications involving DAF yeast display.
3.Relationship between expression of Kiss-1 and nm23 and lymph node metastasis in breast cancer

Background and purpose:Kiss-1 and nm23 have been identifi ed as tumor metastasis suppressor genes,and they have been associated with the metastatic potential of breast cancer.The purpose of this study was to evaluate the relationship of Kiss-1 and nm23 expression with lymph node metastasis in breast cancer.Methods:The expression of Kiss-1 and nm23 protein was detected by immunohistochemistry in 70 patients with breast cancer.Results:The positive rate of Kiss-1 and nm23 were 62.86% and 68.57%,38.46% and 50.00% in breast cancer patients with lymph node metastasis,markedly lower than the 77.27%,and 79.55% in patients without lymph node metastasis(P

6.Study on genetic microarray for detection of katG mutations associated with Mycobacterium tuberculosis resistance to INH

Objective To establish and evaluate a gene microarray for determination katG mutations of Mycobacterium tuberculosis isolates associated with resistance to isoniazid(INH).Methods A panel of probes were designed and gene chips were prepared by dotting.Mycobacterium tuberculosis isolates resistance to 5 drugs was determined by proportional dilution methods.Amplicons of Mycobacterium tuberculosis isolates were detected by our chip and sequenced.Results The drug resistance rate of the isolates to at least one of the anti-tuberculosis drugs was 70.8%(97/137).45 strains out 137 Mycobacterium tuberculosis isolates was resistant to INH(32.8%).katG was successfully amplified from 100% of the susceptible strains and 88.9%(40/45)resistant strains.4 of 45 INH resistant isolates' katG were deleted.27 of 40(67.5%) katG has been detected to have katG 315 codon mutations.The mutations were 315 AAC(Asn,13/40), ACC(Thr,6/40),ACA(Thr,4/40),ATC(Ile,2/40),AGC(Arg,2/40).The mutation rate of katG analyzed by gene chips we prepared were identical to katG sequencing.Conclusion The gene microarray techniques we developed for determination of Mycobacterium tuberculosis resistance to INH are specific, sensitive and may be used as an alternative in clinical laboratory.
7.Evaluation of left ventricular dyssynchrony in patients with coronary heart disease by speckle tracking imaging

ObjectiveTo assess the clinical values of the longitudinal strain delay index(LSDI) and the delay of time in peak anterospetal wall to posterior wall strain(TAS-POST),which are analyzed by speckle tracking imaging,was discussed in the article.Methods The study included 39 patients with acute myocardial infarction,37 patients with coronary heart disease but not acute myocardial infarction which was confirmed by selective coronary angiography and 30 age-gender-related volunteers.Two-dimensional echocardiography was performed to collect dynamic images of left ventricular apical long axis view,two-chamber view,four-chamber view and parasternal short axis mitral view,papillary view and apical view.The time to minimal systolic longitudinal,radial,circumferential strain (Tssl,Tssr and Tssc) were measured from the start point of QRS wave of electrocardiogram.The left ventricular segmental standard deviation (Tssl-SD,Tssr-SD,Tssc-SD) and maximal difference (Tssl-Dif,Tssr-Dif,Tssc-Dif) were calculated.The left ventricular dyssynchrony was defined as TAS-POST≥130 ms and LSDI≥25%.ResultsThe dyssynchrony parameters in the acute myocardial infarction group were significantly increased compared to the control group ( P <0.001 or P <0.05 ).The parameters in the myocardial ischemia group were also higher than those in the control group ( P <0.05).Linear regression showed that LSDI was positive correlated with TAS-POST( r =0.676,P <0.05).In acute myocardial infarction group,LSDI was more sensitive in the detection of left ventricular dyssynchrony than TAS-POST ( P < 0.05 ).Conclusions Left ventricular dyssynchrony can be evaluate accurately by speckle tracking imaging.LSDI and TAS-POST are quantitative parameters for the evaluation of left ventricular dyssynchrony in coronary heart disease.
8.Effect and safety of Reteplase for Injection combined with reduced glutathione in treatment of acute ST segment elevation myocardial infarction

Objective To research the effect and safety of Reteplase for Injection combined with reduced glutathione in the treatment of acute ST segment elevation myocardial infarction.Methods Patients with acute ST segment elevation myocardial infarction (90 cases) in Jianli People's Hospital from February 2015 to February 2016 were selected and divided equally into A,B and C groups according to different treatment methods.Totally 30 patients in group A were given conventional therapy,30 patients in group B were given reduced glutathione for treatment on the basis of conventional treatment,and the other 30 in group C were given combined therapy of reteplase and reduced glutathione on the basis of conventional treatment.The vascular recanalization rate,improvement of effective indicators including creatine kinase isoenzyme (CKMB),troponin Ⅰ (cTnⅠ),left ventricular end diastolic diameter (LVEDd) and left ventricular ejection fraction (LVEF),oxidative stress kinase including superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px),and incidence of adverse events of patients were compared among the three groups.Results After thrombolysis,the vascular recanalization rate of group C at different time points (2 h,6 h and 12 h) showed significant difference compared with those of groups A and B (P < 0.05).After the treatment,the effective indicators of the three groups were both significantly improved (P < 0.05),and group C improved more significantly than groups A and B (P < 0.05).After the treatment,the SOD and GSH-Px of groups B and C both significantly improved than group A (P < 0.05),but there was no significant difference between groups B and C.After treatment,the incidence of adverse events of group C was significantly lower than those of groups A and B (P < 0.05).Conclusion Reteplase for Injection combined with reduced glutathione has significant curative effect in the treatment of acute ST segment elevation myocardial infarction,which can effectively improve the cardiac function and inhibition of oxidative stress.It is of higher security but with lower incidence of adverse events.
9.Free medial sural artery perforator flap for the tissue defect in the upper or lower limbs

Objective To report 11 cases with the tissue defects of their upper or lower limbs repaired with the anastomsed medial sural artery perforator flaps.Methods The free medial sural artery perforator flaps,with length of 8 cm to 15 cm and width of 6 cm to 14 cm were used for tissue defect reconstruction of the distal upper or lower limbs in 11 cases,including 6 females and 5 males.The flap was harvested from the ho- mo-lateral calf,confined between the posterior-medial edge of the tibia and the middle line of the calf and a- bove the distal half part of the medial sural muscle,with a same axis of this muscle.Results Ten cases survived very well,which was relatively thin,and the donor site can be acceptable.One case resulted in a complete flap necrotized and covered with a split skin graft.No obvious motor function defect was observed of the donor leg.Conclusion The anastomsed medial sural artery perforator flap is alternative donor flap for the upper or lower limb tissue defect repair,especially for the defect in the hand or foot.
10.Expression of human hDAF in CHO cells and its decay-accelerating activity

Objective To obtain Chinese hamsterovary (CHO) cell line expressing human decay accelerating activity (hDAF) stably and to observe the protective effect of hDAF on heterologous cells under the circumstance of complement activation. Methods The eukaryotic expression vector DAF pcDNA3.1 was constructed and then transfected into CHO cells by lipofection. Monoclones of cells expressing hDAF stably were screened by the method of limiting dilution. hDAF expression was detected by flow cytometry. The decay accelerating activity of hDAF was determined by assay of C3 deposition and 51Cr release. Results The expression vector DAF pcDNA3.1 was successfully constructed, and monoclones of cells expressing hDAF were obtained. CHO cells expressing hDAF could decrease C3 deposition and attenuate the killing effect of activation of the complement system. Conclusion We have obtained CHO cell clones expressing hDAF stably, which is helpful for the further studies of the relationship of the structure with the functions of hDAF.
