Objective To evaluate the effects of restraint stress on behaviors of mice with focal cerebral ischemia. Methods Male mice were first subjected to left middle cerebral artery occlusion/reperfusion. Repeated restraint stress (2 hours/day for 2 weeks) was given from 1 week after surgery. At the end of stress scheme, sucrose solution consumption test, open-field test and forced swimming test were performed. The study of quality control of Chinese medicine has always been the hot and the difficulty spot of the development of traditional Chinese medicine (TCM), which is also one of the key problems restricting the modernization and internationalization of Chinese medicine. Multivariate statistical analysis is an analytical method which is suitable for the analysis of characteristics of TCM. It has been used widely in the study of quality control of TCM. Multivariate Statistical analysis was used for multivariate indicators and variables that appeared in the study of quality control and had certain correlation between each other, to find out the hidden law or the relationship between the data can be found,.which could apply to serve the decision-making and realize the effective quality evaluation of TCM. In this paper, the application of multivariate statistical analysis in the quality control of Chinese medicine was summarized, which could provided the basis for its further study.

4.Clinical significance of p170, bcl-2, MRP by flow cytometry in acute leukemia

Objective To explore the role of p170, MRP, bcl-2 in clinical drug- resistance and their correlation. Methods There are 44 acute leukemia(AL) patients, among them ANLL is 29, ALL is 15.The expression of p170, MRP, bcl-2 were detected. At last, the patients were divided into two groups: CR and NR according to efficacy after two periods standard chemotherapy. Results Overexpression of p170, MRP,bcl-2 were found in AL (P <0.05). The expression of p170 and MRP were lower in CR group than that in NR groups in AML and ALL; there were 22 of 24 patients expressed one or two proteins in NR groups and the expression of p170 in patients was concordant with that of MRP to certain extent (P <0.01). The overexpression of bcl-2 was related with the worse prognosis. The expression of bcl-2 was lower significantly in CR group than in CR and NR (P<0.05). Conclusion p170, bcl-2, MRP are all associated with clinical MDR. High expression of these proteins is an unfavorable factor to prognosis. The expression of MRP and p170 showed coincidence, but showed no significance with expression of bcl-2 in untreated patients, it indicated that the drug-resistance mediated by p170 was different from bcl-2.
5.Effect of acupuncture and herbs on risk factors and prognosis of patients with transient ischemic

Objective To observe the effect of acupuncture and herbs on risk factors and prognosis of transient ischemic attack (TIA).Methods 120 patients received treatment from March to May 2011 in Gaomi People's Hospital and the Hospital of Gaomi were randomly divided into three groups,with 40 cases in each.Acupuncture and Chinese medicine group was treated with needling Neiguan and administered with modified Tianmagouteng decoction,3 times/d; Chinese medicine group was treated with modified Tianmagouteng diction exclusively,3 times/d; Western medicine group was treated with Aspirin,0.1 g,1 times/d.The three groups were treated for 30 d of two therapeutic courses.TCM syndrome score,blood pressure,blood lipids,blood rheology,blood sugar changes in the three groups of patients were observed before and after the treatment.And follow-up of elapse rate and the incidence of stroke in a year were observed.Results After treatment,① in the acupuncture and Chinese medicine group: the total cholesterol was (5.21 ± 0.97)mmol/L,whole blood viscosity was [(4.26±0.66) 200 mPa · s,(5.69±0.15) 30 mPa · s,(9.72±2.33) mPa · s],fibrinogen was (2.12± 0.46)g/L,systolic blood pressure was (124.13±9.96) mm Hg,diastolic blood pressure was (79.23±7.50) mm Hg,blood glucose was (5.59 ±0.78) mmol/L; in the Chinese medicine group,the total cholesterol was (5.43± 1.04) mmol/L,whole blood viscosity was [(4.25±0.25) 200 mPa · s,(5.46±0.55) 30 mPa · s,(9.81± 2.01) 5 mPa · s],fibrinogenwas (2.32±0.33) g/L,systolic blood pressure was (129.78±14.25) mm Hg,diastolic blood pressure was (79.54± 7.65) mm Hg,blood glucose was (5.60± 0.75) mmol/L; in the western medicine group,the total cholesterol was (5.52± 1.16) mmol/ L,full blood viscosity was [(4.12±0.54) 200 mPa · s,(5.45± 0.65) 30 mPa · s,(9.09 ± 1.11) mPa · s],fibrinogen was (2.41 ± 0.44) g/L,systolic blood pressure was (129.83 ± 14.91) mm Hg,diastolic blood pressure was (80.72±7.47) mm Hg,blood glucose was (5.75 ± 0.61) mmol/L.all these results were improved significantly than those before the treatment in the same group (P<0.05),and the acupuncture and Chinese medicine group improved better than the other two groups (P<0.05).② In 1-year follow-up,TIA recurrence rate of stroke in the acupuncture group were lower than the other two groups.Conclusion Acupuncture and Chinese medicine treatment were not only be able to improve the clinical symptoms of patients with TIA and blood pressure,blood lipids,blood theology,blood sugar and other risk factors for TIA patients significantly,but also can effectively reduced the relapse rate and the incidence of stroke.

