1.Clinical impact analysis of stereopsis establishment for concomitant strabismus children after surgery

AIM: To investigate the effect of binocular vision training on stereopsis establish and visual function in child concomitant strabismus after surgery
METHODS:Ninety-three cases of strabismus children were randomly divided into two groups, making it comparable. The control group (n=46) were treated with conventional surgery and care;the observation group ( n=47) on the basis of routine care as control group, were given binocular vision training. Synoptophore and Titmus stereoscopic view were used to check the function of stereoscopic.
RESULTS:After treatment, simultaneous perception, visual fusion and stereopsis of trail patient were 77%, 62% and 40%, respectively, those of control group were 48%, 35% and 18%, the difference was statistically significant (P<0. 05). After treatment, fovea stereopsis and macular stereopsis children in observation group were accounted for 32% and 26%, respectively, in the control group those were accounted for 18% and 11%, respectively. Those in the observation group were significantly more than the control group. No stereoscopic patients in observation group were 23%, and that was 54% in the control group. That in the observation group was significantly less than the control group. The difference comparison between the two groups was statistically significant (P<0. 05).
CONCLUSION: Binocular vision training can effectively help children to build concomitant strabismus after surgery, for children with strabismus should be treated with conventional visual training to improve visual function and improve the life quality.
2.Establishment of a cell-based filovirus entry inhibitor evaluation system.

Ebola virus, the cause of severe and fatal hemorrahagic fever in humans, belongs to filovirus family. This study was designed to establish a cell-based screening and evaluation system in the pharmacological study of antivirus compounds. Three reporter systems were established with recombinant pseudoviral luciferase of HIV core (pNL4-3.Luc.R(-)E(-)) packed with filovirus glycoprotein (EBOV-Zaire GP/HIV-luc, EBOV-Sudan GP/HIV-luc and Marburg GP/HIV-luc), which are required for virus entry of cells. The level of filovirus entry was determined by the expression of luciferase reporter gene in the infected cells. For screening of filovirus entry inhibitors, the vesicular stomatitis G packed pseudovirions (VSVG/HIV-luc) was used to determine the compound specificity. The results of known filovirus entry inhibitors demonstrated successful establishment of the new model systems, which would be useful in high throughput screening of anti-filovirus drugs in the future.

5.The role of Toll-like receptor 1 in the immunoregulatory regulation of Th17 cells by mesenchymal stem cells

BACKGROUND:Our previous studies have shown that mesenchymal stem cells play an immunomodulatory role in Th17 cells, but the mechanism of action remains to be elucidated, therefore, to explore the role of Tol-like receptor 1 in which wil provide possible experimental basis for the future potential of celltherapy strategies.
OBJECTIVE:To investigate the role of Tol-like receptor 1 in the immunoregulatory function of mesenchymal stem cells on Th17 cells.
METHODS:Separation of adherent bone marrow-derived human embryonic sources of mesenchymal stem cells, immunomagnetic separation of normal CD4+T cells. CD4+T cells were cultured alone or in combination with mesenchymal stem cells co-cultured 4d. Real-time quantitative polymerase chain reaction assay interleukin-17, Tol-like receptor 1 expression levels related genes;number of Th17 cells by flow cytometry. Bone marrow mesenchymal stem cells from human embryos were separated using adherence method, and co-cultured with human CD4+T cells from healthy donors using immunomagnetic separation method for 4 days. The expression of interleukin-17 and Tol-like receptor 1 mRNA was detected by real-time PCR, and the number of Th17 cells was observed by flow cytometry.
RESULTS AND CONCLUSION:Tol-like receptor 1 mRNA was expressed in both CD4+T cells and mesenchymal stem cells. Level of interleukin-17 mRNA was significantly higher in mesenchymal stem cells co-cultured with CD4+T cells than in CD4+T cells cultured alone (relative value 3.59±0.11 vs. 1.14±0.08, P<0.01). Consistent with the expression of interleukin-17 mRNA, increased level of Tol-like receptor 1 mRNA was detected in mesenchymal stem cells co-cultured with CD4+T cells compared with CD4+T cells cultured alone (relative value 6.07±1.79 vs. 1.53±0.63, P<0.01). Furthermore, Flow cytometry analysis showed that the percentage of Th17 cells in the mesenchymal stem cells co-cultured with CD4+T cells was significantly higher than that in CD4+T cells cultured alone, (4.53±1.27)%vs. (2.39±0.80)%(P<0.01). Tol-like receptor 1 might be involved in the immunoregulatory regulation of Th17 cells by mesenchymal stem cells.
6.Two fusion peptides promote healing of scalded mice

