1.Sorafenib inhibits the proliferation of human oral cancer TCA8113 cells through the activation of p38MAPK

Objective:To investigate the effect of sorafenib on the proliferation of human oral cancer TCA8113 cells and to explore the underlying mechanisms.Methods:Mter treated with sorafenib at 2.5,5,10,20 μg/ml respectively for48 h,TCA8113 cell proliferation was examined by MTT and colony formation assay.Western blotting was employed to examine the p38MAPK expression in the cells.TCA8113 cells were pretreated with 10 μmol/L of SB203580 (a specific inhibitor of p38MAPK) for 30 min,and then by different concentrations of sorafenib for 48 h,cell proliferation was tested by MTT assay.Results:Sorafenib significantly inhibited the proliferation of TCA8113 cells in a concentration dependent fashion.Sorafenib also remarkably promoted the activation of p38MAPK of the cells.SB203580 significantly alleviated soiafenib induced TCA8113 cell viability decrease.Conclusion:Sorafenib can inhibit the proliferation of TCA8113 cells,which may be related to the activation of p38MAPK.
2.Genistein inhibits proliferation of human oral cancer TCA8113 cells through suppression of VEGF expression

AIM:To investigate the effect of genistein on the proliferation of human oral cancer TCA 8113 cells and to explore the underlying mechanisms .METHODS:The cell proliferation was examined by MTT assay , cell counting and colony formation assay .Western blotting was employed to examine the protein levels of vascular endothelial growth fac -tor (VEGF), extracellular signal-regulated kinase (ERK) and p-ERK.RESULTS: Genistein significantly inhibited the proliferation of TCA8113 cells in a concentration-dependent fashion .Moreover , genistein dose-dependently decreased the protein levels of VEGF, ERK and p-ERK.The expression of VEGF was also blunted by U 0126, a specific inhibitor of ERK.U0126 and axitinib, a VEGF receptor antagonist , both significantly inhibited the proliferation of TCA 8113 cells. CONCLUSION:Genistein inhibits the proliferation of TCA8113 cells, which may be related to its inhibitory effect on ERK expression and activation , thus subsequently decreasing the expression of VEGF .
3.L-Carnitine inhibits H2 O2-mediated NFATc3 nuclear translocation

AIM: To explore the effect of L-carnitine on nuclear factor of activated T-cells, cytoplasmic 3 (NFATc3) in cardiomyocytes under H2O2 stimulation.METHODS: Primary cultured neonatal rat myocardial cells were stimulated by H2 O2 at concentration of 200μmol/L for 12 h to induce oxidative stress injury.In treatment group, L-carni-tine and cyclosporin A ( CsA) , a specific inhibitor of calcineurin ( CaN) , were administered 30 min prior to H2 O2 stimula-tion.After treatment, total, cytoplasmic and nuclear NFATc3 protein levels were determined by Western blotting.The method of immunofluoresence was used to evaluate the distribution of NFATc3.RESULTS: H2 O2 treatment produced no effect on the expression of total NFATc3, but caused its translocation from the cytosolic to nuclear compartment, which was greatly blunted by L-carnitine pretreatment.CONCLUSION:L-carnitine antagonized oxidative stress injury via alleviating NFATc3 nuclear translocation.
4.Investigation of COPD knowledge among community nursing managers

Objective To evaluate the level of COPD knowledge of community nursing managers, and to provide basis for carrying out targeted training and improving community management capacity of chronic diseases.Methods The method of convenient sampling was used to investigate the self-designed questionnaire of COPD knowledge among 41 community nursing managers in Dongcheng district through the questionnaire network.Results The total score of COPD knowledge was (5.27±2.247), of which score of the basic concept of COPD knowledge was (2.93±1.349) and score of medication guidance knowledge was (1.44±0.776), score of disease care and health guidance knowledge (0.88±0.927). The correlation between knowledge mastery and disease nursing knowledge was the strongest (r=0.403,P<0.01), which had correlation with age, length of work, title, whether receiving the nursing training, and whether having experience to care non-invasive ventilator (P<0.05).Conclusions COPD knowledge of community nursing managers was poor. A variety of targeted and diverse knowledge and skills training should be conducted to improve the education and management ability of community nursing managers.
5.Effects of Baduanjin on dyspnea and activity tolerance among patients with stable chronic obstructive pulmonary disease