Isocitrate dehydrogenase 1 (IDH1), a key rate-limiting enzyme in tricarboxylic acid cycle,is found within the cytoplasm and peroxisomes.In recent years, IDH1 mutation is discovered in most gliomaand its large scale metabolites, 2-hydroxyglutarate, may be involved in tumor as potential carcinogens, and isshown to be closely related to better patients' survival as well.IDH1 mutation is anticipated to be a novel genet-ic biomarker which will guide the treatments of clinical glioma.
7.Spinal muscular atrophy combined with respiratory distress type Ⅰ but no respiratory failure:a case report

Objective To explore the diagnosis and differential diagnosis of spinal muscular atrophy with respiratory distress type Ⅰ (SMARD1). Method The clinical data, results of gene detection, and follow-up information of a girl diagnosed with SMARD1 were retrospectively analyzed, and related literatures were reviewed. Results The girl was born by cesarean section due to oligohydramnios. After birth, she was transferred to neonatology department for poor feeding and response, and diagnosed with neonatal sepsis, infectious shock, disseminated inravascular coagulation and atypical purulent meningitis. She was discharged after one month of treatment. However, at 2 months old, she presented contracture of ankle joint, abnormal liver function, and myocardial damage. At 6 months old, she had obvious reduced muscular tension and development retardation. At 8 months old, the SMA gene was detected and it was normal. At 9 months old, The panel gene of peripheral neuropathy was detected and found 2 heterozygosis mutations in IGHMBP2 gene, exon8 c.1061-2A>G and exon12 c.1708C>T, which came from her father and mother respectively. Locus of exon12 c.1708C>T has been reported to be associated with the disease, and the other is a shear mutation. The diagnosis of SMARD1 was confirmed by the clinical and gene detection. The girl, 2-year-old now, suffered with recurrent respiratory tract infections, but had no respiratory distress or no respiratory failure yet. Conclusion The clinical phenotype of SMARD1 is complex and diverse. This case is the first domestic case comfirmed by gene detection.
8.The impaired activity of sensory afferent nerves in diabetes mellitus of rats in acute myocardial ischemia and reperfusion

Objective The aim of the study was to investigate the diabetic neuro-degeneration and its changes in neuroreaction to myocardial ischemia and reperfusion,by evaluation of the altera-tion of noxious thermal threshold and expression of substance P (SP),calcitonin gene related peptide (CGRP)in dorsal root ganglia in upper thoracic segments (T1-5 )in diabetic rats.Methods Thirty two male Sprague-Dawley rats,weighing 180-200g,were randomly divided into control group (group C)and diabetic group (group DM),1 6 rats in each group.rats in DM group were fed with high sug-ar-fat diet for 14 weeks and were given streptozotocin (STZ,35 mg/mg,i.p.)at the end of the 4 th week,to set up diabetes experimental model.The animals in control group were fed with standard la-boratory diet.Tail flick latency to thermal stimulation was measured weekly.At the end of 10 weeks after administration of STZ,diabetic rats (and rats in control group)were further divided into myo-cardial ischemia-reperfusion group (group IR)and sham operation group (group Sham).The left an-terior descending branch of coronary artery was occluded for 30 min followed by reperfusion for 120 min,establishing myocardial ischemia-reperfusion.The histological immunofluorescence assay and Enzyme-linked immunosorbent assay (ELISA)were carried out to evaluate the changes of the expres-sions of CGRP and SP in the dorsal root ganglia.Results The tail flick latency was significantly in-creased in group DM,compared to the group C (P < 0.01).The immunoreactive materials for CGRP and SP in the sensory neurons in dorsal root ganglia of upper thoracic segments (T1-5 )were markedly declined in group DM (P <0.01 or P < 0.05).Furthermore,levels of SP and CGRP were signifi-cantly lower in the DRG of the group IR after myocardial ischemia-reperfusion,compared to that in the group sham (P <0.01).Conclusion Diabetes causes sensory denervation and obvious reduction of expression of SP and CGRP in the sensory neuron innervating heart during myocardial ischemia-reper-fusion,indicating impairment of adaptive reactivity of neuro-endocrine function of cardiac sensory nerves.

As an important incentive for cardiovascular disease, cancer, diabetes and other chronic diseases, obesity after cardiovascular disease and cancer has been listed as the third largest threat to human health by World Health Organization. Recent research has shown that obesity is associated with colorectal cancer, esophageal cancer, kidney cancer, endometrial cancer, however, it has not been fully disclosed that the mechanism could lead to an increased risk of tumors. Leptin and adiponectin are a kind of hormone secreted by adipose tissue, which involve in the energy metabolism, and play an increasing significant role in the tumors' affecting factors.
10.The effect of miR-21 on the secretions of IFN-γ and IL-17 from PBMC in type 1 diabetic patients

Objective To investigate the effect of miR-21 on the secretion of IFN-γ and IL-17 from peripheral blood mononuclear cells (PBMC) in type 1 diabetic patients (T1D). Methods The expression levels of miR-21 in PBMC from 60 T1D patients and controls by quantitative real-time quantitative polymerase chain reaction (qRT-PCR) were detected and the secretions of IFN-γ and IL-17 from T1D PBMC after transfection with miR-21 were detected by enzyme linked immunosorbent assay. Results The expression of miR-21 in PBMC from T1D patients was significantly lower than the control (P<0.01). The levels of IFN-γand IL-17 from T1D PBMC after transfection with miR-21 were remarkably downregulated compared to the control (P < 0.05 or P < 0.01). Conclusion Compared with that at the healthy control group, the expression of miR-21 in PBMC from T1D patients is decreased. MiR-21 could suppress the secretions of IFN-γand IL-17 from PBMC in T1D patients.