Objective:To study the therapeutic effect of 2 artificially synthesized fusion peptides on scalded mice. Methods: Two new fusion peptides, M-T508 and M-T?1, were synthesized based on amino acid sequences selected according to the characteristics of scald (including 3 stages: inflammation, infection and repair by cell proliferation). The 2 peptides were applied for second degree scald burn in mice and the healing of the scald burn was observed. The expression ofⅠcollagen, vascular endothelial growth factor (VEGF), proliferating cell nuclear antigen (PCNA), and cytokeratin (CK) was detected by immunohistochemical staining. Serum TNF-? concentration was determined by ELISA in the animals and the blood vessel permeability was analyzed by Evans blue staining. Results: The healing periods of scald burns in the 2 peptide groups were 1 week earlier than that in the control group. Immunohistochemical results showed that the expression of collagenⅠ, VEGF, PCNA and CK was all positive in the 2 peptide groups 10 days after scald and negative in the control group. Examination of blood vessel permeability and serum TNF-? concentration in the M-T?1 and M-T508 groups showed that the 2 fusion peptides had anti-inflammation and anti-infiltration effects in scalded mouse. Conclusion: The synthesized fusion peptide M-T?1 and M-T508 can promote the healing of scald burns in mice.
7.Monitoring of Medical Staff's Hand Contamination after Change for Infected Wound and Managing Hand Hygiene

OBJECTIVE To monitor the hand contamination after dressing change for the infected wound and to explore an effective way for hand hygiene. METHODS Caregivers were divided into three groups after applying dressing on infected wound:group 1(no hand-washing),group 2(regular hand-washing),and group 3(standard hand-washing),the pathogens in hands were monitored and the effects of two ways of hand-washing were compared. RESULTS The rate of hand-carrying pathogens after dressing change for the infected wound was 100.0%,the qualified rate of regular hand-washing was only 47.9% and that of standard hand-washing (six-steps method)was 97.9%. CONCLUSIONS The standard hand-washing is more effective than regular hand-washing. Correct hand hygiene should be emphasized to prevent and control nosocomial infection.
8.Characteristics of internal fixation with cervical lateral mass plate in posterior approach of cervical spine fracture dislocation

To discuss the character and application of internal fixation with cervical lateral mass plate fixation in posterior approach of cervical spine operation. Twelve patients with cervical fracture dislocation treated with posterior cervical lateral mass fixation and bony fusion were selected. The patients received spinal decompression or reduction and decompression according to different diseases. A suitable 3.5 mm bi-cortical screw was inserted using Magerl technique. The results showed that solid bony fusion could be obtained at 3-5 months after the surgery, and the intervertebra heights as well as physiological curves were kept well without screw loosening or breaking. Neurological function was recovered to different extent. Cervical lateral mass plate fixation has the advantages of easily operation technique, wide indications, strong stability and short segment fixation.
9.Effects of hemin on the expression of hemeoxygenase in human umbilical vein endothelial cells

Objective To observe the effects of hemin on the expression of hemeoxygenase(HO) on the human umbilical vein endothelial cells.Methods The hemin and zinc protoporphyrin-Ⅸ were added in the cultured endothelial cells to induce and inhibit the expression of HO-1,respectively.The expressions of HO-1 and HO-2 mRNA were detected using reverse transcription polymerase chain(RT-PCR) and the relative amount of CO released into the media was quantitated as carboxyhemoglobin(COHb) by enzyme linked immunosorbent assay(ELISA).The proliferation of endothelial cells was detected by methyl thiazolyl tetrazolium test(MTT).Results The expression of HO protein was mainly detected in the cytoplasm of endothelial cell.The expression of HO-1 mRNA in endothelial cell and the amount of COHb in the media increased significantly with the treatment of hemin.Conclusion The expression of HO-1 and the increase of endogenous CO are the molecular bases of the proliferation of endothelial cells,and the HO/CO system may play an important role in the development of cardiovascular diseases.
10.Progress in evidence-based medicine for the treatment of optic neuritis

Optic neuritis is a common optic neuropathy,which typically presents with sudden loss of vision.Visual deficit of optic neuritis varies in severity and generally can be improved spontaneously over several-month duration.The most convincing eight evidences from level Ⅰ and Ⅱ evidence of evidence-based medicine( EBM ) about the treatment of all the optic neuritis were collected by searching theCochrane Controlled TrialsRegister (CENTRAL),MEDLINE,EMBASE and reference lists of identified trial reports since the last decade.At present,the latest findings showed that there is no conclusive evidence of benefit in terms of recovery to normal visual acuity,visual field or contrast sensitivity with either intravenous or oral corticosteroids at the doses evaluated in trials included in this review.Either no treatment or intravenous corticosteroid therapy followed by oral corticosteroids is appropriate,and intravenous corticosteroid may benefit the patient in terms of faster recovery to normal vision.There may be of different treatment of optic neuritis in China compared to Western countries due to geographical,racial and genetic background that may cause different etiology and clinical features of optic neuritis.Therefore,in China,it is imperative to carry out a large randomized control trial(RCT) of optic neuritis as soon as possible to get the best evidence for the management of optic neuritis patients.
