1.Therapeutic effect of microwave radiation combined with liposome Adriamycin transarterial embolization on liver VX2 tumor in rabbits

Objective To assess the therapeutic effect of microwave radiation combined with Adriamycin liposome on liver VX2 tumor in rabbits. Methods VX2 tumor pieces were successfully implanted into liver lobes of rabbits with liver tumors formation. All rabbits were divided randomly into 4 groups (each n=8). Group A underwent transcatheter arterial chemoembolization (TACE) with Adriamycin, group B were treated with Adriamycin liposome, group C were treated with microwave radiation combinate with Adriamycin, and group D were treated with microwave radiation combinate with Adriamycin liposome. Spiral CT scans was performed to measure size of liver tumors at 1 day before operation and 14 days after operation, and liver function was tested at 1 day before operation, 1 day and 7 days after operation. Breathing frequency, heart rate and body temperture were measured before microwave radiation processing of microwave radiation, at 30 min after microwave radiation and 1 h after microwave radiation. All experimental animals were scarficed on the 15th day after operation and followed by pathologic and histologic examination of the tumor and after the operation with correlative comparisons. Results There was no significant difference in volumes and liver function among 4 groups at 1 day before operation. At 14 days after operation, the average tumor volume was (6.02±1.21) cm3 in group A, (5.74±1.43) cm3 in group B, (3.26±0.37) cm3 in group C and (1.89±0.14) cm3 in group D. There was no significant difference of liver function among 4 groups. After microwave radiation the rabbit's vital signs were stable. The intratumoral necrosis was more significant in group D than in the others (P<0.05). Conclusion Adriamycin liposome treatment for liver tumor via vascular interventional method combined with microwave radiation can significantly inhibit the growth of liver VX2 tumor in rabbits. Adriamycin liposome is a kind of effective pracputium in interventional chemoembelization, and the united therapy is safe and acceptable.

Objective To study the effects of Tripterygium Hypoglaucum Hutch(THH) and its extract(THW-4) on neointimal formation of rabbit carotid arteries after balloon angioplasty.Methods After injury of the common carotid arteries was set up by balloon angioplasty,the rabbits were divided into 3 groups and were given different treatment protocol including gastrogarage of THH(n=9),associated gastrogavage of THH with local administration of THW-4 into the injured arteries(n=9) and 0.25% DMSO saline local administration as the control(n=9).Neointimal fomation was evaluated on days after the procedure by proliferating all nuclear antigen(PCNA) and Evans Blue staining,and quantitative histomorphometric analysis was performed on 28 days.Results The PCNA-positive index in the neointima at day 14 after the procedure was reduced in the THH gastrogavage group and the THH/THW-4 group compared with the control(17.68%?5.27%,6.60%?2.52% vs 40.37%?7.12%,P
4.Angiogenesis induced by micropocket assay on rat cornea

AIM: To explore the skills and characteristics of corneal neovascular model in rat induced by micropocket assay. ·METHODS: Nine eyes of nine Sprague-Dawley rats were studied. Pellets made of vascular endothelial growth factor (VEGF), poly-2-hydroxylethylmethacrylate and sucralfate were implanted into the corneal stroma nocloser than 1mm from the limbus. Biomicroscopic features of corneal neovascular were observed on 1,3, 5, 7th day after the implantation. ·RESULTS: On day 1 after operation, the limbal vessels were dilated, with no angiogenesis appeared. On day 3, angiogenesis began to invade peri-cornea with a brush shape, the area of CNV was (2.23±0.59) mm2. On day 5, new vessels reached the lower margin of pellet densely, the area of CNVwas (6.81±1.35)mm2. On day 7, new vessels continued to elongate, parts of them extended as loops toward the pellet, the area of CNV was (8.92± 1.79)mm2. Neither hyphema or other complications occurred.·CONCLUSION: Corneal neovascular induced by micropocket assay in rat grows steadily, with no complication, and is suitable for quantitative researches.
5.Cytological study on rat corneal new blood vessel after alkali burn

