Objective:To detect the expression of chemokine receptor CCR5 in human invasive ductal breast carcinoma and explore the role of CCR5 in breast cancer.Methods: Thirty samples of invasive ductal breast carcinoma and 10 samples of breast fibroadenoma were analyzed separately by immunohistochemical staining for CCR5 expression. MIP1?,MIP1? and RANTES concentrations were measured by ELISA in the sera of patients. Results: The expression of CCR5 was detected in invasive ductal breast carcinoma (the positive rate was 53.3%) and was not detected in breast fibroadenoma. 2. Influence of occlusal force change on apoptosis of periodontal ligament fibroblast and expression of proliferating nuclear antigen

Objective: To study the influence of occlusal force change on apoptosis of periodontal ligament fibroblast (PDLF) and expression of proliferating nuclear antigen (PCNA) in the fibroblasts, so as to explore the possible mechanism for remodeling of periodontal ligament. Methods: Animal model of occlusal force change was established by extracting the right first, second and third maxillary molars in male SD rats. The rats were sacrificed at 6, 12 hours and day 1, 2, 3, 5, 7, 14, and 28 d after extracting the teeth(n = 6), and their alveolar bone tissues were harvested. H-E staining was used to observe the morphologic changes of the alveolar bone tissues. TUNEL method and immunohistochemistry were used to detect apoptosis and PCNA expression in PDLF. The rats with normal occlusal forces were used as control (n=6). The results were assigned a mean score based on the percentage and the intensity of cells positively stained for TUNEL and PCNA. Results: Animal model of occlusal force change was successfully established. H-E staining showed that the control group had more pycnotic periodontal ligament, orderly fibres, and flatter alveolar bone than the model group. TUNEL result showed that the cell apoptosis reached its peak at 12 h after extracting the teeth (252.67±6.62), significantly higher than that-of control group (7.17±2.32, P<0.01), then gradually decreased to normal level on day 28 after extraction (8.50±1.87). Immunohistochemical staining showed that the expression of PCNA had a similar tendency of apoptosis:PCNA expression peaked on day 3 after extraction (246.00±7.21) and then gradually decreased to normal level (5.67±2.16, P<0.01). Conclusion: Changes of occlusal force can facilitate the PDLF apoptosis and promote PDLF proliferation, contributing to the remodeling of periodontal ligament.
3.Cardioprotection of anisodamine in patients with right coronary artery acute myocardial infarction during percutaneous coronary intervention

Objective To investigate the protective efficacy of anisodamine against heart injury after ischemia reperfusion in patients with right coronary artery acute myocardial infarction during percutaneous coronary intervention,and to explore the possible mechanism. Methods Totally 80cases with ST elevation myocardial infarction (STEAMI) were received primary percutaneous coronary intervention (PCI).All patients were randomly divided into 2 groups:anisodamine group (40 cases),5 mg anisodamine was administered when the blood flow recovery after the balloon extension,and control group (40 cases) received saline. The rhythm and rate of heart were observed through the electrocardiogram (ECG) monitor in 30 min after the blood flow recovery.The pressure of aorta were observed through the pressure sensor connected to aorta through catheter.TIMI flow grade (TFG)and corrected TIMI frame count (cTFC) were used to evaluate coronary flow during PCI.Electrocardiography was performed to analysis ST segment resolution 30 min and 24 h after PCI.Venous blood was obtained before and 30 min,1 h,2 h,4 h,24 h and 48 h after PCI to test the levels of superoxide dismutase (SOD),malonyldialdehyde (MDA),creatine kinase isozymes (CKMB) and cardiac troponin I(cTNI) in plasma. Results There were 20 cases(50.0%)of sinus bradycardia,7cases(17.5 %) of high degree atrioventricular block (AVB),20 cases (50.0 % )of frequent ventricular premature,14 cases (35.0%) of paroxysmal supraventricular tachycardia, 5 cases (12.5%) of ventricular fibrillation,19 cases(47.5 %) of hypotension,3 cases (7.5 %)of no-reflow phenomenon in anisodamine group which were lower than control group,31 cases (77.5%)of sinus bradycardia,14 cases(35.0%) of high degree AVB,17 cases(72.5%) of paroxysmal supraventricular tachycardia,8 cases (42.5%) of ventricular fibrillation,30 cases (75.0%) of hypotension,6 cases of no-reflow phenomenon (P<0.05).There were more ST fall>50% after rise in anisodamine group than control group after PCI 30 min,but no difference was found after 24 h. The CKMB and cTNI peak value occurred earlier in anisodamine group than control group (P<0.05).The level of SOD was increased,MDA decreased in anisodamine group compared with control group after PCI 30 min and 24 h (both P<0.05). Conclusions The use of anisodamine as the blood flow recovery after the balloon extension can reduce no-reflow phenomenon,various of arrhythmia and hypotension during PCI.The underlying mechanism involves the inhibition of oxygen radicals and the improvement of microcirculation.

