LRRC4, a novel member of LRR (leucine-rich repeat) superfamily, is cloned by expressed sequence tag (EST)-mediated positional cloning strategy combined with 5′-RACE technology. Normal expression of LRRC4 is highly specific for brain, whereas absent or significantly down-regulated in primary tumors including glioma, meningioma and pituitary adenoma. LRRC4 is a functional gene in neural development and axon growth, and associated with glioma grade progression. LRRC4 expression is gradually reduced, even absent accompany with glioma grade increase. Absent expression of LRRC4 is involved in the late event of malignant glioma progression.The reexpression of LRRC4 can decrease a series of growth factors/neurotrophic factors (IGF, EGF, PDGF, CNTF, bFGF,GDNF and BDNF) or receptors gene expression to regulate RTK-mediated many signaling transduction pathway, such as K-Ras/c-Raf/ERK/MAPK, PI-3K/AKT/NF-?B, p70S6/PKC, STAT3 and JNK2/c-Jun/mp53, which block U251 cells in late phase of G1 to inhibit glioma cells proliferation and invasion. This inhibitory effect of LRRC4 is dependent on its LRR domain. LRRC4 induces glioma cells to differentiate into astrocyte-like cells more than apoptosis.

[Method] To study the analgesic effect of Wulian compound extract on mice with hot-plate test, writhing test, and formaldehyde stimulation. To study the anti-inflammatory effect of Wulian compound extract by mice ear-swol en induced by xylene and abdominal cavity capil ary Permeability text .To study the anti-diarrhea effect of the Wulian compound extract with the method of diarrhea induced by castor oil and senna. [Results] Wulian compound extract can inhibit the pain caused by high fever, acetic acid and formaldehyde. Inhibit the xylene induced ear swel ing of mice and increase of celiac capil ary permeability induced by acetic acid. At the same time Wulian compound extract can prolong diarrhea latency induced by the senna and castor oil, reduce the frequency of diarrhea. [Conclusion]Compound Wulian extracts have significant antidiarrheal effect, anti-inflammatory, analgesic.

[Objective] To observe effects of HQSM on CK, LDH and AST activity in serum of acute myocardial ischemia rats caused by injecting pituitrin.[Method] SD rats were divided into several groups at random: normal control group,model control group, three HQSM groups(210mg/kg,150mg/kg,105mg/kg), positive control group(Huangqi Shengmaiyin and Diao Xinxuekang).The rats were treated with respective drugs by ig for 30 days, and pituitrin was injected respectively by caudal vein 1h after administering at the 29th and 30th days.Blood serum was taken out to determinate the activity of CK, LDH and AST at 1h and 24h after the last injection. [Result] The activity of LDH and AST in serum increased at 1h after injecting pituitrin, the activity of CK in serum increased,meanwhile LDH and AST was still higher than normal control group at 24h. Recent studies have shown that metabolism in the tumor tissue is far more complicated than we previously knew. Despite vigorous glycolysis, in fact, the tumor tissue still retains mitochondrial aerobic metabolism. Mitochondria is one of the main sites of the biosynthesis process of tumor cells. Recent studies revealed that abnormal fatty acid metabolism. Amino acid metabolism plays a key role in the tumorigenesis. The metabolic network in tumor cells was reprogrammed, leading to nutrition lfux reorganization and re-direction. Metabolism reprogramming in tumor cells facilitates the balance between the needs of energy supply and the synthesis of biological macromolecules. Targeting cell metabolism is not intended to interfere the energy supply of tumor cells but to affect the synthesis of metabolic rate, thereby inhibiting the proliferation of the tumor. hTe review focuses on the importance of metabolic reprogramming in tumor development and cancer therapy. We summarize what is currently known about metabolic reprogramming and establish a framework to understanding its contribution to the altered metabolism of cancer cells.

Nasopharyngeal carcinoma (NPC) is a polygenetic inheritance tumor. It has very high incidence in southern China and among immigrants and their offspring from south Chinese, and has done a great harm to the health of south Chinese. As a malignant tumor with obvious tendency of familial aggregation and regional difference, it is suggested that the etiology and pathogenesis of NPC are relevant to the coeffects of genetic and environmental factors. It is the main point to focus on the elucidating of the pathogenesis of PNC as a polygenetic inheritance tumor, primarily establishing of the concept of susceptibility gene groups in NPC and the carcinogenesis model of the multi-step Domino effect involved in the molecular mechanisms of NPC dominated by susceptibility gene groups in order to lay out a substantial foundation for finding the high-risk susceptibility factors of NPC, screening the high-risk population of NPC and exploring the targetic and individual measures in diagnosis and treatment of NPC.

