1.Immune-related gene expression profile in multiple sclerosis with gene chips

Objective To construct the immune genes expression profile database of multiple sclerosis (MS) and to screen the high immune related candidate genes by using microarray analysis Methods Polymerase chain reaction (PCR) products on behalf of 484 immune related genes from mixed tissue library were assembled on the modified Oako glass slide The general RNA from MS patients and normal persons lymphocytes was transcripted to cDNA by RT PCR and labelled with cy 3 and cy 5 The two probes were mixed and hybridized with the above mentioned gene chips Scan array 4000 was used for scanning the hybridizing signals and GenePix Pro3 0 for date analysis Results Of the total 484 double genes monitored,22 genes of group one showed changes in expression level of the ratio outside 0 5 and 2.0;27 genes differ in group 2;72 genes change in group 3 With the same consistent gene ID of the double genes, the group one was 6,the group tow was 9 and the group three was 30 Conclusions These results show that the different expression of immune related genes might occur between MS patients and normal persons It might also afford a view of the changes that these genes should be probably related to the occurrence, development or progress of MS The cell immune related genes might be the important and most immune related genes involved in MS pathogenesis The results suggest the deeper insights into the mechanism,relapsing and treatment of MS
2.Gene expression profile in mice model of experimental autoimmune encephalomyelitis using cDNA microarrays

Objective To construct the genes expression profile database of experimental autoimmune encephalomyelitis (EAE) mice model and to screen the high candidate genes with microarray analysis. Methods Polymerase chain reaction (PCR) products on behalf of 4 096 genes from mixed mice tissue library were assembled on the modified Oako glass slide. The general RNA from 6 EAE mice model and 6 normal mice head was transcripted into cDNA by RT-PCR and labelled with cy-3 and cy-5.6 groups each consist one EAE mice and a control. These two probes were mixed and hybridized with the above mentioned gene chips. Scan array 4000 was used for scanning the hybridizing signals and GenePixPro 3.0 for date analysis. Results Of the total 4 096 genes monitored, 43 genes in group one showed changes in expression level of the ratio outside 0.5 and 2, 30 genes differed in group two, 176 genes changed in group three, 76 in group four, 294 in group five and 129 in group six. Conclusions The results showed the consistent different genes expression throughout the EAE mice model. The genes related to immune, cell structure, cell cycle, ion channel, signal transduction, protein synthesis and metabolism are involved in EAE pathogenesis. The results provide deeper insights into the mechanism of EAE and multiple sclerosis.
3.Effects of Different Doses of Dexmedetomidine Hydrochloride Intrathecal Injection on Ropivacaine Spinal Block

Objective To investigate the effects of intrathecal different doses of dexmedetomidine hydrochloride in spinal block by ropivacaine hydrochloride . Methods Forty lower limb surgery scheduled for elective under spinal anesthesia, were randomly divided into 2 groups (n = 20 each): the control and the treatment groups.The control and the treatment group were intrathecally injected with 4,12 μg dexmedetomidine hydrochloride respectively.The 0.75% ropivacaine hydrochloride 1.5 mL was injected for spinal anesthesia.SBP,DBP,HR,SpO2 and Ramsay Sedation Score were recorded before the spinal anesthesia conduct and thereafter every five minutes. And the onset and duration of block were recorded,adverse reaction like nausea, vomiting and respiratory depression were also observed. Results Compared with the control group,the onset of sensory block was shorter [(6.9±2.6) min vs (8.7±2.9) min] (P<0.05),and the duration of sensory and motor block was longer in the treatment group[(130.8±30.1) min vs (115.9±23.9) min] (P<0.05) and [(145.9±29.0) min vs (130.0±30.1) min] (P<0.05). Conclusion Intrathecal dexmedetomidine hydrochloride at 12 μg improves anesthesia via shortening the sensory block onset and prolonging sensory and motor block,which maintains hemodynamically stable,and does not generate adverse reactions as nausea,vomiting,bradycardia and respiratory depression.
4.Higher expression of AdPLA in orbital adipose tissue of patients with thyroid associated ophthalmopathy of Ⅲ level and stationary phase

Objective To detect the expression levels of adipose-specific phospholipase A2 (AdPLA) mRNA in orbital adipose tissue of thepatients with thyroid associated ophthalmopathy and the normals.Methods Sixteen patients with TAO of m level and stationary phase underwent orbital decompression,and 29 normals underwent ocular plastic surgery in Shenzhen Eye Hospital between August,2015 and October,2016.Orbital fat samples were collected from one eyes of these patients during surgery.The age,gender,height,weight,body mass index (BMI),exophthalmos degree,orbital fat of the patients with TAO and the normals were recorded and calculated.Using real time PCR,the AdPLA mRNA were detected from these orbital fat samples.Results There was no significant difference between the patients with TAO and the normals in age,gender,and BMI (all P > 0.05).TAO group had more exophthalmos degree (20.406 ± 1.369)mm than the normals (14.207 ± 1.146) mm.TAO group had more orbital fat (32.162 ± 1.923) mL than the normals (24.279 ± 1.070) mL.The average expression level of AdPLA in patients with TAO was 0.039 42 ± 0.009 85,and 0.004 42 ± 0.001 36 in the normal.There was significant difference between two groups (P < 0.05).Conclusion The patients with TAO of Ⅲ level and stationary phase have more exophthalmos degree and orbital fat than the normals.AdPLA mRNA is higher expressed in orbital adipose tissue of the patients with TAO of Ⅲ level and stationary phase than the normals.The high expression of AdPLA may reduce lipolysis in the orbital adipose tissue,lead to fat accumulation in orbits,and aggravate exophthalmos of patients with TAO.
5.Effect of rhDCN on proliferation of leukemic cell line K562