Background Microvessels are composed of two interacting cell types: endothelial cells and pericytes. Over the past decades, studies of corneal angiogenesis have concentrated mainly on endothelial cells, while interest in pericytes has lagged behind. Objective The present study aimed to investigate the recruitment of vascular endothelial cells and pericytes in rat corneas after alkali burn. Methods Corneal alkali burn models were established in the right eyes of 36 adult SPF SD rats by putting 4 mm medicators containing a 1% 1 mol/L NaOH solution at the central corneas for 30 seconds, and 3 matched normal rats were used as controls. Corneas were excised 1,2,3,5,7 and 10 days after surgery. Frozen sections that parallel with the corneoscleral limbus were constructed. Double immunofluorescence staining was used to observe the dynamic expression of CD31 and α-smooth muscle actin (α-SMA) in corneal tissue for the evaluation of the number of endothelial cells and pericytes. The pericyte coverage index (PCI) was calculated to quantify the recruitment of pericytes to neovascular sites. The use of experimental animals followed the Statement of Association for Research in Vision and Ophthalmology. Results CD31 was expressed in the superficial stromal layer of the cornea on the 1 st day, showing the presence of red fluoresence. The positive cell number for CD31 was gradually increased with the passage of time and proceeded into the deep stromal layer from days 2 through 5 but decreased after that. However,α-SMA was positively expressed on the 2nd day in the cornea after alkali burn with the presence of green fluorescence. The positive cell number for α-SMA was less than those of CD31 throughout the experimental period. The PCI was 0, 16.07%, 11.95%, 43.84%, 73.97% and 86. 21% , respectively, 1,2,3,5,7 and 10 days after surgery. Conclusion Pericytes recruitment to corneal new vessels may play a key role in the stabilization and maturation of angiogeneis.
6.Emphasizing clinical investigation on anti-inflammatory therapy of dry eye

Inflammation is a defensive reaction and the most common pathological manifestation of dry eye.In addition,excessive inflammatory response is considered to be the most common pathogenic factor and main cause of dry eye.Currently,the active mechanism of anti-inflammatory drugs has been well-known,and topical antiinflammatory therapy for dry eye is exerting a role at certain extend.However,some adverse responses of these drugs are emerging during the treating procedure.Therefore,it is emphasized that a large sample size of and multicenter randomized-controlled clinical trial is needed to identify the different effects of various anti-inflammatory drugs for different types of dry eye diseases,which will offer a basis for standardized anti-inflammatory treatment for dry eye.
7.Public distribution analysis for clinic features of the patients with Budd-Chiarri syndrome from 1997 to 2006

Objective:To study the clinic epidemic of Budd Chiari Syndrome in Shandong Province,so as to research the causes and pathogenesis.Material and Methods:To collect the credible clinic data of 1592 patients in Shandong Province from 1997 to 2006.According to the pathologic location,patients were divided into four types—hepatic small vein(HSV),hepatic main vein( HMV),inferior vena cava(IVC) and HMV combined with IVC(CHI).And the age,sex,medical record and clinical feature were descripted.Results:The cases increased in the last decade years,especially in adults .The incidences in male and female were equal.The cases of the IVC type were the most(57.2%,911/1592),then CHI type(28.5%,454/1592),HMV type(13.4 %,214/1592) and HSV type(0.8%,13/1592).The course of the HSV type was acute .In HMV type,most of the course was chronic,and IVC type and CHI type were chronic.The HSV type had clear cause,and 19.6% of the HMV type were related with coagulation abnormalities.Causes of IVC type and CHI type were not clear.Conclusion:In Shandong Province,the main types are IVC type and CHI type,with special clinic character and unknown causes.
8.Chronic prostatitis during puberty and the effects of pelvic floor biofeedback therapy