Objective To summarize the key point of application of acellular dermal matrix(ADM)in operative cooperation of pharyngeal tissue repair.Methods In order to rebuild the air-tight cavity,ADM was sewn to the anterior vertebrae fascia in 7 patients with pharyngeal malignant tumor undergoing total laryngectomy or pharyngolaryngectomy to rebuild the posterior wall of the laryngopharynx.Meanwhile the muscular flap of the pectoralis major Was used to rebuild the anterior and lateral walls of the laryngopharynx.Results There was no reject reaction or pharyngeal fistula in these 7 cases and the wound surface were all mucous membranized.All the incisions reached the primary healing.Conclusions ADM is easy to get and possesses good histocompatibility and compatible thickness.It is easy to be managed during operation and can modify the peripharyngeal defect with the combination of muscular flap of the pectoralis major or any other neck tissue.Strict nursing preparation before operation and perfect cooperation during operation are key steps to complete the surgery successfully.

It had been proved by many evidences that several heat shock proteins (HSPs) expression is up-regulated in tissue-derived primary lung cancer, and HSPs may play important roles in development, in resistant to drugs and in prognosis of lung cancer. However, there have not still systemic research on which HSPs,especially HSP70 can be or not thought as a new biological target in the therapy to lung cancer. In order to address the expression and roles of heat shock protein 70 (HSP70) in lung adenocarcinoma, immunoblotting was performed to detect the expression of HSP70 in tissue specimens from lung adenocarcinoma which were diagnosed unambiguously by branch fibromicroscopy and were excised. It showed that in the normal lung tissues, the expression of HSP70 was less then that in cancer tissues. After down-regulation of HSP70 protein by HSP70 anti-sense oligonucleotides in A549 cell line, MTT assay showed that the proliferation of A549 cells was inhibited remarkably after the treatment with HSP70 antisense oligonucleotides and Act D. There had significant differential in HSP70 antisense treatment group followed by Act D treatment and Act D treatment group. Results of Hoechst33258 staining revealed that HSP70 antisense oligonucleotides could promote Act D-mediated apoptosis in A549 cells with a higher percentage of apoptotic cells (26.91?3.73)% than that of Act D-treated group (16.83?3.41)% (P

Object To probe into the antihypertension activity mechanism of extracts from Compound Jueming (CJ). Methods The method of radioimmunoassay was used to assay the concentration of the related substances on spontaneously hypertensive rats (SHR) such as angiotensin Ⅱ (ANGⅡ), renin activity (RA), endothelin (ET), atrial natriuretic polypeptide (ANP), aldosterone (ALD), and urine. Results All of the three doses (9.5, 7.2, and 4.8 g/kg) of extracts from CJ reduced ANG Ⅱ and RA, but had no influence on ALD, ET, ANP, and urine in SHR. Conclusion The antihypertension effect of extracts from CJ is related to decreasing of renin angiotensin, but not to ALD, ET, ANP, and urine.

Objectives: To establish melanoma models of B16 cell line in ICR mice. Methods: Inoculating B16 Melanoma cells in ICR mice's feet, back, abdomen and caudal vein, to establish transplanted and metastatic melanoma models. We observed the morphologic characteristics and examined the pathological parameters of melanoma. We analyzed the relations between the number of B16 cells inoculated and the genesis of tumors, the influence of tumor growth on mice's behavior,and growth curve of the melanoma. Results:Six days later, sesamoid, linear and nodal even kaulifloweroid melanoma were seen gradually in ICR mice's feet, back, abdomen and lung (anatomid) after inoculating. The bearing tumor rates have a positive relation to the number of inoculated B16 cells. The rates of bearing tumor was near 100%, after inoculating with 5?10 6 B16 cells. Conclusions: The B16 melanoma cell line derived from C57/B16 mice(black ) has high rates of bearing tumor in ICR mice (white). The B16 melanoma models in ICR mice are ideal tumor models that have the characteristics of easy established,easy observing and easy obtaining.