ObjectiveTo investigate the anti-tumorigenesis function of rhDCN on the leukemia K562 cells in vitro and analyze the possible mechanism.Methods Exponential phase of K562 cells were transfected with pcDNA3.1(+)-DCN,and PBS,liposome alone,and pcDNA3.1(+) vector were as control groups.Morphology change of K562 cells was detected by Wright stain,and cell proliferation activity was detected by MTT. Cell cycle and apoptosis of K562 cells were assessed by FCM. The expression of apoptosis-related protein,including bcl-xl,Mcl-1 and Bax were detected by Western blot.ResultsWright stain showed that typical apoptotic morphology of K562 cells were observed in DCN transfected group.There were no morphological changes of apoptosis in other groups. MTT method results showed that proliferation inhibition rate of the transfected cells [24 h (16.14±1.08) %,48 h (14.07±1.01) %,72 h (20.29±1.19) %]was higher than that of the other control groups (P < 0.05).FCM results showed that the apoptosis index (20.15±1.31) %of the DCN transfected group was higher than that of the other groups (P < 0.05),and most of cells arrested in the G0/G1 phase (51.15±0.57) % (P < 0.05).Western blot results showed the expression levels of Bax were increased while bcl-xl and Mcl-1 were decreased in pcDNA3.1(+)-DCN/K562 group.ConclusionrhDCN can inhibit the growth of K562 cells and induce the apoptosis.The effect of DCN on bcl-xl,Mcl-1,Bax may play a role in its mechanism.
6.Comparison of blood pressure control in community hypertensive patients with different management methods

Objective To compare blood pressure control in community hypertensive patients with different management methods.Methods Two neighborhood committees in a community of Pudong were selected as study area using cluster sampling method.A total of 5 166 residents aged ≥35 y were screened for blood pressure; the subjects with high blood pressure and had antihypertensive medication in last 6 months were included,and patients with secondary hypertension was excluded.The patients who entered community hypertension management program and got medication from community were included in community group; those who did not enter in community management program and/or not get medication from community were included in non-community group.Self-designed questionnaire was used for investigation.The medication compliance,awareness of hypertension risk factors and high blood pressure control were compared between two groups.Results Among 5 166 residents 4 763 were surveyed for hypertension with a response rate of 92.2% and hypertension prevalence rate of 23.2% (1 105/4 763).Among 1 012 patients with drug treatment for more than 6 months,there were 878 cases in community (86.8%) and 134 cases (13.2%) in non-community group.There were no significant differences in gender,age,education,working condition between community group and non-community group (P > 0.05).44.3% (389/878) patients in community group had a history of high blood pressure > 10 y and that was 56.7% (76/134) in non-community group (P =0.011) ; 28.6% (251/878) patients in community group were at high risk for risk stratification and that in non-community group was 47.8% (64/134) (P <0.001).The awareness of hypertension risk factors in community group and non-community group was 83.9% and 95.5%,respectively (P < 0.001).The medication compliance and blood pressure control rates in two groups were 93.2 % and 84.3 % (P < 0.001),68.6% and 51.5 %,respectively (P < 0.001).Conclusion The outcomes of hypertension management in terms of medication compliance and blood pressure control in community group are better than those in non-community group.
7.Effects of individualized health education by the form of one-day ward in patients with pregnancy-induced hypertension

Objective To apply the individualized health education by the form of one-day ward to nursing of patients with pregnancy-induced hypertension so as to reduce the blood pressure (BP), anxiety, depression and the incidence of caesarean section.Methods A total of 194 patients with pregnancy-induced hypertension were randomly divided into observation group and control group with 97 patients in each group. Patients in the control group received routine inspection and health education in out-patient clinic. On the basis of nursing in the control group, patients in the observation group adopted individualized health education by the form of one-day ward. And then, the BP, mastery of health related knowledge, the anxiety, depression, mode of delivery, self-efficacy and satisfaction with nursing of patients between two groups were observed when they were admitted to groups and before discharge.Results After the intervention, the systolic pressure and the diastolic pressure [(122.5±9.0) mmHg and (83.4±7.8)mmHg]of patients in the observation group was lower than those [(136.3±10.7) mmHg and (95.1±8.3)mmHg]in the control group with significant differences (t=4.083, 3.769;P<0.001). After the intervention, the scores of anxiety and depression of patients in the observation group were lower than those in the control group; the mastery of health related knowledge and the self-efficacy of patients in the observation group were higher than those in the control group; the incidence of caesarean section in the observation group was lower than that in the control group; the satisfaction with nursing in the observation group was higher than that in the control group; there were all statistically significant differences in them between two groups (P<0.05).Conclusions The individualized health education by the form of one-day ward can significantly reduce the BP, anxiety, depression and the incidence of caesarean section, and improve the self-efficacy and satisfaction with nursing among patients with pregnancy-induced hypertension.
