1.New advances in magnetic resonance imaging of brain metastasis

The growth of brain metastasis needs appropriate microenvironment, and the change of normal brain microenvironment is the basis for exploring metastasis with MRI. Through the local magnetic field changes and the application of contrast agents for improving the discrepancy of the cerebral tissue, MR can accurately detect brain metastasis in position, number, size, and shape. MR with contrast-enhanced technique is the first choice for screening brain metastasis. The application of all kinds of new MR techniques, sequences and intracellular contrast agents can not only further improve the specificity of detection, but also improve the sensitivity of MRI, which is favorable to formulating reasonable treatment and prolonging the lifetime of the patients. The microenvironment of brain metastasis and advances in the new MR techniques were reviewed in this article.
2.Experimental study on the pretreatment technology of human platelets lyophilization

Objective To study the technology of loading trehalose into cytoplasm of human platelets before lyophilization,and to determine the optimal experimental conditions of pretreatment technology.Methods The curves of loading efficiency,the internal trehalose concentration versus incubated time,temperature,and external trehalose concentration were drawn up to screen the optimal loading parameters.Results The loading efficiency was linear to the incubated time(2 hours later) and incubated temperature(range from 30℃ to 40℃),respectively.The loading efficiency almost reached 60 percent when the platelets were incubated at 37℃ for 4h.The intracellular trehalose concentration was increased with the raise of the external trehalose concentration(
3.Effect of glucocorticoid on the calmodulin mRNA of hypothalamus-pituitary-adrenal axis

AIM To study the effect of glucocorticoid acetic acid cortisone on expression of CaM mRNA in hypothalamus-pituitary-adrenal axis (HPAA). METHODS Using reverse transcription polymerase chain reaction (RT-PCR),the expression of CaM mRNA was measured in HPAA of rats which were injected with acetic acid cortisone. RESULTS Acetic acid cortisone induced the expression of CaM mRNA in hypothalamus and adrenal respectively, but it had no effect significantly in pituitary. CONCLUSION These data suggest that CaM mRNA plays important role in the modulation of GC in HPAA.

5.Clinical Observation on the Treatment of Rheumatoid Arthritisin Early Stage with TongLuoShu Liquid

Objective To investigate the clinical efficacy of TongluoShu Liquid in treating rheumatoid arthritis at early stage. Methods 100 cases were randomly recruited into a control group and a treatment group with 50 cases in each group. The treatment group was given TongluoShu liquid, 150ml / time, twice a day; and the control group was given Diclofenac Sodium Sustained Release Tablets (Deegan), one granule for each time, twice a day, and Methotrexate 7.5mg / time, once per week. Results In the treatment group, symptoms, signs and biochemical indicators were significantly improved, and the swelling of the soft tissue around the joints in stage Ⅰ and Ⅱ were also reduced. Besides, osteoporosis was controlled and the progress of destruction to the cartilage of stage Ⅲ was detained, though there was no statistical difference between the two groups after the treatment. Conclusion ShuTongluo liquid is a safe and effective medicine with less adverse reactions in treating rheumatoid arthritis.
6.Evaluation on effect of ultrafiltration technology on endotoxin removal from recombinant MUC1-MBP fusion protein

Objective To evaluate the effectiveness of ultrafiltration technology in endotoxin removal from purified recombinant MUC1-MBP fusion protein (MUC1-MBP)and to demonstrate the effect of ultrafiltration on endotoxin removal.Methods CM Sepharose FF weak cation exchange (CM)(CM group), CM combined with Phenyl Sepharose 6 FF exchange (C6)(CM+C6 group),CM combined with ultrafiltration (CM+ultrafiltration group), and CM combined with C6 and ultrafiltration (CM+C6+ultrafiltration group)were used to purify the MUC1-MBP from E.coli. and remove endotoxin;the expression level of endotoxin was detected by Chromogenic End-point Tachypleus Amebocyte Lysate.Results There was a single band at the expected molecular weight of 62 000 by SDS-PAGE analysis.and the purity>96% by Quantity One analysis.The endotoxin levels in CM group and CM +C6 group were quite high and there was no significant difference between two groups (P>0.05 );the endotoxin level in CM+ultrafiltration group was significantly lower than that in CM group, and there was significant difference (P<0.01);the endotoxin level in CM+C6+ultrafiltration group was significantly decreased compared with CM+C6 group (P<0.01);there was no significant differences of endotoxin level between CM+ultrafiltration group and CM +C6+ultrafiltration group (P>0.05 ). Conclusion The effects of CM or CM combined with C6 on endotoxin removal are quite poor, especially C6;CM combined with ultrafiltration are quite effective on endotoxin removal,and ultrafiltration plays an important role in endotoxin removal.
7.Inductive effect of recombinant MUC1-MBP fusion protein combined with R848 on immune activity of T cells in mice

