1.Effects of interleukin 1β on the expression of prostaglandin E_2 in cultured airway fibroblasts

Objective To explore the role of IL-1β in inhalation injury repairment through its effects on the expression of prostaglandin E_2 in cultured airway fibroblasts of mouse. Methods The cultured airway fibroblasts in vivo from male Kunming species mouse were randomly divided into control group and treatment group. The superna-rant and cells were collected at different time after treated with IL-1β. The expressions of PGE_2, COX-2 and mPGES1 protein in supernatant and cells were measured by ELISA and Western Blot. Results ①PGE_2 levels in the cultured airway fibroblast supematant at 8 h(148.2±28.3) ng/L,16 h(267.6±45.4)ng/L,24 h(210.5±38.6) ng/L and 48 h(198.7±36.5) ng/L were higher than that in the control group (P < 0.01), and peaked at 16 h;COX-2 ex-pressions at 8 h(0.478±0.029) COX-2/β-actin, 16 h(0.672±0.047) COX-2/β-actin,24 h(0.617±0.036) COX-2/β-antin, and 48 h (0.593±0.034) COX-2/β-actin were higher than that in control group (P < 0.01), and peaked at 16 h;mPGES1 at 8 h(0.300±0.018) mPGES1/β-actin, 16 h(0.549±0.034) mPGES1/β-actin, 24 h (0.497±0.030)mPGES1/β-actin and 48 h(0.443±0.026)mPGES1/β-actin were higher than that in the control group(P < 0.01), and peaked at 16 h. ②PGE_2 in 0.1 μg/L IL-1β group (142.9±2.3) ng/L, 1.0 μg/L IL-1β group (267.6±45.4) ng/L and 10.0 μg/L IL-1β group (587.3±106.9) ng/L were higher than those in the control group (58.5±16.0) ng/L (P < 0.01), and there was significant difference among the three groups (P < 0.01);COX-2 in 0.1 μg/L IL-1β group(0.525±0.031) COX-2/β-actin, 1.0 μg/L IL-1β group(0.672±0.047) COX-2/ β-actin and 10.0 μg/L IL-1β group(1.012±0.064)COX-2/β-actin were higher than that in the control group (0.309±0.019) COX-2/β-actin (P < 0.01), and there was difference among the three groups (P < 0.01);mPG-ES1 in 0.1 μg/L IL-1β group(0.380±0.021) mPGES1/β-actin, 1.0 μg/L IL-1β group (0.549±0.034) mPG-ES1/β-actin, and 10.0 μg/L IL-1β group(0.879±0.045) mPGES1/β-actin were higher than those in the control group(0.199±0.012)(P<0.01),and there was difference among the three groups(P<0.01).Conclusions IL-1β may upregulate PGE_2, COX-2 and mPGES1 expressions in airway fibroblast, indicating that IL-1β influences PGE_2 synthesis through COX-2 and mPGES1 expression and participates in airway inhalation injury course. Airway fibroblasts may be a main source cell of inflammatory mediators in injury repair.

Inhalation injury seriously threatens the survival and quality of life in burn and trauma patients. So far there is no breakthrough in the treatment of inhalation injury. A significant advance has been witnessed in the experimental study of the use of stem cells in the treatment of lung injury in recent years. In this paper, according to the results of our study in the systemic transplantation of bone marrow mesenchymal stem cells for the treatment of inhalation injury, the effect of mesenchymal stem cells on anti-inflammatory process and repair of lung tissues in inhalation injury, and its possible mechanisms are reviewed.

3.The effects of enteral nutrition with recombinant human growth hormone onimmunological function and inflammatory responses in severe burned rats

Objective: To investigate the effects of enteral nutrition with recombinant human growth hormone (rhGH) on immunological function and inflammatory responses in severe burned rats. Methods: Sixty-four SD rats inflicted by 30% TBSA Ⅲ degree scalds were randomly divided into enteral nutrition with rhGH (ENGH) and enteral nutrition (EN) groups. Two groups animal were fed isocaloric enteral nutrition. Lymphocyte subsets, serum albumin, transferrin, serum levels of immunoglobulins,endotoxin,IL-6,TNF-? were determined on the 1st,4th,7th and 10th day postburn. 4.Gastrointestinal stromal tumors: A pathomorphologic and immunohistochemical study of 77 cases