Objective?To explore the effects of health Qigong Buduanjin on dyspnea and activity tolerance among patients with stable chronic obstructive pulmonary disease (COPD). Methods?From January to December 2015, 73 patients with stable COPD in Beijing Dongcheng District Dafangjia Community Health Service Station were selected by convenience sampling. All of the patients were divided into observation group (n=37) and control group (n=36) with the random number table. Patients of two groups all accepted drug therapy and health education. In addition, patients of control group did exercise such as walking, brisk walking, riding a bicycle, lifting dumbbell according to their habits and ability. On the basis of exercises of the control group, patients of observation group practiced health Qigong Buduanjin last 12 months. The modified version of the British Medical Research Council respiratory questionnaire (mMRC) and 6-minute walk distance (6MWT) were used to evaluated patients' dyspnea and activity tolerance before and after intervention. The number of patients who were admitted to hospital for equal or more than one time because of acute exacerbation was counted to evaluate the risk of increasing serious disease condition. Results?At last,35 patients in observation group and 31 patients in control group completed the study. After intervention, there were statistical differences in the level of mMRC, 6MWT and the risk of increasing serious disease condition between observation and control group (Z=-0.525, t=5.950, χ2=4.705;P< 0.05). Conclusions?Health Qigong Buduanjin can reduce the severity of dyspnea of patients with stable COPD, improve patients' activity tolerance and reduce patients' risk of increasing serious disease condition.
6.Identification of the small supernumerary marker chromosomes in two patients with Turner syndrome.

OBJECTIVETo identify the small supernumerary marker chromosomes (sSMC) and guide the genetic counseling and medical treatment in two patients with Turner syndrome.
METHODSHigh resolution GTG and C banding, SRY amplification by PCR and fluorescence in situ hybridization (FISH) on metaphase chromosomes were performed to the two patients.
RESULTSThe karyotypes of the two patients were 45, X [29]/46,X, +mar[31] and 45,X[71]/46,X, +mar[29] respectively. SRY test indicated SRY-positive for patient 1, whose sSMC was originated from chromosome Y. The karyotype was confirmed as 45,X[29]/46,X,idic(Y)(q10)[31]. ish idic(Y)(q10)(RP11-115H13x2) (SRY+) by FISH. While in patient 2, the sSMC was originated from chromosome X, whose karyotype was determined as 45, X[71]/46,X, r(X)(p11.23q21)[29]. ish r(X) (p11.23q21)(AL591394.11xAC092268.3).
CONCLUSIONUsing cytogenetic and molecular cytogenetic analyses, we have identified the sSMCs in two patients with Turner syndrome, which was helpful to the clinical diagnosis and treatment.

7.Alpha-wave Cerebral Cortex Functional Networks of Magnetic Stimulation on Acupoints of Different Meridians

Objective To study the coordination mechanism of brain induced by stimulating on acupoints of different meridians by means of constructing cerebral cortex functional networks reflecting actual brain functional connections. Methods A 128-lead electroencephalogram (EEG) recording and analysis system was used to collect the EEG signals of 14 healthy subjects (eight males and six females) aged 21 to 25 in resting state and magnetic stimulation on the acupuncture points of Guangming (GB37) and Neiguan (PC6) located in different meridians from October to November, 2017. Then EEG sources were localized by group independent component analysis and standard low-resolution brain electromagnetic tomography, and the statistical relationships of EEG components were calculated. Finally, alpha-wave cerebral cortex functional networks were constructed and analyzed based on complex network theory. Results The connections of brain regions associated with movement, vision increased in the stimulation of Guangming acupoint. The brain regions associated with movement, attention and working memory in the stimulation of the Neiguan acupoint were reduced in the network. Some common brain areas were activated and some changes of functional connections were similar. Conclusion The changes in the topological structure of brain networks are basically consistent with the efficacy of the acupuncture point during magnetic stimulation on Guangming and Neiguan of different meridians, however, it causes some similar changes in functional connectivity connections of some brain regions.