Objective:To study the inductive effect of recombinant MUC1-MBP fusion protein combined with R848 on the immune activity of T cells in the mice,and to provide experimental basis for searching the suitable adjuvants of recombinant MUC1-MBP fusion protein.Methods:A total of 21 C57BL/6 mice were randomly divided into control group(treat with normal saline),MUC1-MBP+R848 group (treated with MUC1-MBP+R848),and MUC1-MBP+baillus calmette guerin(BCG) group(treated with MUC1-MBP+BCG)(n=7).After immunized for 4-7 d,the spleen tissue was taken and the spleen indexes of the mice in various groups were measured;the stimmulus index(SI) of the mice was detected by lymphocyte proliferation response;the levels of tumor necrosis factor-γ(TNF-γ) and interleukin-4(IL-4) were detected by ELISA method;the proprotions of T lymphocyte subsets in spleen cells were analyzed by flow cytometry.Results:Compared with control group,the spleen indexes,SI,and TNF-γ levels of the mice in MUC1-MBP+ R848 and MUC1-MBP+BCG groups were significantly increased (P<0.05 or P<0.01);the indexes metioned above of the mice in MUC1-MBP+ R848 were higher than those in MUC1-MBP+BCG group;the IL-4 levels had no significant difference(P>0.05).Compared with control group,the proprotions of CD3+T,CD4+T,and CD8+ T cells of the mice in MUC1-MBP+ R848 group and MUC1-MBP+BCG group were significantly increased(P<0.05 or P<0.01).Conclusion:Both MUC1-MBP fusion protein combined with R848 and BCG can induce the Th1 type of immune activity in the mice,and R848 is the potential candidate adjuvant for MUC1-MBP.
8.Detection of serum amyloid protein and polypeptide growth factor in patients with dementia

AIM and METHODS: To detect the changes of amyloid protein (?-AP) and polypeptide growth factor levels and study their role in the possible pathogenesis of Alzheimers disease (AD) and vascular dementia (VD). We detected serum ?-AP, transforming growth factor- ?(TGF-?) and insulin-like growth factor (IGF-Ⅱ) levels by radio immunoassay in 8 patients with AD,15 patients with VD and 63 patients with ischemic cerebrovascular disease(ICVD) and 38 cases healthy volunteers. RESULTS: In AD and VD groups, serum ?-AP, TGF-? and IGF-Ⅱ were significantly higher than that in ICVD and control group; Serum ?- AP,TGF-? and IGF-Ⅱ contents in ICVD group were obviously higher than that in normal control, the highest increase occurred in sequel of cerebral infarction(SCI) and vertebrobasislar ischemia(VBI) groups. There are positive correlation among ?- AP, TGF-? and IGF-Ⅱ in AD and VD groups.CONCLUSION: ①?- AP is a risk factor in the pathogenesis of AD and VD. ② TGF- ? and IGF-Ⅱ play roles in the neurotoxin effect that lead to dementia. ③ ?-AP plays a important role in formation of senile plaque.
9.Huangqintang in Treatment of Ulcerative Colitis: A Review

Huangqintang comes from Treatise on Cold Damage Diseases (《伤寒论》) and is regarded as the traditional prescription for treating dysentery. It is composed of four herbs: Radix Scutellariae, Radix Paeoniae Alba, Fructus Jujubae and Radix Glycyrrhizae, with the effect of clearing heat and stopping dysentery, and is a classic prescription for clinical treatment of ulcerative colitis (UC). Both experimental research and clinical practice show Huangqintang has the characteristics of multiple pathways and multiple targets in treating UC. At present, research on the mechanism of Huangqintang in the treatment of UC mainly focuses on reducing intestinal inflammation, repairing epithelial cell barrier, improving microflora disorder, maintaining immune balance, relieving oxidative stress, regulating mitochondrial autophagy, and inhibiting cell pyroptosis and ferroptosis. In clinical application, Huangqintang and its modified prescription combined with conventional western medicine have a clear effect on UC, which can significantly alleviate abdominal pain, diarrhea, purulent bloody stool and other symptoms, effectively control the condition and improve the quality of life of patients, with few adverse reactions and high safety. However, the experimental methods and research programs of Huangqintang in treating UC need to be further improved, and the related mechanisms need to be further studied. Through retrieval and sorting of relevant literature, this paper systematically summarized and comprehensively analyzed the mechanism and clinical research results of Huangqintang in treating UC in recent years, and proposed suggestions on the shortcomings in order to provide theoretical and data support for the further application of Huangqintang. This had practical guiding significance for interpreting the connotation of Huangqintang and clinical scientific application.
10.Evaluation of internal doses to workers and monitoring of 131I concentration in air in nuclear medicine workplaces

Objective:To asscentain the 131I activity concentration in 131I treatment workplaces and to explore the method of estimating the internal dose to workers by air sampling and to analyze its influencing factors. Methods:Air sampling method was used to collect aerosols containing radioactivity in 10 randomly selected workplaces in Zhengzhou where 131I therapy was performed. Aactivity concentration of 131I in treatment workplace was measured for gamma emitters by gamma-ray spectrometry. The internal dose due to 131I inhalation was estimated based on measurement result and field investigation result. Results:The activity concentration of 131I in air samples from 19 subpacking rooms ranged from 0.087 to 570 Bq/m 3, with an average of (51.04 ± 128.58) Bq/m 3. Those from 11 wards ranged from 0.162 to 54.6 Bq/m 3, with an average of (7.97 ± 15.89) Bq/m 3. In terms of the work hours recommended by the national standard GBZ 129-2016 Specifications for individual monitoring of occupational internal exposure, the estimated annual effective dose to radiation workers due to the inhalation of 131I ranges from 0.002 to 10 mSv, with an average of (0.61 ± 1.80) mSv, below the dose limit specified in the national standards. Conclusions:The samples with high 131I activity concentration in nuclear medicine workplaces of 10 medical institutions selected in Zhengzhou are mostly distributed in tertiary class hospitals operating large amount of radionuclide with large numbers of thyroid cancer patients adimitted. The result ing internal dose to radiation workers cannot be ignored. Estimating the internal dose based on the measurement result of air samples has a large uncertainty.However, air sampling method can promptly detect radioactive contamination in case of abnormal events or accidents, providing early warning for workers to carry out dose measurement from external exposure and internal exposure assessment.