5.Packing effects of different hemostatic materials during functional endoscopic sinus surgery: histocompatibility comparison

BACKGROUND:At present, the most common materials for clinical hemostasis are absorbable tampons and Vaseline gauze. OBJECTIVE:To compare the packing effects of absorbable tampons and Vaseline gauze in functional endoscopic sinus surgery. METHODS:Totaly 100 sinusitis patients, 57 males and 43 females, aged 16-61 years, with a history of 5 months to 23 years, were randomized into two groups: patients in the observation group were treated with absorbable tampons and patients in the control group treated with Vaseline gauze during functional endoscopic sinus surgery. Patient's discomfort, bleeding when the filer was extracted at 48 hours, and nasal bleeding within postoperative 24 hours were compared between the two groups. RESULTS AND CONCLUSION: No adverse reaction related to absorbable tampons occurred in the observation group, indicating that the absorbable tampon has better histocompatibility. Patient's discomfort, bleeding when the filer was extracted at 48 hours after operation, and nasal bleeding within postoperative 24 hours were significantly improved in the observation group compared with the control group (P < 0.05). These findings suggest that the absorbable tampons has a better hemostatic function, which can effectively reduce postoperative nasal bleeding and relieve postoperative discomfort in patients with sinusitis.
6.New drug delivery systems and the function of camptothecin

Camptothecin is known to possess a broad spectrum of anticancer activities, however due to its high toxicity and low solubility in water, camptothecin is severely limited in clinic. To optimize the effects of camptothecin in the treatment of malignancies, besides the structural modification and reconstruction of camptothecin, the establishment of new drug delivery systems becomes a hot topic in the camptothecin research. This paper aims to summarize the advanced progress in the delivery system and function of camptothecin at recent years. The advances in improving the poor solubility, lactone ring instability, controlled release systems and targeting delivery systems of camptothecin are also reviewed. As the related methods and technologies are improved and developed, camptothecin will be a group of prospective anticancer drugs.

Objective To investigate the TNF-? expression in hepatic tissue of Otsuka Long-Evans Tokushima Fatty(OLETF) rats(an insulin resistance model) with abnormalities of glucose-lipid metabolism and effect of metformin on TNF-? expression.Methods OLETF rats were randomly assigned to two groups(OLETF and OLETF/M).The OLETF/M group was treated with metformin [100 mg/(kg?d)],the OLETF group and age-matched LETO(nondiabetic Long-Evans Tokushima Otsuka rats) group were fed with placebo.Twenty-two weeks after treatment,we analyzed hepatic triglyceride level and measured expression of TNF-? by western-blot(protein) and quantitative RT-PCR(mRNA).Results Twenty-two weeks after treatment,the level of fasting glucose,insulin,triglyceride,free fatty acids(FFA) and hepatic triglyceride increased significantly in OLETF group as compared with LETO group.Western-blot method showed upregulated expression of TNF-? protein in OLETF group(1.57-fold vs LETO group,P

Objective This paper was conducted to study the preparation methods and the hypoglycemic effects of oral sodium alginate-insulin nanoparticles (INS-NPs) on the blood glucose level in Streptozotocin-induced diabetic Wistar rats. Methods The INS-NPs were prepared by an ionic gelation method. The changes of the morphology and size of the INS-NPs were examined by transmission electron microscope and Zetasizer 3000HS. The hypoglycemic effects of the INS-NPs were evaluated by monitoring the blood glucose levels in streptozotocin-induced diabetic Wistar rats. Results The INS-NPs were spherical or ellipsoidal in shape with a diameter of 236.4?19.3nm. The entrapment efficiency and load efficiency of INS-NPs were 78.5%?6.1% and 22.6%?4.4%, respectively. In vivo hypoglycemic study showed the levels of blood glucose of diabetic Wistar rats declined at 7h after oral administration of INS-NPs (26U/kg). Their hypoglycemic effects were maintained for 12h and the levels of blood glucose were kept with normal range for 6h (less than 7.0mmol/L). Conclusion The INS-NPs have the hypoglycemic effect on the blood glucose level of streptozotocin-induced diabetic rats.
10.The effect of aminophylline to strenthen diaphragmatic function in dogs

After the bilateral phrenic nerves of the dogs were stimulated,the effect of amino-phylline on the function of normal and fatigued diaphragm was observed with the determination of transdiaphragmatic pressure-stmulation frequency curve (Pdi-F).and the analysis of the power spectrum of diaphragmatic electromyogram (DMGdi) at spontaneous breathing.After the administration of 10 and 20 mg/kg of aminophylline ,the PDi pf the normal diaphragm was increased and that of the fatigued diaphragm recovered.The Pdi-F curve of the fatigued diaphragm was shifted to the left,and the central frequency,the medium frequency,and the high/low frequency ratio were markedly increased.It is concluded that aminophylline at the therapeutic dosage can imporve the generation of the strength and enhance the recovery of the fatigued diaphragm.