Purpose To investigate the pathomorphological and immunohistochemical features of gastrointestinal stromal tumors (GIST). Methods Light microscopy was used to study the morphologic characteristics of 77 cases of GIST. The expression of c-kit(CD117), CD34, vimentin, SMA and S-100 protein were detected in all cases with S-P immunohistochemical method. Results GIST tended to have a higher degree of cellularity and lesser degree of cytoplasmic eosinophilia than observed in classic leiomyomas. The neoplastic cells of GIST were spindle, epithelioid or both in different proportion. Focal to prominent cytoplasmic vacuolization was often seen. The histological spectrum included interlacing fascicles, diffuse sheets, storiform and palisading pattern. The formation of cell clusters was rather special. Stromal and vascular hyalinization was common. GIST primary in the mesentery tended to have a higher potentiality of malignancy. The positivity of CD117 and CD34 were 90%, 92%, respectively. Conclusion The characteristic histological features exist in GIST. The detection of CD117 and CD34 is helpful to differential diagnosis.
5.Effect of lung protective ventilation strategy on oxygenation and pulmonary inflammatory response in dogs with severe smoke inhalation injury

Objective To observe the effect of lung protective ventilation strategy on oxygenation and pulmonary inflammatory response in dogs with severe smoke inhalation injury.Methods Twelve local healthy male dogs were selected and anesthetized underwent endotracheal intubation, the time controlled smoke was applied to replicate the model of severe smoke inhalation injury, and they were divided into two groups according to the random number table: conventional ventilation group (CV group) and protective ventilation group (PV group), each receiving corresponding ventilation mode for 8 hours respectively. The blood gas analyses were detected before injury, immediately after injury and at ventilation for 2, 4, 6, 8 hours. The contents of tumor necrosis factor-α (TNF-α), interleukin-10 (IL-10) in serum were detected by the enzyme-linked immunosorbent assay (ELISA) at each time point. Animals were killed after 8-hour ventilation, and different parts of the lung tissues were obtained for pathological examinations of lung tissues and evaluation of injury scores. The levels of inflammatory factors as TNF-α and IL-10 in lung homogenates were measured by ELISA.Results The arterial partial pressure (PaO2) levels in CV and PV groups were significantly decreased after injury compared with those before injury [mmHg (1 mmHg = 0.133 kPa): 57±19 vs. 128±31, 58±15 vs. 126±22, bothP < 0.01]. Compared with those before injury, the pH values and arterial carbon dioxide partial pressure (PaCO2) levels after injury in two groups had no statistically significant differences at each time point (bothP > 0.05). At 6 hours ventilation, PaO2 level in PV group was significantly higher than that in CV group (mmHg: 121±11 vs. 105±11,P < 0.05). The comparisons of PaO2 levels, pH values and PaCO2 levels at 2, 4, 8 hours of ventilation between the two groups showed no statistically significant differences (allP > 0.05). The histopathological changes revealed that there were alveolar tissue edema and inflammatory cells infiltration in both groups, the degree of severity in CV group was more prominent and its pulmonary tissue injury score was higher than that in PV group (3.68±0.22 vs. 3.27±0.35, P < 0.05). The serum levels of TNF-α and IL-10 after smoke injury were significantly increased in CV and PV groups [TNF-α (μg/L): 4.32±1.13 vs. 0.35±0.11, 4.51±2.02 vs. 0.41±0.08; IL-10 (ng/L): 16.73±2.31 vs. 4.27±0.56, 18.39±3.15 vs. 4.03±1.07, allP < 0.01]. Compared with CV group, the levels of TNF-α were significantly lower at 6 hours and 8 hours of ventilation [6 hours (μg/L): 2.62±0.34 vs. 3.65±1.08, 8 hours (μg/L): 3.02±0.31 vs. 4.21±1.27, bothP < 0.05), while the contents of IL-10 were obviously increased in PV group [6 hours (ng/L): 21.07±2.95 vs. 16.11±3.02, 8 hours (ng/L): 23.57±2.69 vs. 18.28±3.21, bothP < 0.05]. The content of TNF-α in lung homogenate in CV group was significantly higher than that in PV group (μg/L: 5.85±2.57 vs. 3.08±1.17,P < 0.05), but the content of IL-10 in lung homogenate of CV group was markedly lower than that in CP group (ng/L: 19.64±3.16 vs. 24.05±2.09, P < 0.05).Conclusion Lung protective ventilation strategy can effectively improve oxygenation and pulmonary inflammatory response in dogs with severe smoke inhalation injury, thus the lung injury is alleviated and the strategy has protective effect on damaged lung tissues.