Objective To investigate the features of chronic prostatitis during puberty(CPP)and the effects of pelvic floor biofeedback therapy.Methods Totally,25 CPP children (mean age,16 years) and 15 children (mean age,16 years) with normal lower urinary tract as controls were included.In CPP group,NIH-CPSI scores were evaluated,expressed prostatic secretions (EPS) were examined,and bacterial culture was done;and CPP patients were categorized based on the definitions of NIH types.In both groups, urodynamic examination was performed,including evaluation of uroflow curve,maximum flow rate (Q_(max)), post-voiding residual urine (PVR),detrusor-sphincter dyssynergia (DSD),maximum detrusor pressure (P_(det,max))and maximum urethral closure pressure (MUCP).CPP patients underwent biofeedback therapy, and 10 weeks later the effects were assessed.Results In CPP group,NIH typing showedⅡ,ⅢA andⅢB in 1,3 and 21 cases,respectively.Before treatment in CPP and control groups,the incidence of staccato voiding (20 cases vs 1 case),DSD (22 cases vs 1 case),Q_(max)(10.7?3.7 vs 15.0?4.3ml/s),PVR (7.7?4.1vs 3.2?2.6ml),P_(det,max)(115.1?33.6vs 76.8?16.6cm H_2O)and MUCP(176.5?45.7 vs 86.2?28.5cm H_2O)all showed significant differences between the 2 groups(P<0.05).In CPP group,the differences in pain(4.6?2.2 vs 2.1?1.6),urination (7.9?2.0vs 2.2?1.7),life impact (9.4?2.2vs 2.6?2.1)and total scores(22.0?5.2vs 7.0?4.2) of NIH-CPSI and Q_(max)(10.7?3.7 vs 14.9?5.6) between pre-and post-biofeedback were significant (P<0.05).Conclusions The main type of CPP is categoryⅢB.The primary symptom is voiding disorder,which leads to greater psychological stress in patients.Children with CPP have pelvic floor dysfunctions and multiple abnormal urodynamic param- eters.The short-term effect of biofeedback strategies for CPP is satisfactory.

10.The role of negative-regulatory factors of toll-like receptors signal pathways in the immunological pathogenesis of Kawasaki disease

Objective To investigate the role of negative-regulatory factors of toll-like receptors (TLRs)signal pathways in immunological pathogenesis of Kawasaki disease(KD).Methods Thirty-two chil- dren with Kawasaki disease and 16 age-matched healthy children were studied.Reverse-transcription PCR (RT-PCR)and real-time PCR were used to evaluate the mRNA expression levels of toll-like receptor 4(TLR4), MD-2,MyD88,IRAK-4,TRAF6,T1/ST2,IRAK-M,Triad 3A,and proinflammatory factors such as IL-1?, IL-6,IL-8 and TNF-?,in peripheral blood monocytes/macrophages(MC).The expression of TLR4 protein in MC was analyzed by flow cytometry.Results①Compared with the control group,the mRNA levels of TLR4, MD-2,MyD88,IRAK-4 and TRAF6 in KD group were up-regulated significantly(P<0.01),and the expression level of TLR4 protein was also found to be up-regulated in KD group during acute phase.It was detected that expression levels of TLR4 protein in KD with coronary artery lesion(KD-CAL~+)was significantly higher than that of KD without coronary artery lesion(KD-CAL-)[flow cytometry:(6.5?1.7)% vs(11.9_+2.4)%,P<0.01].②The expression level of negative-regulatory factors such as IRAK-M and Triad3A were significantly up-regulat- ed in acute phase of Kawasaki disease,while the mRNA levels of IRAK-M and Triad3A in KD-CAL~+ group was found to be significantly lower than those of KD-CAL~- group(P<0.01).No difference of T1/ST2 mRNA expres sion level was detected among all groups(P>0.05).③The expressions of proinflammatory eytokines such as IL-1?, IL-6,IL-8 and TNF-?in monoeytes/macrophages during acute phase of Kawasaki disease were higher than those of the control group(P<0.01),and expression of proinflammatory cytokines in KD-CAL~+ group was significantly higher than that of KD-CAL~- group.Conclusion Relative insufficient expression of negative-regulatory factors, such as IRAK-M and Triad3A,maybe correlate with immunological pathogenesis of Kawasaki disease.
