dosage ( P<0.01).Conclusion UVA can inhibit the proliferation activity of human skin fibroblasts. It might be related to the up-regulation of iNOS gene expression and the over-secretion of NO induced by UVA.
2.Effect of triptolide on the proliferation and apoptosis of human epidermal squamous cell carcinoma cell line A431 in vitro

Objective To investigate the effect of triptolide on the proliferation and apoptosis of human epidermal squamous cell carcinoma cell line A431 in vitro. Methods Human epidermal squamous cell carcinoma cell line A431 was cultured. After the treatment with triptolide, the inhibi-tion of cellular growth was determined by measuring MTT dye absorption of the living cells. Light mi-croscope showed morphological changes. The cell cycle and apoptosis rate were assessed by flow cy-tometry. Results Triptolide could significantly inhibit the proliferation of A431 cells in a dose- and time-dependent manner. Triptolide could also cause cell morphological changes ( the number of float-ing cells and nuclear pyknosis increase), induce cell apoptosis, and change the distribution of cell cycle phase in A431 cells. Compared with the control group, the G0/G1 phase A431 cell rate in-creased and the rate of S phase cell decreased in TP-treated group. Cell cycles were obviously inhibi-ted by triptolide in G0/G1 phase (both P<0.05). Conclusion TP could play an anti-tumor role by effectively inducing cell apoptosis and inhibiting the proliferation of A431 cells.
3.Rhesus monkey P53 gene silencing at the cellular level

Objective In order to establish a rhesus monkey model of p53 gene silencing, firstly we screened and determined the effective silencing targets of p53 gene at the cellular level in rhesus monkey.Methods The expression of p53 gene was detected in COS-7 cells ( derived from the kidney of the African Green Monkey, Cercopithecus aethiops).Three small hairpin RNA ( shRNA) sequences targeting rhesus monkey p53 gene were designed, analysed by bioinformatics, and inserted into lentivirus-based gene silencing constructs FUGW-TDT.The plasmids of p53-RNAi and control vector were transfected into the COS-7 cells, respectively.The suppression of p53 mRNA was detected by real-time PCR, and the changes of p53 protein expression were detected by Western blot assay.Results p53 gene expression was detected in COS-7 cells.Bioinformatics analysis showed that three gene-silencing sequences were screened which lied in the open reading frame ( ORF) region and targeted 238 -258bp, 681 -701bp, 169 -189bp of the rhesus monkey p53 mRNA.At 48 hrs after transfection of the three silencing constructs, p53 mRNA was suppressed by(87.17 ±4.03)%, ( 72.62 ±4.11)% and(76.22 ±0.98 )%, and p53 protein was suppressed by ( 84.44 ±2.18 )%, ( 71.04 ±1.18)% and ( 74.17 ±0.95 )%, respectively. Conclusions We obtained three effective target sequences showing high efficiency in p53silencing, which can be used in further studies on gene silencing in rhesus monkey.
4.Development and reliability and and validity testing of a nutritional literacy scale for patients with inflammatory bowel disease

Objective:To develop a nutritional assessment scale for patients with inflammatory bowel disease and examine its reliability and validity for assessing nutritional literacy in patients with inflammatory bowel disease.Methods:Based on the Nutbeam health literacy stratification model and knowledge-attitude-practice model, a preliminary scale was developed through literature review, semi-structured interviews, expert consultation and pre-surveys. A convenient sampling method was used to select 376 inflammatory bowel disease patients admitted to Tenth People′s Hospital Affiliated to Tongji University from September, 2022 to April, 2023 for questionnaire surveys, and reliability and validity tests were conducted to form the final scale.Results:The nutritional assessment scale for inflammatory bowel disease included 39 items. Exploratory factor analysis identified five common factors: nutritional knowledge, nutritional attitudes, nutritional practices, information interaction ability, and information evaluation ability. These factors explained 65.431% of the total variance. The content validity index of the scale was 0.857, and the item-level content validity index ranged from 0.800 to 1.000. The Cronbach α coefficient of the scale was 0.869, and the Cronbach α coefficients of each dimension ranged from 0.847 to 0.922. Conclusions:The developed nutritional assessment scale for inflammatory bowel disease demonstrates good reliability and validity, allowing for effective evaluation of patients′nutritional status.
